Abstract:The surface hydrophobicities of bovine serum albumin (BSA) and bovine immunoglobulin G (bIgG) were studied by a fluorescence method with 8-anilino-1-naphthalene sulfonate (ANS) as the probe. Effects of factors including pH, temperature and additives on the surface hydrophobicity indexes (S0) were investigated. The results show that pH can significantly affect the determination of S0, as ANS is an anionic fluorescent probe. Solution pH slightly higher than isoelectric point of proteins is suitable. Temperature has different effects on S0 of BSA and bIgG. S0 of BSA decreases from 50℃ to 80℃, while that of bIgG increases. Salt addition could enhance the surface hydrophobicity of both proteins, and bIgG has much greater variation than that of BSA. Depending on the denaturation mechanisms, surface hydrophobic properties and molecular structure of proteins, the denaturants have different impacts on S0 of the two proteins. With the increase of sodium caprylate (NaCA) concentration, S0 of BSA declines and that of bIgG remaines unchanges. This result is consistent with the impacts of NaCA on the separation of BSA and bIgG in hydrophobic interaction chromatography. This study demonstrates that the ANS fluorescence probe method can be used to investigate the effects of liquid conditions on protein surface hydrophobicity and study the hydrophobic interactions between specific additives and proteins.
Protein adsorption kinetics of mixed-mode adsorbent with benzylamine as functional ligand

Abstract:Adsorption kinetics of bovine serum albumin (BSA) to mixed-mode adsorbent with benzylamine ligand was studied via stirred-batch uptake experiments. The effects of liquid-phase conditions, such as salt concentration and pH, on the uptake rates of BSA were evaluated by effective pore diffusivity (De) derived from the pore diffusion model (PDM). The results indicated that when electrostatic attractive interactions exist between protein and ligand, De increases firstly with increasing salt concentration and then turns to decreasing after reaching the maximum. When there are electrostatic repulsion protein/ligand interactions, it seems that the adsorption process is patch controlled, and the specific salt concentration to result in a minimum adsorption capacity could be found. The value of De showed a similar tendency to the change of adsorption capacities. With the increase of salt concentration, the increase in De value was considered due to the decrease of electrostatic repulsion interactions and increase of hydrophobic interactions between BSA and ligand. Since BSA has the most compact structure at the isoelectric point, which leads to lower hydrodynamic forces, it is reasonable that the maximum of effective diffusion coefficient could be found around the isoelectric point.

Abstract:The adsorption mechanisms of bovine serum albumin (BSA) on mixed-mode adsorbents with benzylamine as the functional ligand were discussed in terms of the isotherm adsorption behavior and chromatographic retention under various conditions. The adsorbents showed the salt-tolerant and pH-dependent properties for protein binding. The results indicated that the electrostatic interactions might be an important contributor to the protein adsorption under the conditions of electrostatic attractive protein–ligand interactions. While there are some amounts of electrostatic repulsion protein–ligand interactions, it seems that the adsorption process is patch controlled, and the specific salt concentration to result in a minimum adsorption capacity could be found. Accordingly the isocratic retention factors have a minimum value under a certain salt concentration, and showed the typical “U” shape curve as the function of salt concentration. In addition, the total number of released water molecules during protein adsorption was analyzed using the preferential interaction theory, and was found to be consistent with the estimated hydrophobic contact area between the bound protein and adsorbent surface.

Abstract:The self-assembly adsorption of bovine serum albumin (BSA) on the hybrid surface including the discontinuous gold nano film and silica under liquid condition was studied in situ by atomic force microscope (AFM) at room temperature. To investigate the adsorption of BSA on silica and gold nano film simultaneously, the surface with discontinuous gold nano film and visible blank block on silica by physical vapor deposition (PVD) and the ultrasonic cleaning technology was fabricated. Then we observed the variation on the substrate surface before and after injecting BSA aqueous solution with a 0.05 mg/ml concentration at real time by AFM. The experimental results indicated that the adsorption strength of BSA on the surface of gold nano film is larger than that on silica surface, and further implied that the adsorption capability of the hydrophobic surface is larger than that of the hydrophilic, which is consistent with those reported. Moreover, it could be easily observed that the adsorption of BSA on gold nano film has great tendency to form a ridgeway-like topography.

