Abstract:A novel one-pot protocol for the synthesis of valuable 3,4-dihydropyridin-2-ones from the condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds in the presence of imidazole was developed. A series of aldehydes and 1,3-dicarbonyl compounds were employed to examine the scope of substrates for this protocol. This reaction proceeded through the formation of one ring and four new bonds (two C?C, one C?N, one C?C) via the sequence involving Knoevenagel condensation, Michael addition and intramolecular cyclization with moderate to excellent yields. All new compounds were characterized by IR, 1H NMR, 13C NMR and HRMS.

Abstract:A metal- and hypervalent iodine reagent-free electrochemical oxidative dearomatization-induced [5+2] cycloaddition/pinacol rearrangement cascade reaction was described. The electrosynthetic method showed strong tolerance for vinylphenols, ethynylphenols, and allenylphenols, which thus enabled the rapid assembly of diversely functionalized bicyclo[3.2.1]octanes in 41-95% yields and up to >20:1 dr. This protocol could be scaled up to gram amounts and should find wide application in complex natural product synthesis.

Abstract:A metal-free cascade of α-acyloxylation/carboxamidation of I^((III))/S^((VI))-ylides, carboxylic acids, and isonitriles via a Passerini-like multicomponent reaction is reported. Abstract:As an important building block, developing efficient and green synthesis strategy of cyclohex-2-enones is of great importance. In this present work, a general approach to the mild synthesis of substituted cyclohex-2-enones derivatives starting fro m simple aldehydes and acetone have been achieved via D-aminoacylase-initiated Aldol condensation/Robinson annulation cascade reaction using imidazole as an additive in organic media. The influences of reaction conditions including solvents, enzyme concentration, additives type, molar ratio of enzyme to additive, and substrate scopes were systematically investigated. Furthermore, some experiments were designed to explore the catalytic roles of D-aminoacylase and imidazole in the multistep cascade process, and one possible mechanism was proposed.

Abstract:A convenient Ni(II)-catalyzed C-C and C-N cascade coupling reaction was developed to directly access various 2,4-disubstituted imidazoles. Abstract:A catalytic asymmetric [3+2] cycloaddition of glycine ketimines with -CF3,-disubstituted enones was realized in the presence of a chiral copper(I)/Ming-Phos complex. This method provides an access to construct highly functionalized pyrrolidines bearing three contiguous stereocenters, which including a trifluoromethylated all-carbon quaternary stereocenter. The features of this reaction include high chemo-, diastereo-, enantioselectivity (up to >20:1 cr, >20:1 dr, 98% ee), readily available starting materials, well functional-group tolerance and mild reaction conditions. Control experiments demonstrate that the fluoro-substituent is crucial for the reactivity.

Abstract:A Cu(I)-catalyzed asymmetric [3 + 3] cycloaddition of azomethine ylides with 2-indolylnitroethylenes is described. Challenges confronted in this reaction include chemoselectivity between [3 + 2] cycloaddition and [3 + 3] cycloaddition, reactivity and stereoselectivity of intramolecular Friedel–Crafts reaction, and enantioselective control for constructing tetrahydro-γ-carbolines bearing multiple stereocenters. In the presence of copper(I)/Ph-Phosferrox complex, an array of chiral tetrahydro-γ-carboline compounds was generally obtained in moderate to high yields (up to 92%) with moderate to high chemoselectivities (up to 94:6 cr) and a high level of stereoselectivity (up to >98:2 dr, >99% ee in most cases). [Here, cr is the chemoselectivity ratio, dr is the diastereoisomeric ratio, and ee is enantiomeric excess.]
