Abstract:Adsorption kinetics of bovine serum albumin (BSA) to mixed-mode adsorbent with benzylamine ligand was studied via stirred-batch uptake experiments. The effects of liquid-phase conditions, such as salt concentration and pH, on the uptake rates of BSA were evaluated by effective pore diffusivity (De) derived from the pore diffusion model (PDM). The results indicated that when electrostatic attractive interactions exist between protein and ligand, De increases firstly with increasing salt concentration and then turns to decreasing after reaching the maximum. When there are electrostatic repulsion protein/ligand interactions, it seems that the adsorption process is patch controlled, and the specific salt concentration to result in a minimum adsorption capacity could be found. The value of De showed a similar tendency to the change of adsorption capacities. With the increase of salt concentration, the increase in De value was considered due to the decrease of electrostatic repulsion interactions and increase of hydrophobic interactions between BSA and ligand. Since BSA has the most compact structure at the isoelectric point, which leads to lower hydrodynamic forces, it is reasonable that the maximum of effective diffusion coefficient could be found around the isoelectric point.

Abstract:The adsorption mechanisms of bovine serum albumin (BSA) on mixed-mode adsorbents with benzylamine as the functional ligand were discussed in terms of the isotherm adsorption behavior and chromatographic retention under various conditions. The adsorbents showed the salt-tolerant and pH-dependent properties for protein binding. The results indicated that the electrostatic interactions might be an important contributor to the protein adsorption under the conditions of electrostatic attractive protein–ligand interactions. While there are some amounts of electrostatic repulsion protein–ligand interactions, it seems that the adsorption process is patch controlled, and the specific salt concentration to result in a minimum adsorption capacity could be found. Accordingly the isocratic retention factors have a minimum value under a certain salt concentration, and showed the typical “U” shape curve as the function of salt concentration. In addition, the total number of released water molecules during protein adsorption was analyzed using the preferential interaction theory, and was found to be consistent with the estimated hydrophobic contact area between the bound protein and adsorbent surface.
Adsorption of IgG and BSA on Two Chromatographic Resins—Poly(ethylenimine)-4FF Resin and Tetrapeptide-poly(ethylenimine)-4FF Resin

Abstract:Poly(ethylenimine) (PEI) as a long flexible chain has been well used in protein chromatography with the advantages of high spatial utilization and "chain delivery" effect. In this work, two resins, i.e., poly(ethylenimine)-4FF resin (PEI-4FF resin) and Ac-YFRH-PEI-4FF resin by grafting tetrapeptide to PEI-4FF resin, were prepared for the protein adsorption and chromatographic separation. The pH-dependent adsorption properties of PEI-4FF resin and Ac-YFRH-PEI-4FF resin were discovered by getting the data of adsorption capacities of bovine serum albumin (BSA) and immunoglobulin G (IgG) under different pH conditions and buffer systems. The results showed high binding capacities of BSA on PEI-4FF resins across a range of pH values (5.0-8.9) with almost very little adsorption of IgG over a pH range from 5.0 to 8.0, indicating that IgG could be separated by flow-through method using PEI-4FF resin. Since PEI has the advantage of having many flexible branched chains and Ac-YFRH (Ac-Try-Phe-Arg-His) as a peptide ligand has the advantages of comparatively high stability and low immunogenicity for antibody adsorption, Ac-YFRH-PEI-4FF resins were prepared by using PEI as the spacer arm and Ac-YFRH as the peptide ligand. The isotherm adsorption of the Ac-YFRH-PEI-4FF resin showed that the maximum binding capacity of IgG (q(m) = 123.6 mg/g resin, K-d = 0.10 mg/mL) was far more than that of BSA (q(m) = 62.0 mg/g resin, K-d = 0.25 mg/mL) at pH 8.0. Meanwhile, this tetrapeptide resin also showed high binding capacity of BSA (q,, = 131.0 mg/g resin, K-d = 0.05 mg/mL) and low binding capacity of IgG (q(m) = 2.0 mg/g resin, K-d = 0.01 mg/mL) at pH 5.0, indicating that the separation of BSA and IgG could be achieved with the big differences in adsorption behaviors.
Influences of ligand density and pore size on BSA adsorption on agarose-based DEAE-ion-exchange resins

