Abstract:Dearomatization reactions provide the most efficient method for the synthesis of spiro- or fused-ring systems from readily available compounds. This review summarizes the recent developments in dearomatization reactions of indoles, pyridines, quinolines, isoquinolines, and some other heteroaromatic compounds. The applications of these methods in total synthesis of natural products are also briefly introduced.

Abstract:Dearomative reactions allow for concise assembly of structurally complex, highly functionalized three‐dimensional molecules by breaking the plane of two‐dimensional aromatic starting materials. With the growing interest in N‐heterocyclic carbene (NHC) organocatalysis as a powerful strategy for synthesizing diversiform molecules, mainly by polarity‐reversal (umpolung) approaches, NHC‐catalyzed dearomatization reactions have recently been successfully used for the construction of the skeletons of natural products and drugs. By generating various key intermediates (i.e., Breslow intermediates, acyl azolium intermediates, homoenolate intermediates, enolate intermediates, and azolium dienolate intermediates), NHCs can readily destroy aromaticity by electrophilic or nucleophilic interactions with aromatic compounds. In this overview, progress in NHC‐catalyzed dearomative reactions is summarized; the reactions are grouped according to reaction modes and categories of intermediates, and representative mechanisms are provided. We expect this review to serve as a comprehensive reference for work in the emerging field of organocatalytic dearomatization.

Abstract:Abstract This study reports the first synthesis of tetrahydroquinaldic and tetrahydropicolinic amides using readily available quinolines and pyridines through dearomative difunctionalization and hydrolysis under transition‐metal‐ and reductant‐free conditions. The prepared amide products could be easily oxidized to the corresponding quinaldic and picolinic amides. Furthermore, the chemical reactivity of the amide group has been explored to develop convenient methods for their conversion to other common organic functional groups.

Abstract:Dearomatization has emerged as a powerful manifold for rapid construction of 3D molecular architectures from simple, abundant, and planar (hetero)arenes. The field has evolved beyond the simple dearomatization driven by new synthetic technology development. With the renaissance of photocatalysis and expanding the activation mode, the last few years have witnessed impressive developments on innovative photochemical dearomatization methodologies, enabling skeletal modifications of the dearomatic structures. They offer truly efficient and useful tools for facile construction of highly complex structures in synthesis of nature products and drug discovery. In this review, we aim to provide a mechanistically insightful overview on these innovations based on the degree of skeletal alternation, categorized into dearomative functionalization and skeletal editing, and highlight their synthetic utilities.

Abstract:Dearomatization reactions provide a rapid approach to construct complicated molecules that are difficult to synthesize by traditional methods from simple aromatic compounds. Herein, we report an efficient dearomative [3+2] cycloaddition reaction of 2-alkynyl pyridines with diarylcyclopropenones, leading to the synthesis of densely functionalized indolizinones in moderate to good yields under metal-free conditions. In addition, this strategy can also be employed in dearomative cyclization of isoquinolines to access a variety of benzo-fused indolizinones. Density functional theory (DFT) calculations revealed that an appropriate substituent at the 2-position of pyridine is crucial to the dearomatization process.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:Pyridones/isoquinolones are a kind of important nitrogen-containing heterocyclic compounds, which have good biological activity and unique chemical properties, and play an important role in synthesis chemistry, functional materials, biomedicine and so on. Recently, transition-metal-catalyzed C-H activation has emerged as a more atom- and step-economical strategy for the construction of functionalized pyridones/isoquinolones, and it has attracted the attention of many organic chemists. Herein, the recent advances in transition-metal-catalyzed C-H activation of pyridone/isoquinolone derivatives are summarized according to the types of metal catalysis. An emphasis on the discussion of catalytic system, reaction mechanisms, substrate scopes and synthetic applications is also introduced. Finally, the limitations and development trend of this research field are analyzed and prospected.

