Abstract:Evidence from clinical trials and animal experiments has shown that estrogen has anti-atherosclerotic effects when administered to young women or experimental animals. The mechanisms involve the modulation of vascular inflammation, growth factor expression, and oxidative stress injured arteries. However, whether estrogen modulates the foam cell formation in plaque remains unknown. Here, we investigated the effects of 17β-estradiol (E2) on cholesterol efflux in vivo and in vitro. ApoE null mice underwent an ovariectomy at 5(th) week of age and then were treated with E2 or vehicle for the following 8 weeks. Compared with the vehicle-treated mice, the serum total cholesterol level, atherosclerotic plaque size, and lipid deposits were decreased and meanwhile ATP-binding cassette transporter A1 (ABCA1) expression in the plaque was increased in mice with E2 treatment. E2 also increased suppressor of cytokine signaling 3 (SOCS3) expression in the atherosclerotic plaques and in RAW264.7 cells. In vitro, E2 treatment reversed janus kinase/signal transducers and activators of transcription (JAK/STAT)-inhibited ABCA1 expression in RAW264.7 cells but had no effect on ABCA1 expression in SOCS3 knockdown cells. SOCS3 overexpression elevated ABCA1 expression through the inhibition of JAK2/STAT3 phosphorylation. Finally, we also found that E2 enhanced the cholesterol efflux to apoA I in RAW264.7 cells. In summary, E2 reduces atherosclerosis in ApoE null mice associated with upregulating ABCA1 expression and modulating the cholesterol efflux, which are dependent on SOCS3 upregulation. These results provide new insight into the athero-protective effects of estrogen.

Abstract:Previous studies indicated that estrogen could improve endothelial function. However, whether estrogen protects vascular complications of diabetes has yet to be clarified. The study was designed to investigate the action of 17ß-estradiol on vascular endothelium in streptozotocin (STZ)-induced diabetic rats. Ovariectomized female Sprague-Dawley rats were administered with streptozotocin to produce an ovariectomized-diabetic (OVS) model which manifested as dysfunction of aortic dilation and contraction ability. Meanwhile, OVS animals with 17ß-estradiol supplementation significantly improved aortic function. Accordingly, nitric oxide synthase-3 (NOS-3), Akt, PI3K and estrogen receptor α (ERα) protein expression in aorta declined in the OVS group. Such effects were partially restored by estrogen replacement. The presence of 17ß-estradiol similarly counteracted the reduction of cyclic guanosine monophosphate (cGMP), the enhanced expression of inducible NOS (NOS-2) and NO metabolites (nitrite and nitrate), as well as the increase of matrix metalloproteinase-9/tissue inhibitor of metalloproteinase-1 (MMP-9/TIMP-1), which is an index of arterial compliance. 17ß-estradiol could also decrease ROS production in vascular endothelium. In EA hy 926 cells we found that ER antagonist, wortmannin and Akt inhibitor could block improvement effects of 17ß-estradiol. These results strongly suggest that functional impairment of the ERα/NOS-3 signaling network in OVS animals was partially restored by 17ß-estradiol administration, which provides experimental support for estrogen recruitment to improve vascular outcomes in female diabetes after endogenous hormone depletion.
17 Beta-Estradiol Promotes Cholesterol Efflux From Vascular Smooth Muscle Cells Through A Liver X Receptor Alpha-Dependent Pathway

Abstract:Estrogen has pleiotropic effects on the cardiovascular diseases, yet the underlying mechanisms remain incompletely understood. Cholesterol efflux is a key mechanism through which to prevent foam cell formation and the development of atherosclerosis. Recent studies highlight the role of vascular smooth muscle cell (VSMC)-derived foam cells in atherogenesis. However, it remains unclear whether estrogen promotes cholesterol efflux from VSMCs and inhibits VSMC-derived foam cell formation. In the present study, we demonstrated that 17 beta-estradiol (E2) markedly enhanced cholesterol efflux to apolipoprotein (apo)A-1 and high-density lipoprotein (HDL) and attenuated oxidized low-density lipoprotein (ox-LDL) induced cholesteryl ester accumulation in VSMCs, which was associated with an increase in the expression of ATP-binding cassette transporters ABCA1 and ABCG1. The upregulation of ABCA1 and ABCG1 expression by E2 resulted from liver X receptor (LXR)alpha activation, which was confirmed by the prevention of the expression of ABCA1 and ABCG1 after inhibition of LXR alpha with a pharmacological inhibitor or small interfering RNA (siRNA). Furthermore, E2 increased LXR alpha, ABCA1 and ABCG1 expression in VSMCs via the estrogen receptor (ER), and the involvement of ER beta was confirmed by the use of selective ER alpha or ER beta antagonists (MPP and PHTPP) and agonists (PPT and DPN). These findings suggest that E2 promotes cholesterol efflux from VSMCs and reduces VSMC-derived foam cell formation via ER beta- and LXR alpha-dependent upregulation of ABCA1 and ABCG1 and provide novel insights into the anti-atherogenic properties of estrogen.
The effects of 17 alpha-estradiol to inhibit inflammation in vitro

