Abstract:Enantioselective hydrogenation of a broad spectrum of N-, O-, and S-containing aromatic benzoheterocycles or nonaromatic unsaturated heterocycles has been realized by using an Ir/SpinPHOX (SpinPHOX=spiro[4,4]-1,6-nonadiene-based phosphine-oxazoline) complex as the catalyst, affording an array of the corresponding chiral benzoheterocycles (30 examples) with excellent enantioselectivities (>99 % eein most cases) and turnover numbers up to 500.

Abstract:A novel class of chiral glycosyl-triazole-based P,N ligands were synthesized by 'click chemistry' and were applied to the Rh-catalyzed asymmetric hydrosilylation of a range of substituted acetophenones. Enantioselective hydrosilylation of acetophenone gave (S)-1-phenylethanol in moderate enantioselectivity (72% ee) and in good conversion (93%).

Abstract:Optically active medium-sized cyclic carbonyl compounds bearing an -chiral carbon center are of interest in pharmaceutical sciences and asymmetric synthesis. Herein, SpinPhox/Ir-I catalysts have been demonstrated to be highly enantioselective in the asymmetric hydrogenation of the CC bonds in the exocyclic ,-unsaturated cyclic carbonyls, including a broad range of -alkylidene lactams, unsaturated cyclic ketones, and lactones. It is noteworthy that the procedure can be successfully used in the asymmetric hydrogenation of the challenging -alkylidenelactam substrates with six- or seven-membered rings, thus affording the corresponding optically active carbonyl compounds with an -chiral carbon center in generally excellent enantiomeric excesses (up to 98%ee). Synthetic utility of the protocol has also been demonstrated in the asymmetric synthesis of the anti-inflammatory drug loxoprofen and its analogue, as well as biologically important epsilon-aminocaproic acid derivatives.

Abstract:The first series of chiral phosphine-imidazole carbene ligands based on a 1,1'-binaphthyl framework were synthesized from (R)-2-amine-2'-(diphenylphosphino)-1,1'-binaphthyl (1) in a four-step pathway. After deprotonation of these phosphine-imidazolium salts with (LiOBu)-Bu-t, and subsequent complexation with [Ir(COD)Cl](2) and anion exchange with NaBArF, phosphine-carbene chelated iridium complexes (R)-6a and (R)-6b were obtained. Their structures have been characterized by NMR and X-ray diffraction analysis. The NHC-phosphine rhodium complex (R)-6c has been also obtained by a similar synthetic method. These iridium complexes have been applied to catalyze the asymmetric hydrogenation of alkenes to give the corresponding products in moderate to excellent conversion (up to 99%) and moderate enantioselectivities under mild conditions (up to 61% ee).

Abstract:The first asymmetric hydrogenation of 3-ylidenephthalides has been developed using the IrI complex of a spiro[4,4]-1,6-nonadiene-based phosphine-oxazoline ligand (SpinPHOX) as the catalyst, affording a wide variety of chiral 3-substituted phthalides in excellent enantiomeric excesses (up to 98 % ee). The utility of the protocol has been demonstrated in the asymmetric synthesis of chiral drugs NBP and BZP precursor, as well as the natural products chuangxinol and typhaphthalide.

Abstract:New chiral phosphine-oxazoline ligands (7, SIPHOX) with a rigid and bulky spirobiindane scaffold were synthesized, starting with optically pure 7-diphenylphosphino-7'-trifluoromethanesulfonyloxyl-1,1'-spirobiindane, in four steps in 40-64% overall yield. Iridium complexes of 7, the chiral analogues of the Crabtree catalyst, were generated by coordination of ligands 7 and [Ir(COD)Cl](2) in the presence of sodium tetrakis-3,5-bis(trifluoromethyl)phenylborate. The complexes were characterized by NMR, ESI-MS, and X-ray diffraction analysis. The Ir-SIPHOX complexes can catalyze the hydrogenation of acyclic N-aryl ketimines under ambient pressure with excellent enantioselectivities (up to 97% ee) and full conversions. This result represents the highest enantioselectivity and the first example of the hydrogenation of imines catalyzed by chiral analogues of the Crabtree catalyst at ambient pressure. Studies on the stability of the catalysts revealed that the catalysts Ir-SIPHOX are very stable and resistant to the formation of inactive trimers under hydrogenation conditions. On the basis of the X-ray diffraction analysis of the structures of catalysts and amine products, a rational explanation for the enantiocontrol of the chiral catalysts in the hydrogenation of imines is proposed.

Abstract:An iridium catalytic system with a ferrocene-based phosphine ligand bearing a modular and tunable unsymmetrical vicinal diamine scaffold was developed for the asymmetric hydrogenation of aryl ketones. This approach provided a powerful tool for the enantioselective synthesis of diverse chiral alcohols with excellent reactivity and enantioselectivity (up to 99% yield, up to 99% ee, and up to 50,000 turnover number). The substituents and chirality of unsymmetrical diamines in ligands played an important role in the satisfactory results.

Abstract:A new type of chiral phosphine–quinoline ligand bearing a spiro[4,4]-1,6-nonadiene scaffold was designed and synthesized. Its cationic iridium complex has been characterized by X-ray diffraction analysis and evaluated in the catalytic asymmetric hydrogenation of alkene and imine derivatives, giving the corresponding hydrogenation products with high activity albeit moderate enantioselecitivity.

Abstract:A new type of ferrocene-based phosphine-oxazoline ligand has been prepared over a few simple steps. An iridium complex of this ligand is air stable and exhibits excellent performance for the asymmetric hydrogenation of simple ketones (up to 98% yield, up to 99% ee, and 20 000 S/C). Exo-alpha,beta-unsaturated cyclic ketones could be regiospecifically hydrogenated to give chiral allylic alcohols with good results. This study indicates that P,N-ligands can also efficiently promote Ir-catalyzed asymmetric hydrogenation without NH-hydrogen-bonding assistance.

Abstract:An Ir-catalyzed asymmetric hydrogenation of 2-substituted 1,4-benzodioxines was developed for the preparation of chiral 1,4-benzodioxanes, which are present in numerous biologically active compounds and natural products. Our tropos biphenyl phosphine-oxazoline ligand is essential for obtaining good ee. A broad range of substrates were tolerable to the reaction conditions and gave the corresponding hydrogenation products in excellent yields and with moderate to good enantioselectivities using the Ir-complex of our tropos phosphine-oxazoline ligand.
