Abstract:Mechanistic and computational studies were conducted to elucidate the mechanism and the origins of enantiocontrol for asymmetric chiral phosphoric acid-catalyzed spiroketalization reactions. These studies were designed to differentiate between the S(N)1-like, S(N)2-like, and covalent phosphate intermediate-based mechanisms. The chiral phosphoric acid-catalyzed spiroketalization of deuterium-labeled cyclic enol ethers revealed a highly diastereoselective synselective protonation/nucleophile addition, thus ruling out long-lived oxocarbenium intermediates. Hammett analysis of the reaction kinetics revealed positive charge accumulation in the transition state (rho = -2.9). A new computational reaction exploration method along with dynamics simulations supported an asynchronous concerted mechanism with a relatively short-lived polar transition state (average lifetime = 519 +/- 240 fs), which is consistent with the observed inverse secondary kinetic isotope effect of 0.85. On the basis of these studies, a transition state model explaining the observed stereochemical outcome has been proposed. This model predicts the enantioselective formation of the observed enantiomer of the product with 92% ee, which matches the experimentally observed value.

Abstract:An enantioselective intramolecular chiral phosphoric acid-catalyzed cyclization of unsaturated acetals has been utilized for the synthesis of functionalized chiral piperidines. The chiral enol ether products of these cyclizations undergo subsequent in situ enantioenrichment through acetalization of the minor enantiomer. A new computational reaction exploration method was utilized to elucidate the mechanism and stereoselectivity of this transformation. Rather than confirming the originally postulated cyclization proceeding directly through a vinyl oxocarbenium ion, simulations identified an alternative two-step mechanism involving the formation of a mixed chiral phosphate acetal, which undergoes a concerted, asynchronous S(N)2'-like displacement to yield the product with stereoselectivity in agreement with experimental observations.

Abstract:Density functional theory (DFT) calculations were performed to study the mechanism and factors affecting the enantio-, regio-, and chemoselectivities in the palladium/Xu-Phos-catalyzed cascade Heck/remote C(sp2)-H alkylation reaction. The active catalyst is found to be able to sustain coordination with P and S atoms and can adapt its coordination mode to accommodate the significant steric hindrance between the ligand and substrate, unlike previous findings that showed coordination with P and O atoms. The reaction is established to occur in sequence through the oxidative addition of the aryl iodide to Pd(0), intramolecular alkene insertion, C(sp2)-H bond activation, and C(sp2)-C(sp3) bond reductive elimination. The C(sp2)-C(sp3) bond reductive elimination is identified as the rate-determining step, and the intramolecular alkene insertion as the enantioselectivity-determining step. The high enantioselectivity originates from the stronger electronic interaction between the catalyst and substrate; the exclusive 5-exo-regioselectivity is due to the stronger nucleophilicity of the terminal alkene carbon atom, and the chemoselectivity of C-H activation over carboiodination is driven by thermodynamics.

Abstract:A novel biscinchona alkaloid-catalyzed highly enantioselective desymmetrization reaction of bisphenol compounds with achiral Morita-Baylis-Hillman carbonate agents was developed. Through the asymmetric allylic alkylation strategy, a broad range of optically active P-stereogenic phosphine oxides were generated with excellent to good yields (up to 99%) and high enantioselectivities (up to 98.5:1.5 e.r.). The reaction was further investigated by the linear free energy relationship (LFER) analysis. A possible transition state was proposed and furthered verified by theoretical calculations.

Abstract:AbstractThe discovery of proper ligands to simultaneously modulate the reactivity and effectively control the stereoselectivity is a central topic in the field of enantioselective C−H activation. Herein, we reported the synthesis of axially chiral biaryls by Pd‐catalyzed atroposelective C−H olefination. A novel chiral spiro phosphoric acid, STRIP, was identified as a superior ligand for this transformation. A broad range of axially chiral quinoline derivatives were synthesized in good yields with excellent enantioselectivities (up to 98 % ee). Density functional theory was used to gain a theoretical understanding of the enantioselectivities in this reaction.

Enantioselective Arbuzov Reaction Enabled by Catalytic Ion-Pair Reorganization

Abstract:The stereocontrolled synthesis of stereogenic-at-phosphorus compounds is a long-standing challenge in organic chemistry that has received heightened research attention in recent years. None of the catalytic approaches taken to date have leveraged the rich manifold of transformations proceeding through nucleophilic dealkylation of phosphonium ion intermediates (e.g. Michaelis–Arbuzov, Pudovik, and Appel reactions). Here, we report enantioselective hydrogen-bond-donor-catalyzed Michaelis–Arbuzov reactions of dialkylphosphonites with hydrogen chloride to afford H-phosphinates, which are versatile P-chiral building blocks. Mechanistic and computational investigations reveal that the catalyst diminishes the reactivity of the chloride nucleophile, yet accelerates the rate-determining dealkylation step by preorganizing the phosphonium chloride resting state into a geometry that is primed to enter the SN2 transition state.