Abstract:Background: 17 Alpha-estradiol (17 α-E2) is a natural, non-feminizing stereoisomer of 17 beta-estradiol (17 β-E2). Whereas much is known about the physiological effects of 17 β-E2, much less is known about 17 α-E2. For example, 17 β-E2 exerts anti-inflammatory effects in neurons and adipocytes through binding and activation of estrogen receptor alpha (ERα); however, if 17 α-E2 has similar effects on inflammation is currently unknown. Methods: To begin to address this, we analyzed the ability of 17 α-E2 and 17 β-E2 to suppress lipopolysaccharide (LPS)-induced inflammation in vitro using embryonic fibroblast cells (MEF) from wild type and total body ERα (ERKO) male and female mice. Additionally, we further probed if there were sex differences with respect to the effects of E2s using primary pre-adipocyte cells from C57BL/6J male and female mice. Also, we probed mechanistically the effects of E2s in fully differentiated 3T3-L1 cells. Results: Both E2s decreased LPS-induced markers of inflammation Tnf-α and Il-6, and increased the anti-inflammatory markers Il-4 and IL-6 receptor (Il-6ra) in MEF cells. To begin to understand the mechanisms by which both E2's mediate their anti-inflammatory effects, we probed the role of ERα using two methods. First, we used MEF cells from ERKO mice and found reductions in ERα diminished the ability of 17 α-E2 to suppress Tnf-α in female but not in male cells, demonstrating a sexual dimorphism in regard to the role of ERα to mediate 17 α-E2's effects. Second, we selectively reduced the expression of ERα in 3T3-L1 cells using siRNA and found reductions in ERα diminished the ability of both E2s to suppress Tnf-α and Il-6 expression. Lastly, to determine the mechanisms by which E2s reduce inflammation, we explored the role of NFκB-p65 and found both E2s decreased NFκB-p65 expression. Conclusions: In conclusion, we demonstrate for the first time that 17 α-E2, as well as 17 β-E2, suppresses inflammation through their effects on ERα and NFκB-p65.
17Β-Estradiol Suppresses Carboxylesterases by Activating C-Jun/ap-1 Pathway in Primary Human and Mouse Hepatocytes.

Abstract:In order to study the influence of estrogen on carboxylesterases, we investigated the effects of 17β-estradiol on CES1 (Ces1d) and CES2 (Ces1e) in human and mouse hepatocytes. After being treated with 17β-estradiol, the mRNA levels of CES1 and CES2 decreased by 29–39% and 28–55%, respectively, in the human hepatocytes from four donors. Consistently, the hydrolysis of para-nitrophenylacetate decreased markedly by 32% induced by 17β-estradiol. Moreover, 17β-estradiol decreased CES1 and CES2 by 45% and 47% respectively at protein levels. The response of altered expression of Ces1d (CES1) and Ces1e (CES2) to 17β-estradiolin in mouse hepatocytes was very similar to that in the human hepatocytes. Further, the decreased Ces1d and Ces1e expression induced by 17β-estradiol could be abolished by SP600125, an inhibitor of AP-1, both at mRNA and protein levels. Likewise, the increased c-Jun expression induced by 17β-estradiol could almost be abolished by SP600125. In vivo, the expression of Ces1d, Ces1e and the hydrolytic activity of liver were higher in the ovariectomized female mice(OVX) than those in control mice(SHAM). However, when 17β-estradiol was administrated, the expression of Ces1d, Ces1e and the hydrolytic activity of liver in the ovariectomized female mice (OVX+E2) became restored to their normal levels. Taken together, 17β-estradiol suppresses carboxylesterases by activating c-Jun/AP-1 pathway in primary human and mouse hepatocytes. The findings can offer the potential gains in the safety and efficacy of pharmacotherapy for women, especially for pregnant and menopausal women.

