Abstract:An effective synthetic method to achieve difluoromethylated oxazolidin-2-imine has been developed via visible-light promoted three-component tandem reaction of aryl allylamines, 2-BTSO2CF2H (BT = Benzothiazole) and isocyanates. This method features mild reaction conditions and good functional group tolerance, and the reaction mechanism was confirmed by experiments and interpreted by quantum chemical calculations.

Abstract:A simple protocol for amide bond construction using β‐keto sulfonyl fluorides (BKSFs) as the acyl surrogates has been achieved. The reaction of BKSFs with a range of amines could be performed in presence of N‐bromosuccinimide (NBS), affording various amides with diverse functionalities. Preliminary mechanistic studies revealed that the dibromo‐substituted sulfonyl fluorides could be the key intermediate in this amidation process.

Abstract:Fluorinated small molecules are commonly utilized in the realm of functional small-molecule chemistry, and N-difluoromethyl (N-CF2H) compounds are especially fascinating because of their unique and unexplored physiochemical properties. However, despite limited progress, a general applicable methodological approach to the synthesis of N-CF2H compounds remains elusive. Herein, guided by computation, we present an operationally simple and practical protocol to access N-CF2H amides and related carbonyl derivatives. The protocol utilizes a one-pot conversion of thioformamides via desulfurization-fluorination and acylation, providing N-difluoromethylcarbamoyl fluoride building blocks that can be further diversified to a diversity of unexplored N-CF2H carbonyl compounds with rich functionality. Furthermore, preliminary studies on their properties and stability showcased their application in pharmaceuticals and agrochemicals.

Abstract:An efficient and practical method for the synthesis of medicinally important acridines from readily available o-aminoaryl ketones and arylboronic acids was developed using copper(II)-mediated relay reactions that involve intermolecular Chan-Lam cross-coupling and subsequent intramolecular Friedel-Crafts-type reactions. A sole promoter, i.e., Cu(OTf)2, was used; therefore, strongly acidic and basic conditions, nonreadily available or expensive substrates, additives, and noble-metal catalysts were not needed.

Abstract:BF3 center dot OEt2 (Boron trifluoride etherate), an inexpensive and commercially easily available Lewis acid stoichiometrically employed for Beckamann rearrangement in general, was now found to efficiently catalyze Beckmann rearrangement of ketoximes into their corresponding amides (up to 99% yield) in anhydrous acetonitrile under reflux temperature.

Abstract:Tandem Meinwald rearrangement and nucleophilic substitution of oxiranenitriles was realized. Arylacetic acid derivatives were readily synthesized from 3-aryloxirane-2-carbonitriles with amines, alcohols, or water in the presence of boron trifluoride under microwave irradiation, and the designed synthetic strategy includes introducing a cyano leaving group into arylepoxides and capturing the in situ generated toxic cyanide with boron trifluoride, making the reaction efficient, safe, and environmentally benign. The reaction occurs through an acid-promoted Meinwald rearrangement, producing arylacetyl cyanides, followed by an addition-elimination process with nitrogen or oxygen-containing nucleophilic amines, alcohols or water. The current method provides a new application of the tandem Meinwald rearrangement.

Abstract:Fluorination using chiral catalytic methods could result in a direct access to asymmetric fluorine chemistry. However, challenges in catalytic asymmetric fluorinations, especially the longstanding stereochemical challenges existed in BF3·Et2O-based fluorinations, have not yet been addressed. Here we report the catalytic asymmetric nucleophilic fluorination using BF3·Et2O as the fluorine reagent in the presence of chiral iodine catalyst. Various chiral fluorinated oxazine products were obtained with good to excellent enantioselectivities (up to >99% ee) and diastereoselectivities (up to >20:1 dr). Control experiments (the desired fluoro-oxazines could not be obtained when Py·HF or Et3N·3HF were employed as the fluorine source) indicated that BF3·Et2O acted not only as a fluorine reagent but also as the activating reagent for activation of iodosylbenzene.

