Abstract:Objective:To study the method of primary culture of SD rat gastric smooth muscle cells by co-collagenase digestion method,and develop a 3-dimensional scaffold culture model for tissue engineering.Methods:Smooth muscle layer of rat was digested by 0.2% 1-type collagenase and 0.05% trypsin.The cultured cells were observed and identified.Expanded cells of P3 were seeded into PLGA constructs with fibrin matrix.Results:The smooth muscle cells were successfully cultured passaged in 2 weeks.The passaged smooth muscle cells grew with typical peak-valley pattern.Im munohistochemicalmethod (S-P method) showed strong expression of α-smooth muscle actin.Seeding of the expanded PC was technically feasible.The 3-dimensional scaffold culture technique provided homogeneous cell distribution without sig nificant cell loss.Conclusion:Our primary cultured SD rat gastric smooth muscle cells with the co-collagenase method can stably grow.The model of 3D PLGA seeded is technically feasible and applies a promising method for creation of new tissue-engineered tissue in vitro.

Abstract:Objective To explore the effects of gastrin on the rat stomach mucous membrane.Methods Totally 60 Wistar rats were divided into gastrin and control groups.In the control group(22 rats),pure water was feed without a limitation.In the gastrin group(38 rats),the rats received subcutaneous injection of gastrin once a day for 30 days(300 μg/kg).The rats were killed every 4 weeks since week 12 until week 56,2 rats in the control and 3 rats in the gastrin group were killed each time.Tissue samples from the stomach of the rats were obtained and stained with hematoxylin-eosin and then the mucosal thickness were measured.The expression of gastrin and enterochromaffin-like cells(ECL) marker histidine decarboxylase(HDC) were determined with immunohistochemical staining.Results Compared with the control,Gastrin group displayed a thickening of the mucosa.In the gastrin group there were more gastrin and HDC positive cells.Conclusions By combining to its specific receptors,gastrin promotes the growth of gastric mucosal,especially promotes enterochromaffin-like(ECL) cells proliferation,which suggests that ECL cell proliferation is one of the mechanisms of the mucous membrane thickening.

Abstract:To study regulative action of mica monomer granule preparation on gastrin (GAS), somatostatin (SS) and G cells as well as D cells of gastric mucosa in experimental chronic atrophic gastritis (CAG) rat.CAG rats were treated with mica monomer granule preparation with three different dosages--high, moderate and low level respectively. Changes of blood serum GAS, blood plasma SS and G cells as well as D cells of gastric mucosa in CAG rats were observed and detected with ELISA method, RIA method and immunocytochemistry method.Mica monomer granule of three different dosages could increase the quantity of G cells as well as D cells of gastric mucosa and the concentration of blood serum GAS and decrease the content of blood plasma SS in CAG rat at different level respectively. It was more effective in high and moderate dosage groups.Mica has the pharmacological action of protecting gastric mucosa, promoting the palingenesis of gastric gland and enhancing blood stream of gastric mucosa consequently to abate the inflammation reaction of gastric mucosa. Its effective mechanism is associated with the neuroendocrine regulative mechanism of promoting the secretion of gastric acid and gastric pepsin by increasing the amount of G cells as well as D cells and the concentration of blood serum GAS, and reducing inhibiting action on GAS secretion and enhancing the secretion of GAS by decreasing the content of SS.
Differentiation of Pig Gastric Primary Cells into Mucus Producing Epithelial Cells

Abstract:There is a growing interest in the development of in vitro models that mimic the intrinsic characteristics of cells in vivo to replace and/or reduce the use of experimental animals. The stomach is lined with mucus secreting epithelial cells, creating a thick mucus layer that protects the underlying epithelial cells from acid, pathogens, and other harmful agents. Mucins are a main component of the mucus layer, and their secretion is an important protective feature of epithelial cells in vivo. Here, we present a method that differentiates pig gastric primary cells into mucin secreting epithelial cells by culturing the cells on polyester membranes under semi-wet interface for 14 days, using differentiation medium containing the N-[(3,5-difluorophenyl)acetyl]-L-alanyl-2-phenyl]glycine-1,1-dimethylethyl ester (DAPT) in the basolateral compartment for the first 7 days and subsequent 7-day culture in non-differentiation medium. The in vitro mucosal surfaces created by these cells are harvested 2 weeks post confluence, and two preservation methods are described to fix the monolayers for further analysis.
The study of regeneration gene I in an atrophic gastritis rat model