Abstract:Estrogen shows a vasoprotective role through inhibiting the proliferation and migration of vascular smooth muscle cells (VSMCs). The mechanism underlying the effect of estrogen, however, is not completely understood. Here, we explored the role of peroxisome proliferator-activated receptor-gamma (PPARγ) coactivator-1alpha (PGC-1α) in estrogen-mediated vasoprotection. Firstly, we showed that oleic acid (OA) decreased PGC-1α expression while stimulating VSMC proliferation and migration. In contrast, administration of VSMCs with 17β-estradiol (E2, 1 or 10nM) significantly restored OA-decreased PGC-1α expression, treatment with 10nM E2 almost completely abolished OA-induced VSMC proliferation and migration. Secondly, by using PGC-1α siRNA, the inhibitory effect of E2 on VSMC growth is strongly reduced via suppressing PGC-1α expression, indicating that E2 may exert its role through restoring PGC-1α. Finally, E2 (10nM) treatment inhibits OA-induced extracellular signal-regulated kinase 1/2 (ERK1/2) phosphorylation, however, suppression of PGC-1α expression abolishes this inhibitory effect of E2. Our findings demonstrate for the first time that in OA-stimulated rat VSMCs, treatment with E2 (1 or 10nM) diminishes VSMC proliferation and migration via restoring OA-decreased PGC-1α expression. This observation offers a novel molecular basis of the vasoprotective effect of estrogen.

Abstract:Introduction: 17β-Estradiol (E2) has been shown to attenuate the inflammatory response to vascular injury in young animals. Clinical trials that enrolled older women reported increased cardiovascular risk in women treated with menopausal hormones. We hypothesize that E2 effects on vascular inflammation are age-dependent. Methods and Results: Young (10 wk) and aged (52 wk) C57BL/6 intact female mice were used as source for primary cultures of bone marrow derived macrophages (BMMs). Real-time quantitative RT-PCR showed that pro-inflammatory mediators were expressed at low levels in vehicle treated cells, and that mRNA levels increased markedly at 4 h after C-reactive protein (CRP) treatment (Table). Abstract:Objectives Evidence from clinical trials and animal experiments has shown that oestrogen has anti-atherosclerotic effects that are independent of its cholesterol-lowering activity; these activities include reducing the proliferation of smooth muscle cells and leukocyte adhesion and increasing the synthesis of nitric oxide. However, whether oestrogen can regulate foam cell formation remains unknown. Here, we investigated the effects of 17β-oestradiol (E2) on cholesterol efflux in vivo and in vitro. Methods ApoE-null mice (B6.129P2-ApoEtm1Unc) underwent an ovariectomy (OVX) or Sham operation at 5 weeks of age and then were treated with E2 or vehicle for the following 8 weeks. After above treatment, serum and aorta of mice were harvested and then lipid level, plaque size and composition were detected. RAW267.4 cells were pretreated with or without interferon-gamma (IFN-gamma) for 12 h then treated with E2 for additional 12 h, then ATP-binding cassette transporter A1 (ABCA1), ATP-binding cassette transporter G1 (ABCG1) and CD36 expression and cholesterol efflux rate to apoA I and HDL were detected. Results Compared with the vehicle-treated mice, the serum total cholesterol level were decreased by 27.72% in the Sham group and 35.32% in the OVX+E2 group p<0.01 for both), and the serum triglyceride level was decreased by 22.64% in the Sham group and 23.84% in the OVX+E2 group p<0.05 and p < 0.01, respectively). Compared with the OVX group, the size of the plaque area was significantly reduced by 22.03% in the Sham group and by 21.84% in the OVX+E2 group p<0.01 for both). Oil Red O staining showed that the lipid deposits in the plaques decreased by 20.57% and 30.36%, respectively p<0.01 for both). The immunohistochemical staining showed the percentage of ABCA1-positive areas in the plaque were 1.61 and 1.57 times higher p<0.01 for both) in the Sham and OVX+E2 groups, respectively, than in the OVX group. According to above results, compared with the OVX group, the mRNA expression of ABCA1 in the aorta also increased by 2.09 and 2.43 times, respectively p<0.01 for both). In vitro, E2 treatment alone did not influence ABCA1 expression, but reversed the IFN-gamma inhibited ABCA1 expression of RAW 264.7 cells. Compared with the untreated cells, ABCA1 mRNA levels decreased by 35.29% in the presence of IFN-gamma and were reversed to 87.25% p<0.01, for both) by the E2 treatment, no change was found in ABCG1 or CD36 expression. Finally, we also found that E2 enhanced the cholesterol efflux to apoA I (28.66±1.76% for E2 vs 15.80±3.77% for control, p<0.01) in RAW264.7 cells, but not to HDL. Conclusions In summary, E2 increased ABCA1 expression and cholesterol efflux, both in vivo and in vitro. Thus, E2 delayed the atherosclerotic plaque progression in ApoE-null mice. These results provide new insight into the athero-protective effects of oestrogen.
Effect of estradiol on cholesterol metabolism in J774a.1 mouse mononuclear/macrophage cells]

