Abstract:Previous studies estimating gastrin release have measured the product of gastrin activity, e.g.gastric acid secretion, or more recently measured circulating gastrin levels (which may reflect changes in rates of gastrin disposal as well as gastrin release).In the present study gastrin release from antral mucosa under controlled conditions was directly measured in response to chemical stimulation by cholinergic agents (Urecholine and acetycholine with neostigmine) and modifications of calcium ion concentrations, including calcium-free solutions and those simulating normal and increased ionized calcium levels.Portions of freshly procured guinea pig and human antral tissue (approx.100 mg) were placedonmillipore filters (5\1 pore size) in closed chambers which were perfused at a constant rate (0.4 ml/min), at 37°C, under 95% 02 -5% C02, with balanced physiological solutions.Gastrin content of sequential perfusion fractions, collected at 5 or 10 min.intervals in response to perfusion of specific test agents, were measured by radioimmunoassay, and these were compared with simultaneously performed control perfusions.Antral gastrin release was promptly stimulated by (l) bethanechol chloride (Urecholine) 100 \lg/mg, and by (2) aceytlcholine chloride 10 mg/ml with neostigmine 10 \lg/ml.Constant perfusion with Urecholine for 30 min.produced peak increases in gastrin release (mean increase -585 ± 137 SEM pg gastrin/min/100mg antral tissue) at 10 to 15 minutes, returning to or toward prestimulation levels before termination of perfusion.Increasing perfusate ionized calcium concentration from 4.8 to 6.5 mg % increased antral gastrin release from 930 ± 37 to 1545 ± 33 pg gastrin/min/100mg gastric antrum (p < 0.001).This study describes a technique for the direct demonstration and measurement of gastrin release by antral mucosa under controlled conditions in response to defined chemical stimulation.Utilizing this system antral gastrin release in response to cholinergic stimulation and to increased calcium ion concentration was directly demonstrated.
