Abstract:Considerable efforts have been focused on shifting the wavelength of aequorin Ca2+-dependent blue bioluminescence through fusion with fluorescent proteins. This approach has notably yielded the widely used GFP-aequorin (GA) Ca2+ sensor emitting green light, and tdTomato-aequorin (Redquorin), whose bioluminescence is completely shifted to red, but whose Ca2+ sensitivity is low. In the present study, the screening of aequorin mutants generated at twenty-four amino acid positions in and around EF-hand Ca2+-binding domains resulted in the isolation of six aequorin single or double mutants (AequorinXS) in EF2, EF3, and C-terminal tail, which exhibited markedly higher Ca2+ sensitivity than wild-type aequorin in vitro. The corresponding Redquorin mutants all showed higher Ca2+ sensitivity than wild-type Redquorin, and four of them (RedquorinXS) matched the Ca2+ sensitivity of GA in vitro. RedquorinXS mutants exhibited unaltered thermostability and peak emission wavelengths. Upon stable expression in mammalian cell line, all RedquorinXS mutants reported the activation of the P2Y2 receptor by ATP with higher sensitivity and assay robustness than wt-Redquorin, and one, RedquorinXS-Q159T, outperformed GA. Finally, wide-field bioluminescence imaging in mouse neocortical slices showed that RedquorinXS-Q159T and GA similarly reported neuronal network activities elicited by the removal of extracellular Mg2+. Our results indicate that RedquorinXS-Q159T is a red light-emitting Ca2+ sensor suitable for the monitoring of intracellular signaling in a variety of applications in cells and tissues, and is a promising candidate for the transcranial monitoring of brain activities in living mice.
Transient Expression of Apoaequorin in Zebrafish Embryos: Extending the Ability to Image Calcium Transients During Later Stages of Development

Abstract:When aequorin is microinjected into cleavage-stage zebrafish embryos, it is largely used up by ~24 hours. Thus, it is currently not possible to image Ca(2+) signals from later stages of zebrafish development using this approach. We have, therefore, developed protocols to express apoaequorin, i.e., the protein component of aequorin, transiently in zebrafish embryos and then reconstitute intact aequorin in vivo by loading the coelenterazine co-factor into the embryos separately. Two types of apoaequorin mRNA, aeq-mRNA and aeq::EGFP-mRNA, the latter containing the enhanced green fluorescent protein (EGFP) sequence, were in vitro transcribed and when these were microinjected into embryos, they successfully translated apoaequorin and a fusion protein of apoaequorin and EGFP (apoaequorin-EGFP), respectively. We show that aeq::EGFP -mRNA was more toxic to embryos than equivalent amounts of aeq-mRNA. In addition, in an in vitro reconstitution assay, apoaequorin-EGFP produced less luminescence than apoaequorin, after reconstitution with coelenterazine and with the addition of Ca(2+). Furthermore, when imaging intact coelenterazine-loaded embryos that expressed apoaequorin, Ca(2+ )signals from ~2.5 to 48 hpf were observed, with the spatio-temporal pattern of these signals up to 24 hpf, being comparable to that observed with aequorin. This transient aequorin expression approach using aeq-mRNA provides a valuable tool for monitoring Ca(2+ )signaling during the 2448 hpf period of zebrafish development. Thus, it effectively extends the aequorin-based Ca(2+) imaging window by an additional 24 hours.

Abstract:Voltage imaging allows mapping of the membrane potential in living cells. Yet, current intensity-based imaging approaches are limited to relative membrane potential changes, missing important information conveyed by the absolute value of the membrane voltage. This challenge arises from various factors affecting the signal intensity, such as concentration, illumination intensity, and photobleaching. Here, we demonstrate electronic preresonance hyperspectral stimulated Raman scattering (EPR-hSRS) for spectroscopic detection of the membrane voltage using a near-infrared-absorbing microbial rhodopsin expressed in E. coli. This newly developed near-infrared active microbial rhodopsin enables electronic preresonance SRS imaging at high sensitivity. By spectral profiling, we identified voltage-sensitive SRS peaks in the fingerprint region in single E. coli cells. These spectral signatures offer a new approach for quantitation of the absolute membrane voltage in living cells.