Abstract:To explore the anti-atherosclerotic mechanism of estrogen and especially observe the effect of estradiol on the content of cholesterol in J774a.1 mouse mononuclear/macrophage-derived foam cells which were incubated with oxidized low-density lipoproteins (ox-LDL). J774a.1 mouse mononuclear/macrophages were incubated with ox-LDL or with both ox-LDL and estradiol (1, 0.1 or 0.01 micromol x L(-1)). Oil red O staining was used to observe the formation of foam cells, and cholesterol oxidase fluorometric was used to determine the content of cellular cholesterol content. Western blotting and RTFQ-PCR were used to observe the expressions of scavenger receptor class B type I (SR-B I ) in J774a.1 foam cells. Compared with the control cells, J774a.1 mouse mononuclear/macrophage-derived foam cells showed significantly increased contents of total cholesterol and cholesterol ester (P < 0.001) and decreased SR-B I mRNA expression (P < 0.01). Estradiol treatment significantly lowered the contents of total cholesterol and cholesterol ester (P < 0.05), and increased SR-B I protein and mRNA expression (P < 0.01) in the foam cells in a dose-dependent manner. Estradiol can inhibit the formation of mononuclear/macrophage-derived foam cells by decreasing the contents of total cholesterol and cholesterol ester and up-regulating the expression of SR-B I in the foam cells.

Abstract:Objectives Several epidemiological and experimental evidences indicate that oestrogen is intimately involved in the pathogenesis of atherosclerosis. The anti-proliferatory, pro-apoptotic, anti-migratory, anti-oxidative, and anti-inflammatory effects of oestrogen on vascular smooth muscle cells (VSMCs) have been demonstrated in vitro . However the role of oestrogen and its underlying mechanism in lipid accumulation and reverse cholesterol transport in VSMCs are poorly understood. The aim of the study was to investigated whether 17β-estradiol (E2) attenuates lipid accumulation in VSMCs via an increased cholesterol efflux as well as to determine the involved molecular mechanisms. Methods Mouse VSMCs were pretreated with indicated concentrations of E2 (10-9-10-7M) for 2h and then stimulated with oxLDL (50 μg/ml) for 72h. The intracellular lipid droplets were stained by oil red O staining and hematoxylin was used as a counterstain. Cellular free cholesterol and cholesteryl ester were quantified by an enzymatic method. The effect of E2 on cholesterol efflux was investigated using NBD-labelled cholesterol efflux. Real-time PCR and western blotting were used to detect the effect of E2 on ATP-binding cassette transporters (ABCA1), ABCG1 and liver X receptor (LXRα). Western blotting and NBD-labled cholesterol efflux were also used to examine the effects of oestrogen receptor (ER) and LXRα on E2-induced ABCA1 and ABCG1 expression and cholesterol efflux in VSMCs. Results E2 treatment dose-dependently reduced lipid droplets and celluar cholesteryl ester accumulation in VSMCs. E2 markedly enhanced apoA-1- and HDL-mediated cholesterol efflux from VSMCs in a dose-dependent manner, which was associated with an increase in ABCA1 and ABCG1 mRNA and protein expression. Nonselective ER inhibitor ICI-182,780 abolished the upregulation of ABCA1 and ABCG1 expression by E2. Remarkably, ERβ agonist DPN dose dependently upregulated ABCA1 and ABCG1 protein expression, whereas ERα agonist PPT had no effect, indicating upregulation of ABCA1 and ABCG1 by E2 is selectively mediated by ERβ. Both E2 and DPN promoted LXRα mRNA and protein expression in VMSCs. Moreover, inhibition of LXRα with a pharmacological inhibitor GGPP or siRNA suppressed the stimulatory effects of E2 on ABCA1 and ABCG1 expression and cholesterol efflux. Conclusions ERβ- and LXRα-dependent upregulation of ABCA1 and ABCG1 might mediate the atheroprotective effect of E2, which ameliorated the oxLDL-mediated lipid accumulation in VSMCs-derived foam cell formation.
β-estradiol adjusts intestinal function via ERβ and GPR30 mediated PI3K/AKT signaling activation to alleviate postmenopausal dyslipidemia

