Abstract:As in any other economically important crop, the possibility of producing fully homozygous, doubled haploid lines in cucumber allows for faster and cheaper breeding. At present, the fastest way to doubled haploidy is the production of cucumber haploid plants and duplication of their chromosomes to make them doubled haploid. In this chapter, we describe a complete protocol to successfully produce cucumber doubled haploid plants, including the evaluation of their ploidy level by flow cytometry. Briefly, this protocol involves a first step of anther culture to induce microspores to divide and proliferate forming calli. The calli produced are isolated from anthers and transferred first to a liquid medium and then to a solid medium to induce organogenesis. Organogenic shoots will eventually give rise to entire DH plants.

Abstract:Heterosis is one of the important approaches to increase tobacco yield and improve tobacco quality,and combining ability breeding is one of the main breeding methods for utilization of heterosis.To quickly and accurately predict the heterosis of F1 and to promote selecting good hybrid combinations with strong advantages,using NCⅡ genetic design method to analysis of combining ability of 24 hybrid combinations,which were crossed by four self-breeding tobacco double haploid lines(DH-line) GDH84,GDH88,GDH64 and GDH02 and six foreign introduced tobacco varieties K326,Va116,NC82,G-28,K730 and Coker147.The combining ability effects of some main characters of the 10 parents and 24 F1 were discussed to select the ideal combinations of parents and strong advantage and to provide references for correct selecting parents for hybrid breeding of tobacco.

Tobacco lines resistant to TMV and PVY obtained by maternal-derived haploid breeding methods

Abstract:Doubled haploid(DH) technique can accelerate traits stabilization in hybridization offspring. In the breeding of variety resistant to Tobacco mosaic virus(TMV) and Potato virus Y(PVY), haploids were obtained from F1 hybrids between Coker176 and NC55 crossing as female to Nicotiana african. TMV and PVY resistant haploids were selected by seedling resistance identification and molecular marker test. DH plants were regenerated from leaf mid-vein culture method. Artificial inoculated resistance evaluation, markers detection, and field comparison were conducted. The third generation seed of DH lines which showed stable resistance and botanical characters were harvested. Compared to conventional hybrid breeding methods, DH breeding method showed greater variation between offspring lines and objective traits were relatively more stable.

OPTIMIZATION OF DOUBLED HAPLOID PRODUCTION IN BURLEY TOBACCO ( Nicotiana tabacum L.)

Abstract:OF A DISSERTATION OPTIMIZATION OF DOUBLED HAPLOID PRODUCTION IN BURLEY TOBACCO (Nicotiana tabacum L.) Doubled haploidy (DH) is a plant breeding technique that is often utilized by plant breeders to minimize the time required to reach homozygosity in breeding lines. The first objective of this study was to compare two methods of generating DH lines in tobacco (Nicotiana tabacum L.). Inbred burley tobacco varieties TN 90LC and GR 149LC were used to produce both androgenic derived doubled haploids (ADDH) and maternally derived doubled haploids (MDDH). The relative agronomic performance of TN 90LC and GR 149 LC ADDH and MDDH lines was compared when used either as pure-line cultivars or when used for the production of the KT 204LC and TN 97LC hybrid cultivars, respectively. The ADDH method was more efficient than the MDDH method in generating large numbers of haploid plants. On average the ADDH TN 90LC population was statistically inferior to the inbred TN 90LC for several agronomic traits; this inferiority of the ADDH method was not observed in the GR 149LC populations. For both genotypes, the MDDH populations were comparable to the inbred parental genotypes. The ADDH method was inferior for TN 90LC, but several individual TN 90LC ADDH lines were equal or superior to the inbred source. The agronomic variability observed in both ADDH and MDDH lines was decreased when they were used to produce hybrid cultivars. Less variation was observed in the DHderived hybrids KT 204LC and TN 97LC compared to the ADDH and MDDH TN90LC and GR149LC parental lines, respectively. The significant inferiority of ADDH TN 90 lines in comparison to inbred TN 90LC was not observed in the ADDH derived KT 204 population compared to KT 204LC. The second objective of this study was to compare DH Lines derived from an F1 breeding population versus DH lines derived from a segregating F2 population where plants used for DH were pre-screened for quantitatively inherited resistance to soil-borne diseases black shank (Phytophthora nicotianae) and/or Fusarium wilt (Fusarium oxysporum f. nicotianae). There was a clear difference in susceptibility to black shank between the F1 and F2 derived DH populations, both in terms of average disease incidence, and more importantly, in the percentage of individual lines displaying high disease resistance. For two different burley crosses, DH lines derived from the F1 generation were considerably more susceptible to black shank than DH lines derived from the F2 generation. No differences in the incidence of Fusarium wilt were observed between DH lines of F1 and F2 generations; this was likely due to low overall disease incidence. Although delaying the DH process in tobacco from the F1 to the F2 generation could add time to the development of homozygous breeding lines, the delay may be offset by having to screen fewer finished DH lines to identify superior lines.