Abstract:Objective We established an chronic atrophic gastritis(AG) rat model to detect the expression of regeneration gene Ⅰ(RegⅠ) and the proliferation of mucus gland.Methods The AG rat model was established by administration intermixture of 2% sodium salicylate and 30% alcohol,20mmol/L sodium desoxycholate,0.1% ammonia in stead of water,combining irregular feeding.RegⅠ and RegR mRNA expression of gastric mucosa were estimated by real-time quantitative PCR(Q-PCR) and western blot was used to detect the RegⅠ protein expression.The cell proliferation in gastric glands was estimated by immunohistochemistry staining of PCNA.Results We successfully established an AG rat model.The RegⅠ protein expression was increased in gastritis tissue in rats.The Q-PCR showed there was no significant difference in RegⅠ mRNA expression between two groups(P>0.05),however,the Reg-R was significantly elevated in model rats(P<0.05).The immunohistochemistry staining of PCNA suggested cell proliferation in gastric glands was increased in the model rats.Conclusion RegⅠ is overexpressed in the chronic AG rat model,combined with the cell proliferation of gastric glands.Ⅰt suggests that RegⅠ may increase the damaged mucus glands repair in AG rat.
The DHE cell line as a model for studying rat gastro-intestinal mucin expression: effects of dexamethasone

Abstract:The expression of mucin genes was evaluated in rat intestinal cell lines in order to establish an in vitro model for investigating the regulation of intestinal mucin expression in this species. Two rat intestinal cancer cell lines (DHE, LGA) and three nontumoral rat intestinal cell lines (IEC6, IEC17, IEC18) were screened. The mRNA expression of rMuc1, rMuc2, rMuc3, rMuc4, and rMuc5AC mucin genes was studied by semiquantitative RT-PCR, real-time RT-PCR and Northern-blot analysis. Results were correlated with immunohistochemical expression of rat gastric and intestinal mucin proteins, and secretion of glycoconjugates was examined by enzyme-linked lectin assay. We showed that mRNA of rMucl and rMuc2 were constitutively expressed in all IEC cell populations but periodic acid Schiff staining of these cells did not reveal the presence of glycoproteins. DHE cells expressed rMuc1-5AC mRNA and LGA expressed the same mucins but the level of rMuc4 was much lower. Mucin mRNA expression also differed in relation with the length of cultivation. Immunocytochemical studies revealed the presence of gastric and intestinal mucins in the two tumoral cell lines. Functional experiments showed that bethanechol, A23187 and PMA stimulated release of glycoconjugates in DHE but not in LGA cells. Treatment of DHE cells with dexamethasone (10(-7) mol/l) enhanced rMuc2 mRNA but decreased rMuc1 and rMuc5AC mRNA. Real-time RT-PCR showed that the expression of rMuc1 and rMuc5AC genes was reduced by more than tenfold after 24 h. The increased expression of rMuc2 gene was confirmed by Northern blot analysis. In conclusion, DHE cells provide a valuable cellular model for research on rat mucin secretion and expression.
Hepatocyte growth factor regulates the development of highly pure cultured chief cells from rat stomach by stimulating chief cell proliferation in vitro

Abstract:The physiology of gastric epithelial cells is often studied by using cancer cell lines, which may or may not provide information relevant to normal cells. Because few models exist to study chief cell physiology in vitro, our purpose was to develop primary cultured chief cells from rodent species that are structurally and functionally similar to native chief cells. For this, isolated chief cells from the rat stomach, purified by counterflow elutriation and density gradient centrifugation, were grown in media with growth factors. Purity and the continuity of tight junctions were determined, and permeability, viability, transepithelial resistance (TER), cell number and proliferation, and pepsinogen secretion in response to carbachol were measured. When plated in media alone or with basic fibroblast growth factor, the isolated chief cells attached by 2 days and were confluent by 4 days after seeding. However, tight junctions were discontinuous, TER was less than 300 Omega cm(2), and permeability was high. In contrast, chief cells incubated with hepatocyte growth factor (HGF) were confluent in 3 days and had a TER greater than 2,000 Omega cm(2), continuous tight junctions, and low permeability. EGF was intermediate. HGF facilitated monolayer development by increasing cell number, which occurred by the proliferation of chief cells. Chief cell cultures, grown with HGF, consisted of more than 99% gastric intrinsic factor-expressing cells and showed robust pepsinogen secretion. Coexpression studies for neck and chief cell markers suggest that the cultures are a mixture of mature, immature, and transitional zone cells. This model will be useful for investigating mechanisms that regulate chief cell physiology in health and disease.

