Abstract:The pollen-mediated transformation approach could not only circumvent the tedious tissue culture procedures like in Agrobacterium infection, but also circumvent the incertitude of the pollen tube path-way. So,its transformants combined from the transformed pollen and eggs would not be chimera.At last this approach is simple, easy to operate, and could be widely used in practice. For the transformation happened in the pollen culture medium, the solution with a suitable solute has to be selected. This study showed the medium consisting of (0.1%) H_3BO_3 、(0.3%) Ca(NO_3)_2、(0.2%) MgSO_4·7H_2O、(0.1%) KNO_3 and 45% sucrose could maintain normal turgidity of the pollens and facilitate them to be germinated. There are more than 95% pollens don't dispose in this medium and the germinating rate was up to 58.5%. A transient GUS expression assay and the exploded pollen rate proved that the transformation method with 1‰ Tween-20 in the medium using vacuum-infiltration was a better method. Finally, four transformed seedlings were confirmed by Kanamycin selection, GUS expression and Northern blot. Therefore this transformation approach is available.

Abstract:Anther culture is an effective breeding method for improving desirable traits, since it allows fast achievement of genetically homozygous lines. The present studies were carried out to evaluate the response of flue-cured tobacco anthers for high frequency pollen embryo induction in different culture media, and the effects of colchicine treatment on tobacco chromosome doubling. The results showed that different media caused significant changes in pollen embryo induction, in which the most obvious change was found in B medium with induction rates of 23.28%, 26.64%, and 28.07% for TMK-12 x Jiyan 5, K326 x Shangkang 70, and NC71 x Jingyehuang, respectively. In addition, shoot tips of the haploid aseptic seedlings cultivated from anthers were treated with 4 g/L colchicine for 48 h, and the ploidy levels were then checked by using flow cytometry and the number of chloroplasts in stomatal guard cells. The results suggested that colchicine treatment not only doubled the chromosomes of sterile haploid seedlings, but also caused some sterile haploid seedlings to die. The survival rate of TMK-12 x Jiyan 5, K326 x Shuangkang 70, and NC71 x Jingyehuang seedlings were 33.29%, 35.11%, and 17.61%, respectively. Similarly, TMK-12 x Jiyan 5 and K326 x Shuangkang 70 also showed a relatively high chromosome doubling rate compared with NC71 x Jingyehuang; their chromosome doubling rates were 21.53%, 18.54%, and 7.98%, respectively. In general, our work provides a theoretical basis for accelerating the breeding and improvement of new flue-cured tobacco varieties.
Obtaining Mutant Pollen for Phenotypic Analysis and Pollen Tube Dual Staining.

Abstract:Mutant phenotype observation is the most useful and important method to study which biological process a gene-of-interest is involved in. In flowering plants, excessive pollen grains land and germinate on the stigma, then pollen tubes grow through the transmitting tract to reach the ovules, eventually enter the micropyle to complete double fertilization. First, for mutants whose homozygotes could not be obtained due to pollen tube defects, it is difficult to observe the defect phenotype since the pollen grains of different genotypes are mixed together. Here, we provide a detailed protocol to pick out mutant pollen grains from the heterozygous mutant plants in Arabidopsis thaliana. By using this method, we could obtain sufficient mutant pollen grains for phenotypic analysis. Second, it is difficult to compare the pollen/pollen tube behavior of two different genotypes/species in vivo in a same pistil. Here, we develop a new dual staining method which combines GUS staining with aniline blue staining. By using this method, we can analyze the competence of the two different pollen tubes in the same pistil.

Abstract:Double fertilization is a unique characteristic trait of flowering plants.Sperm cells have lost mobility and are transported from the stigma to the female gametophyte via the pollen tube to achieve double fertilization.Pollen tube growth and guidance is largely governed by the maternal sporophytic tissues of the stigma,style and ovule.However,the last phase of the pollen tube path is under female gametophytic control.Many genes involved in different stages of pollen tube guidance have been isolated.In this paper,mechanisms of cellular development,genes and chemical compounds involved in the process of sporophytic control and gametophytic control of pollen tube growth and guidance were reviewed.Differences of pollen tube growth and guidance between Poaceae and Brassicaceae plants were also compared.

