Abstract:A simple protocol for amide bond construction using β‐keto sulfonyl fluorides (BKSFs) as the acyl surrogates has been achieved. The reaction of BKSFs with a range of amines could be performed in presence of N‐bromosuccinimide (NBS), affording various amides with diverse functionalities. Preliminary mechanistic studies revealed that the dibromo‐substituted sulfonyl fluorides could be the key intermediate in this amidation process.

Abstract:In the presence of KI and (diacetoxyiodo)benzene (PIDA), primary amines bearing an alpha-hydrogen can be efficiently converted to fluoroalkylated imidazoles by reacting with fluoroalkylated alkynes and sodium azide via a cascade Michael addition/azidation/cycloamination process. KI plays a crucial role in this reaction by generating iodine(I) species to promote the cyclization. Substrate scope was explored, and the application of this protocol was demonstrated. Additionally, a plausible reaction mechanism was proposed.

Abstract:The regioselective ring-opening of 1,2- and 1,3-sulfamidates with fluorine anion is reported. Direct construction of monofluoro-substituted amines and amino acid derivatives using inexpensive and easily available potassium fluoride (KF). The monofluorination process only needs 35 minutes under heating, which will be an attractive route for the synthesis of the PET fluorine-18 radiotracer. The application of the product can be extended to the construction of fluorinated polypeptides after simple deprotection treatment.

Abstract:A new and convenient procedure was developed for the preparation of sulfonamides and sulfonic esters using N-fluorobenzenesulfonimide(NFSI) as a novel phenylsulfonyl group transfer reagent. In this protocol, a broad range of functional groups were tolerated to give the corresponding sulfonamides or sulfonic esters in moderate to excellent yields. The synthetic strategy for sulfonamides formation proceeded efficiently even under base-, metal- and additive-free conditions with the advantages of operational simplicity, mild reaction conditions as well as short reac-tion time.

Abstract:Amines are a fundamentally important class of biologically active compounds and the ability to manipulate their physicochemical properties through the introduction of fluorine is of paramount importance in medicinal chemistry. Current synthesis methods for the construction of fluorinated amines rely on air and moisture sensitive reagents that require special handling or harsh reductants that limit functionality. Here we report practical, catalyst-free, reductive trifluoroethylation reactions of free amines exhibiting remarkable functional group tolerance. The reactions proceed in conventional glassware without rigorous exclusion of either moisture or oxygen, and use trifluoroacetic acid as a stable and inexpensive fluorine source. The new methods provide access to a wide range of medicinally relevant functionalized tertiary β-fluoroalkylamine cores, either through direct trifluoroethylation of secondary amines or via a three-component coupling of primary amines, aldehydes and trifluoroacetic acid. A reduction of in situ-generated silyl ester species is proposed to account for the reductive selectivity observed.

Abstract:The reactivity of model amidines with general structure (NBu 4 )[B 12 H 11 NHC(CH 3 )HNCH(CH 2 OTos) 2 ] under nucleophilic fluorination conditions has been investigated in the study. The obtained products have been characterized using multinuclear NMR and IR spectroscopies, RP-HPLC, and high-resolution mass spectrometry. The results obtained can be applied in the synthesis of similar derivatives containing 18 F atoms for studying the pharmacokinetic parameters of BNCT agents using the PET method.

Abstract:N -(Difluoropropenyl)amides/amines are an important class of fluorinated compounds. Here, we report an efficient method for synthesizing these compounds without the use of transition metals. Under simple base-promoted conditions, 3-bromo-3,3-difluoroprop-1-ene reacts with N -methylanilines or N -arylacrylamides, with the elimination of one molecule of HBr, to give the target compound. Another efficient method for synthesizing difluoroalkenes is the reaction of 2-bromo-3,3,3-trifluoroprop-1-ene with indole or its analogues. Abstract:Fluorinated aziridines are very important structural motifs, but methods access to such structures are still very rare. In this paper, we report a mild, catalyst-free and operationally simple strategy for the direct aminofluoroalkylation of olefins driven by the noncovalent interaction between N-allylanilines and fluoroalkyl iodides. This photochemical transformation features synthetic simplicity, mild reaction conditions, without the use of any photoredox catalyst, and high functional group tolerance. Moreover, the structures can be used as important precursors for the synthesis of beta-fluoroalkylated alkylamine derivatives, such as fluorinated amino acids and fluorinated 2-pyrrolidinones.

Abstract:A facile route to synthesise N-aminosulfonamides through a palladium-catalyzed coupling reaction of aryl nonaflates, sulfur dioxide, and hydrazines is reported. This transformation proceeds in the presence of Pd(OAc)2/XantPhos, and TBAB in 1,4-dioxane at 80 °C, leading to the corresponding N-aminosulfonamides in moderate to good yields. The reaction scope has been demonstrated, and good functional tolerance is observed. A plausible mechanism is proposed through the insertion of sulfur dioxide.

