Abstract:A novel approach was developed for the preparation of 4.6-disubstituted-1.2-dihydro-2-oxo-3-pyridinecarboxylic ester in moderate to good yields. This route involves a reaction sequence of Michael addition. transformation to ene-lactam, and aromatization, featuring easily available material, variable substituents, and good functional compatibility.

Abstract:In sharp contrast to the well researched and well developed aminocarbonylation of alkenes, the dearomative carbamoylation of abundant arene pi-systems has scarcely been studied despite its great potential to enrich the diversity-oriented synthesis of high-value amides. The formidable challenges associated with such dearomatization include the low reactivity of stable aromatic pi-systems and intricate selectivity issues. Herein, we disclose a general approach toward highly selective dearomative carbamoylations of arene pi-bonds under CO-gas-free strated by tolerating a broad range of nucleophilic partners with high yields and excellent selectivities, thus providing modular access to the divergent synthesis of beta-functionalized primary amides. In addition, diverse downstream derivatizations including a for-mal C-H 1,2-olefination/carbamoylation reaction were conducted, exhibiting great potential in syn-thetic and medicinal chemistry.

Abstract:Herein, a general strategy for the regioselective dearomative 1,2-hydrocyanoalkylation of chromium-bound (hetero)arenes with simple alkylnitriles as pronucleophiles was disclosed, providing rapid access to 1,3-cyclohexadienes possessing useful alkylnitrile groups. The versatility of this methodology further enabled a selective dearomative deuteration reaction. Finally, synthetic applications of the method in the formal synthesis of natural products, including erysotramidine, demethoxyerythratidinone, and morphine, were demonstrated.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:This investigation presents a novel, cost-effective approach for creating a 2-pyridone core substituted with an ester group through intramolecular rearrangement. This one-step transformation involves an amide-mediated intramolecular transformation of a pyranone to pyridone scaffold, followed by alcoholic-mediated ester formation, in 75–85% yield. Deuterated alcohol was used to confirm the esterification under alcoholic conditions. The reaction requires a nucleophile source from the solvent, which was observed by performing the reaction of three derivatives in CD 3 OD to obtain the desired carboxylate compounds, replacing -OR with -OCD 3 . All synthesized compounds were characterized by spectroscopic analysis, and six selected compounds were further confirmed by single-crystal X-ray diffraction studies. Abstract:A novel Pd/Cu-catalyzed chemoselective aerobic oxidative N-dealkylation/carbonylation reaction has been developed. Tertiary amines are utilized as a "reservoir" of "active" secondary amines in this transformation, which inhibits the formation of undesired by-products and the deactivation of the catalysts. This protocol allows for an efficient and straightforward construction of synthetically useful and bioactive (E)-α,β-unsaturated amide derivatives from easily available tertiary amines, olefins and CO.

Abstract:An unprecedented approach for the synthesis of imine derivatives was achieved via a Pd(II)-catalyzed dearomatization reaction of N-aryl ureas with internal alkynes. The corresponding spirocyclic imine derivatives were obtained with 20 examples in good to excellent yields. Mechanistic investigation indicated that an interesting 1,3-palladium migration process led to the α-regioselective dearomatization when 2-naphthyl ureas were used as the substrates. Fused ring products could also be obtained to further prove this migration process.

Abstract:Efficient copper-catalyzed aerobic oxidative C-H and C-C functionalization of 1-[2-(arylamino)aryl]ethanones leading to acridones has been developed. The procedure involves cleavage of aromatic C-H and acetyl C-C bonds with intramolecular formation of a diarylketone bond. The protocol uses inexpensive Cu(O2CCF3)2 as catalyst, pyridine as additive, and economical and environmentally friendly oxygen as the oxidant, and the corresponding acridones with various functional groups were obtained in moderate to good yields.

Abstract:We report herein the development of palladium-catalyzed deacylative deuteration of arylketone oxime ethers. This protocol features excellent functional group tolerance, heterocyclic compatibility, and high deuterium incorporation levels. Regioselective deuteration of some biologically important drugs and natural products are showcased via Friedel-Crafts acylation and subsequent deacylative deuteration. Vicinal meta-C–H bond functionalization (including fluorination, arylation, and alkylation) and para-C–H bond deuteration of electro-rich arenes are realized by using the ketone as both directing group and leaving group, which is distinct from aryl halide in conventional dehalogenative deuteration.

Abstract:A nickel-catalyzed asymmetric decarboxyarylation of NHP esters via reductive cross-coupling has been established. Utilizing the NHP of amino acid esters as radical precursors furnishes a new protocol in which structurally diverse chiral benzylamines could be accessible. This method has demonstrated excellent catalytic efficiency, high enantioselective control, mild conditions, and good functional group tolerance, thus enabling the late-stage modification of bioactive molecules and pharmaceuticals.

Abstract:Herein, a novel protocol was constructed for the synthesis of highly functionalized 2,4-diarylpyridines. In this reaction, α,β-unsaturated ketoxime acetates and enaminones were used as substrates in a one-pot cascade reaction, which included a Michael reaction, intramolecular cyclization, dehydrogenation, and the removal of HOAc. This reaction resulted in the formation and cleavage of two bonds by refluxing a mixture of the starting materials in toluene and in the presence of the catalyst Fe(OTf)3. As a result, a series of functionalized pyridines were produced through a complex cascade reaction. This protocol can be used to synthesize functionalized asymmetrical 2,4-diarylpyridines. This approach is suitable for the combinatorial and parallel syntheses of pyridine derivatives in a one-pot reaction rather than traditional tedious multi-step reactions.

Abstract:AbstractA novel rhodium‐catalyzed dearomatization of O‐substituted pyridines to access N‐substituted 2‐pyridones has been developed. A computational study suggests a mechanism involving the formation of a pyridinium ylide followed by an unprecedented 1,4‐acyl migratory rearrangement from O to C. Furthermore, the chiral dirhodium complexes serve as the catalyst for the asymmetric transformation with excellent enantioselective control. DFT calculations indicate the chirality is transferred from axial chirality to the central stereogenic centre. The stronger π–π interaction and CH–π interaction account for the high enantioselectivity.