Abstract:Since haploid embryoids were first obtained from microspore culture of Brassica napus there has been a rapid progress in the application of this technique in genetic studies and breeding utilization of rapeseed.The relative simplicity of the culture protocol and high-frequency recovery of doubled haploid plants makes microspore culture a potentially powerful technique for varietal development.Nevertheless, there are still some obstacles in the efficient use of this technique such as recalcitrant genotypes, secondary embryogenesis of embryoids, and the lack of efficient chromosome doubling.The present experiment is attempted to improve the production efficiency of doubled haploid rapeseed plants.

Abstract:Abstract Exploitation of biotechnological tools in conventional breeding strategies is the need of the hour for overcoming limitations in rice production and productivity. In addition, improvement in quantity and quality along with resistance to climatic and disease stress in rice require immediate attention. Anther culture has proven its efficiency by instantaneously fixing homozygosity through diploidization of regenerated haploid plants. Therefore, androgenesis provides an efficient platform for developing inbred lines in a short period of time. Although anther culture shows its efficiency in speeding up breeding in several crop species, including rice, associated limitations still prevent the exploitation of its optimum potential. Although anther culture is well exploited in japonica rice breeding, its application in indica rice is limited because of inherent recalcitrant genetic backgrounds. The success of anther culture is determined by several factors that limit the efficiency of androgenesis. Identified constraints are early anther necrosis, poor-callus response, and proliferation, and low green-plant regeneration, along with the most frustrating albinism associated with indica rice, which has been considerably clarified. This chapter details the method of androgenesis and scope for improving the applicability of anther culture producing doubled haploids of rice in order to use it as a complementary tool for precision breeding.

Abstract:This paper compared the methods of anther and microspore-isolated cultures in Chinese cabbage.The results showed that the difference of genotype between varieties was the key factor affecting production of the pollen embryo.The embryo inducement efficiency of microspore-isolated culture was 10 times greater than that of pollen culture.But the regeneration rate of pollen plants of microspore-isolated culture was much lower than that of pollen culture.Many key techniques of microspore-isolated culture should be improved in order to widely apply to breeding work.

Abstract:Double haploid technology (DH) is an essential tool in plant breeding, enabling the rapid production of homozygous lines. However, Cannabis sativa L. has been categorized as recalcitrant to DH induction. In this study, we evaluated the potential to generate DH C. sativa plants via anther culture and indirect de-novo organogenesis. We examined two cultivars, a THCA-dominant cultivar and a CBDA-dominant line. Callus induction success varied, with 29.48% for the THCA cultivar and 71.08% for the CBDA genotype with a regeneration success of 14.45% in 17 weeks for the latter. Mixoploidy in the callus indicated spontaneous genome doubling, while genetic testing confirmed DH nature of the regenerants. This is the first report documenting the successful induction of DH C. sativa plants through de-novo indirect organogenesis. These findings have profound implications for the C. sativa breeding sector by potentially improving efficiency of genome editing and hybrid development in this economically significant species.

Abstract:The doubled haploid technique aims to generate pure inbred lines for basic research and as commercial cultivars. The doubled haploid technique first generates haploid plants and is followed by chromosome doubling, which can be separated in time or overlapped, depending the procedure for each species. For a long time, much effort has been focused on haploid production via androgenesis, gynogenesis, or parthenogenesis. The obtention of haploid plants has frequently required more optimization and has lagged behind research and improvements in chromosome doubling methods. Nevertheless, chromosome doubling has recently been of renewed interest to increase the rates and efficiency of doubled haploid plant production through trialing and optimizing of different procedures. New antimitotic compounds and application methods are being studied to ensure the success of chromosome doubling once haploid material has been regenerated. Moreover, a haploid inducer-mediated CRISPR/Cas9 genome-editing system is a breakthrough method in the production of haploid plant material and could be of great importance for species where traditional haploid regeneration methods have not been successful, or for recalcitrant species. In all cases, the new deployment of this system will demand a suitable chromosome doubling protocol. In this review, we explore the existing doubled haploid and chromosome doubling methods to identify opportunities to enhance the breeding process in major crops.

