Abstract:We applied the PCR-RFLP method on a Chinese random population of 1 000 to detect their genotypes on the SDF1-3^A site. We obtained the distribution frequencies of SDF1-3^A in the Chinese population and observed an obvious diversity of the frequencies from the south to the north.

Abstract:OBJECTIVE:To investigate the FCGR3B gene frequencies and FCGR3 variants in a Chinese population from Zhejiang Province.METHODS:DNA was extracted from the blood specimens of 487 healthy blood donors from Zhejiang Province. The FCGR3B gene frequencies were determined by polymerase chain reaction with sequence-specific primers (PCR-SSP). The 19 specimens with 3 different FCGR3B genotypes underwent FCGR3 gene cloning and sequencing.RESULTS:The gene frequencies of FCGR3B(*1), FCGR3B(*2), and FCGR3B(*3) in these 487 individuals were 0.564, 0.429, and 0.000 respectively. The genotype frequency of FCGR3B(null) was 0.62% (3/487). Sequencing of FCGR3 revealed that in 7 out of the 19 Chinese individuals variants caused by single nucleotide exchanges at one or more of the polymorphic positions 141, 147, 227, 266 and 277 in exon 3 also existed in this Chinese population.CONCLUSION:FCGR3B(*1) gene is more frequent in a Chinese population from Zhejiang Province than the FCGR3B(*2) gene, and the FCGR3B(*3) gene seems to be absent. Gene variants caused by single nucleotide exchanges are found in addition to the well known forms, but the reason for this remains unclear.

Abstract:OBJECTIVE:To investigate the polymorphism of HLA class II alleles in the Han population in Hubei province.METHODS:The alleles of DRB1*(n=168), DQB1*(n=160) and DPB1*(n=93) were typed by using the polymerase chain reaction/sequence specific primer(PCR/SSP) and polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) techniques.RESULTS:39 HLA-DRB1, 15 DQB1 and 17 DPB1 alleles were found. Alleles with higher frequencies are DRB1*0901 (GF=14%), *1501(GF=11.3%), *0301(GF=7.1%), *0803(GF=4.8%); DQB1*0301(GF=18.8%), *0303(GF=18.4%), *0201(GF=10%), *0302(GF=8.4%); DPB1*0501(GF=31.2%), *0401(GF=15.1%), *0201(GF=14%), *0402(GF=11.8%), respectively. As compared with Caucasians, the Han people in Hubei province have higher gene frequencies of DRB1*0901, *1001, *0803; DQB1*0303, *0502 and DPB1*0501, *0202; and lower gene frequencies of DRB1*0301, *0401, *1301; DQB1*0201, *0603 and DPB1*0401, suggesting that Hubei Hans have their own layout of HLA class II allele frequencies.CONCLUSION:These data may serve as normal reference values for the Han population in south China.
[Polymorphism Analysis of D6S366 STR Locus].

Abstract:The polymorphic STR locus D6S366 was analyzed using PCR and PAG electrophoresis and 7 alleles were detected from 200 unrelated individuals in Wuhan. The allete distribution of the locus was in accordance with Hardy-Weinberg equilibrium. The H, Dp, P, E, and PIC of the D6S366 were calculated in this paper.

Abstract:CCR5-Δ32, CCR2-64I, mutants in two chemokine receptors and SDF1-3′A, mutant in a ligand gene, can delay AIDS pathogenesis. The distribution of the three polymorphic loci was studied in 1 046 DNA samples from 26 provinces (cities) in China. No CCR5-Δ 32 was observed. CCR2-64I and SDF1-3′ A had reverse distribution cline from south to north in China, with average frequency of 20.8% and 24.8% respectively. Relative hazard was evaluated. Important information to the epidemiology of AIDS and the origin and spread of these polymorphic loci in Chinese was provided.