Abstract:Ion exchange chromatography (IEC)is a common and powerful technique for the purification of proteins. The ligand density and pore size of the resins have significant effects on the separation behaviors of protein. In the present work, three kinds of crossed-linked agarose gel with different agarose concentrations were used as the matrices to represent various pore sizes, and anionic ligand, diethylaminothyl (DEAE), was coupled with different ligand densities. The preparation conditions, including the reaction temperature, time, DEAE concentration and NaOH concentration, were optimized to control the ionic exchange capacity. A series of DEAE resins with different ligand densities and pore sizes were obtained. The adsorption isotherms and kinetics of bovine serum albumin (BSA) were measured on these resins. The saturated adsorption capacity increased with the increasing of ligand densities and the decreasing of pore sizes at the range tested. For the adsorption kinetics, the pore size influenced significantly the effective pore diffusivities. The results demonstrated that both the pore size and ligand density affect the static and dynamic adsorption. The pore size dominates the diffusion and mass transfer of protein, while the ligand density affects the interactions between ligand and protein. © All Rights Reserved.

Abstract:The self-assembly adsorption of bovine serum albumin (BSA) on the hybrid surface including the discontinuous gold nano film and silica under liquid condition was studied in situ by atomic force microscope (AFM) at room temperature. To investigate the adsorption of BSA on silica and gold nano film simultaneously, the surface with discontinuous gold nano film and visible blank block on silica by physical vapor deposition (PVD) and the ultrasonic cleaning technology was fabricated. Then we observed the variation on the substrate surface before and after injecting BSA aqueous solution with a 0.05 mg/ml concentration at real time by AFM. The experimental results indicated that the adsorption strength of BSA on the surface of gold nano film is larger than that on silica surface, and further implied that the adsorption capability of the hydrophobic surface is larger than that of the hydrophilic, which is consistent with those reported. Moreover, it could be easily observed that the adsorption of BSA on gold nano film has great tendency to form a ridgeway-like topography.

Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:The resin mixed PES-based fibrous adsorbents with open pore structure on the surface were prepared, and their adsorption behavior with BSA was studied. The adsorption isotherm demonstrated that the adsorption was well fitting the Langmuir model. The maximum adsorption capacity was about 139 mg BSA/g adsorbent. The resin Lewatit CNP80ws mixed PES-based fibrous adsorbent with more open pore structure on its surface had much more adsorption capacity with high adsorption rate for BSA. The effective diffusion coefficient of BSA in different resin mixed PES-based fibrous adsorbents was in the range of 1.82 x 10(-14) similar to 8.7 x 10(-14) m(2)/s. The values of diffusion coefficient of BSA in these fibrous adsorbents were comparable with those in resins reported in the literatures. The influence of operation parameters on the behavior of adsorption and desorption of BSA for the weak acidic type cation exchange resin mixed PES-based fibrous adsorbent in different structures was also investigated in this work. The pH of BSA buffer solutions was a very important parameter when the ion-exchange adsorbent was used for separation of proteins. The result indicated that the adsorption capacity was very low when the pH of BSA solutions was above the isoelectric point of BSA.

Abstract:In this work, the influence of particle size and surface functional groups on the adsorption behavior of bovine serum albumin (BSA) by three types of oxide nanoparticles (NPs), TiO(2) (50±5 nm), SiO(2) (30±5 nm), and Al(2)O(3) (150±5 nm for α type and 60±5 nm for γ type) was investigated in deionized water, in order to explore their interaction mechanisms without competitive influence of other ions. Bulkparticles (BPs) were also used for comparison with NPs. BSA adsorption maxima on oxide particles were controlled by the surface area and hydrogen content, while adsorption process was primarily induced by electrostatic interaction, hydrophobic interaction and ligand exchange between BSA and oxide surfaces. With the increase of hydrogen content, the BSA adsorption mechanism switched from mainly hydrophobic interaction to hydrogen bonding and ligand exchange. Calculations, based on surface area and BSA size, suggested that a multilayer of BSA covered on α-Al(2)O(3), and single layer on the other oxide particle surfaces. BPs led to greater conformational change of BSA molecules after the adsorption on the surfaces of oxide particles though NPs adsorbed more BSA than BPs.
Immobilization of bovine serum albumin onto spherical polyelectrolyte brushes in aqueous solution