Abstract:The isotherm adsorption behavior of bovine serum albumin on the mixed-mode cellulose-stainless steel powder composite adsorbent with benzylamine as the functional ligand (named Cell-SSP-BA) was measured using batch experiments. Four separate adsorbents with different ligand densities were studied under varying salt concentrations and pH. The mixed-mode adsorbent Cell-SSP-BA showed the salt-tolerant and pH-dependent properties for protein binding. An empirical modification of the Langmuir equation has been proposed to describe the effects of both salt concentration and pH on the isotherm behaviors of mixed-mode adsorption. The results indicated that the modified Langmuir equation could correlate the protein adsorption at different salt concentrations and pH with only one set of parameters.

Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:In this work, the influence of particle size and surface functional groups on the adsorption behavior of bovine serum albumin (BSA) by three types of oxide nanoparticles (NPs), TiO(2) (50±5 nm), SiO(2) (30±5 nm), and Al(2)O(3) (150±5 nm for α type and 60±5 nm for γ type) was investigated in deionized water, in order to explore their interaction mechanisms without competitive influence of other ions. Bulkparticles (BPs) were also used for comparison with NPs. BSA adsorption maxima on oxide particles were controlled by the surface area and hydrogen content, while adsorption process was primarily induced by electrostatic interaction, hydrophobic interaction and ligand exchange between BSA and oxide surfaces. With the increase of hydrogen content, the BSA adsorption mechanism switched from mainly hydrophobic interaction to hydrogen bonding and ligand exchange. Calculations, based on surface area and BSA size, suggested that a multilayer of BSA covered on α-Al(2)O(3), and single layer on the other oxide particle surfaces. BPs led to greater conformational change of BSA molecules after the adsorption on the surfaces of oxide particles though NPs adsorbed more BSA than BPs.

Abstract:The protein adsorption is a crucial but complex phenomenon in many applications, like biosensors, biomaterials. The amount, orientation and biological activity of adsorbed proteins are heavily dependent upon the structure of protein, like widely-applied Bovine Serum Albumin (BSA), and the properties of the substrate. At some extent, to explore the physicochemical properties and dynamics interaction process between protein / multi-proteins with crystal/biomaterial surface, it has been presented that how to dig out the potential ability of the apparatus and other methods/tools for removing the block of studying interactions and measuring these complex interactions with crystal environments efficiently. In our recent works, it was demonstrated how to detect the BSA under aqueous conditions by steering the AFM tip in contact mode to sample more abundant topographical clues. The results indicated that this approach should be available for exploring more complex interaction system and acquiring more useful interaction information without altering the current apparatus platforms.
Influences of ligand density and pore size on BSA adsorption on agarose-based DEAE-ion-exchange resins

Abstract:Ion exchange chromatography (IEC)is a common and powerful technique for the purification of proteins. The ligand density and pore size of the resins have significant effects on the separation behaviors of protein. In the present work, three kinds of crossed-linked agarose gel with different agarose concentrations were used as the matrices to represent various pore sizes, and anionic ligand, diethylaminothyl (DEAE), was coupled with different ligand densities. The preparation conditions, including the reaction temperature, time, DEAE concentration and NaOH concentration, were optimized to control the ionic exchange capacity. A series of DEAE resins with different ligand densities and pore sizes were obtained. The adsorption isotherms and kinetics of bovine serum albumin (BSA) were measured on these resins. The saturated adsorption capacity increased with the increasing of ligand densities and the decreasing of pore sizes at the range tested. For the adsorption kinetics, the pore size influenced significantly the effective pore diffusivities. The results demonstrated that both the pore size and ligand density affect the static and dynamic adsorption. The pore size dominates the diffusion and mass transfer of protein, while the ligand density affects the interactions between ligand and protein. © All Rights Reserved.

