Fluorescence-Labeled Imperacalcin Binds to Open Ryr2 Channels in Mouse Ventricular Myocytes

Abstract:Imperacalcin (IpCa) engages cardiac ryanodine receptors (RyR2 channels) with high affinity (Kd=5∼10nM), inducing a reversible and long-lasting substate, thereby releasing Ca2+ from sarcoplasmic reticulum. Here we investigated the in situ binding properties of IpCa to RyR2s using a fluorescent derivative of IpCa (Alexa546-2IT-IpCa) in mouse ventricular myocytes. Methods: Ventricular myocytes were permeabilized with 0.5% saponin in internal solution containing 10nM Ca2+. Specific binding of fluorescence-labeled IpCa to RyRs was calculated by subtracting the fluorescence of Alexa546-2IT-IpCa at the M band (FM) from that at the Z-line (FZ). Different combinations of Ca2+, ryanodine, caffeine, thapsigargin and tetracaine were used to characterize the IpCa-RyR2 binding properties. We also tested the effect of IpCa on FRET between Alexa488-FKBP12.6 and Alexa568-CaM. Results: Binding of Alexa546-2IT-IpCa (500nM) to RyR2s at Z-lines in 100nM Ca2+ was inhibited by 5µM IpCa (FZ-FM, 13.2±4.6 vs. 1.5±3.1 a.u., n=10, p<0.01), indicating specific binding, and enhanced by Ca2+ (FZ-FM at 10 and 100nM Ca2+ was 6.3±1.5 vs. 13.3±4.1 a.u., respectively. n=10, p<0.01). At 10nM Ca2+, when RyR2 channels are mostly closed as indicated by the presence of scattered sparks, both 20nM or 100µM ryanodine plus 5mM caffeine (which open RyR2 channels) increased the binding of Alexa546-2IT-IpCa to RyR2 significantly. However, at 100nM Ca2+ and in the presence of discrete Ca2+ waves, there was no further increase of binding by ryanodine plus caffeine combined. On the other hand, the RyR2 channel blocker tetracaine (2mM) plus thapsigargin (2.5µM) decreased Alexa546-2IT-IpCa binding at 100nM Ca2+(FZ-FM, 0.3±1.4 vs. 11.5±3.4a.u., n=10, p<0.01). No effect of IpCa on FRET between Alexa488-FKBP12.6 and Alexa568-CaM was detected. Conclusion: Alexa546-2IT-IpCa binds specifically to RyR2s at Z-lines under conditions that favor RyR2 openings. IpCa does not compete with CaM or FKBP12.6 for binding to RyR2s.