Abstract:Abstract Blood specimens were collected from unrelated volunteer donors. DNA was extracted from blood specimens using Chelex-100 (1). DNA typing was carried out by PCR. The components of PCR were: target DNA 20 ng, primer 0.2 µmol/L, dNTPs 200 µmol/L, KCl 50 µmol/L, Tris-HCl (pH 8.3) 10 mmol/L, MgCl2 1.5 mmol/L, Taq 1U. Primer sequences: D21S1435: 5'-CCC TCT CAA TTG TTT GTC TAC C-3', 5'-ATG GCA CTG AAA TCT CTT GC-3'; D21S2055: 5'-AAC AGA ACC AAT AGG CTA TCT ATC-3', 5'-TCT CCT ACC AAG TGA TTT ACT GTA-3'.PCR conditions: D21S1435: start at 94°C for 3 min, 30 cycles consist of 35s at 94°C, 45s at 61°C, 55s at 72°C followed by a 5 min extension at 72°C. D2152055: start at 94°C for 3 min, 30 cycles consist of 35 s at 94°C, 45 s at 61°C, 55 s at 72°C followed by a 5 min extension at 72°C. The amplified products were electrophoresed in 6% polyacrylamide gel by using 100 bp ladder and allelic markers for both D21S1435 and D21S2055 as size marker, followed by sliver staining. Data were analyzed by The Promega Software, POWERSTATS. Calculating of Chi-square test was carried out for Hardy-Weinberg equilibrium test.

Abstract:The chemokine receptors and ligand CCR5, CXCR4 and SDF-1 play important roles in the entry of HIV-1 into host cells. Genetic polymorphisms such as CCR5-Δ32 and SDF-1 3′A have been reported to be associated with HIV-1 susceptibility and the progression of AIDS. Considering the remarkable difference in CCR5-Δ32 allele frequency among worldwide populations, we aimed to survey the genetic variations in CCR5, CXCR4 and SDF-1 in different Chinese populations. The open reading frames and regulatory regions of CCR5, CXCR4 and SDF-1 were sequenced in 141 Chinese individuals from three ethnic groups: Han, Mongol and Uyghur. Ninety-six variants were identified, 41 of which were newly identified (NI) in Chinese populations. A novel non-synonymous variant c.459 C>T (Trp153Cys) within CCR5 was identified in one Han individual. Of NI variants, 11 were common polymorphisms with a minor allele frequency (MAF) >5%. The polymorphism CCR5-Δ32 was found in three Uyghur individuals but was absent in Han and Mongol groups. The linkage disequilibrium (LD) analysis of CCR5 and SDF-1 and frequency of CCR5 haplotypes showed considerable divergence among three ethnic groups. Our results show the great genetic heterogeneity within CCR5, CXCR4 and SDF-1 in Chinese ethnic populations.

Abstract:Four Chinese populations have been studied by the polymerase chain reaction (PCR) for obtaining the data on genetic polymorphisms of D19S400 locus in four populations in China and for evaluating its use in forensic practice. EDTA-blood samples were collected from 356 unrelated individuals in Nanning (Zhuang), Dali (Bai), Lasa (Tibetan); Hailaer (Mongolian), DNA samples were extracted by Phenol/Chloroform method. The PCR products were analyzed by PAG vertical electrophoresis. Nine alleles and 42 genotypes were found at D19S400 locus in these four populations. The heterozygosity was between 0.75 and 0.89 and the discriminating power between 0.9274 and 0.9625. Significant difference was observed in the distribution of allele frequencies among the four Chinese populations. According to the results obtained in this study, D19S400 locus is a useful marker for individual indentification, paternity testing and for genetics study.