Abstract:The effects of different conditions such as light intensity, humidity, weather and stigma life on the seed setting rate of tea trees were studied. The response surface model is used to study the influence relationship between different factors. The results show that the weather is sunny, the temperature and humidity are suitable, and it is suitable for morning hybridization. Pollination once can eliminate pollination; sunny hybridization is best; When shedding, the flowers on the robust lines should be selected for hybridization, and the phosphorus and potassium fertilizers should be appropriately increased.
Optimization of Airflow Velocity of Pneumatic Pollination for Hybrid Rice Breeding

Abstract:Pollination is a key link in hybrid rice seed production. The pollination efficiency can be significantly improved through the method of pneumatic pollination under large-scale planting mode. In order to explore the effect of air velocity on pollen distribution in the process of pollination of hybrid rice and its better parametersï¼The self-made pneumatic pollination test platform was used to carry out field experiments and collect pollen. Based on the pollen distribution of single pollination, the objective functions of pollen quantity, variation rate and air velocity of each paternal plant row were established by response surface method. The multi-objective optimization of airflow velocity is carried out by genetic algorithm. The results showed that when the airflow acted on the paternal plant in single row, the propagation distance of pollen was short and the pollen accumulated near at low air flow velocity. The moderate increase of airflow velocity can promote the growth of pollen and reduce the variation rate. Due to the blocking effect of lateral paternal plants, the transmission of medial pollen is seriously limited. The pollen distribution under different airflow velocity combinations was obtained. When the airflow velocity range of the paternal plants in the first, second and third rows near maternal plants was 22.4-24m/s, 23.1-27m/s and 23.5-24.1m/s, respectively, pollination was sufficient and uniform. This study can provide a reference for the theory of pneumatic pollination and the optimization of working parameters under large-scale planting mode.

Abstract:Pollination is the key process in the hybrid rice seed production. To realizing mechanical pollination of hybrid rice, this paper designed and trial-manufactured a hybrid rice pollination machine combined collision and air-blowing method. Flow field distribution of two types of the pollination tube on the machine, including air-blow nozzle type and air-blow hole type were simulated using CFD technology. And the flow velocity out of each nozzle or hole on pollination tube was measured, and the pollination experiments in the paddy field using pollination machine prototype were carried out. In the experiment, the pollen distribution produced by mechanical pollination and manual pollination was measured adopting gravity slide method, and the flow field model of the pollen distribution was established using Gambit software and Fluent software. The k-ε standard turbulence model was adopted during the simulation of solving the control equations. The inlet boundary condition was set as velocity inlet, and the outlet boundary condition was set as pressure outlet, and the others were set as tube wall. The results of numerical simulation showed that under the same condition of the inlet velocity, the airflow outside of the pollination tube with air-blow hole type is slant, and there are some dead zones where air could not reach outside of the pollination tube. The airflow direction outside the pollination tube with air-blow nozzle type is almost the same, which can provide uniform and stable airflow field for pollination. So the pollination tube with air-blow nozzle type is the ideal structure for hybrid rice pollination machine. In order to verify the simulation results, a trial-produced pollination machine and two types of pollination tubes with air nozzles were built for experiments. The velocities out of the blower on the machine were measured by an anemograph under different engine gears. These velocities were also the inlet velocities of pollination tube and they were 5.40 m/s, 10.00 m/s and 13.50 m/s respectively. Then the velocity out of each air nozzle on pollination tube was measured by the same anemograph under the above three inlet velocities. The results showed that the variation trends of air velocity both by measured and simulated are the same. The average errors between simulation value and measured value are 2.14%, 2.40% and 3.43% respectively, and the uniformity variation coefficient of air flow distributing along the pollination tube is 14.76%, 15.93%, 16.31% respectively in simulation. The pollination experiments in paddy field were conducted by the trial-manufactured pollination machine when the outlet velocity was 7.30 m/s. The pollen was collected by glass slide coated with Vaseline and the acquisition points were set on the four lines of female parent against the male parent. The same experiment method was used to collect pollen during manual pollinating. The results of field pollination experiments showed that the pollen amount on each line and pollen distribution uniformity among lines in mechanization pollination are better than that of manual pollination when the outlet velocity of air-blow nozzle is 7.30 m/s. The pollination efficiency of the mechanical pollination is higher than that of manual pollination, which provides practical machine model for mechanization pollination of hybrid rice.
Development and Innovation of Haploid Induction Technologies in Plants.