Abstract:The effect of rat submaxillary extract on the growth of rat C6 glioma cells in serum-free culture has been examined. Extracts (10-15 microgram/ml) of submaxillary glands from both male and female rats markedly enhanced the growth of serum-deprived C6 cells and, in combination with insulin, transferrin, and NIH-LH (a source of fibroblast growth factor), were able to stimulate C6 cell growth to an extent comparable to that achieved with an optimal amount of fetal calf serum. The mitogenic activity of rat submaxillary extracts was found to be heat-labile, acid-stable, and partially inactivated by protease and 2-mercaptoethanol. Under our assay conditions, biologically active preparations of purified mouse submaxillary gland epidermal growth factor (EGF) or nerve growth factor (NGF) were not mitogenic for C6 cells, nor was the mitogenic activity of rat submaxillary extracts inhibited by antiserum to these mouse submaxillary gland growth factors. These results suggest that the active component(s) of rat submaxillary extracts is unrelated to either EGF or NGF. The growth-enhancing effect also appears unrelated to esteropeptidase activity present in these extracts since the mitogenic activity was unaffected by several protease inhibitors. Moreover, two purified mouse submaxillary gland arginylesteropeptidases, EGF-binding protein and gamma-subunit of 7 S NGF, were unable to elicit a comparable growth response even when added to cell culture medium at unreasonably high concentrations. The C6 cell mitogenic activity of crude submaxillary extracts could be separated into two biologically similar components by either gel filtration on Sephadex G-100, preparative isoelectric focusing in a pH gradient of 3-10, or adsorption to DEAE-cellulose followed by elution with a sodium chloride gradient. One of the active components was acidic in nature and had an apparent molecular weight of 40,000, while the other was near neutral in charge and possessed a molecular weight of approximately 20,000. The relationship between these two C6 cell mitogenic components and the rat submaxillary gland component responsible for stimulating Balb/c-3T3 cell growth in serum-free, factor supplemented medium (McClure et al., 1979, J. Cell Biol. 83:96a) is also discussed.

Abstract:In the literature on gastric secretion, the capacity of mucus to lower the free acid in the stomach is repeatedly suggested. While working on the “Relationship between Gastric Secretion and Basal Secretion of the Stomach” Lim determined that the concentration of the mucus in the stomach is highest at the lower secretion rates. The problem here was to determine the effect of increasing the mucus content of the stomach upon the free HCl. To evaluate the antacid effect of gastric mucin, it was necessary to either stimulate the secretion of mucin or to administer mucin by mouth. In these experiments attempts at stimulation failed to give consistent results, therefore a neutral preparation of hog mucin was prepared. Two ounces of this mucin in the stomach of Pavlov pouch dogs, after stimulation with 1 mgm. of histamine, was found sufficient to keep the stomach free of free HCl despite the fact that the pouches in these dogs showed adequate gastric secretion. Free HCl would always be present if the same experiment were performed and egg albumen, gelatin or meat substituted for the mucin. When half an ounce of mucin mixed with a pound of meat was fed the dogs, no free HCl was present during an observation period of 5 to 7 hours. Since January, 1930, twelve patients with definite ulcer histories and typical roentgen findings of peptic ulcer were available for study. In addition to the customary bland diet, one ounce of powdered mucin was added to each meal and about 1 gm. of mucin in tablet form given hourly. All these patients were relieved of subjective symptoms within 3 days of treatment and as yet there has been no recurrence of the pain in observation periods of 2 to 5 months.

Helicobacter pylori induces pepsinogen secretion by rat gastric cells in culture via a cAMP signal pathway