Abstract:Organocatalytic synthesis of chiral allene can be successfully achieved through the reaction between propargylic alcohol and 3-phenylindole catalyzed by chiral phosphoric acid with high yield and high enantioselectivity. The detailed reaction mechanism and origin of enantioselectivity were uncovered by quantum mechanical calculations. The results indicate that the reactions undergo the dehydration process, the C–C bonding reaction, the hydrogen abstraction and the regeneration of catalyst. Eight possible pathways were found and the most favorable reaction paths leading to the major and minor products were evaluated which were consistent with the experimental results. The distortion/interaction analyses of the transition states in enantioselectivity-determining steps confirm the most favorable pathway in leading the major product. QTAIM and IGM analyses are used to illustrate the role of intermolecular interactions. EDA analysis is also employed to understand the origin of selectivity. This work would provide insightful viewpoints on the asymmetric organocatalytic reaction by chiral phosphoric acid.

Abstract:Chiral diphosphines' are the ligands of choice for a wide range of enantioselective transition-metal-catalyzed processes.* While chirality may be independently incorporated on either the linking carbon chain or the phosphorus centers of these ligands, only a limited number of P-chiral diphosphines have been reported due to the difficulties associated with the synthesis of enantiomerically enriched phosphorus stereo center^.^ The purpose of this Communication is to disclose a convenient approach to the synthesis of Cz-symmetric P-chiral diphosphines4 that relies upon successive enantioselective deprotonation of aryldimethylphosphine-boranes or aryldimethylphosphine sulfides (eq 1) and subsequent oxidative coupling to the diphosphine precursors (eq 2) with high overall enantioselectivity (296%) for the two-step process.

Abstract:A detailed theoretical mechanistic investigation on the chiral phosphoric acid (CPA)-catalyzed Paal-Knorr reactions, with and without the presence of a Lewis acid, for the synthesis of N−N axially chiral atropisomers is described herein. Density functional theory (DFT) studies elucidate that in the absence of a Lewis acid, CPA-catalyzes both the initial cyclization and subsequent dehydroxylation processes, irresolutely identified as the rate-determining step in the reactions. Conversely, when a Lewis acid participates in the reaction, it facilitates the second dehydroxylation process with a significant lower energy barrier, thereby reversing the rate-determining step to the initial cyclization step. It is noteworthy that in the case of N-aminoindoles, both the S-configurational transition state TS1 in the cyclization step and TS2 in the dehydroxylation process are favourable. In contrast, for the synthesis of bispyrrole, R-configurational TS1 and S-configurational TS2 are dominant. Therefore, the enantiodivergence observed is essentially induced by the reversed rate-determining steps in the absence or presence of a Lewis acid in the bispyrrole case. Furthermore, the non-covalent interaction (NCI) and atoms-in-molecules (AIM) analysis of the TS structures reveal that the non-covalent interactions play a pivotal role in determining the enantiodivergence observed in these reactions.

Abstract:P-Stereogenic phosphinamides represent important structural elements in chiral organocatalysts and bioactive compounds. Herein, we report Pd(II)-catalyzed enantioselective C-H alkynylation using cheap commercially available l-pyroglutamic acid as a chiral ligand. A range of structurally diverse P-stereogenic phosphinamides was prepared in good yields with high enantioselectivities via desymmetrization and kinetic resolution. A tailor-made congested directing group, N-ethyl-N-(3-methylpyridin-2-yl)amino, was crucial for the reactivity.

Abstract:Efficient access to two enantiomers of one chiral compound is critical for the discovery of drugs. However, it is still a challenging problem owing to the difficulty in obtaining two enantiomers of one chiral catalyst. Here, we report a general method to obtain both enantiomeric products via fine tuning the hydrogen-bonding interactions of phosphonium salts. Amino acid derived phosphonium salts and dipeptide derived phosphonium salts exhibited different properties for controlling the transition state, which could efficiently promote the Michael addition reaction to give opposite configurations of products with high yields and enantioselectivities. Preliminary investigations on the mechanism of the reaction and applications of the products were also performed.