Abstract:Ratiometric indicators with long emission wavelengths are highly preferred in modern bioimaging and life sciences. Herein, we elucidated the working mechanism of a standalone red fluorescent protein (FP)-based Ca2+ biosensor, REX-GECO1, using a series of spectroscopic and computational methods. Upon 480 nm photoexcitation, the Ca2+-free biosensor chromophore becomes trapped in an excited dark state. Binding with Ca2+ switches the route to ultrafast excited-state proton transfer through a short hydrogen bond to an adjacent Glu80 residue, which is key for the biosensor’s functionality. Inspired by the 2D-fluorescence map, REX-GECO1 for Ca2+ imaging in the ionomycin-treated human HeLa cells was achieved for the first time with a red/green emission ratio change (ΔR/R0) of ~300%, outperforming many FRET- and single FP-based indicators. These spectroscopy-driven discoveries enable targeted design for the next-generation biosensors with larger dynamic range and longer emission wavelengths.

Abstract:This review updates the imaging of free cytosolic calcium with the chemiluminescent aequorins. Basic principles of chemiluminescence are discussed and the biochemistry of aequorins is briefly described. The review provides practical tips on handling and microinjecting aequorins and describes available ultra low light imaging systems. It is argued that aequorin‐based calcium imaging is the method of choice for exploratory studies, since it is extremely sensitive, can detect a broad range of calcium concentrations, and allows for continuous recording during long periods of time. Abstract:4596 Background: One in seven men over age 60 will be diagnosed with prostate cancer. Improvements in screening and diagnostic procedures allow clinicians to better identify men at high risk of having prostate disease, but no established means of preventing prostate cancer are currently available. Statins are a group of drugs used to control high cholesterol through inhibiting HMG CoA reductase and interfering in the mevalonate pathway. Recent research has demonstrated that in a number of tumor types, statins also induce apoptosis and growth arrest. The purpose of this pilot study was to evaluate the association between statin use and prostate cancer risk in a population of older veterans enrolled in a case-control study of diet and prostate cancer risk. METHODS Cases were recruited upon referral for prostate biopsy and frequency age-matched; PSA normal controls were identified from the VA primary care clinic. Statin use was determined from medical record review. Years of use, type of statin, dose and any changes were recorded. Years of use were categorized as > or <= 3 years of use of any single or combined statin. Dose was categorized as > or <= 40mg/day, based on the highest dose prescribed. RESULTS Records from 47 incident prostate cancer cases and 142 PSA normal clinic controls were reviewed. After adjusting for age and body mass index, men with any recorded statin use had a 56% lower risk of prostate cancer as opposed to non-users (odds ratio (OR) 0.44, 95% confidence interval (CI) (0.22-0.88)). When statin use was grouped by duration and dose, those with the longer duration of use (>3 years) and higher dose (>40mg/day) were at lower risk of prostate cancer as compared to non-users (OR=0.35, 95% CI=0.12-1.00). CONCLUSIONS Results of this pilot study suggest that any statin use is associated with a reduced risk of prostate cancer and that use for a longer period of time at a higher dose may confer the most risk reduction. This study is limited by a small sample size and lack of control for additional comorbid factors. If these results are confirmed in a larger prospective study they may provide necessary evidence to consider the use of statin drugs in prostate cancer prevention. Abstract:Calcium ions (Ca2+) play central roles in cellular physiology. Fluorescent indicators for Ca2+ ions revolutionized our ability to make rapid, accurate, and highly parallel measurement of Ca2+ concentrations in living cells. The use of ratio-based imaging with one particular indicator, fura-2, allowed practitioners to correct for a number of experimental confounds, including dye bleaching, variations in sample thickness, and fluctuations in illumination intensity. Ratio-based imaging with fura-2 was the most accurate and reliable method for measuring Ca2+ concentrations. Two drawbacks to fura-2 exist. First, it requires ultraviolet (UV) excitation, which is more toxic to living cells than visible light. Second, our ability to use fura-2 for accurate, stable, ratio-based determinations of Ca2+ concentration in living cells is fast becoming a method of the past. This is due, in part, because modern microscopes are phasing out the use of mercury arc lamps that provide the UV excitation needed for fura-2 imaging. To address this problem, we describe the design, synthesis, and cellular application of benzo[b]phosphole-based fluorescent Ca2+ indicators for ratio-based imaging of Ca2+ in living cells that can be used with modern light emitting diode (LED)-equipped fluorescence microscopes. We show that isoCaRed-1Me has absorbance spectra in the visible range, shows Ca2+ selectivity over Mg2+, and displays a Ca2+-dependent excitation spectra. These unique properties enable ratio-based imaging in immortalized cell lines, primary mammalian hippocampal neurons, and human induced pluripotent stem cell-derived cardiomyocytes. These data show that isoCaRed-1Me will be useful for ratio-based Ca2+ imaging using modern microscopes.