Abstract:Infection with Helicobater pylori (H. pylori) is associated with various stomach diseases such as chronic gastritis, peptic ulcer, and gastric carcinoma. In order to investigate the mechanisms of enhanced production of pepsinogen by H. pylori in cultured rat gastric cells that have the potential to produce pepsinogen, secretion and synthesis of pepsinogen in the cells exposed to H. pylori extract were determined by measuring the hydrolysis of hemoglobin. Various drugs were used to study the mechanisms of effects of H. pylori on the cells. Exposure of the gastric cells to H. pylori extract caused a significant increase in pepsinogen secretion into the culture medium within 30-180 min in a dose-dependent manner, accompanied by a significant increase in pepsinogen synthesis in the gastric cells after 60 min of incubation. Heat treatment of the H. pylori sonicate at 100 degrees C for 10 min completely abolished the stimulatory effect of H. pylori on pepsinogen secretion. 2',3'-Dideoxyadenosine (50 microM), a specific adenylate cyclase inhibitor, abolished the effect of H. pylori-induced pepsinogen secretion. Puromycin (10 microg/ml), a protein synthesis inhibitor, and nicorandil (0.1 mM), a specific intracellular calcium antagonist, reduced the H. pylori-induced pepsinogen secretion by 37% (p<0.01) and 25% (p<0.05), respectively. On the other hand, actinomycin D (1 microg/ml), an RNA synthesis inhibitor, did not affect the H. pylori-induced pepsinogen secretion. Consequently, dibutyryl cAMP potentially stimulated the pepsinogen secretion from gastric epithelial cells in a dose-dependent manner. H. pylori induces pepsinogen secretion and synthesis by gastric epithelial cells through an increase in the intracellular cAMP and mobilization of the intracellular calcium. In addition, H. pylori affects pepsinogen synthesis at the translational level.
The prophylactic use of lamivudine can maintain dose-intensity of adriamycin in hepatitis-B surface antigen (HBs Ag)-positive patients with Non-Hodgkin's lymphoma who receive cytotoxic chemotherapy.

Abstract:We investigated the effectiveness of lamivudine to prevent hepatitis flare up due to reactivation of hepatitis-B virus (HBV) in hepatitis-B surface antigen (HBsAg)-positive patients with Non-Hodgkin's lymphoma (NHL) during cytotoxic chemotherapy. HBsAg-positive patients with NHL were identified from the lymphoma database of the Asan Medical Center from January 1995 to August 2002, and their medical records were reviewed. We found that 31 patients were received cytotoxic chemotherapy among 41 NHL patients with HBsAg-positive during same period. We divided them into 2 groups of HBsAg patients with NHL as follows: Group A who received cytotoxic chemotherapy with lamivudine 100 mg daily; Group B without any prophylactic antiviral therapy. There were no significant differences between Group A and B in several clinical variables. Seventeen patients (85%) in group B and one patient (9%) in Group A had hepatitis due to reactivation of HBV (p<0.001), with one hepatic failure related death in Group B and none in group A. The mean dose intensity of adriamycin actually delivered was 13.3 mg/m2/week (80% Relative Dose intensity (RDI)) in Group A and 9.1 mg/m2/week (55% RDI) in Groups B (p<0.001). Our data suggest that the frequency of chemotherapy-related HBV reactivation may be significantly decreased by lamivudine prophylaxis with maintenance of the dosage of adriamycin.
Morphological and Pathologic Changes of Experimental Chronic Atrophic Gastritis (CAG) and the Regulating Mechanism of Protein Expression in Rats

Abstract:Objective: To study the pathologic change and molecular regulation in cell proliferation and apoptosis of gastric mucosa in rats with chronic atrophic gastritis (CAG), and evaluate the possible mechanisms. Methods: Rats were administered with 60% alcohol or 2% salicylate sodium, 20 mmol/L deoxycholate sodium and 0.1% ammonia water to establish chronic atrophic gastritis (CAG) models. The gastric specimens were prepared for microscopic view with hematoxylin and eosin (H-E) and alcian blue (A-B) stain. The number of infiltrated inflammatory cells, the thickness of the mucosa gland layer (μm) and the number of gastric glands were calculated. The damage of barrier in mucosa with erosion or ulceration, and the thickness of mucin were examined by scanned electron microscope (SEM). The levels of PGE2, EGF (epiderminal growth factor) and gastrin in the serum were measured with radioimmunoassay or ELISA method. The immunohistochemistry method was used to observe the number of G cells, the expression of protein of EGFR (EGF receptor), C-erbB-2, p53, p16 and bel-2 in gastric tissue. Results: Under SEM observation, the gastric mucosa was diffused erosion or ulceration and the thickness of mucin was decreased. Compared with normal rats, the grade of inflammatory cell infiltration in CAG rats was elevated, whereas the thickness and number of gastric gland were significantly lower (P<0.05). Compared with normal level of (0.61±0.28) μg/L, EGF in CAG (2.24±0.83) μg/L was significantly higher (P<0.05). The levels of PGE2 and gastrin in serum were significantly lower in CAG rats than that in normal rats (P<0.05). Immunohistochemistry detection showed that the number of G cell in antrum was lower in CAG group (P<0.05). Immuno-stain showed EGFR protein expression in the basal and bilateral membrane, and the cytoplasma in atrophic gastric gland, while negative expression was observed in normal gastric epithelial cells. Positive staining of p53 and p16 protein was localized in the nucleus of epithelial cells. The former was higher positively expressed in atrophic gland, while the later was higher positively stained in normal gastric tissue. bel-2 protein was positively stained in the cytoplasma in atrophic gastric gland, while very weakly stained in normal gastric tissue. Conclusion: The pathological findings in gastric gland accorded with the Houston diagnostic criteria of antrum-predominant CAG. CAG in rats was related with the damage of barrier in gastric mucosa and the misbalance of cell proliferation and apoptosis. There was high protein expression of oncogene, while inhibitor of suppressor gene in CAG rats indicated high trend of carcinogenesis.

