Abstract:Hypervalent iodine-promoted dearomatization of phenols has received intense attention. This mini-review summarizes recent computational mechanistic studies of phenolic dearomatizations promoted by hypervalent iodine(III) reagents or catalysts. The first part of this review describes mechanisms of racemic dearomatization of phenols, paying special attention to the associative and dissociative pathways. The second part focuses on mechanisms and selectivities of diastereo- or enantio-selective dearomatization of phenols.

Abstract:Phenols are important readily available synthetic building blocks and starting materials for organic synthetic transformations, which are widely found in agrochemicals, pharmaceuticals, and functional materials. The C–H functionalization of free phenols has proven to be an extremely useful tool in organic synthesis, which provides efficient increases in phenol molecular complexity. Therefore, approaches to functionalizing existing C–H bonds of free phenols have continuously attracted the attention of organic chemists. In this review, we summarize the current knowledge and recent advances in ortho-, meta-, and para-selective C–H functionalization of free phenols in the last five years.

Abstract:A direct catalytic asymmetric multiple dearomatization reaction of phenols was disclosed, which provides expedient access to a series of architecturally complex polycyclic compounds bearing four stereogenic centers in high enantiopurity. The key to achieve such a transformation is the combination of a dearomative 1,8-addition of beta-naphthols to para-quinone methides generated in situ from propargylic alcohols and a subsequent intramolecular dearomative Diels-Alder reaction. Noteworthily, this protocol enrichs not only the diversity of dearomatized products but also the toolbox of dearomatization strategies.

Abstract:Abstract Substituted aromatic compounds play important roles in materials, biological agents, dyes, etc. Thus, the synthesis of substituted aromatic compounds has been a hot topic throughout the history of organic chemistry. Traditionally, the Friedel–Crafts reaction was a powerful tool for synthesizing substituted aromatic compounds. In recent decades, metal-catalyzed cross-coupling reactions were well developed via carbon–heteroatom bond cleavage, however, having difficulties towards some strong bonds, such as C(Ar)–OH. To overcome such challenges, newer strategies are needed. Abstract:Dearomative reactions allow for concise assembly of structurally complex, highly functionalized three‐dimensional molecules by breaking the plane of two‐dimensional aromatic starting materials. With the growing interest in N‐heterocyclic carbene (NHC) organocatalysis as a powerful strategy for synthesizing diversiform molecules, mainly by polarity‐reversal (umpolung) approaches, NHC‐catalyzed dearomatization reactions have recently been successfully used for the construction of the skeletons of natural products and drugs. By generating various key intermediates (i.e., Breslow intermediates, acyl azolium intermediates, homoenolate intermediates, enolate intermediates, and azolium dienolate intermediates), NHCs can readily destroy aromaticity by electrophilic or nucleophilic interactions with aromatic compounds. In this overview, progress in NHC‐catalyzed dearomative reactions is summarized; the reactions are grouped according to reaction modes and categories of intermediates, and representative mechanisms are provided. We expect this review to serve as a comprehensive reference for work in the emerging field of organocatalytic dearomatization.

Abstract:Oxidative reactions provide an effective strategy for phenol transformation via conjugated addition, C–H functionalization, and cyclization reactions, etc. It has enabled broad utilization of phenolic substrates as versatile chemical feedstocks in the preparation of various oxygenated molecules valuable for pharmaceutics, agricultural chemicals, materials, and so on. Many of the reactions are accomplished via quinone intermediates. Despite being versatile synthons in organic synthesis, quinones have to be generally pre-synthesized using stoichiometric metal oxides and stabilized by electron-withdrawing groups to circumvent dimerization or degradation. Their structural diversity, sustainable accessibility, and synthetic potentials remain to be further exploited. This review aims at giving an overview of oxidative phenol transformations in nature and synthetic chemistry, including reactions involving the generation of quinone intermediates, oxidative coupling for biaryl formation, and oxidative cyclization into various heterocycles. It offers clear, concise, and illustrated guidelines for synthetic chemists seeking inspiration through catalytic oxidative reactions.