Abstract:The adsorption of bovine serum albumin (BSA) in spherical poly(acrylic acid) brushes (SPB) was observed by the methods of turbidimetric titration, rheology, dynamic light scattering (DLS), Zeta potential measurement, and UV spectroscopy. Turbidimetric titration and DLS were used to examine the binding boundaries of protein-polyelectrolyte. The critical pH values where adsorption was initiated (pH <sub xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">c</sub> ) and aggregation occurred (pHφ) were determined by turbidimetric titration and DLS, respectively. Adsorption of BSA by SPB occurred at range of 4.6 <; pH <; 7.0 as observed by both turbidimetric titration and DLS. High ionic strength can damage the binding between BSA and SPB. Upon decreasing pH value, the viscosity, the elastic modulus G' and viscous modulus G” of SPB binding of BSA increased significantly. The binding amount of BSA in SPB was determined by UV spectroscopy.

Abstract:Carbon-coated Fe3O4 (Fe3O4/C) microspheres activated with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC) were prepared, characterized and applied to adsorb bovine serum albumin (BSA). The prepared magnetic microspheres had spherical core-shell structure with a uniform and continuous carbon coating coupled with activation by EDC, and possessed superparamagnetic characteristics. The experimental results showed that the adsorption amount of BSA on the EDC-activated Fe3O4/C (Fe3O4/C-EDC) microspheres was higher than that on the Fe3O4/C microspheres. The maximum adsorption of BSA on Fe3O4/C-EDC microspheres occurred at pH 4.7, which was the isoelectric point of BSA. At low concentrations (below 1.0 M), salt had no noticeable effect on BSA adsorption. The BSA adsorption of Fe3O4/C-EDC microspheres had a better fit to the Langmuir model than the Freundlich isotherm and Temkin isotherm model, and the kinetic data were well described by the pseudo-second-order model. The adsorption equilibrium could be reached within 20 min. High desorption efficiency (97.6%) of BSA from Fe3O4/C-EDC microspheres was obtained with 0.5 M Na2HPO4 (pH 9.4) as the desorbent.

Abstract:The cation-exchange resin-mixed polyethersulfone (PES)-based fibrous adsorbents were developed to study their adsorption behavior with bovine serum albumin (BSA). A fibrous adsorbent with an open pore surface had much better adsorption behavior with a higher adsorbing rate. The adsorption capacity of the Lewatit CNP80ws resin or SP112ws mixed PES-based fibrous adsorbents with an open pore surface was in the range of 68.2–93.2 mg BSA/g adsorbent for 50% resin loading, which was comparable with the resin static adsorption capacity. Compared to a thick fibrous adsorbent, a fibrous adsorbent with a thin diameter had the higher adsorption capacity with BSA. The results showed that the adsorption for cation-exchange resin-mixed PES-based fibrous adsorbents were controlled by a diffusion process whether in matrix pores of the adsorbent or that of resins.

Abstract:Two kinds of pH-dependent magnetic nanoadsorbent based on silica-coated MnFe2O4 nanoparticles (SMNPs) and amino-modified silica-coated MnFe2O4 nanoparticles (AS-MNPs) have been synthesized using sol–gel method. X-ray diffraction, Fourier transform infrared spectroscopy and transmission electron microscopy have been employed to characterize the structure and morphology of the nanoadsorbents. The magnetic properties and zeta potential were also investigated. These nanoadsorbents were used to adsorb bovine serum albumin (BSA). The adsorption capacity of BSA largely depended on the pH and ionic strength of solution. In the best result, BSA was adsorbed on AS-MNPs at a high value of 164mgg−1, which is much higher than that of SMNPs (100mgg−1). This may due to the increased surface amino that can be conjugated to BSA by a chemical bond and the electrostatic attraction between BSA and magnetic nanoparticles.
Adsorption Behavior of Bovine Serum Albumin by Histidine Grafted Chitosan Porous Scaffolds