Abstract:Abstract Although fluorescent indicators have been broadly utilized for monitoring bioactivities, fluorescence imaging, when applied to mammals, is limited to superficial targets or requires invasive surgical procedures. Thus, there is emerging interest in developing bioluminescent indicators for noninvasive mammalian imaging. Bioluminescence imaging (BLI) of neuronal activity is highly desired but hindered by insufficient photons needed to digitalize fast brain activities. In this work, we develop a luciferase prosubstrate deliverable at an increased dose and activated in vivo by nonspecific esterase. We further engineer a bright, bioluminescent indicator with robust responsiveness to calcium ions (Ca 2+ ) and appreciable emission above 600 nm. Integration of these advantageous components enables the imaging of the activity of neuronal ensembles in awake mice minimally invasively with excellent signal-to-background and subsecond temporal resolution. This study thus establishes a paradigm for studying brain function in health and disease.

Abstract:Ca ions passing through a single or a cluster of Ca -permeable channels create microscopic, short-lived Ca gradients that constitute the building blocks of cellular Ca signaling. Over the last decade, imaging microdomain Ca in muscle cells has unveiled the exquisite spatial and temporal architecture of intracellular Ca dynamics and has reshaped our understanding of Ca signaling mechanisms. Major advances include the visualization of “Ca sparks” as the elementary events of Ca release from the sarcoplasmic reticulum (SR), “Ca sparklets” produced by openings of single Ca -permeable channels, miniature Ca transients in single mitochondria (“marks”), and SR luminal Ca depletion transients (“scraps”). As a model system, a cardiac myocyte contains a 3-dimensional grid of 10 spark ignition sites, stochastic activation of which summates into global Ca transients. Tracking intermolecular coupling between single L-type Ca channels and Ca sparks has provided direct evidence validating the local control theory of Ca -induced Ca release in the heart. In vascular smooth muscle myocytes, Ca can paradoxically signal both vessel constriction (by global Ca transients) and relaxation (by subsurface Ca sparks). These findings shed new light on the origin of Ca signaling efficiency, specificity, and versatility. Abstract:Calcium ion is a versatile second messenger for diverse cell signaling in response to developmental and environmental cues. The specificity of Ca(2+)-mediated signaling is defined by stimulus-elicited Ca(2+) signature and downstream decoding processes. Here, an Aequorin-based luminescence recording system was developed for monitoring Ca(2+) in response to various stimuli in Arabidopsis. With the simple, highly sensitive, and robust Ca(2+) recording, this system revealed stimulus- and tissue-specific Ca(2+) signatures in seedlings. Cellular Ca(2+) dynamics and relationship to Aequorin-based Ca(2+) recording were explored using a GFP-based Ca(2+) indicator, which suggested that a synchronous cellular Ca(2+) signal is responsible for cold-induced Ca(2+) response in seedlings, whereas asynchronous Ca(2+) oscillation contributes to osmotic stress-induced Ca(2+) increase in seedlings. The optimized recording system would be a powerful tool for the identification and characterization of novel components in Ca(2+)-mediated stress-signaling pathways.
Simultaneous dual-color calcium imaging in freely-behaving mice