Abstract:Decreases in estrogen secretion and estrogen receptor function lead to an increase in the incidence of dyslipidemia and cardiovascular disease (CVD) in postmenopausal women. We previously reported that β-estradiol has a significant regulatory effect on lipids in ApoE -/- mice with bilateral ovariectomy. In the present study, we investigated how β-estradiol regulates intestinal function via estrogen receptors to alleviate postmenopausal dyslipidemia. Ovariectomized ApoE -/- mice were treated with β-estradiol for 90 days, and we found that β-estradiol reduced TC, TG, LDL-c, IL-1β and IL-18 levels in serum and decreased lipid accumulation in the liver. β-estradiol reduced injury and inflammation in the jejunum in ovariectomized mice, and promoted the expression of tight junction-related proteins. Moreover, β-estradiol increased ERα, ERβ, GPR30 and ABCG5 protein expression, and decreased the levels of NPC1L1 and SR-B1 in the jejunum of ovariectomized mice. In Caco-2 cells incubated with cholesterol, β-estradiol up-regulated PI3K/AKT signaling, reduced cholesterol accumulation, suppressed inflammatory signaling, and increased the expression of tight junction-related proteins. ERβ or GPR30 inhibition decreased the protective effect of β-estradiol on cholesterol accumulation, tight junctions, and inflammation in cholesterol incubated Caco-2 cells, while silencing both ERβ and GPR30 completely eliminated the protective effect of β-estradiol. PI3K/AKT inhibition abolished the protective effect of β-estradiol on cholesterol accumulation, tight junction-related protein expression, and inflammation, but had no influence on ERα, ERβ or GPR30 expression in cholesterol incubated Caco-2 cells. Our results provide evidence that β-estradiol regulates intestinal function via ERβ and GPR30 mediated PI3K/AKT signaling activation to alleviate postmenopausal dyslipidemia.

Abstract:AIM: To investigate the pathway via which 17 beta-estradiol (beta-Est) exerts suppressive effects on rat hepatic fibrosis.METHODS: In vivo study was done in CCl4-induced female hepatofibrotic rats. Fibrosis-suppressive effect of beta-Est (20 mu g/kg . d) was evaluated in intact and ovariectomized rat models. Six weeks after the treatment, all the rats were sacrificed and specimens of serum or liver tissue were collected for the studies. Serum liver enzymes, fibrosis markers and estradiol levels were determined by standard enzymatic methods, ELISA and RIA, respectively. Degrees of fibrosis and areas of hepatic stellate cells (HSCs) positive for alpha-smooth muscle actin (alpha-SMA) in the liver were determined by van Gieson (VG) stain and immunohistochemistry. In vitro studies, HSCs were isolated by a combination of pronase-collagenase perfusion and density gradient centrifugation. First-passage HSCs were randomly divided into 10 groups, and different concentrations of beta-Est, 2-hydroxyestradiol (2OHE) or 2-methoxyestradiol (2MeOE) were separately added to the cell groups. After incubation for 72 h, the degree of cell proliferation, collagen production, alpha-SMA or estrogen receptor (ER) expression was determined by MTT assay, ELISA and immunohistochemistry, respectively.RESULTS: beta-Est treatment reduced aspartate aminotransferase (AST), alanine aminotransferase (ALT), hyaluronic acid (HA) and type IV collagen (C IV) in sera, suppressed hepatic collagen content, decreased the areas of HSCs positive for alpha-SMA significantly in both intact and ovariectomized female hepatofibrotic rats. There was a negative correlation between the percentage of fibrotic area of liver tissue and the serum estradiol level; the calculated correlation coefficient was -0.57 (P<0.01). beta-Est and its metabolites concentration-dependently (10(-9) mol/L-10(-7) mol/L) inhibited HSC proliferation and collagen synthesis. At the concentration of 10(-7) mol/L, they could inhibit alpha-SMA expression. The order of potency was 2MeOE>2OHE>beta-Est.CONCLUSION: beta-Est may suppress hepatic fibrosis probably via its biologically active metabolites.