Abstract:Chemokine receptor CCR2 and stromal-derived factor (SDF-1) are involved in HIV infection and AIDS symptom onset. Recent cohort studies showed that point mutations in these two genes, CCR2-64I and SDF1-3'A, can delay AIDS onset > or = 16 years after seroconversions. The protective effect of CCR2-64I is dominant, whereas that of SDF1-3'A is recessive. SDF1-3'A homozygotes also showed possible protection against HIV-1 infection. In this study, we surveyed the frequency distributions of the two alleles at both loci in world populations, with emphasis on those in east Asia. The CCR2-64I frequencies do not vary significantly in the different continents, having a range of 0.1-0.2 in most populations. A decreasing cline of the CCR2-64I frequency from north to south was observed in east Asia. In contrast, the distribution of SDF1-3'A in world populations varies substantially, and the highest frequency was observed in Oceanian populations. Moreover, an increasing cline of the SDF1-3'A frequency from north to south was observed in east Asia. The relative hazard values were computed to evaluate the risk of AIDS onset on the basis of two-locus genotypes in the east Asian and world populations.

Abstract:OBJECTIVE:To describe the distribution of reduced folate carrier gene (RFC1)genotype and allele frequency between southern and northern, female and male Chinese population. METHOD:RFC1 (A80G) genotype was detected, using polymerase chain reaction-restriction fragment length polymorphism (RFLP-PCR) on 720 blood spot DNA from the normal subjects. RESULTS:The frequencies of the northern population with AA, GG and GA genotypes were 22.28%, 31.09% and 46.63%, and the frequencies of the southern population were 18.56%, 22.75% and 58.68%, respectively. Findings showed that there were significant differences between southerners and northerners in RFC1 (A80G) genotype (P < 0.01). There was no significant difference between G allele frequency of the northern (52.10%) and southern population (54.40%). The frequencies of male with RFC1 (A80G) AA, GG and GA genotype were 24.88%, 25.85% and 49.27%, and among female were 18.83%, 27.77% and 53.40%, respectively. There were no significant differences between male and female in RFC1 genotype (P > 0.05), or between G allele frequency in female (50.49%) and that in male (54.47%). CONCLUSIONS:The distribution of RFC1 genotype seemed to be consistent with neural tube defects (NTDs) while its prevalence among the northerners was higher than that of southerners, with female having a higher NTDs prevalence. This study provided genetic epidemiological data for etiological hypothesis between RFC1 and diseases relative to folate metabolism.

Abstract:OBJECTIVE:To determine whether there are any associations between the -258T/G polymorphism of the promoter and the IVS3 -20T/C polymorphism in parkin gene and Parkinson's disease (PD) from a Han population in Sichuan province.METHODS:Polymerase chain reaction (PCR), restriction fragment length polymorphism, denaturing high performance liquid chromatography(dHPLC) and sequence analysis were used to determine the genotype of each subject. The -258T/G polymorphism and IVS3 -20T/C polymorphism were analysed in 198 patients with sporadic PD and 187 healthy controls, matched for age and gender.RESULTS:There were significant differences in allele frequency of the -258T/G polymorphism between PD patients and controls, with the G allele more common in cases than controls (52.5% vs 43.3%; chi square is 6.17, P< 0.025, OR is 1.45, 95% CI 1.04-1.86). There were also significant differences in G allele frequency between PD patients with onset age over 50 years old and controls(chi square is 9.048, P< 0.01, OR is 1.57, 95% CI:1.08-2.06). The frequency of TG+GG genotype was significantly higher in PD patients than in controls (78.79% vs 69.51%; chi square is 3.854, P< 0.05, OR is 1.63, 95% CI:0.88-2.38). In addition, there were significant differences in age of onset between PD patients with different genotypes (P< 0.05). The average age of onset in group of GG genotype was later about 5 years compared with the group of TT or TG genotype. The frequency of CC genotype in IVS3 -20T/C polymorphism was much higher than that of TC genotype. No TT genotype was found.CONCLUSION:This study suggests that the parkin promoter -258T/G polymorphism might be a risk factor for late onset PD in Sichuan. CC genotype for IVS3 -20T/C polymorphism is common in Sichuan Han population. No TT genotype for IVS3 -20T/C polymorphism is found in Sichuan Han population.
Association of FVIII Val34Ldu with Coronary Heart Disease