Abstract:The stereoselective preparation of functionalized [1,2,4]triazolo[4,3-a]pyridines from N-tosylhydrazones and pyridines was developed through the dearomatization of pyridines. The current transformation features good step- and atom-economy, high diastereoselectivity, and the efficient formation of four new carbon-heteroatom bonds in the corresponding product tetrahydro pyridines.

Abstract:Diazo compounds had been widely employed as a class of environment friendly and highly efficient coupling reagents in transition-metal-catalyzed C-H bonds insertion reactions.Recently, C-H alkylation of various arenes and heteroarenes has been achieved enormous successes using diazo compounds as the alkylation reagents under a new catalytic model.The recent progress in diazo compounds mediated transition-metal-catalyzed C-H alkylation and tandem C-H alkylation/cyclization is reviewed, including reaction mechanism and synthetic applications.Furthermore, the prospects of this reaction are also discussed.

Abstract:Accessing fascinating organic and biological significant indolines via dearomatization of indoles represents one of the most efficient approaches. However, it has been difficult for the dearomatization of the electron deficient indoles. Here we report the studies leading to developing a photoredox mediated Giese-type transformation strategy for the dearomatization of the indoles. The reaction has been implemented for chemoselectively breaking indolyl C=C bonds embedded in the aromatic system. The synthetic power of this strategy has been demonstrated by using structurally diverse indoles bearing common electron-withdrawing groups including (thio)ester, amide, ketone, nitrile and even aromatics at either C2 or C3 positions and ubiquitous carboxylic acids as radical coupling partner with high trans-stereoselectivity (>20:1 dr). This manifold can also be applied to other aromatic heterocycles including pyrroles, benzofurans and benzothiophenes. Furthermore, enantioselective dearomatization of indoles has been achieved by a chiral camphorsultam auxiliary with high diastereoselectivity.

Abstract:Herein, based on dearomatization-rearomatization strategy, a straightforward construction of poly-substituted and fused 4H-quinolizin-4-ones via direct ortho C-H bond functionalization of pyridines in metal-free conditions has been developed. This approach offers a convenient method for the modular synthesis of a diversity of functionalized 4H-quinolizin-4-ones from simple pyridines, methyl 2-bromoacetates, and alkenes. Mechanistic studies indicated that the reaction included pre- or in-situ activation of pyridines to N-(2-Methoxy-2-oxoethyl) pyridinium salts, dearomative [3+2] cycloaddition, and rearomative ring expansion processes. Importantly, without the ester group, an interesting C-H bond allylation of pyridine was observed. Finally, based on the experimental results, an aromatization-driven β cleavage of relatively stable pyridine radical intermediate was proposed as key process for the construction of the desired 4H-quinolizin-4-ones over the traditional indolizines.

Abstract:A regioselective one‐pot approach to the synthesis of C4‐alkylated tetrahydropyridines (THPs) and tetrahydroquinolines (THQs) by dearomatization of pyridines and quinolines has been developed. The transformation is achieved by the BF 3 •OEt 2 ‐mediated, C4‐selective addition of Grignard reagents, followed by a cascade of enamine protonation and the subsequent Et 3 SiH reduction of the resulting iminium ions. This protocol not only provides a facile synthetic approach to C4‐substituted THPs and THQs, but also offers a new strategy for the dearomatization of heteroaromatics.

Abstract:The review summarizes the known data on various methods for saturation and dearomatization of heterocyclic moieties, forming diverse aromatic compounds. Since the classical hydrogenation of substrates with molecular hydrogen has a number of fundamental drawbacks that hinder both its use at the laboratory level and its widespread implementation in industrial production, the main attention is paid to non-classical methods described in the literature over the last 20-25 years. The methods of ammonia-free Birch reduction, transfer hydrogenation, reduction by Hantzsch esters and some other reactions involving heterocyclic compounds are summarized and systematized. It is shown that such methods of reduction or dearomatization of heterocycles can in some cases be more efficient than classical procedures. The bibliography includes 173 references.
