Abstract:Blood samples were obtained from 120 healthy unrelated males from the Han ethnic group in Chengdu of China. The DNA was extracted by using Chelex 100 protocol as described by Walsh et al. (1). The allelic variation at the three Y-STR loci, named as DYS447, DYS449 and DYS450, were analyzed by PCR amplification system. Each PCR reaction was performed in a 37.5 µL containing 2-10 ng DNA, 1 X Taq buffer, 1.5 mM MgCl2, 200 RM each dNTP (Pharmacia Biotech, Sweden), 1.5 U Taq polymerase (NEB, UK), 0.3 RM each primer in a Perkin-Elmer 9600 thermocycler (ABI, Foster City, CA). The PCR products were analyzed by horizontal non-denaturing polyacrylamide gel electrophoresis with discontinuous buffer system and visualized by silver staining (2). Alleles were designated according to recommendation of the International Society of Forensic Genetics (3). The gene diversities, the haplotypes diversity and the standard errors of diversity were calculated in accordance with Hou's method (4).

Abstract:OBJECTIVE:To obtain the population genetic data of 17 Y-chromosomal short tandem repeat (Y-STR) in the Han population in Chengdu of Sichuan Province.METHODS:The 17 Y-STR loci were amplified from the blood samples of 111 unrelated Chengdu Han individuals using the AmpFlSTR Yfiler system. The PCR products were genotyped with an ABI 3130 genetic analyzer.RESULTS:In the loci of in DYS456, DYS389I, DYS390, DYS389II, DYS458, DYS19, DYS385a/b, DYS393, DYS391, DYS439, DYS635, DYS392, Y-GATA-H4, DYS437, DYS438, and DYS448, 3 to 8 alleles were detected in the Han population in Chengdu, and 36 alleles were detected in the locus DYS385a/b, with the minimal gene diversity (GD) value of 0.3970 (DYS391) and maximal value of 0.9561 (DYS385a/b). The DNA samples of 16 women and 7 different species of animals were amplified, but no specific products were found for the 17 Y-STR loci. No mutations of the 17 Y-STR alleles were observed in 20 father-son pairs as confirmed by autosomal STR analysis.CONCLUSION:The 17 Y-STR loci are highly polymorphic and are suitable for personal identification, paternity testing, population genetics and anthropology studies.

Abstract:In this study, 20 Y-specific short tandem repeat (STR) loci (DYS434, Y-GATA-A10, Y-GATA-H4, DYS438, DYS439, DYS443, DYS444, DYS446, DYS447, DYS448, DYS456, DYS458, DYS460, DYS520, DYS531, DYS557, DYS622, DYS630, DYS635(Y-GATA-C4), and DYS709) were analyzed in 158 unrelated healthy men from southeast China by three fluorescence-labeled multiplex polymerase chain reaction systems. The Y-STR multiplexes developed have followed the published nomenclature and International Society for Forensic Genetics (ISFG) guidelines for STR analysis. Gene diversity ranged from 0.2506 at DYS434 to 0.8034 at DYS447. A total of 157 different haplotypes were observed, and among these, 156 were unique, while 1 was found two times. The haplotype diversity value calculated from all 20 loci combined was 0.9997, which is informative. Furthermore, 80 father–son pairs, previously confirmed by autosomal STR analysis, were typed using the same 20 Y-STR loci, and four mutation events were identified at the Y-GATA-H4, DYS439, DYS456, and DYS458 loci, giving an average mutation rate of 0.25% per locus per generation (95% confidence interval 0.09–0.54). These results including the haplotype data at 20 Y-STR loci would enrich Chinese genetic informational resources and provide useful information in forensic practice.

Abstract:The PCR products were analyzed by the polyacrylamide gel electrophresis and DNA sequencing. 102 unrelated, healthy, male individuals of Henan province's Han population were investigated, and the allelic frequency distribution and haplotype diversity of 4 Y chromosomal STRs were explored. Results showed that twenty-four alleles were detected, whose distribution frequencies ranged from 0. 0098～0.7745. Out of the total of 102 individuals, 55 haplotypes were found in 4 Y-STR. The haplotype diversity for 4 Y-STR loci was 0.8939.

Abstract:To investigate haplotype distribution of the Y-chromosome STRs (DYS19, DYS390 and DYS389 locus) in Han population in Guangdong area. The STRs were typed by poplymerase chain reaction followed by discontinuous PAGE system. Among 130 unrelated males, 81 different haplotypes were observed, 52 out of them were found only one time. The haplotype genetic diversity, discrimination power and non-father exclusion chance are 0. 9989, 0. 9824, 0. 9824, respectively. The high informative haplotypes make these STRs useful for the forensic individual identification and paternity testing.