Abstract:We present a Rolling-Circle-Enhance-Enzyme-Activity-Detection (REEAD) system with potential use for future point-of-care diagnosis of malaria. In the developed setup, specific detection of malaria parasites in crude blood samples is facilitated by the conversion of single Plasmodium falciparum topoisomerase I (pfTopI) mediated cleavage-ligation events, happening within nanometer dimensions, to micrometer-sized products readily detectable at the single molecule level in a fluorescence microscope. In principle, REEAD requires no special equipment and the readout is adaptable to simple colorimetric detection systems. Moreover, with regard to detection limit the presented setup is likely to outcompete standard gold immuno-based diagnostics. Hence, we believe the presented assay forms the basis for a new generation of easy-to-use diagnostic tools suitable for the malaria epidemic areas in developing countries.
Studies of the Pathologic Change of Gastric Mucosa and Regulating Factors of Cell Proliferation in Rats with Experimental Chronic Atrophic Gastritis

Abstract:Objective To assess pathologic and molecular regulating changes of cell proliferation of gastric mucosa in rats with chronic atrophic gastritis (CAG) and approach the possible mechanisms of the disease. Methods Rats were administered with 60% alcohol (2% Salicylate Sodium), 20 mmol/L sodium deoxycholate and 0.1% ammonia water to establish CAG model. The pathological change and mucin secretion were evaluated. The level of the serum prostaglandin E (PGE)2 and endothelial growth factor (EGF) were measured with radioimmunoassay. The immunohistochemistry was used to detect the expression of EGF receptor (EGFR), C-erbB2, P53, P16 and Bcl-2. Results Compared with normal rats, the grade of inflammatory cell infiltration in gastric mucosa in CAG rats was elevated, whereas the thickness and number of gastric gland were significant lower. Scanning electromicroscopy showed the cells rupture and injury of barrier of gastric mucosa. The levels of haxosamine in gastric mucosa and PGE2 in serum were lower in CAG rats than in normal rats (P0.05). Compared with normal level of (0.61±0.28) μg/L, EGF in CAG in serum (2.24± 0.83) μg/L was significant higher (P0.05). The immunohistochemistry studies showed that the expression of EGFR and C-erbB2 were 45% and 70% respectively in CAG rats, while no expression in normal rats (P 0.05). The expression rate of P16 was 75% in normal rats,which was 10% higher than that in CAG rats (P0.05). Compared with the rats in normal group, Bcl-2 expression was higher in CAG (10% vs. 50% P0.05). There was no significant difference in expression of P53 between CAG and normal group (30% vs. 10%, P0.05).Conclusion The pathogenesis of CAG in rats was related with the damage of barrier in gastric mucosa and the imbalance of cell proliferation and apoptosis.

Abstract:Submandibular glands of 4-week-old rats were dissociated by a procedure involving digestions with collagenase and hyaluronidase, chelation with ethylenediaminetetraacetic acid, and mechanical force. The isolated cells were purified by centrifugation in a Ficoli solution and were maintained in culture for 36 hours. On the basis of trypan blue exclusions, about 70 per cent of the dissociated cells were viable. Electron microscopic observations indicated that the isolated acinar cells and intercalated and striated duct cells retained their essential in situ ultrastructural characteristics. During a 36-hour culture period the number of viable cells declined to about 40 per cent, and the various cell types formed mixed aggregates. The ultrastructural features of the intercalated and duct cells changed relatively little, but the acinar cells revealed several structural alterations. These included a decrease in the number of the secretory granules, fusions of the secretory granules, and an increase in the rough surfaced endoplasmic reticulum. In general, the polarity of acinar cells became less distinct. The endogenous peroxidase activity in the acinar cells gradually diminished during the culture. Isoproterenol when added to the cultured cells failed to stimulate the incorporation of radioactive thymidine or the discharge of the secretory material from the acinar cells.