Abstract:Introduction: 17β-Estradiol (E2) offers cardiovascular protection in young female animals and peri-menopausal women. Clinical trials carried out in older women deprived of E2 for many years have shown harm or no cardiovascular benefit. We have recently demonstrated that E2 attenuates C-reactive protein (CRP)-driven inflammatory response in vascular smooth muscle cells and macrophages derived from young but not aged mice and reduces neointima formation in injured carotid arteries of young but not aged CRP transgenic mice (CRPtg). Hypothesis: After prolonged E2 deprivation, E2 will not attenuate neointimal response to vascular injury. Methods and Results: C57BL/6 non-transgenic (NTG) and CRPtg (10 wk old) mice underwent ovariectomy and then received vehicle (veh), uninterrupted E2 for 97 days (E2), or veh for 90 days followed by E2 for 7 days (E2, deprivation) delivered using subcutaneous pellets. Mice were then subjected to right common carotid artery ligation. At 28 days after injury, the carotid arteries were harvested, fixed, sectioned, stained, and subjected to computer-assisted morphometric analysis to assess the extent of the injury response. In veh treated mice, neointima formation was exaggerated in CRPtg compared to NTG (Figure). E2 treatment, whether continuous or after prolonged deprivation, was associated with greatly diminished neointima formation in both genotypes (2-way ANOVA interaction not significant). Importantly, the extent of neointima formation was not different between the two E2-treated groups (E2 vs. E2, deprivation). Conclusions: In mice, E2-induced vasoprotection abolishes CRP-mediated vascular injury and this effect is preserved even after prolonged E2 deprivation. In combination with our previous finding that the vasoprotective effects of E2 are age-dependent, the current data suggest that the lack of benefit associated with E2 seen in clinical trials is associated with advanced age rather than prolonged E2 deprivation.

Abstract:During their reproductive years, females have a lower risk for atherosclerosis as compared with age-matched males, although the mechanism behind this are not clearly understood. Cytokines, including TNF-alpha, play an important role in the pathogenesis of atherosclerosis. We therefore evaluated whether or not there was any difference between 17 beta -estradiol and testosterone in modulating TNF-alpha release from murine bone marrow-derived macrophages (BMM) in vitro. Cells were incubated with or without physiological concentrations (10(-10)-10(-8) M) of 17 beta -estradiol or testosterone for 48 h, followed by an additional 6 h in the absence or presence of lipopolysaccharide (LPS: 10 mug/ml). The amount of TNF-alpha released into the culture medium was determined with radioimmunoassay. We found that 17 beta -estradiol or testosterone alone did not affect TNF-alpha release from BMM as compared to untreated controls. Preincubation with 17 beta -estradiol significantly inhibited LPS-induced TNF-alpha release by 18.15% (P<0.05), 25.28% (p<0.05) and 40.93% (p<0.01) for 10(10), 10(9) and 10(8) M of 17<beta>-estradiol, respectively, as compared to LPS alone. In contrast, testosterone tested for 3 concentrations did not significantly effect TNF-alpha release induced by LPS. The results indicate that 17 beta -estradiol, but not testosterone, inhibits TNF-alpha release from LPS-stimulated macrophages, which may be one of the mechanisms by which estrogen protects against atherosclerosis. Abstract:Menopause is associated with dyslipidemia and an increased risk of cardiovascular disease, the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen. In this study, we find that estrogen mediates an atherosclerotic-protective action via estrogen receptor alpha/SREBP-1 signaling. Increased lipid accumulation and low-density lipoprotein (LDL)-uptake in HepG2 cells and THP-1 macrophages were induced by treatment of mixed hyperlipidemic serum from postmenopausal women; 17β-estradiol [estrogen (E2)] (10 nM) administration significantly improved hyperlipidemic profiles, relieved fatty-liver damage and attenuated the plaque area in the heart chamber of high-fat diet (HFD)-fed ovariectomized (OVX) Apo E –/ – mice. Expression of sterol regulatory element-binding protein (SREBP)-1 mRNA of circulating leukocytes in postmenopausal women was strongly correlated to the serum E2 level. Exploration of data from the Gene Expression Profiling Interactive Analysis (GEPIA) database revealed that expression of SREBP-1 protein correlated to expression of estrogen receptor (ESR)α protein in the liver, blood and in normal tissue. Genetic overexpression/inhibition of ESRα resulted in increased/decreased SREBP-1 expression as well as attenuated/deteriorated lipid deposition in vitro . An inhibitor of the protein kinase B/mammalian target of rapamycin (AKT/mTOR) pathway, AZD8055, abolished ESRα-induced SREBP-1 expression in HepG2 cells. Moreover, E2 and statin co-treatment significantly reduced lipid accumulation in vitro and hindered the progression of atherosclerosis and fatty-liver damage in OVX Apo E –/ – mice. Collectively, our results suggest that estrogen could exerted its atherosclerotic-protective action via ESRα/SREBP-1 signaling. E2 might enhance the cellular sensitivity of statins and could be used as a novel therapeutic strategy against atherosclerotic disorders in postmenopausal women.

