Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:In this study, we focused on the study of pharmacodynamic effects for 6 major bioactive lignans of Schisandra chinensis, namely deoxyschizandrin, schisandrin B, schisandrin C, schisandrin, schizandrol B and schisantherin A. A compound-gene-pathway network, which contained 124 related genes and 88 pathways, was constructed by collecting drug genes through mining relevant literatures and network pharmacology analysis. Based on the network analysis, 32 pathways and 80 related genes were associated with inflammation, which implied that anti-inflammatory might be the major pharmacodynamic effects of these compounds. All lignans except schizandrol B reduced LPS-induced NO production in RAW264.7 cells, which validated the anti-inflammatory hypothesis generated from network analysis. Furthermore, we investigated the effects of deoxyschizandrin, schisandrin C, schisandrin and schisantherin A on the secretion of inflammatory cytokines TNF-α, IL-1β, IL-6, PGE2 and protein expressions of iNOS, COX-2. As a result, deoxyschizandrin showed the strongest anti-inflammatory activity with inhibitory effect on all 4 inflammatory cytokines secretions and protein expressions of iNOS, COX-2. This study provided evidences for systematic exploration on the pharmacolgical actions and mechanisms of schisandra.

Abstract:Four new tetrahydrofuran lignans, schpropinrins A-D ( 1–4 ), together with five known ones, were isolated from the leaves and stems of Schisandra propinqua var. sinensis . Their structures, including absolute configurations, were characterized by means of spectroscopic analysis and ECD calculation. Compounds 1–4 featured a ketal or hemiketal substructure at C-7 and all of the isolates were tested for their anti-HIV integrase activity.

Abstract:Chemical investigation into the stems of the medicinal plantSchisandra sphaerandraled to the isolation and identification of a new dibenzocyclooctadiene lignan sphaerandrin A (1) and 11 known ones gomisin B (2), schirubrisin B (3), kadsuphilin B (4), schizandrin (5), benzoylgomisin Q (6), angeloylgomisin Q (7), gomisin G (8), schisanwilsonin O (9), isogomisin O (10), schisantherin D (11), and wuweizisu C (12). The structure of the new compound was elucidated by comprehensive spectroscopic methods including 1 D/2D NMR, HRESIMS, and CD spectrometry. To the best of our knowledge, compounds2 - 11were obtained from this species for the first time. All the compounds were evaluated for the cytotoxic activity against the triple-negative breast cancer cell lines MDA-MB-231 and HCC-1937.

Abstract:Six new lignans (1-6), as well as five known ones (7-11) were isolated from the leaves and stems of Schisandra chinensis. The structures of 1-6 were established on the basis of spectroscopic methods including 1D-and 2D-NMR techniques and CD experiments. Compound 1 was the first example of naturally occurring N-containing lignans featuring a nicotinoyl group. All the new compounds were evaluated for their anti-HIV-1 activities and showed EC50 values in the range 17.89-138.23 mu g/mL.

Abstract:AIM: To study the chemical constituents of the seeds of Schisandra sphenanthera. METHOD: The chemical constituents of Schisandra sphenanthera were isolated with chromatographic techniques, and their chemical structures were elucidated by spectral analysis. RESULT and CONCLUSION: Sphenanlignan(1), a new 2,3-dimethyl-1,4-diarylbutane lignan was isolated with 6 known lignans (+)-anwulignan(2), schisantherin A(3), deoxyschizandrin(4), tigloylgomisin O(5), 6-O-benzoylgomisin O(6) and schisandrone(7). 6-O-benzoylgomisin O(6) was obtained from Schisandra sphenanthera for the first time.