Abstract:Miniaturized fluorescence microscopes (miniscopes) enable imaging of calcium events from a large population of neurons with single-cell resolution in freely behaving animals. Traditionally, miniscopes have only been able to record from a single fluorescence wavelength. Here, we present a new open-source dual-channel Miniscope that simultaneously records two wavelengths in freely behaving animals, providing more flexibility and function. To enable simultaneous acquisition of two fluorescent wavelengths, we incorporated two CMOS sensors with two independent sets of emission filters into a single Miniscope. This enables us to acquire images at the full frame rate that the CMOS sensor supports while simultaneously eliminating crosstalk between channels, which allows imaging fluorophores that heavily overlap in their emission spectrums (for example, GFP and tdTomato). We adopted a design that enables adjusting the position of one imaging sensor along the light path to match the focal plane of the other sensor, providing additional flexibility for calibration. To validate our dual-channel Miniscope in vivo, we imaged hippocampal CA1 region that co-expressed a dynamic calcium indicator (GCaMP) and a static nuclear signal (constitutively active tdTomato) while mice ran on a linear track. We used GCaMP signals to investigate spatial coding in the hippocampus and tdTomato signals as a landmark for registration of neurons across recording sessions. Our results suggest that, even when neurons were registered across recording sessions using tdTomato signals, hippocampal spatial coding changes over time as previously reported. In conclusion, our novel dual-channel Miniscope enables imaging of two fluorescence wavelengths in the same focal plane with minimal crosstalk between the two channels, opening the doors to a multitude of new experimental possibilities.

Abstract:Abstract Mitochondrial Ca 2+ dynamics are involved in the regulation of multifarious cellular processes, including intracellular Ca 2+ signalling, cell metabolism and cell death. Use of mitochondria-targeted genetically encoded Ca 2+ indicators has revealed intercellular and subcellular heterogeneity of mitochondrial Ca 2+ dynamics, which are assumed to be determined by distinct thresholds of Ca 2+ increases at each subcellular mitochondrial domain. The balance between Ca 2+ influx through the mitochondrial calcium uniporter and extrusion by cation exchangers across the inner mitochondrial membrane may define the threshold; however, the precise mechanisms remain to be further explored. We here report the new red fluorescent genetically encoded Ca 2+ indicators, R-CEPIA3 mt and R-CEPIA4 mt , which are targeted to mitochondria and their Ca 2+ affinities are engineered to match the intramitochondrial Ca 2+ concentrations. They enable visualization of mitochondrial Ca 2+ dynamics with high spatiotemporal resolution in parallel with the use of green fluorescent probes and optogenetic tools. Thus, R-CEPIA3 mt and R-CEPIA4 mt are expected to be a useful tool for elucidating the mechanisms of the complex mitochondrial Ca 2+ dynamics and their functions.

Effect of Ca²⁺ on the steady-state and time-resolved emission properties of the genetically encoded fluorescent sensor CatchER.

Abstract:We previously designed a calcium sensor CatchER (a GFP-based Calcium sensor for detecting high concentrations in the high calcium concentration environment such as ER) with a capability for monitoring calcium ion responses in various types of cells. Calcium binding to CatchER induces the ratiometric changes in the absorption spectra, as well as an increase in fluorescence emission at 510 nm upon excitation at both 395 and 488 nm. Here, we have applied the combination of the steady-state and time-resolved optical methods and Hydrogen/Deuterium isotope exchange to understand the origin of such calcium-induced optical property changes of CatchER. We first demonstrated that calcium binding results in a 44% mean fluorescence lifetime increase of the indirectly excited anionic chromophore. Thus, CatchER is the first protein-based calcium indicator with the single fluorescent moiety to show the direct correlation between the lifetime and calcium binding. Calcium exhibits a strong inhibition on the excited-state proton transfer nonadiabatic geminate recombination in protic (vs deuteric) medium. Analysis of CatchER crystal structures and the MD simulations reveal the proton transfer mechanism in which the disrupted proton migration path in CatchER is rescued by calcium binding. Our finding provides important insights for a strategy to design calcium sensors and suggests that CatchER could be a useful probe for FLIM imaging of calcium in situ.