Abstract:To investigate the distribution fo coagulation factorⅧ(FⅧ)Val34Leu polymorphism in Chinese and the relationship between the polymorphism and coronary heart disease (CHD) or myocrdial infaarcion. A total of 195 patients with angiraphically confimed CHD and 203 controls were genotyped for the Val34Leu polymorphism by polmerase chain reaction-single strand conformational polyorphism (PCR-SSCP) analysis. The FⅧ Val34Leu was found in 19 out of all 398 samples (4.8%) representing a Leu allele frequency of 2.4%. The distribution of FⅧ genotype and allele was not signifcantly different between all patients and controls (P>0.05). The Val/Leu genotype an Leu allele frequencies in subjects without MI were signifcantly higher than that in subjects with MI (P<0.05). There is FⅧVal34Leu polymporhism in Han ethnic group.

Abstract:Allele frequency data and statistical parameters for D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818 and FGA loci were determined in a sample of 120 healthy unrelated individuals of Chinese Yi ethnic minority group living in Yunnan province, China. We observed 132 alleles with allelic frequencies ranging from 0.0042 to 0.5333. The forensic statistical parameters from the data of all the loci showed high values. All loci were in accordance with Hardy-Weinberg equilibrium (p>0.05). The obtained frequency distributions were compared with previously published other population data, and significant differences were found between Yi population and Korean, Chinese Tibetan, Uigur, Ewenki, Han, Hui population at some STR loci. Our results of present study were valuable for forensic application and Chinese population genetic studies. These population data enriched Chinese genetic informational resources.

Abstract:The restriction fragment length polymorphisms distribution and frequency of dystrophin gene in Chinese were studied by using 14 subclones of the entire 14kb cDNA for the dystrophin as hybridization probes. Allelic fragments were detected in hybridization patterns of PvuII/la, Taq I /2b-3, Taq I/5b-7, arid Xba l /10. Among them, the allelic fragments (26kb and 3.8kb) in PvuII/2b-3 pattern and the allelic fragments (10.0kb and 8.4kb) in Taq I/5b-7 patterns had never been reported previously. Compared with the data from Caucasians and Japanese, it indicated that there was a significant difference (P<0.01) of the allelic fragment frequency in Taq I/2b-3 and Xba I/10 patterns between Chinese and Caucasians. The frequencies of allelic fragments A2 (5.6kb) in Taq I/8 and A2 (10.7kb) in EcoR V/9 were high in Caucasians, yet had not been detected in Chinese, the differences were also highly significant. But in Chinese and Caucasians, the B1B2 allelic frequencies in Taq I/5b-7 are the same. As to the frequency of the allelic fragments A1A2 and B1B2 in Pvu II/la, there was no significant difference between Chinese and Japanese.

Abstract:Chemokine receptors (CCR5, CXCR4 and CCR2) have been shown to be important co-receptors for HIV infection. Mutations at CCR5 (CCR5-Delta 32), CCR2 (CCR2-641), and stromal-derived factor SDF1 (SDF1-3'A), a primary ligand for CXCR4, are known to have protective effects against HIV-1 infection and the onset of AIDS symptoms. We studied the three-locus genotype frequency distributions in 70 worldwide populations from a sample of 2341 individuals without any known history of HIV-1 infection and AIDS symptoms. From these data, we estimated the risk of AIDS onset (relative hazard, RH) of each population. This survey shows that the substantial allele frequency differences of each of these mutations translate into an extensive variation in relative hazards for AIDS in worldwide populations. However, no evidence of natural selection against the mutant gene carriers is detected. Finally, the combined three-locus genotype data predict the highest relative hazard (RH) in South-East Asia and Africa where AIDS is known to be more prevalent.
Genetic analysis of STR markers on chromosome 21 in a Han population from southeast China.