Abstract:Two new dibenzocyclooctadiene lignans angeloyl-(+)-gomisin K(3) (1) and methylisogomisin O (2), together with six known ones, isogomisin O, angeloylisogomisin O, gomisin O, angeloygomisin O, benzoylgomisin O, epigomisin O, and four 1,4-bis(phenyl)-2,3-dimethylbutane type lignans, pregomisin, meso-dihydroguaiaretic acid, isoanwulignan, and sphenanlignan were isolated from the aerial parts of Schisandra propinqua var. propinqua. Abstract:Schisandra chinensis (Schisandraceae) is a medicinal plant widely used in traditional Chinese medicine. Under the name Wu Wei Zi, it is used to treat many diseases, especially as a stimulant, adaptogen, and hepatoprotective. Dibenzocyclooctadiene lignans are the main compounds responsible for the effect of S. chinensis. As a part of ongoing studies to identify and evaluate anti-inflammatory natural compounds, we isolated a series of dibenzocyclooctadiene lignans and evaluated their biological activity. Furthermore, we isolated new sesquiterpene 7,7-dimethyl-11-methylidenespiro[5.5]undec-2-ene-3-carboxylic acid. Selected dibenzocyclooctadiene lignans were tested to assess their anti-inflammatory potential in LPS-stimulated monocytes by monitoring their anti-NF-κB activity, antioxidant activity in CAA assay, and their effect on gap junction intercellular communication in WB-ras cells. Some S. chinensis lignans showed antioxidant activity in CAA mode and affected the gap junction intercellular communication. The anti-inflammatory activity was proven for (−)-gomisin N, (+)-γ-schisandrin, rubrisandrin A, and (−)-gomisin J.

Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:Schsphenines A ( 1 ) and B ( 2 ), two new lignans were isolated from the fruit of Schisandra sphenanthera , together with six known ones respectively identified as schisanhenol ( 3 ), methylgomisin O ( 4 ), wuweilignan E ( 5 ), neglschisandrin E ( 6 ), schisandrin A ( 7 ) and schisandrin B ( 8 ). The structures of isolated compounds were determined by UV, IR, HR-ESI-MS, NMR, ORD analysis and quantum chemical calculation. The in vitro cardioprotective activities of all compounds were studied on H 2 O 2 -injury H9C2 cells using the MTT method. Compound 1 exhibited a considerable protective effect with cell viability in 79.94 ± 2.36%. This research provided useful information for the utilisation of S. sphenanthera as the TCM.

Abstract:Phytochemical investigation of an ethanolic extract of stems of Schisandra neglecta led to the isolation and identification of two new dibenzocyclooctadiene lignans, designated neglschisandrins E (1) and F (2), and thirteen known lignans. All structures and stereochemistries were determined by spectroscopic methods, including 2D-NMR techniques. The isolates were evaluated for in vitro cytotoxic activity. Among them, compounds 2–6 exhibited moderate to weak cytotoxicity against the human colorectal carcinoma HCT-8 cell line with EC50 values of 7.33~19.8 μg/mL. In addition, compounds 2–4 also exhibited marginal cytotoxicity against the human lung carcinoma A549 cell line with EC50 values of 11.8~15.0 μg/mL.

Abstract:Two rare 7,8-seco-lignans (1, 2), three new lignan glycosides (3, 4a, 4b), and 10 known lignans (5-14) were isolated from the fruit of Schisandra glaucescens Diels. The absolute configurations of 1 and 2 were determined by comparing their experimental and calculated electronic circular dichroism spectra. The molecular structures of the new compounds (3, 4a, and 4b), including their absolute configurations, were determined using various spectroscopic methods and hydrolysis reactions. The antioxidant activities of the isolated compounds were tested using 2,2-diphenyl-1-picrylhydrazyl and ferric reducing antioxidant power assays. Compounds 4, 7, 8, 10, 11, and 12 exhibited antioxidant activities of varying potential in both assays. Of these compounds, 7 showed the strongest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity, with IC50 values of 15.7 (150 μM DPPH) and 34.6 μM (300 μM DPPH), respectively, and 4, 12, and 7 displayed higher total antioxidant activities than Trolox in the ferric reducing antioxidant power assay. The neuroprotective effects of these compounds against Aβ25-35-induced cell death in SH-SY5Y cells were also investigated. Compounds 1, 2, 6, 7, 8, 11, and 12 exhibited statistically significant neuroprotective effects against Aβ25-35-induced SH-SY5Y cell death compared with the group treated only with Aβ25-35.