Abstract:Haploid induction is one of the main techniques for breeding new varieties of major crops, and its key steps are improving the haploid induction rate and simplifying the induction procedure. With the development and innovation of plant haploid induction technologies, haploid breeding has been widely used in varietal improvement of important crops, showing the advantages of rapid homozygosity of heterozygous genes, shortening breeding period, and improving breeding efficiency. The combination of haploid breeding with crossing breeding, mutation breeding, reverse breeding, and molecular marker-assisted selection will greatly improve the effectiveness of crop breeding. Haploids and doubled haploids have demonstrated their usefulness in production of genetic populations, characterization of gene functions, and transgenic and cytological studies in plants. In this review, we summarize the progress of haploid induction technologies in view of various haploid induction techniques and applications of haploids and double haploids. In particular, the advances on the haploid induction in several major crops by genome editing were briefly described. Finally, we discuss current issues and future perspectives in this field, so as to promote the application of the haploid induction techniques, especially the techniques of creating haploid inducer lines by genome editing in crop breeding.

Semi-In Vivo Assay for Pollen Tube Attraction

Abstract:In flowering plants, each pollen tube delivers two sperm cells into the ovule to complete double fertilization. During the process, pollen tubes need to be navigated into the ovule, where accurate and complex pre-ovule guidance and ovule guidance are required. In recent years, different methods have been established to study those genes involved in the regulation of pollen tube guidance. Semi-in vivo ovule targeting mimics in vivo pollen tube micropylar guidance, and the semi-in vivo ovule targeting assay has been used to investigate function of genes involved in micropylar guidance. Moreover, the ovule targeting assay is the best way to do live cell imaging, which facilitates observation of pollen tube reception, synergid cell degeneration, and semi-in vivo gamete fusion. Meanwhile, semi-in vivo pollen tube attraction assay is another useful method to directly determine whether a certain molecule has pollen tube attraction activity.

Abstract:HANCOCK, WESLEY GRAHAM. Interspecific Hybridization in Nicotiana: Various Roles in a Modern Applied Tobacco Breeding Program. (Under the direction of Dr. Ramsey Lewis). Tobacco (Nicotiana tabacum L.) is one of the most economically important crops grown in North Carolina and worldwide. In a modern tobacco breeding program, continued efforts are undertaken to increase yields while maintaining or improving leaf quality and disease resistance. Two challenges are low levels of genetic diversity within flue-cured germplasm pools and the need for improved methods of doubled haploid breeding. The role of interspecific hybridization in dealing with these challenges was investigated in this research. Although favorable genetic variation affecting important quantitative traits likely exists within diploid Nicotiana relatives, their use in breeding programs presents a number of challenges. Work was conducted to evaluate the feasibility of using synthetic tobaccos as an alternative source of favorable alleles influencing key traits. The first objective of this research was to evaluate yield and growth rate and the relative amounts of heterosis for these traits in tobacco hybrids involving a set of diverse materials including two synthetic tobaccos. Cultivated tobacco  synthetic tobacco F2 populations and selected F2:3 families were also evaluated for a series of quantitative traits to determine realized heritability for growth rate on a single plant basis. Results showed significant mid-parent heterosis for hybrids involving the synthetic tobaccos for most traits. A range of phenotypes were observed in F2 populations and the realized heritability for growth rate was found to be very low (h = 0.14). Genome-wide transmission genetics, recombination frequencies, and preliminary marker-trait associations for measured traits in a N. tabacum × 4x(N. sylvestris  N. otophora) F2 population were examined and the relative importance of dominance, overdominance, and epistasis affecting heterosis for these traits was also investigated. We observed an overall reduction in pairwise marker distances and overall map length relative to those distances previously determined for populations derived from N. tabacum x N. tabacum crosses. Several QTL affecting growth rate, yield, leaf number and plant height were identified. Marker loci exhibited a range of d/|a| ratios, with the majority suggesting the importance of overdominance as a contributor to heterosis for yield, growth rate, and plant height, while epistasis and overdominance were important contributors to days to flower and leaf number. Synthetic tobaccos may be a useful source of favorable alleles affecting yieldrelated traits in a mediumto long-term breeding program. Conventional methods of inbreeding require a significant amount of time. An alternative inbreeding method used by tobacco breeders is doubled haploid breeding. In this method, haploid plants are identified, characterized, and subsequently chromosome-doubled to produce inbred lines in a greatly reduced amount of time. One method to produce N. tabacum haploids is using interspecific hybridization between N. tabacum and N. africana. This is a semi-lethal cross, where greater than 99% of the progeny die at the cotyledonary stage due to an interspecific lethality reaction. The remaining surviving plants are mixtures of maternal haploids of N. tabacum and aneuploid interspecific F1 hybrids. An efficient method is needed to distinguish maternal N. tabacum haploids from aneuploid hybrids during the seedling stage, however. The objective of this research was to investigate the use of N. africana engineered with a green fluorescent protein (gfp) transgene to assist in the discrimination between aneuploid hybrids and haploid plants resulting from this interspecific cross. Results demonstrate gfp expression to be a useful visual phenotypic marker for identification of maternal haploids at the seedling stage. DNA marker genotyping, flow cytometry, and chromosome counts of surviving non-haploid plants suggest that one or more genes on N. tabacum chromosome H play a role in the interspecific lethality reaction. Microsatellite marker genotyping and chromosome counts of three plants with low DNA contents indicated that these plants were likely the result of incomplete chromosome elimination, leading to the suggestion that chromosome elimination may play at least a partial role in haploid production in the N. tabacum  N. africana interspecific cross.