Abstract:Short tandem repeats (STRs) are highly polymorphic sequences and have been extensively used as genetic markers in mapping studies, disease diagnosis, and human identity testing. In this study, 11 STR markers on chromosome 21, including D21S1432, D21S11, D21S1246, D21S1412, D21S1437, D21S1442, D21S2039, D21S1270, D21S1435, D21S1409, and D21S1446, were analyzed in 740 unrelated Han individuals from southeast China. A total of 132 alleles, ranging from 7-21 for each locus, were named according to the guidelines of the International Society for Forensic Haemogenetics. The distributions of allelic frequencies for the 11 STRs and population genetic parameters were determined. All 11 STR markers showed high polymorphism and heterogeneity in the southeast Han population, with polymorphism information content of 0.61-0.87, heterogeneity of 64.5-86.1%, and power of discrimination of 0.835-0.973. Among the 11 STR markers, D21S1412, D21S1270, D21S11, and D21S1442 showed relatively higher heterogeneity. Their combination was relatively informative and was used in a quantitative fluorescence-polymerase chain reaction assay to diagnose Down syndrome (trisomy 21) in a southeast Chinese Han population. The genetic information and population data for these 11 STRs may be used not only in quantitative fluorescence-polymerase chain reaction assays but also in forensic studies and other genetic tests.

Abstract:Objective: To investigate the polymorphism distribution of Y-chromosome haplotypes in Miao population. Methods: PCR-RFLP was used to detect the frequency of Y - chromosome haplotypes consisted of thirteen single nucleotide polymorphism alleles in Miao people. Results: Nine Y -chromosome haplotypes (H1, H2, H5, H6, H7, H8, H9, H11, H1 2) were found in the samples. The frequency distribution of these haplotypes in Miao people were very similar to that in other southern populations in China, including Han, Tujia, and Yi people. Conclusion: It indicates that there is a closer genetic relationship among southern Han, Tujia, and Yi populations implicating a common ancestry.
[Sequence analysis and genetic polymorphisms of D4S1647 and D6S2414 loci in Chinese Han, Mongolian and Tibetan populations]

Abstract:Objective: To investigate the polymorphisms of two short tandem repeats (STR) loci D4S1647 and D6S2414 in Chinese Han, Mongolian and Tibetan populations. Methods: Blood and saliva samples were collected from 308 unrelated individuals in the three populations. DNA was extracted by chelex-100 method and amplified by the polymerase chain reaction (PCR). The PCR products were analyzed by nondenaturing polyacrylamide gel electrophoresis and silver staining. Results: The genotype distributions of D4S1647 and D6S2414 loci were in accordance with Hardy-Weinberg equilibrium. The frequencies of alleles showed no significant differences among the three populations. Analyses of Han families indicated that the alleles agreed with Mendelian inheritance law. Conclusion: This study yielded information on the genetic polymorphisms of the two STR loci in Chinese Han, Mongolian and Tibetan populations.
SDF1-3′A polymorphism is associated with size but not occurrence of abdominal aortic aneurysm in a Chinese population

Abstract:Previous studies have suggested that stromal-derived factor 1 (SDF1) plays a pivotal role in abdominal aortic aneurysm (AAA) development. The SDF1-3'A polymorphism (G to A, rs1801157), located in the 3' untranslated region, could influence the expression of SDF1. The aim of our study was to investigate the relationship of SDF1-3'A polymorphism with the risk of AAA occurrence and size.A total of 205 AAA patients and 216 age- and sex-matched controls were recruited. The SDF1-3'A polymorphism was evaluated by polymerase chain reaction, followed by restriction enzyme analysis.The frequency of A allele was similar between groups. The genotype distribution also displayed no statistically significant differences under both the dominant model (odds ratio [OR], 0.922; P = .678) and the recessive model (OR, 1.617; P = .300). The multiple logistic regression analysis indicated that the association between the polymorphism and AAA remained insignificant when applied to the dominant model. However, AAA patients with GG genotype were more likely to develop aneurysms larger than 50 mm (OR, 0.497; P = .014). AAA patients with SDF1-3'A allele have reduced plasma SDF1α levels. In addition, plasma SDF1α levels were positively correlated with AAA size.Our study suggests that SDF1-3'A polymorphism is associated with size but not occurrence of AAA, providing further evidence that SDF1 is implicated in AAA progression.