Abstract:The distribution of 17 Y-chromosome STR loci DYS456, DYS389I, DYS390, DYS389II, DYS458, DYS19, DYS385a/b, DYS393, DYS391, DYS439, DYS635, DYS392, Y-GATA-H4, DYS437, DYS438, and DYS448 haplotypes was determined in a population sample of 222 unrelated Chinese Han from Shanxi Province, Northern China. A total of 219 haplotypes were observed, and of these, 216 were unique, while 3 were found two times. The overall haplotype diversity was 0.9999 and the discrimination capacity was 0.9865, indicating a high potential for differentiating between male individuals in this population. Comparison analysis via Analysis of Molecular Variance (AMOVA) and construction of MDS plot revealed that Shanxi Han sample clusters with Chinese origin populations and stands far apart of the non-Chinese populations, justifying the establishment of local databases in Shanxi Han population for any future forensic and genetic epidemiology efforts in this region.

Abstract:We analyzed haplotypes for 24 Y chromosomal STRs (Y-STRs), including 17 Yfiler loci (DYS19, DYS385a/b, DYS389I/II, DYS390, DYS391, DYS392, DYS393, DYS437, DY438, DYS439, DYS448, DYS456, DYS458, DYS635 and Y-GATA-H4) and 7 additional STRs (DYS388, DYS444, DYS447, DYS449, DYS522 and DYS527a/b) in 1100 unrelated Chinese Han individuals from Henan Province using AGCU Y24 STR kit systems. The calculated average gene diversity (GD) values ranged from 0.4105 to 0.9647 for the DYS388 and DYS385a/b loci, respectively. The discriminatory capacity (DC) was 72.91% with 802 observed haplotypes using 17 Yfiler loci, by the addition of 7 Y-STRs to the Yfiler system, the DC was increased to 79.09% while showing 870 observed haplotypes. Among the additional 7 Y-STRs, DYS449, DYS527a/b, DYS444 and DYS522 were major contributors to enhancing discrimination. In the analysis of molecular variance, the Henan Han population clustered with Han origin populations and showed significant differences from other Non-Han populations. In the present study, we report 24 Y-STR population data in Henan Han population, and we emphasize the need for adding additional markers to the commonly used 17 Yfiler loci to achieve more improved discriminatory capacity in a population with low genetic diversity.

Abstract:We investigated the distributions of haplotypes for four Y-STR loci, named as DYS434, DYS438, DYS439, and A10, and developed a multiplex amplified for these Y-STR markers. A total of 100 samples were analyzed. Each sample was amplified utilizing multiplex PCR for four Y-STR loci. The PCR products were analyzed by non-denaturing polyacrylamide gel electrophoresis and visualized by silver staining. A total of 37 different haplotypes were found, and 16 haplotypes were being unique. Gene diversity ranged from 0.3945 at DYS434 to 0.7051 at DYS439. The haplotype diversity, chance of exclusion and discrimination power of four Y-STR loci was 0.9628, SE 0.00429. The results indicated that they were suitable Y linked markers for forensic applications in our population studied.

Abstract:OBJECTIVE:To investigate the distribution characteristics of Y chromosome haplogroups in Sichuan Han population.METHODS:Using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), denatured high performance liquid chromatography (DHPLC) and DNA sequencing methods, 18 Y-chromosome bialletic markers were detected to type Y chromosome haplogroups in 341 unselected men from Sichuan Han population.RESULTS:A total of 14 haplogroups were observed, in which haplogroups H2 and H4 were identified for the first time in Sichuan population, and haplogroups H14 and N* were found firstly in Chinese. There was a significant frequency difference of Y haplogroups between Sichuan Han population and Southern Han population.CONCLUSION:More comprehensive frequency distribution data of Y chromosome haplogroups are obtained in Sichuan Han population, which would be helpful for understanding the association of Y chromosome background and the susceptibility to male specific diseases such as spermatogenic failure, prostate cancer, testical cancer and so on in present population.

Abstract:We have co-amplified and analyzed 17 Y-chromosomal STRs loci including European ‘extended haplotypes’ (DYS19, DYS389 I, DYS389 II, DYS390, DYS391, DYS392, DYS393, DYS385a,b, DYS438, and DYS439) and also six additional loci DYS437, DYS448, DYS456, DYS458, DYS635 and YGATA H4 in a population sample of 167 healthy unrelated autochthonous male individuals of Chinese Tibetan ethnic minority group residing in Qinghai province of China using a multiplex PCR system. The gene diversity values for the Y-STRs loci ranged from a minimum 0.3581 for DYS391 locus to a maximum of 0.8702 for DYS385a,b loci in Tibetan population. A total of 163 haplotypes were identified in the set of Y-STR loci, of which 159 were unique and 4 found in two individuals. The overall haplotype diversity for the set of Y-STRs loci was 0.9998, and the discrimination capacity was 0.9938. Research results will be valuable for human identification and paternity tests in the region and for Chinese population genetic study in the future.