Abstract:Six type of histidine grafted chitosan(NHCS)porous scaffolds were synthesized by changing the mole ratio of histidine/chitosan and the molecular weight of the chitosan.Adsorption behavior of bovine serum albumin(BSA)by NHCS in simulated body fluid(SBF)was investigated.Ultraviolet-visible spectrophotometer was used to determine the adsorptive equilibrium curve at different initial concentrations and the adsorptive dynamic curve at different temperatures.The results indicate the adsorption capacity of 50kD-NHCS-3(S4, Mw(CS)is 50×103,n(histidine)/n(chitosan)is 3∶1)is the best of all studied NHCS porous scaffolds,the maximum adsorption capacity is 820.90 mg/g at 37.0℃,while the initial concentration of BSA solution is 2.5 mg/mL,The adsorption kineticsis in good agreement with pseudo first-order kinetic model,whereas,the adsorption isotherms could be well described by Langmuir equation.The adsorption of BSA is a spontaneous,entropy increasing,endothermic and physical process.50kD-NHCS-3 is a good reusable matrix for adsorption and separation of BSA.
Penetrable silica microspheres for immobilization of bovine serum albumin and their application to the study of the interaction between imatinib mesylate and protein by frontal affinity chromatography

Abstract:In the current study, novel featured silica, named penetrable silica, simultaneously containing macropores and mesopores, was immobilized with bovine serum albumin (BSA) via Schiff base method. The obtained BSA-SiO 2 was employed as the high-performance liquid chromatographic (HPLC) stationary phase. Firstly, d - and l -tryptophan were used as probes to investigate the chiral separation ability of the BSA-SiO 2 stationary phase. An excellent enantioseparation factor was obtained up to 4.3 with acceptable stability within at least 1 month. Next, the BSA-SiO 2 stationary phase was applied to study the interaction between imatinib mesylate (IM) and BSA by frontal affinity chromatography. A single type of binding site was found for IM with the immobilized BSA, and the hydrogen-bonding and van der Waals interactions were expected to be contributing interactions based on the thermodynamic studies, and this was a spontaneous process. Compared to the traditional silica for HPLC stationary phase, the proposed penetrable silica microsphere possessed a larger capacity to bond more BSA, minimizing column overloading effects and enhancing enantioseparation ability. In addition, the lower running column back pressure and fast mass transfer were meaningful for the column stability and lifetime. It was a good substrate to immobilize biomolecules for fast chiral resolution and screening drug-protein interactions. Abstract:Molecular imprinting of bovine serum albumin (BSA) on silica surface is described using a covalently bonded template method. For polymerization, two kinds of organic silane ( 3 -aminopropyltn'methoxysilane: APS and n-octyltrimethoxysilane: OMS) were used. Aminopropylsilanized silica gel was first aldehyde functionalized with glutaraldehyde and then reacted with BSA by forming imine bonds to covalently immobilize the template protein. Polymerization occurred for siloxane-silica in the presence of the template protein. After removal of the template with oxalic acid, the BSA imprinted polymer on silica surface was obtained. Adsorption experimental results indicate that the surface imprinted polymer show high selective binding for the template BSA over the other tested proteins. One of the main advantages of the method is its binding sites located on or close to the surface and thus having good site accessibility towards the target protein.

Abstract:An epoxy functionalized organic-silica hybrid monolith was successfully synthesized via one-step method with tetramethoxysilane (TMOS) and gamma-glycidyloxypropyltrimethoxysilane (GPTMS) as precursors. The volume of reactive monomer was optimized and the capillary monolithic columns with good permeability and mechanical strength were obtained. Scanning electron microscope characterization demonstrated that a structure of continuous bed was formed. Then bovine serum albumin (BSA) was covalently immobilized on capillary monolith inner surface by Schiff base method. The preparation condition was studied and it was found that the appropriate concentration of BSA for preparation of BSA-modified hybrid monoliths was 8.0 g/L. Meanwhile, the influence factors on the sample separation were investigated, including running buffer pH (25 mmol/L Tris, pH 7.4), applied voltage (10 kV), capillary temperature (15 degrees C) and sample condition (2 kV x 3 s). The BSA-modified hybrid monolith exhibited good chiral separation ability to (+/-)-tryptophan and achieved satisfactory run-to-run, day-to-day and column-to-column repeatability. Compared with BSA-immobilized silica-based monolithic column, this work presented better chiral recognition ability and also simplified preparation procedures and shortened preparation period.