Abstract:Experiment was conducted with Shuguang(Prunus persical.Batsch)for studying the applying of Pollen-tube path way in peach.Results showed:(1)24 h after pollination in vivo,pollens germinated on the stigma;After 48 h,96 h,144 h plenty of pollen-tubes respectively reached to the upper,middle,bottom of style.(2)The best time to inoculate was 144 h after pollination;The injection method of adding the crosscut on the stigma was not suitable for peach;The rate of fruit setting was better than other concentration when was 0.2 mg/l.
Optimizing Pollen Germination and Subcellular Dynamics in Pollen Tube of Torreya grandis

Abstract:Torreya grandis, a dioecious Taxaceae species of significant economic value in southeast China, presents challenges for natural pollination due to asynchronous maturation of its sex organs and low pollen vitality. In order to enhance fertilization success through artificial pollination of T. grandis, this study investigated the optimal conditions for in vitro pollen germination and pollen tube growth of T. grandis. The optimal in vitro growth medium was found to contain 29mM sucrose, 0.8mM H3BO3, 0.72mM CaCl2, and 0.32mM MgSO4, supplemented with 4μM NAA, 2μM GA3, and 5μM 2,4-D at pH=5.6. Under these conditions, we achieved a maximum pollen germination ratio of 69.99 ± 5.17% and a pollen tube length of 34.38 ± 6.04µm after 6 days germination at 28°C. FM4-64 dye and Mitotracker Red staining revealed highly dynamics of vesicles and mitochondria during germination, which were accumulated at the tip of pollen tube and exhibited biphasic movement patterns. The total number, motion rate, and movement velocity of vesicles as well as mitochondria showed an initially increase followed by a gradual decrease pattern. The presence of sucrose in the medium significantly increased the dynamics and metabolic activity of both vesicles and mitochondria, which may relate with higher pollen germination ratio and faster pollen tube growth compared to sucrose-depleted conditions. Thus, these findings shed light on the physiological characteristics of Torreya pollen germination and provide scientific information for improving Torreya fruit yield through artificial pollination.
The extent of hybridization between Helicoverpa armigera (Hübner) and H. assulta (Guenée) and a molecular method for identifying these species and their hybrids