Abstract:Haplotypes and allele frequencies of 17 Y-short tandem repeats (STRs) included in the PowerPlex1 Y systems kit (DYS19, DYS389 I, DYS389 II, DYS390, DYS391, DYS392, DYS393, DYS385, DYS438, DYS439, DYS437, DYS448, DYS456, DYS458, DYS635 and Y GATA H4) were examined in a population sample of 276 unrelated Han male living in Henan province. A total of 272 haplotypes were observed in the 276 individuals studied, of which 268 were unique and four were found in two individuals. The gene diversity values ranged from 0.5019 (DYS391) to 0.9579 (DYS385a, b). The overall haplotype diversity for the 17 Y-STR loci was 0.9998.

Abstract:In this study, we investigated the genetic polymorphisms of 24 Y-chromosomal short tandem repeat (Y-STR) loci in 885 unrelated Chinese Han male individuals from Guangdong Province, using a domestic AGCU Y24 STR kit. A total of 878 different haplotypes were observed at the 24 Y-STR loci; among them, 871 haplotypes were unique and 7 haplotypes occurred twice. The overall haplotype diversity was 0.99998 and the discrimination capacity was 99.2%. The gene diversity values ranged from 0.4354 at DYS438 to 0.9606 at DYS385a/b. Population relationships between the Guangdong Han population and seven other published Chinese populations were evaluated by Rst values and visualized in a two multi-dimensional scaling plot. The results showed the 24 Y-STR loci are highly polymorphic in Guangdong Han population and of great value in forensic application.

Abstract:The allelic distribution of 15 short tandem repeats (STRs) loci (D8S1179, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D2S1338, D19S433, vWA, TPOX, D18S51, D5S818 and FGA) were tested in 497 unrelated Yunnan Han individuals by the AmpFl STR® Identifiler™ kit. No deviation from Hardy–Weinberg equilibrium was observed except D7S820 locus treated by Bonferroni correction. Genetic parameters of forensic interests were calculated and the genetic differences between Yunnan Han and other eight populations were analyzed.

Abstract:OBJECTIVE:To acquire the population genetic data of fifteen short tandem repeat (STR) loci in Chengdu Han population.METHODS:A total of 210 EDTA-blood specimens were collected from the unrelated individuals in Chengdu Han population. The DNA samples were extracted with Chelex method and amplified by multiplex PCR technique. The PCR products were analyzed by an automatic genetic analyzer; the relative fragment's lengths of PCR products were calculated by gene scan analysis software and afterward genotyped by genotype software.RESULTS:Fifteen STR loci of the 210 samples showed a successful result of genotyping. The heterozygosities of the fifteen STR loci in Chengdu Han population were found to be 0.529-0.881; the combined exclusion probability and discrimination power for the fifteen STR loci in Chengdu Han population were determined to be 0.999998 and 7.3 x 10 (-17); respectively.CONCLUSION:The distinct genotype of fifteen STR loci and the sex of sample could be unveiled just through PCR and electrophoresis once, and a higher measured value could be obtained for both the combined discrimination power and the exclusion probability; the fifteen STR loci can meet the needs of the parentage testing and personal identification in forensic medicine.

Abstract:OBJECTIVE:To obtain population genetic data of loci D11S4951, D11S4957, GATA193H05, D2S2951, and D6S2421 in Han population in Chengdu area and to validate the value of their forensic application.METHODS:Blood samples were collected in EDTA tubes from unrelated individuals. DNAs were extracted with Chelex-100 and were analyzed by PCR and horizontal PAGE followed by silver staining.RESULTS:Alleles 7, 10, 8, 6 and 8 were found in 5 STR loci, respectively. No deviations from Hardy-Weinberg balance were observed. The heterozygosities observed were 0.743, 0.772, 0.833, 0.650 and 0.800, respectively. The chances of exclusion were 0.497, 0.549, 0.662, 0.356 and 0.599, and the discrimination powers were 0.863, 0.912, 0.947, 0.829 and 0.931.CONCLUSION:All of the five loci studied may be useful markers for individual identification and paternity testing.

Abstract:Haplotypes and allele frequencies for 12 Y-STR loci were determined in Sichuan Han ethnic population samples of 237 unrelated males living in west China. Only one haplotype was encountered in triple. Twelve of the haplotypes were encountered in duplicate, while 210 haplotypes were unique. The gene diversity was 0.9952. This study provides an essential precondition for identification of male DNA and tracing of paternal lineages.