Abstract:Magnetic sandwich structured mesoporous silica microspheres containing a silica-coated Fe3O4 core and a layered mesoporous silica shell have been successfully synthesized by multi-step reactions. The as-prepared Fe3O4/SiO2/mesoporous SiO2 (Fe3O4/SiO2/mSiO(2)) microspheres were characterized via X-ray diffractometry (XRD), transmission electron microscopy (TEM), nitrogen adsorption-desorption and vibrating sample magnetometry (VSM). The results indicate that these materials possess superior porosity with high surface areas and large pore volumes, while still maintaining the microspheres morphologies, which is beneficial for enrichment and purification applications. The microspheres were applied as adsorbents for bovine serum albumin (BSA). The adsorption equilibrium was well explained in the entire experimental region by the Langmuir isotherm model and the maximum adsorption amounts of BSA were calculated as high as 289 mg g(-1). This work provides a promising approach for the design and synthesis of multifunctional microspheres, which can be used for potential applications in a variety of biomedical fields including drug delivery and biosensors.

Abstract:Viscosities of aqueous solutions of bovine serum albumin (BSA) at different temperatures were carefully measured in a common glass capillary Ubbelohde viscometer in the concentration range from dilute down to extremely dilute concentration regions. The adsorption effect occurred in viscosity measurements were theoretically analyzed and discussed. The theory based on the Langmuir isotherms could adequately describe the existing data. The influence of temperature on the adsorption of BSA solution was discussed and a simple adsorption model was proposed. Tunable Adsorption of Bovine Serum Albumin by Annealed Cationic Spherical Polyelectrolyte Brushes

Abstract:By combining turbidimetric titration, dynamic light scattering (DLS), and zeta potential methods, we demonstrated that the adsorption of bovine serum albumin (BSA) in annealed cationic spherical polyelectrolyte brushes (SPB) can be controlled by modulating the pH, ionic strength, BSA concentration of the mixed solution, and SPB thickness. The SPB consist of a polystyrene core with a diameter around 80 nm and a dense shell of poly (2-aminoethylmethacrylate hydrochloride) (PAEMH) with a thickness from 10 to 60 nm covalently attached on the core surface. Results revealed the existence of three pH regions, corresponding to (1) adsorption of BSA in SPB, (2) aggregation of SPB induced by BSA adsorption, and (3) desorption of BSA from SPB. The extent of the pH regions can be modulated by ionic strength, BSA concentration, or SPB thickness. Adsorption measurements demonstrated that the adsorbed amount of BSA in SPB was affected by pH, ionic strength, and SPB thickness. These findings lay the foundation for protein separation by SPB.

Abstract:The mode of adsorption of bovine serum albumin (BSA) on porous polyethylene (PE) membrane was studied as a function of time and concentration, which may contribute to the surface coverage. An improved physical model for adsorption is initiated based on the results of the adsorptional and desorptional measurements, FTIR analysis, and AFM observations as well as streaming potential measurements. The results obtained indicate that the adsorptional mode depend on both time and concentration. It is shown that a critical concentration (about 1000 ppm here) exists in the adsorptional process. Below this concentration, the adsorption seems to be conducted in a normal side-on way but time elapse gives rise to greater conformational change than concentration increase; above this concentration, the aggregation of protein molecular plays a decisive role and the adsorption is in an aggregation way, which is similar to end-on, but a relative large gap between the adsorbed molecules exists due to aggregation. This conclusion is general and can be expected to apply in other globular protein–hydrophobic porous surface systems.