Abstract:HANCOCK, WESLEY GRAHAM. Interspecific Hybridization in Nicotiana: Various Roles in a Modern Applied Tobacco Breeding Program. (Under the direction of Dr. Ramsey Lewis). Tobacco (Nicotiana tabacum L.) is one of the most economically important crops grown in North Carolina and worldwide. In a modern tobacco breeding program, continued efforts are undertaken to increase yields while maintaining or improving leaf quality and disease resistance. Two challenges are low levels of genetic diversity within flue-cured germplasm pools and the need for improved methods of doubled haploid breeding. The role of interspecific hybridization in dealing with these challenges was investigated in this research. Although favorable genetic variation affecting important quantitative traits likely exists within diploid Nicotiana relatives, their use in breeding programs presents a number of challenges. Work was conducted to evaluate the feasibility of using synthetic tobaccos as an alternative source of favorable alleles influencing key traits. The first objective of this research was to evaluate yield and growth rate and the relative amounts of heterosis for these traits in tobacco hybrids involving a set of diverse materials including two synthetic tobaccos. Cultivated tobacco  synthetic tobacco F2 populations and selected F2:3 families were also evaluated for a series of quantitative traits to determine realized heritability for growth rate on a single plant basis. Results showed significant mid-parent heterosis for hybrids involving the synthetic tobaccos for most traits. A range of phenotypes were observed in F2 populations and the realized heritability for growth rate was found to be very low (h = 0.14). Genome-wide transmission genetics, recombination frequencies, and preliminary marker-trait associations for measured traits in a N. tabacum × 4x(N. sylvestris  N. otophora) F2 population were examined and the relative importance of dominance, overdominance, and epistasis affecting heterosis for these traits was also investigated. We observed an overall reduction in pairwise marker distances and overall map length relative to those distances previously determined for populations derived from N. tabacum x N. tabacum crosses. Several QTL affecting growth rate, yield, leaf number and plant height were identified. Marker loci exhibited a range of d/|a| ratios, with the majority suggesting the importance of overdominance as a contributor to heterosis for yield, growth rate, and plant height, while epistasis and overdominance were important contributors to days to flower and leaf number. Synthetic tobaccos may be a useful source of favorable alleles affecting yieldrelated traits in a mediumto long-term breeding program. Conventional methods of inbreeding require a significant amount of time. An alternative inbreeding method used by tobacco breeders is doubled haploid breeding. In this method, haploid plants are identified, characterized, and subsequently chromosome-doubled to produce inbred lines in a greatly reduced amount of time. One method to produce N. tabacum haploids is using interspecific hybridization between N. tabacum and N. africana. This is a semi-lethal cross, where greater than 99% of the progeny die at the cotyledonary stage due to an interspecific lethality reaction. The remaining surviving plants are mixtures of maternal haploids of N. tabacum and aneuploid interspecific F1 hybrids. An efficient method is needed to distinguish maternal N. tabacum haploids from aneuploid hybrids during the seedling stage, however. The objective of this research was to investigate the use of N. africana engineered with a green fluorescent protein (gfp) transgene to assist in the discrimination between aneuploid hybrids and haploid plants resulting from this interspecific cross. Results demonstrate gfp expression to be a useful visual phenotypic marker for identification of maternal haploids at the seedling stage. DNA marker genotyping, flow cytometry, and chromosome counts of surviving non-haploid plants suggest that one or more genes on N. tabacum chromosome H play a role in the interspecific lethality reaction. Microsatellite marker genotyping and chromosome counts of three plants with low DNA contents indicated that these plants were likely the result of incomplete chromosome elimination, leading to the suggestion that chromosome elimination may play at least a partial role in haploid production in the N. tabacum  N. africana interspecific cross.

Abstract:The microspore culture technique has its wide applications in plant genetic research and breeding programmes in oilseed Brassicas due to its relative simplicity, efficiency in haploid and doubled haploid production, mutation and germplasm generation, and gene transformation. Various factors could influence microspore embryogenesis and haploid production including donor plant genotype, donor plant physiology, microspore developmental stage, culture conditions, culture environment and pretreatments. Stress is also an essential component during embryogenesis induction in microspore culture. Efficient plant regeneration from microspores mostly occurs through direct embryogenesis ensuring minimal occurrence of cytogenetic abnormalities. Appropriate stress conditions such as chilling, partial desiccation, cotyledon excision, and successive subculture of microspore-derived embryos could promote plant development in oilseed rape. Medium renovation, phytohormones and plant growth regulators, and chromosome doubling agents such as colchicine treatment also affect plant regeneration in Brassica species. Compared to colchicine treatments of microspore-derived embryos and plants, immediate colchicine treatment of isolated microspores results in high embryogenesis and diploidisation and low chimeric percentages. The ploidy level of microspore-derived plants of Brassica species could be estimated by different methods at various stages. Mutation breeding techniques are widely used in plant breeding for producing useful mutants and variants. Microspore culture also provides an ideal method for mutation because the mutated traits can be fixed in homozygous condition by chromosome doubling, which can enforce to obtain target mutation traits efficiently. Ultraviolet irradiation, mutagenic agents ethyl methane sulphonate and sodium azide could be applied to isolated microspores and the derived embryos of rapeseed. Utilization of microspore-derived embryos for production of desired traits such as the altered fatty acids, disease resistance and glucosinolate compositions through mutagenesis and selection is advancing and also discussed.