Abstract:We report the development of an azanide (−NH2) surrogate which enables the facile conversion of (hetero)aryl halides into primary N‐aryl amines under transition‐metal‐free conditions. The designed amidine reagent is easy to prepare, bench stable, and undergoes facile N‐arylation under basic conditions at 40 °C. Intermediate N‐aryl amidines are readily cleaved to form N‐aryl amines in situ through hydrolysis or base‐promoted elimination. The developed surrogate is a safer and more selective alternative to existing anionic N‐nucleophiles, such as alkali metal amides or azide salts.

Abstract:N-Perfluoro-tert-butyl (N-PFtB) secondary amines, harboring a unique 19F-reporting moiety linked directly to nitrogen, are highly attractive due to their diverse potential applications. However, their mild and facile synthesis remains a significant challenge. Herein, we present a safe and efficient strategy for the direct synthesis of N-perfluoro-tert-butyl secondary amines from readily available N-trifluoromethyl secondary amines. Experiments and theoretical calculations demonstrate that this novel protocol encompasses three main processes: the elimination of hydrogen fluoride from the N-trifluoromethyl precursor, consecutive addition-elimination conversion of difluoromethyl imine (R-N=CF2) to hexafluoropropyl imine (R-N=C(CF3)2), and final addition of R-N=C(CF3)2 with the in situ generated fluoroform (HCF3). Key advantages of this reaction include the utilization of a single trifluoromethyl source and the N-trifluoromethyl starting material itself as the hydrogen source. Notably, the elimination of hydrogen fluoride, facilitated by CsF, is critical for the success of this approach. This method is compatible with a broad range of functional groups, including heterocyclic compounds. 19F MRI experiments suggest promising prospects for PFtB-labeled secondary amines as 19F MRI contrast agents.

Abstract:An efficient method for the metal-free deaminative borylation of alkylamines, using bis(catecholato) diboron as the boron source, to directly synthesize various alkyl potassium trifluoroborate salts is introduced. The key to this high reactivity is the utilization of pyridinium salt activated alkylamines, with a catalytic amount of a bipyridine-type Lewis base as a promoter. This transformation shows good functional-group compatibility (e.g., it is unimpeded by the presence of a ketone, indole, internal alkene, or unactivated alkyl chloride) and can serve as a powerful synthetic tool for borylation of amine groups in complex compounds. Mechanistic experiments and computations suggest a mechanism in which the Lewis base activated B(2)cat(2) unit intercepts an alkyl radical generated by single-electron transfer (SET) from a boron-based reductant.

Firstly, the example of ring-opening hydrofluorination of azetidines was reported. Then, the Olah's reagent also provided a promising method for the construction of enantioenriched β-fluoro-α-amino acid derivatives, which could be used for the preparation of peptide-based bioactive molecules.

Abstract:The first intermolecular three-component aminofluorination of alkenes with electronically rich amino sources has been developed via an umpolung strategy. In this reaction, electrophilic N-bromodialkylamines are employed as amine electrophiles, whereas multifunctional AgF serves as both a nucleophilic fluoride source and a reductive catalyst, for the first time, to initiate a single-electron transfer with N-bromodialkylamines. The addition of hexafluoroisopropanol as a cosolvent is crucial to promote protonated alkylaminyl radical formation and maintain the nucleophilicity of fluoride ion. This protocol exhibits excellent functional-group tolerance toward various aryl and unactivated alkenes, furnishing the corresponding β-fluoroamines with high regioselectivity. Mechanistic investigation reveals an aziridinium intermediate via a radical pathway. The synthetic utility of this transformation was demonstrated by the aminofluorination of several bioactive molecules, including an efficient route to a Rizatriptan analog. We anticipate that our finding holds promise for new discoveries in medicinal chemistry and the amination reactions.

Abstract:The development of an efficient alternative to the widely employed Balz‐Schiemann deaminative fluorination method (i.e. without using aryl diazonium tetrafluoroborate salt) is a challenging task. Herein, we report a convenient one‐pot method for the FeCl3‐nanocellullose mediated mechanochemical synthesis of fluoroarenes through the selective substitution of an aromatic amino group by fluorine group using pyrylium tetrafluoroborate (Pyry‐BF4) and sodium fluoride (NaF) via in situ formation of pyridinium salt intermediate. The scope of the present protocol includes synthesis of twenty‐eight organofluorine compounds with excellent yields via a selective substitution (SNAr) of an amino group by fluorine. The presented concise methodology opens a pathway to access new chemical spaces for the late‐stage functionalization in pharmaceutical industries.