Abstract:The development of synthetic methods for the synthesis of organic azides is highly important, given their critical role in advancing click chemistry over the last twenty years. We report a reagent‐economical, reliable, and scalable synthesis of alkyl azides from primary and secondary alcohols. This robust click method capitalizes on the synergistic interaction between Sulfur Fluoride Exchange (SuFEx) reagents—specifically, benzene‐1,3‐disulfonyl fluoride (BDSF) or benzene‐1,3,5‐trisulfonyl fluoride (BTSF)—and trimethylsilyl azide (TMSN3). The method offers procedural ease, accommodates a wide array of substrates, and enables late‐stage functionalization. Additionally, we demonstrate the protocol's adaptability by validating a straightforward one‐pot deoxyazidation‐CuAAC sequence for drug discovery applications.

Abstract:Herein, we report a straightforward practical and simple method for efficiently synthesizing a BF3- containing unsymmetrical diboron salts. This method involves the direct desymmetrization of commercially available diboron (4). This desymmetrization is based on a selective B-masking strategy via nucleophilic trifluorination, providing the elusive diborons bearing a trifluoroborate group. The synthetic utility of these salts is demonstrated through various examples of (sequen-tial) B-ligand interconversions, enabling the synthesis of unsymmetrical diboron derivatives. These products, which serve as valuable building blocks, are obtained in high yields.

Abstract:gem-Difluoroalkenyl boronates are attractive synthons for constructing diverse gem-difluoroalkenes and organoboron compounds. However, the strategies for the construction of gem-difluorohomoallyl boronates has scarcely been described. Herein, we develop an efficient protocol for the construction of gem-difluorohomoallylic boronates through a Ni-catalyzed radical-promoted defluoroalkylborylation of α-trifluoromethyl alkenes with α-haloboronates under mild conditions. Abstract:Herein, we demonstrate a method for the syntheses of sulfinamides and sulfinate esters using a novel sulfur( IV ) fluoride exchange reaction with organometallic reagents. Our strategy involves the addition of an amine or alcohol nucleophile to thionyl fluoride, acting as a S( IV ) SuFEx hub, followed by an organometallic reagent. This approach allows efficient access to sulfinamides (45–91% yields) and sulfinate esters (44–82% yields) in only 30 minutes. The sulfinamide and sulfinate esters also can be readily derivatized to the corresponding S( VI ) sulfonamides, sulfonate esters, sulfonimidamides, and sulfonimidates without isolation of the intermediates.

Abstract:Boron difluoride (BF2) complexes with asymmetrical N,N'‐bidentate ligands have received increasing attention due to their fascinating properties and broad applications. They are generally constructed in two steps: ligand formation, followed by boron complexation. This review focuses on categorizing these BF2 complexes based on the key synthetic strategies that have been applied in the ligand formation steps. The post‐functionalization, properties and applications of different types of BF2 complexes are presented. Their challenges and opportunities are also discussed. This should help the future rational design and synthesis of BF2 complexes with intriguing properties and practical applications.

Abstract:gem-Difluoroalkenes have steric and electronic profiles similar to those of ketones, aldehydes, and esters, and consequently have been used widely as carbonyl isosteres in modern drug discovery. Although many attempts have been made to achieve gem-difluoroalkenes, the induction of enantioselectivity at the a position of a gem-difluorovinyl group still remains a challenge. Herein, an efficient method for the construction of gem-difluoroallylboronates with high enantiomeric excess via a copper-catalyzed defluoroborylation of 1-(trifluoromethyl) alkenes with B(2)pin(2) is described. The reaction conditions were mild, and a variety of common functional groups, such as ether, fluoride, chloride, bromide, iodide, ester, cyano, sulfide, amino, and indoyl groups, were well tolerated. Furthermore, we not only applied this developed system as a powerful synthetic tool for the late-stage modification of complex compounds but also highlighted the utility of the formed compounds in synthesis.

Abstract:HBpin is used as a boronizing reagent and formal dehydrating agent is reported for the first time.CsF was used as a catalyst to achieve the dehydration and conversion of primary amides,and various nitrile compounds were obtained with good to excellent yields.This method is easy to operate,and the yield of the target product in the gram scale amplification experiment is 96％.The catalytic system exhibits good compatibility with various functional groups,such as halogen,trifluo-romethyl,heterocycle,and amino group.These results indicate that this method has good application potential.Mechanism studies have shown that the in-situ generation of N-boroamide intermediates and H2 is the key to the smooth occurrence of the reaction.And,the formation of thermodynamically stable B-O-B anhydride and nitrile product is the driving force of this transformation.