Abstract:Understanding protein adsorption behaviors on solid surfaces constitutes an important step toward development of efficacious and biocompatible medical devices. Both surface charge and wettability have been shown to influence protein adsorption attributes, including kinetics, quantities, deformation, and reversibility. However, determining the dominant interaction in these surface-induced phenomena is challenging because of the complexity of inter-related mechanisms at the liquid/solid interface. Herein, we reveal the dominant interfacial forces in these essential protein adsorption attributes under the influence of a combination of surface charge and wettability, using quartz crystal microbalance with dissipation monitoring and atomic force microscopy-based force spectroscopy on a series of model surfaces. These surfaces were fabricated via layer-by-layer assembly, which allowed two-dimensional control of surface charge and wettability with minimal cross-parameter dependency. We focused on a soft globular protein, bovine serum albumin (BSA), which is prone to conformational changes during adsorption. The information obtained from the two techniques shows that both surface charge and hydrophobicity can increase the protein-surface interaction forces and the adsorbed amount. However, surface hydrophobicity triggered a greater extent of deformation in the adsorbed BSA molecules, leading to more dehydration, spreading, and resistance to elution by ionic strength changes regardless of the surface charge. The role played by the surface charge in the adsorbed protein conformation and extent of desorption induced by changes in the ionic strength is secondary to that of surface hydrophobicity. These findings advance the understanding of how surface chemistry and properties can be tailored for directing protein-substrate interactions.

Abstract:The mode of adsorption of bovine serum albumin (BSA) on porous polyethylene (PE) membrane was studied as a function of time and concentration, which may contribute to the surface coverage. An improved physical model for adsorption is initiated based on the results of the adsorptional and desorptional measurements, FTIR analysis, and AFM observations as well as streaming potential measurements. The results obtained indicate that the adsorptional mode depend on both time and concentration. It is shown that a critical concentration (about 1000 ppm here) exists in the adsorptional process. Below this concentration, the adsorption seems to be conducted in a normal side-on way but time elapse gives rise to greater conformational change than concentration increase; above this concentration, the aggregation of protein molecular plays a decisive role and the adsorption is in an aggregation way, which is similar to end-on, but a relative large gap between the adsorbed molecules exists due to aggregation. This conclusion is general and can be expected to apply in other globular protein–hydrophobic porous surface systems.

Abstract:The adsorption of a model protein such as bovine serum albumin (BSA) on a magnetic material based on montmorillonite (MtMag) was studied. Kinetic data, equilibrium isotherms, and thermodynamic calculations were performed and analyzed to obtain information about the rate-limiting step of the adsorption process and propose a possible adsorption mechanism. Equilibrium studies showed optimal adsorption around pH 4.5, the maximum adsorption capacity being around 231.9 mg g -1 at 25 °C. The latter behavior may be ascribed to the most packed protein conformation at pH conditions near the BSA isoelectric point. In addition, the BSA adsorption capacity decreased with the ionic strength (10 –3 −10 –1 M) and increased with the temperature (15–30 °C). In all cases, the Sips model yielded the best fit to the experimental results. The thermodynamic analysis revealed an isosteric heat dependence on the coverage, which allowed ruling out the Langmuir model. The kinetic data were adequately fitted by the IPD model, which suggests that the rate-limiting step of the adsorption process is the diffusion of BSA molecules into the internal sites.

Abstract:Interactions between a series of N-alkyl-N,N-di(2-hydroxyethyl)-N-methylammonium bromides with alkyl chain from dodecyl to cetyl and bovine serum albumin (BSA) in Tris-HCl buffer solution (pH=7.1) were studied by fluorescence spectroscopy at 298 K. Effects of the surfactant structure and BSA concentration on the interactions were examined. The interaction mechanism between BSA and surfactants at a relatively low concentration range was discussed using the Stern-Volmer equation and a model of the pseudo association constant. Each of the quaternary ammonium surfactants has a fluorescence quenching effect on BSA and leads to a blue shift of the maximum emission wavelength. The Stern-Volmer quenching constant and the pseudo association constant increase, which indicates that the interactions between surfactants and BSA become stronger, as the hydrophobic chain lengths increase.