Abstract:The risk of atherosclerosis (AS) ascends among post-menopausal women, while current hormone replacement therapy exerts several adverse effects. Alisol B 23-acetate (AB23A), a tetracyclic triterpenoid isolated from the rhizome of Alisma orientale, was reported to show multiple physiological activities, including regulating lipid metabolism. According to molecular docking analysis, it was predicted to bind with estrogen receptor α (ERα). In this study, we aimed to observe the effect of AB23A on preventing post-menopausal AS and explore whether the mechanism was mediated by ERα. In vitro, free fatty acid (FFA) was applied to induce the abnormal lipid metabolism of L02 cells. In vivo, the ApoE−/− mice were ovariectomized to mimic the cessation of estrogen. The high-fat diet was also given to induce post-menopausal AS. We demonstrated AB23A attenuated the accumulation of total cholesterol and triglyceride induced by free fatty acids in hepatocytes. In high-fat diet-ovariectomy-treated ApoE−/− mice, AB23A eliminated lipids in blood and liver. AB23A not only reduced the synthesis of proprotein convertase subtilisin/kexin type 9 (PCSK9) through sterol-regulatory element binding proteins (SREBPs) but also suppressed the secretion of PCSK9 through silent information regulator 1 (SIRT1). Notably, AB23A promoted the expression of ERα in vivo and in vitro. The both ERα inhibitor and ERα siRNA were also applied in confirming whether the hepatic protective effect of AB23A was mediated by ERα. We found that AB23A significantly promoted the expression of ERα. AB23A could inhibit the synthesis and secretion of PCSK9 through ERα, lower the accumulation of triglyceride and cholesterol, and prevent post-menopausal AS.

Abstract:OBJECTIVES: Cardiovascular disease is rare in premenopausal women, but increases after the menopause when hormone replacement therapy reduces coronary events. Vascular smooth muscle cell (SMC) proliferation and migration occur in atherosclerosis, restenosis and venous graft disease. We studied the effects of 17 beta-estradiol on SMC proliferation and migration.METHODS: SMC were cultured from saphenous veins of postmenopausal women and age-matched men. Cell growth was determined by 3H-thymidine incorporation and cell counting. Migration of SMC was assessed in 4-well chambers. SMC were seeded in one corner and PDGF-BB in filter paper glued onto the opposite wall.RESULTS: PDGF-BB (5 ng/ml for 24 h) similarly stimulated 3H-thymidine incorporation in female (511 +/- 57%; n = 8) and male (528 +/- 62%; n = 12) SMC. This was reduced by 17 beta-estradiol (10(-8)-10(-6) M; female 313 +/- 52%; male 337 +/- 54%; P < 0.05). PDGF-BB increased the number of SMC (P < 0.0001 at 10 days) obtained from females (153 +/- 3%; n = 5) and males (150 +/- 4%; n = 5), which was inhibited by 17 beta-estradiol (10(-6) M; female 134 +/- 7%; male 128 +/- 5%; P < 0.05). Similar results were obtained with basic fibroblast growth factor. In contrast to 17 beta-estradiol, another steroid (dexamethasone) had no effects on 3H-thymidine incorporation in these cells stimulated with PDGF-BB, PDGF-BB (0.01-1 ng) stimulated SMC migration (P < 0.05) which was inhibited by 17 beta-estradiol (10(-10)-10(-6) M; n = 5; P < 0.005).CONCLUSION: 17 beta-Estradiol inhibits growth-factor-induced SMC proliferation and migration regardless of gender. These effects of 17 beta-estradiol may contribute to its cardiovascular protective properties in postmenopausal women during replacement therapy.