Abstract:Of the ions involved in myocardial function, Ca 2+ is the most important. Ca 2+ is crucial to the process that allows myocardium to repeatedly contract and relax in a well-organized fashion; it is the process called excitation–contraction coupling. In order, therefore, for accurate comprehension of the physiology of the heart, it is fundamentally important to understand the detailed mechanism by which the intracellular Ca 2+ concentration is regulated to elicit excitation–contraction coupling. Aequorin was discovered by Shimomura, Johnson and Saiga in 1962. By taking advantage of the fact that aequorin emits blue light when it binds to Ca 2+ within the physiologically relevant concentration range, in the 1970s and 1980s, physiologists microinjected it into myocardial preparations. By doing so, they proved that Ca 2+ transients occur upon membrane depolarization, and tension development (i.e., actomyosin interaction) subsequently follows, dramatically advancing the research on cardiac excitation–contraction coupling.

Abstract:We report an intensiometric, near-infrared fluorescent, genetically encoded calcium ion (Ca2+) indicator (GECI) with excitation and emission maxima at 678 and 704 nm, respectively. This GECI, designated NIR-GECO1, enables imaging of Ca2+ transients in cultured mammalian cells and brain tissue with sensitivity comparable to that of currently available visible-wavelength GECIs. We demonstrate that NIR-GECO1 opens up new vistas for multicolor Ca2+ imaging in combination with other optogenetic indicators and actuators.

Abstract:Three new red-excitable monomeric fluorescent proteins obtained by structure-guided mutagenesis of mNeptune are described in this work. The authors show the use of one of them, mCardinal, to visualize the differentiation of myoblasts into myocytes in living mice.

Emerging Trends in Fluorescence Bioimaging of Divalent Metal Cations Using Small‐Molecule Indicators

Abstract:Visualization of cation dynamics inside a living system represent a major breakthrough at the crossroad of chemistry and cellular physiology. Since the inception of BAPTA-based cellular calcium indicators in the 1980s, generations of chemical and genetically encoded ion indicators spanning the visible spectrum have been developed. In this article, we bring up three emerging concepts in this field: 1. red-shifting cation indicators towards far-red and near-infrared (NIR) channels; 2. directing the indicators to various subcellular localizations; 3. lowering the phototoxicity of indicators for long term recording. These initiatives collectively echo the advocate of 4D cellular physiology, where biological processes within living systems can be panoramically unveiled under 3D, long-term, and multi-channel imaging with unprecedented spatial and temporal resolution. This outlook poses exciting challenges and opportunities for chemists to upgrade the toolkit of fluorescent indicators as key enablers for a new era of imageomics.

Abstract:Fluorescent protein (FP)-based indicators revolutionized our ability to spy on the otherwise invisible world of intracellular signalling. The most impactful class of FP-based indicators are the Ca2+ indicators that change their fluorescence intensity or color in response to a change in Ca2+ concentration. These indicators are frequently used in combination with optogenetic actuators to enable simultaneous control and visualization of cellular signalling with precise spatial and temporal resolution, particularly in the area of neuroscience. In this seminar I will describe our most recent efforts to use protein engineering to make a new generation of genetically encoded Ca2+ indicators with improved properties and an expanded range of potential applications. Specifically, I will present our efforts to engineer red and near-infrared Ca2+ indicators that are suitable for use in combination with blue-light activatable optogenetic actuators. In addition, I will describe our recent success at exploiting FPs for optogenetic control of protein activity and gene expression.

Abstract:Eating attitudes and behaviors, body image, and psychological functioning were evaluated in 98 female college students: 36 African-Americans, 34 Asian-Americans, and 28 Caucasians. African-Americans had significantly higher body mass index than either Asian-American or Caucasians. In contrast, Caucasians reported greater levels of disordered eating and dieting behaviors and attitudes and greater body dissatisfaction than did Asian-Americans and African-Americans who differed little on these measures. The nature of variability in these eating behaviors and attitudes and body image was also examined within each of the three groups. A generally consistent pattern emerged within each racial group: low self-esteem and high public self-consciousness were associated with greater levels of problematic eating behaviors and attitudes and body dissatisfaction. A history of being teased about weight and size was associated with problematic eating behaviors and attitudes and body dissatisfaction in African-Americans and Caucasians but not in Asian-Americans. The findings suggest that there exist important racial differences on various aspects of eating, dieting, and body image in college women. Contrary to hypothesis, the degree of acculturation and assimilation within the African-American and Asian-American groups was unrelated to variability in these domains.