Haploid induction plays a fundamental role in the production of DH lines. Haploid induction has been realized and applied in diploid plants using DMP genes. However, it has yet to be elucidated whether haploid induction could be established in polyploid plants. In the current study, three homologues of the DMP genes (NtDMP1, 2, and 3) were identified in the allotetraploid plant Nicotiana tabacum, and the encoded proteins localized in the endoplasmic reticulum. Loss-of-function mutations in all three genes triggered maternal haploids with an induction rate of 1.52–1.75%. Compared with wild-type tobacco, the created haploid inducer exhibited differences in pollen vigor and seed germination rate. Furthermore, to rapidly and easily screen haploids, a visible haploid identification system was established based on a powdery mildew resistance phenotype. Findings from this study lay the foundation for the potential application of haploid inducers in allotetraploid plants such as tobacco.

Abstract:The aim of any breeding process is to fully express the targeted, superior/desirable parent characteristic in the progeny. Hybrids are often used in this dynamic, and complex process for which homozygous parents-which may require up to eight generations of back crossing and selection-are required. Doubled haploid (DH) technologies can facilitate the production of true breeding lines faster and in a more efficient manner than the traditional back crossing and selection strategies. Sunflower is the third most important oilseed crop in the world and has no available double haploid induction procedure/technique that can be efficiently used in breeding programs. A reproducible and efficient doubled haploid induction method would be a valuable tool in accelerating the breeding of new elite sunflower varieties. Although several attempts have been made, the establishment of a sunflower doubled haploid induction protocol has remained a challenge owing recalcitrance to in vitro culture regeneration. Approaches for haploid development in other crops are often cultivar specific, difficult to reproduce, and rely on available tissue culture protocols-which on their own are also cultivar and/or species specific. As an out-crossing crop, the lack of a double haploid system limits sunflower breeding and associated improvement processes, thereby delaying new hybrid and trait developments. Significant molecular advances targeting genes, such as the centromeric histone 3 (CenH3) and Matrilineal (MTL) gene with CRISPR/Cas9, and the successful use of viral vectors for the delivery of CRISPR/Cas9 components into plant cells eliminating the in vitro culture bottleneck, have the potential to improve double haploid technology in sunflower. In this review, the different strategies, their challenges, and opportunities for achieving doubled haploids in sunflower are explored.
Development of an RNA Virus Vector for Non-Transgenic Genome Editing in Tobacco and Generation of Berberine Bridge Enzyme-Like Mutants with Reduced Nicotine Content

Abstract:Tobacco (Nicotiana tabacum) plants synthesize the psychoactive pyridine alkaloid nicotine, which has sparked growing interest in reducing nicotine levels through genome editing aiming at inactivating key biosynthetic genes. Although stable transformation-mediated genome editing is effective in tobacco, its polyploid nature complicates the complete knockout of genes and the segregation of transgenes from edited plants. In this study, we developed a non-transgenic genome editing method in tobacco by delivering the CRISPR/Cas machinery via an engineered negative-strand RNA rhabdovirus vector, followed by the regeneration of mutant plants through tissue culture. Using this method, we targeted six berberine bridge enzyme-like protein (BBL) family genes for mutagenesis, which are implicated in the last steps of pyridine alkaloid biosynthesis, in the commercial tobacco cultivar Hongda. We generated a panel of 16 mutant lines that were homozygous for mutations in various combinations of BBL genes. Alkaloid profiling revealed that lines homozygous for BBLa and BBLb mutations exhibited drastically reduced nicotine levels, while other BBL members played a minor role in nicotine synthesis. The decline of nicotine content in these lines was accompanied by reductions in anatabine and cotinine levels but increases in nornicotine and its derivative myosmine. Preliminary agronomic evaluation identified two low-nicotine lines with growth phenotypes comparable to those of wild-type plants under greenhouse and field conditions. Our work provides potentially valuable genetic materials for breeding low-nicotine tobacco and enhances our understanding of alkaloid biosynthesis.
Transgenic Technology and Its Application in Genetic Improvement of Flowers

Abstract:Transgenic technology played an important role in the genetic modification of flowers.The present situation of the transgenic techniques in the application of modifying the floral characters was introduced.However,some problems were existed in the application of transgenic technology existed,e.g.the issue of biological safety.Due to these matters,some suggestions and layout were made,which could provide the theoretical foundation to cultivate some special flowers and develop the floriculture industry.