Abstract:OBJECTIVE:To determine the protective effect of 17 beta-estradiol on endothelial cells under shear stress.METHODS:cDNA microarray technology was adopted in the analysis of gene expression of cultured human umbilical vein endothelial cells (HUVECs) that had been incubated by 17 beta-estradiol for 48 h and then exposed to low fluid shear stress (4.20 dyne/cm2) for 2 h, Normal static cultured HUVECs were selected as control. Total RNA from normal static cultured HUVECs was labeled by Cy3-dCTP, and the total RNA of HUVECs incubated by 17 beta-estradiol (10(-7) mol/L) and then exposed to low shear stress was labeled by Cy5-dCTP. The expression ratios reported are the average from the two separate experiments.RESULTS:A total of 384 genes (approximately 0.094%) exhibited differential expression. 226 genes were up-regulated and 158 genes were down-regulated. The transcription protein of up-regulated genes related to synthesis of PGI2, macrophage phagocytosis, and so on. The transcription protein of down-regulated genes related to degradation of fibrinogen, and to production of protooncogene.CONCLUSION:Estrin at the physiological concentration could protect impaired endothelial cells. Its protective effect may be related with some genes.

Abstract:To investigate the effects of estrogen treatment on aortic endothelial senescence and atherosclerosis, an ovariectomized female rabbit model was constructed, and human umbilical vein endothelial cells were utilized to explore the potential mechanisms. Twenty-eight female rabbits were randomized into 4 groups (7 each): sham operation, ovariectomized, ovariectomized plus low-dose estradiol treatment, and ovariectomized plus high-dose estradiol treatment. All rabbits were fed on high-cholesterol diet for 12 weeks. Blood samples were collected to determine the serum estradiol, asymmetric dimethyl L-arginine (ADMA), and lipid levels, and the aortas were separated for histopathologic analysis. After ovariectomy and high-fat diet, the concentration of serum estradiols declined significantly (P < 0.01) and the levels of ADMA and serum lipids increased (all P < 0.01) as the area of senescent endothelium and atherosclerotic lesions enlarged (both P < 0.01). However, administration of estradiols reduced the levels of ADMA, total cholesterol, and low-density lipoprotein (LDL) cholesterol and inhibited endothelial senescence and atherosclerosis (all P < 0.01). Simultaneously, the concentration of high-density lipoprotein cholesterol and triglyceride increased (all P < 0.01). In vitro experiments also confirmed that estradiols could decrease the ADMA levels induced by oxidized LDL and inhibited oxidized LDL-induced and ADMA-induced human umbilical vein endothelial cell senescence. These results indicate that estrogens can inhibit endothelial senescence and atherosclerosis with reduced ADMA levels and improved lipid profile.

Abstract:Physiological gender differences in immune capabilities are now well recognized and Suggest that sex steroid hormones such as estrogens may be involved in the regulation of the immunocompetence. In this paper. CID11c-positive murine spleen dendritic cells (SDCs) were treated with various concentrations of 17 beta-estradiol (E2) for 24 h. The viability, phenotype, nuclear factor kappa B p65 (NF-kappa Bp65), endocytosis, stimulatory capacity and cytokine expression were analyzed. Our results showed that E2 increased the viability and MHC-II expression but decreased nuclear NF-kappa Bp65 level and endocytosis of SDCs. E2 also increased the stimulatory capacity of SDCs from low-dose group but decreased it from middle- and high-dose ones. In addition, E2 increased the intracellular expression of IL-6 and IL-10 in SDCs, but no obvious change appeared in IL-12 and TNF-alpha. These data suggested that E2 might influence the immune responses by changing the viability, maturation, NF-kappa Bp65, endocytosis, stimulatory capacity and cytokine expression of SDCs.