Abstract:Objectives] To establish a molecular technique that can reliably distinguish the cotton bollworm, the tobacco budworm, and hybrids between these species, to determine the effect of different kinds of light on hybridization, and the extent of hybridization in a mixed population under greenhouse conditions. [Methods] Hybridization rates under all potential kinds of pairings between 3 day-old virgin cotton bollworm and tobacco budworm adults were recorded under a 0.5 lx black lamp, an incandescent lamp, and complete darkness (control). Microsatellite loci that could reliably distinguish cotton bollworms, tobacco budworms, and their hybrids, were identified and used to identify the resultant progeny. A mixed population of cotton bollworm and tobacco budworm adults were released in a greenhouse and their offspring screened using the previously identified microsatellite markers. [Results] Hybridization could result from all kinds of forced pairings. However, in mixed populations, the proportion of hybrids was just 2.92%, and only one kind of pairing (male cotton bollworm × female tobacco budworm) was observed. There was no significant difference in hybridization rates between the black lamp, incandescent lamp, and dark, treatments. The sizes of the microsatellite loci HarSSR1、HarSSR9, and HarSSR10, differ between the two species. No hybrids were identified among 360 offspring screened during two years research on a mixed, greenhouse population. [Conclusion] Hybridization between the cotton bollworm and tobacco budworm can occur when individuals are not free to choose mates but low intensity light does not promote hybridization. Only one kind of pairing was observed in mixed populations, suggesting that male cotton bollworms are competitively superior to male tobacco budworms in competition for mates. The cotton bollworm, tobacco budworm, and hybrids between these species, could be reliably distinguished with microsatellite markers, which provide a simple and accurate molecular identification method. Hybrids were not identified during two years study of a mixed, greenhouse population, indicating that there is premating reproductive isolation between the two species.
Cytological abnormalities during pollen development in interspecific hybrids of Nicotiana

Abstract:Nicotiana alata is resistant to Tomato spotted wilt virus (TSWV) and of great value in breeding. However, hybrid sterility constrains the application of interspecific genetic resources. Previously, we obtained interspecific hybrids between a cytoplasmic male sterility (CMS) line of Nicotiana tabacum and N. alata , some of which were pollen sterile. In the present research, we studied the cytological abnormalities during pollen development in sterile hybrids (F 1 -D) by comparing pollen development with that in fertile hybrids (F 1 -S) from the same cross. Transmission electron microscopy and DiI staining showed that the membrane structures of microspores and pollen in F 1 -D sterile hybrids were impaired. Carbol fuchsin staining revealed that cytomixis, chromosome loss and asymmetric callose wall formation occurred with high frequency in the microsporocytes and microspores of the sterile hybrids. The cytoplasm and nucleus were lost in the microspores and pollen of sterile hybrids, leading to mature pollen grains that were vacuous and collapsed in the aperture region. In addition, delayed tapetum degradation was detected in the anther of sterile hybrids, and sporopollenin was deposited in the aperture region. Impaired membrane structures of microspores and pollen in F 1 -D sterile hybrids affected the integrity of the cells, and might be associated with chromosome, nuclear and cytoplasm loss, vacuous pollen, and sterility in F 1 -D hybrids. Abnormal tapetum degradation in the anther and irregular sporopollenin deposition in the pollen wall of the F 1 -D sterile hybrids might also be related to the pollen sterility. This study deepens our understanding of the cytological mechanisms of hybrid sterility, and may facilitate the application of TSWV-resistant resources in cultivated Nicotiana species through hybrid fertility restoration and backcross breeding.

Tobacco lines resistant to TMV and PVY obtained by maternal-derived haploid breeding methods

Abstract:Doubled haploid(DH) technique can accelerate traits stabilization in hybridization offspring. In the breeding of variety resistant to Tobacco mosaic virus(TMV) and Potato virus Y(PVY), haploids were obtained from F1 hybrids between Coker176 and NC55 crossing as female to Nicotiana african. TMV and PVY resistant haploids were selected by seedling resistance identification and molecular marker test. DH plants were regenerated from leaf mid-vein culture method. Artificial inoculated resistance evaluation, markers detection, and field comparison were conducted. The third generation seed of DH lines which showed stable resistance and botanical characters were harvested. Compared to conventional hybrid breeding methods, DH breeding method showed greater variation between offspring lines and objective traits were relatively more stable.

Abstract:TAIL-PCR was used to obtain the flanking sequence of T-DNA insertion in transgenic cotton with Bt+Sck developed by pollen-tube pathway-mediated transformation.The results showed that the flanking sequences contained a section of vector sequence and were rich in 'A+T' base pairs bound to nuclear matrices.Furthermore,the left or right border of T-DNA was absent during the T-DNA insertion process.Although the mechanism of pollen-tube pathway-mediated transformation method is very complex,this study will provide practical help for study on the mechanism of this transformation method.