Abstract:Background/Aims: To establish a method for primary culture of adult gastric mucosal epithelialcells. Methods: The resected tissue was incubated with trypsin and collagenase Ⅱ at 4℃ for 8~10hours. The isolated epithelial cells were cultured in DMEM/F12 medium supplemented with 20% fetalcalf serum. Results: Within first 24 hours of culture, the cells attached to the plate and became con-fIuent in 3 days. On phase contrast microscopy, over 90% of the cells possessed epithelial characte-ristics. Immunohistochemical studies showed that over 90% of the cells were positive for anti-cyto-keratin and anti-epithelial mucosal antigen-antibody staining. The active cells could be detected pre-dominantly 4 days after MTT staining. The synthesis of DNA was detected by 3H-TdR involvement andreached the peak on the third day. Conclusions: This method system provides a valuable model forstudies of gastric mucosal epithelial cells in vitro.
Gastrin release in a mucosal organ culture model exposed to a simulated digest of an equine dietary supplement

Abstract:Horses are prone to GI diseases including colic, colitis, and gastric ulcers. Equine dietary supplements are commonly used as a means of supporting optimal equine gastrointestinal health. G’s Formula (manufactured by G’s Organic Solutions INC., BC, CAN) is an equine dietary supplement composed of dried cabbage, carrot, oat flour, and hemp meal, formulated to promote optimal GI health in horses. The objective of the current study was to investigate the impact of a simulated gastric digest of GF on gastrin (Gt) release in an in vitro mucosal organ culture model. Porcine stomachs were collected from a local abattoir (Reist and Weber, ON, CAN). A section of mucosa from the pyloric antrum was isolated and the smooth muscle removed. A 4mm punch biopsy was used to divide the mucosa, and 2 biopsies were placed into each well of a 24‐well culture dish. Each well was filled with 1.5 ml of media supplemented with glutamine. Media was changed at 24h, at which time 100 ul of either a simulated gastric digest of GF (GF), a simulated gastric digest blank (BL), or PBS (CO) was added into sample wells. Media and digests were refreshed at 48h, at which time wells were either stimulated with 10−5M carbachol (GF‐s, n=12; BL‐s, n=12; CO‐s, n=11) or sham stimulated with PBS (GF‐n, n=9; BL‐n, n=13; CO‐n, n=10). This process was repeated at 60h, and the experiment was terminated at 72h. Gt release in the media was assessed using a porcine Gt ELISA (MyBioSource INC., SD, USA) and standardized /g tissue. Data was analyzed using a repeated measures ANOVA in SAS 9.4 using PROC GLIMMIX (SAS Institute, NC, USA). There was a difference between the treatments GF and BL (16534±797 vs 14072±723 pg/ml/g, p=0.03) and a trend towards a difference between GF and CO (14581±794 pg/ml/g, p=0.09). At 60h GF‐n was higher than CO‐n (17475±1784 vs 12164±1688 pg/ml/g) and GF‐s was higher than BL‐s (16903±1543 vs 12434±1544 pg/ml/g). GF‐s was higher at 72h compared to 48h (18289±1543 vs 14051±1543, p=0.03). In conclusion, a simulated gastric digest of GF added into an in vitro organ culture of porcine pyloric antrum appears to enhance tissue Gt release. J.L. MacNicol was funded by NSERC.

Abstract:Objective: To study the regulative action of mica monomer powder preparation on the chief and parietal cells as well as G and D cells in the gastric mucosa of the experimental atrophic gastritis (CAG) rats. Methods: Intervention therapy was given to the experimental CAG rats at three different doses of mica monomer powder preparation to evaluate the changes of chief and parietal cells as well as G and D cells in the gastric mucosa and the histopathological changes of gastric mucosa. Results: Mica monomer powder preparation at three different doses could increase the amount of chief and parietal cells as well as G and D cells in gastric mucosa of the experimental CAG rats and alleviate and control the inflammation of gastric mucosa and the atrophy of gastric mucosa glands. Especially, better effects were shown in the mid and high dose groups. Conclusion: Mica has the pharmacological action of protecting the gastric mucosa, promoting the regeneration of gastric glands, enhancing blood flow of the gastric mucosa, and consequently improving the inflammatory responses of the gastric mucosa. One of the mechanisms is associated with promoting the secretion of gastric acid and gastric pepsin and regulating the neuroendocrine mechanism including gut hormone secretion (gastrin and somatostatin) by increasing the number of chief and parietal cells as well as G and D cells.
