Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:An efficient copper-catalyzed tandem oxidation/alpha-formylation reaction of 2,3-dihydro-1H-pyrroles in an oxygen atmosphere has been developed. The new reaction is conducted in dimethyl sulfoxide (DMSO), which serves as both the carbonyl carbon source and solvent, in the presence of trifluoroacetic acid to directly generate structurally diverse alpha-formylpyrrole derivatives in one pot in moderate to good yields. Sensitive aldehyde groups remain intact in the presence of an oxidant, trifluoroacetic acid and a transition metal salt. A possible mechanism is presented.

Abstract:Dedicated to Professor Brindaban C. Ranu on the occasion of his 75 th birthday. We report an unprecedented synthesis of bis(pyrazolo[1,5- a ]pyrimidin-3-yl)methanes, a new class of di(hetaryl)methanes, from pyrazolo[1,5- a ]pyrimidines by using DMSO as a C1 synthon (methylene source). The transformation is mediated by sodium metabisulfite (Na 2 S 2 O 5 ), which plays a crucial role in the presence of acetic acid as a promoter. A wide variety of bis(pyrazolo[1,5- a ]pyrimidin-3-yl)methanes were synthesized in moderate to good yields of up to 90%. Mechanistic studies suggested that the reaction follows an ionic pathway, probably involving a methyl(methylene)sulfonium ion as an active electrophilic species formed in situ by the reaction of DMSO with Na 2 S 2 O 5 . Abstract:We reported copper-catalyzed the 1,4- or 1,6-conjugate additions of arylidenemalonitriles or para-quinone methides with 2-bromodifluoromethylbenzo-1,3-thiazole for the preparation of benzothiazolyldifluoroalkyl containing (di)arylmethine compounds in moderate to excellent yields, providing a facile route for the synthesis of gem-difluoroalkylatated (di)arylmethine compounds. The synthetic utility of the current method was also demonstrated by a gram-scale reaction.

Abstract:3-Alkyl-5-(diethylaminomethyl)pyrazoles reacted with chloroacetyl chloride to give 1-chloroacetyl derivatives which were subjected to heterocyclization with thiobenzamide, thiourea, and N -phenylthiourea in the presence of triethylamine in ethanol. As a result, previously unknown 3-alkyl-5-(diethylaminomethyl)-1-(1,3-thiazol-4-yl)-1 H -pyrazoles were synthesized.

Abstract:Diethyl chloromalonate was synthesized by chlorination of malonic ester with N-chlorosuccinimide.Then it reacted with formamidine acetate to give 4,6-dihydroxy-5-chloropyrimidine in the presence of alkali catalyst.The total yield was 73.4% based on malonic ester and the purity as high as 99.7%.The structure of the resulting product was confirmed by IR and 1H NMR.

Abstract:Abstract A new method for the preparation of tetrahydroquinolin-2-one derivatives is presented. This approach involves a two-step reaction between enaminones and acylating agents, immediately followed by electrophilic cyclization, all within a single synthesis procedure, eliminating the need to isolate intermediates. The entire process is facilitated by the use of acyl Meldrum’s acids which not only shortens the preparation time of the substrates but also easily extends the range of substituents That can be used. The method’s scope and limitations were evaluated with various reagent combinations thus demonstrating its general applicability to the synthesis of tetrahydroquinolin-2-one core. Interestingly, some exceptions to the regular reaction pathway were observed when a strong EDG (electron donating group) was introduced via acyl Meldrum’s acids. The underlying mechanism of this phenomenon was elucidated during the investigation.

Abstract:5-Amino-3-methylthio-1H-pyrazoles are very important building blocks from which a wide variety of pyrazole derivatives can be prepared. When substituted 5-amino-3-methylthio-1H-pyrazole was treated with CH3I, the methylation occurres at endocyclic two nitrogens at the same time. The ratio of isomers in products was depended upon the nature of 4-position substituent in the pyrazole ring. The products were characterized by X-ray diffraction analysis, and the ratios of isomer were explained by means of the results of ab inito calculation.

Abstract:A series of poly-heterocycles containing hemi-aminal moieties: 6'-hydroxy trifluoromethylated Spiro [indoline-3,4'-pyrazolo [3,4-b]pyridine] derivatives were efficiently synthesized in ethanol catalyzed by NH4OAc via one-pot, three-component reactions of 3-aminopyrazole, ethyl 4,4,4-trifluoroacetoacetate and isatin. Treatment of this hemi-aminal containing products with the dehydrating reagent SOCl2/Pyridine in acetonitrile afforded the corresponding dehydrating trifluoromethylated polyheterocyclic Spiro [indoline-3,4'-pyrazolo [3,4-b] pyridine] derivatives. Furthermore, a more efficient, straightforward approach to formation of their dehydrating products has been developed via one-pot, three-component reactions catalyzed by hydrochloric acid. Meanwhile, the structures of products were fully confirmed by spectroscopic data, two representative products were further confirmed by XRD analysis, respectively.