Abstract:In this research, adsorption properties of bovine serum albumin (BSA) on diatomite clay, which is an oxide mineral, were studied as a function of BSA, sodium phosphate buffer and protein concentration and pH and the thermodynamic parameters of adsorption process were investigated. The BSA adsorption experiment onto diatomite clay indicated that the BSA solution reached the maximum adsorption value at pH 5.5. It was observed that the maximum adsorption capacity (qm) of the data obtained from the adsorption studies showed a great dependence on pH. The maximum amount of adsorption in adsorption experiments can be considered as points where the electrostatic interaction for pH is appropriate. Both structural and electrostatic interaction in regions outside of the isoelectric point may have caused a decrease in BSA absorbance. The structural influences were associated with different conformational states that while BSA molecules accept changes with pH, electrostatic effects can be observed in BSA molecules behaved like soft particles. In this case, it is not possible to explain the independence of the qm–pH curves of the amount of adsorption. The protein molecules at this point are very stable. Because this value is close to the isoelectric point of serum albumin. The surface structural change of BSA and diatomite clay was studied. For this, Fourier transform infrared spectroscopy (FTIR) spectroscopy values were compared before and after the experiment. The diatomite samples used as support material were characterized by FTIR, scanning electron microscopy, thermogravimetric analysis and Brunauer Emmett–Teller surface area analysis. The thermodynamic functions such as enthalpy, entropy, Gibbs free energy and activation energy were investigated in their experimental work. The thermodynamic parameters such as Gibbs free energy (ΔG*), Ea, ΔH* and ΔS* were calculated as − 67.45, 15.41, − 12.84 kJ mol−1 and − 183.28 J mol−1 K−1 for BSA adsorption, respectively. We can deduce that the adsorption process from the data obtained from the thermodynamic parameters is spontaneous and exothermic. The adsorption of the process was investigated using Eyring and Arrhenius equations, and its adsorption kinetic found to be coherent with the pseudo-second-order model. As a result, we reached that the diatomite clay is a suitable adsorbent for the BSA. Experimental results showed that diatomite clay has the potency to be used for rapid pretreatment in the process of identifying proteins.

Abstract:Attenuated total reflection (ATR) FT-IR spectroscopy was used to quantitatively characterize the extent of bovine serum albumin (BSA) adsorbed on the surface-coating-modified poly(ether urethane) (PEU) matrix. The two surface modifying additives (SMA) were respectively a tri-block-coupling-polymer of stearyl poly (ethylene oxide)-4,4'-methylene diphenyl diisocyanate-stearyl poly(ethylene oxide), for short MSPEO, and another similar block-coupling polymer with the Cibacron Blue F3G-A endgroups, for short cibaMPEO. The experiments of static BSA adsorption were composed by two parts. One was static isothermal adsorption, and the other was static adsorption kinetics. The quantitative characterization was based on the optical principles of FT-IR, method of experiment and index of the apparatus, by which the enhancement of BSA adsorption on the SMA-modified surfaces was confirmed.
Protein adsorption at solid-liquid interfaces: Part IV--Effects of different solid-liquid systems and various neutral salts

Abstract:Adsorption isotherms of BSA at the solid-water interfaces have been studied as a function of protein concentration, ionic strength of the medium, pH and temperature using silica, barium sulphate, carbon, alumina, chromium, ion-exchange resins and sephadex as solid interfaces. In most cases, isotherms for adsorption of BSA attained the state of adsorption saturation. In the presence of barium sulphate, carbon and alumina, two types in the isotherms are observed. Adsorption of BSA is affected by change in pH, ionic strength and temperature of the medium. In the presence of metallic chromium, adsorbed BSA molecules are either denatured or negatively adsorbed at the metallic interface. Due to the presence of pores in ion-exchange resins, adsorption of BSA is followed by preferential hydration on resin surfaces in some cases. Sometimes two steps of isotherms are also observed during adsorption of BSA on the solid resins in chloride form. Adsorption of BSA, beta-lactoglobulin, gelatin, myosin and lysozyme is negative on Sephadex surface due to the excess adsorption of water by Sephadex. The negative adsorption is significantly affected in the presence of CaCl2, KSCN, LiCl, Na2SO4, NaI, KCl and urea. The values of absolute amounts of water and protein, simultaneously adsorbed on the surface of different solids, have been evaluated in some cases on critical thermodynamic analysis. The standard free energies (delta G0) of excess positive and negative adsorption of the protein per square meter at the state of monolayer saturation have been calculated using proposed universal scale of thermodynamics. The free energy of adsorption with reference to this state is shown to be strictly comparable to each other. The magnitude of standard free energy of transfer (delta G0B) of one mole of protein or a protein mixture at any type of physiochemical condition and at any type of surface is observed to be 38.5 kJ/mole.
