Abstract:This paper reports a conceptually new and concise synthetic method leading to unprecedented, fully alkyl substituted pyranylidene carbene complexes 2 from M(CO)6 (M = Cr, Mo, W) and the readily available 1-lithio-1,3-butadiene 1, bearing a leaving group, via an intramolecular trapping of metallaacylate intermediates. Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:Cleavage of inert C-N bonds in unstrained azacycles such as pyrrolidine remains a formidable challenge in synthetic chemistry. To address this, we introduce an effective strategy for the reductive cleavage of the C-N bond in N-benzoyl pyrrolidine, leveraging a combination of Lewis acid and photoredox catalysis. This method involves single-electron transfer to the amide, followed by site-selective cleavage at the C2-N bond. Cyclic voltammetry and NMR studies demonstrated that the Lewis acid is crucial for promoting the single-electron transfer from the photoredox catalyst to the amide carbonyl group. This protocol is widely applicable to various pyrrolidine-containing molecules and enables inert C-N bond cleavage including C-C bond formation via intermolecular radical addition. Furthermore, the current protocol successfully converts pyrrolidines to aziridines, γ-lactones, and tetrahydrofurans, showcasing its potential of the inert C-N bond cleavage for expanding synthetic strategies.

Abstract:Despite the significant achievements in dearomatization and C-H functionalization of arenes, the arene ring-opening remains a largely unmet challenge and is underdeveloped due to the high bond dissociation energy and strong resonance stabilization energy inherent in aromatic compounds. Herein, we demonstrate a novel carbene assisted strategy for arene ring-opening. The understanding of the mechanism by our DFT calculations will stimulate wide application of bulk arene chemicals for the synthesis of value-added polyconjugated chain molecules. Various aryl azide derivatives now can be directly converted into valuable polyconjugated enynes, avoiding traditional synthesis including multistep unsaturated precursors, poor selectivity control, and subsequent transition-metal catalyzed cross-coupling reactions. The simple conditions required were demonstrated in the late-stage modification of complex molecules and fused ring compounds. This chemistry expands the horizons of carbene chemistry and provides a novel pathway for arene ring-opening.

Abstract:A photoinduced palladium‐catalyzed ring‐opening hydroarylation of furans to form functionalized indenes was achieved via an intramolecular reductive coupling of aryl bromide with furan using silane as the reductant. Deuterium tracking and DFT analysis revealed a reasonable reaction pathway involving: 1) the homolytic cleavage of the furan C−O bond induced by the α‐allyl radical; 2) a 1,6‐H atom transfer from carbon to oxygen.

Abstract:A regioselective C-C bond cleavage/aminocarbonylation cascade is presented. In recent years, tremendous progress has been made in the alkoxyl radical-mediated C-C bond cleavage of unstrained monocarbocycles. In contrast, the deconstruction and functionalization of bicyclic skeletons has been less developed and has mainly focused on the ring expansion process. Inspired by the aromatization-driven C-C bond cleavage, here we demonstrate a ring-opening/aminocarbonylation cascade under copper catalysis, in which the formation of a stable γ-lactam or succinimide skeleton reverses the selectivity of C-C cleavage. Remarkably, the photo and thermal assistance is not required when the succinimide skeleton is formed during the ring opening process. DFT calculations revealed that this unexpected ring-opening process is thermodynamically and kinetically favourable.

Abstract:A novel iodine promoted [1 + 3 + 2] cleavage cyclization reaction for the synthesis of 1H-pyrazolo[3,4-b]pyridine from aryl methyl ketones, 5-aminopyrazoles and enaminones has been established. This transition metal-free catalysis method has simple reaction conditions and good substrate compatibility, which has been demonstrated in the transformation of alkyl and natural molecule-derived enaminones. Mechanistic studies showed that two cyclization pathways affording different key intermediates were involved, but affording the same target product after site-selective cleavage of the unstrained C–C bond of the acyl group.

Abstract:Organic chemists now can construct carbon-carbon σ-bonds selectively and sequentially, whereas methods for the selective cleavage of carbon-carbon σ-bonds, especially for unreactive hydrocarbons, remain limited. Activation by ring strain, directing groups, or in the presence of a carbonyl or a cyano group is usually required. In this work, by using a sequential strategy site-selective cleavage and borylation of C(aryl)-CH3 bonds has been developed under directing group free and transition metal free conditions. Methyl groups of various arenes are selectively cleaved and replaced by boryl groups. Mechanistic analysis suggests that it proceeds by a sequential intermolecular oxidation and coupling of a transient aryl radical, generated by radical decarboxylation, involving a pyridine-stabilized persistent boryl radical.

Abstract:A direct electrochemical redox reaction involving radical cross‐coupling cyclization for the synthesis of pyrrolo[1,2‐a]quinoxaline derivatives from 1‐(2‐aminophenyl)pyrroles and CH3CN has been developed, which includes the functionalization of C(sp3)‐H bonds as well as the construction of C‐C and C‐N bonds. Notably, the control and deuterium‐labelling experiments suggest that CH3CN in this reaction acts as both a carbon source via C‐C cleavage and solvent. The reaction features metal‐ and oxidant‐free conditions, and various substituted pyrrolo[1,2‐a]quinoxaline derivatives were obtained.

Abstract:An efficient carbo-acylation reaction of 2-arylpyridines with alpha-diketones via Pd-catalyzed C-H bond activation and C-C bond cleavage in the presence of TBHP was developed that generated aryl ketones in good yields. The highly selective formation of aryl ketones was observed when 2-arylpyridines reacted with aromatic/aliphatic alpha-diketones.

Abstract:A novel synthesis method has been developed for the construction of functionalized 2,4-diarylpyridines. The reaction involves several key processes, including a reduction reaction, Michael reaction, decarboxylation, intramolecular cyclization, oxidation, ring-opening and aromatization reaction. All of these processes are facilitated by heating a mixture of the starting materials in the environmentally benign solvent, propylene carbonate (PC), with the oxidant (air) and the catalyst Cu(OAc)2. This cascade reaction not only forms two new bonds but also cleaves three existing bonds in a single pot. The protocol presented in this study is a valuable tool for synthesizing functionalized unsymmetrical 2,4-diarylpyridines.

Abstract:A direct synthesis of multi-substituted pyrrolo[1,2-a]pyrazines via palladium(II)-catalyzed C(sp)–C(sp2) cascade coupling and intramolecular cyclization in the presence of ligand was developed. This reaction originates from phenylboronic acids and readily synthesized 2-carbonyl- or 2-formylpyrroloacetonitriles, and affords products in good to excellent yields for a diversity of substrates. Additionally, a possible mechanism for the transformation is proposed.
