Abstract:<p>Decreases in estrogen secretion and estrogen receptor function lead to an increase in the incidence of dyslipidemia and cardiovascular disease (CVD) in postmenopausal women. We previously reported that β-estradiol has a significant regulatory effect on lipids in ApoE -/- mice with bilateral ovariectomy. In the present study, we investigated how β-estradiol regulates intestinal function via estrogen receptors to alleviate postmenopausal dyslipidemia. Ovariectomized ApoE -/- mice were treated with β-estradiol for 90 days, and we found that β-estradiol reduced TC, TG, LDL-c, IL-1β and IL-18 levels in serum and decreased lipid accumulation in the liver. β-estradiol reduced injury and inflammation in the jejunum in ovariectomized mice, and promoted the expression of tight junction-related proteins. Moreover, β-estradiol increased ERα, ERβ, GPR30 and ABCG5 protein expression, and decreased the levels of NPC1L1 and SR-B1 in the jejunum of ovariectomized mice. In Caco-2 cells incubated with cholesterol, β-estradiol up-regulated PI3K/AKT signaling, reduced cholesterol accumulation, suppressed inflammatory signaling, and increased the expression of tight junction-related proteins. ERβ or GPR30 inhibition decreased the protective effect of β-estradiol on cholesterol accumulation, tight junctions, and inflammation in cholesterol incubated Caco-2 cells, while silencing both ERβ and GPR30 completely eliminated the protective effect of β-estradiol. PI3K/AKT inhibition abolished the protective effect of β-estradiol on cholesterol accumulation, tight junction-related protein expression, and inflammation, but had no influence on ERα, ERβ or GPR30 expression in cholesterol incubated Caco-2 cells. Our results provide evidence that β-estradiol regulates intestinal function via ERβ and GPR30 mediated PI3K/AKT signaling activation to alleviate postmenopausal dyslipidemia.</p>

What problem does this paper attempt to address?
This paper aims to explore how β - estradiol regulates intestinal function by activating the ERβ and GPR30 - mediated PI3K/AKT signaling pathway, thereby alleviating dyslipidemia in post - menopausal women. Specifically, the research focuses on the following aspects: 1. **The effects of β - estradiol on blood lipids and inflammation**: - The study used bilateral ovariectomized ApoE - / - mice as a post - menopausal model and found that the levels of serum total cholesterol (TC), triglyceride (TG), and low - density lipoprotein cholesterol (LDL - c) in these mice were significantly increased, accompanied by an increase in the levels of serum interleukin - 1β (IL - 1β) and interleukin - 18 (IL - 18). - β - estradiol supplementation treatment could significantly reduce the levels of the above - mentioned blood lipid indicators and inflammatory factors, and reduce fat deposition and damage in the liver. 2. **The effects of β - estradiol on intestinal structure and function**: - Ovariectomy led to changes in the intestinal structure of mice, such as shortening of jejunum villus length and deepening of crypts. β - estradiol supplementation treatment could reverse these changes and improve the intestinal structure. - β - estradiol also protected the tight junctions in the jejunum and increased the expression of tight - junction - related proteins (such as Occludin, ZO - 1, and Claudin - 1). - The study also found that ovariectomy led to an increase in the expression of NF - κB p65 in the jejunum of mice, suggesting an intensified intestinal inflammatory response, and β - estradiol supplementation treatment could inhibit this process. 3. **The effects of β - estradiol on the expression of intestinal estrogen receptors**: - Through immunofluorescence and Western blot detection, the study found that ovariectomy led to a decrease in the expression of estrogen receptor α (ERα), estrogen receptor β (ERβ), and G - protein - coupled receptor 30 (GPR30) in the jejunum of mice, and β - estradiol supplementation treatment could significantly increase the expression of these receptors. - The serum estradiol level decreased significantly after ovariectomy, and β - estradiol supplementation treatment could restore this level. 4. **The role of the PI3K/AKT signaling pathway in the protective effect of β - estradiol**: - The study found that ovariectomy led to a decrease in the expression of PI3K and p - AKT in the jejunum of mice, and β - estradiol supplementation treatment could increase the expression of these proteins. - β - estradiol supplementation treatment also inhibited the expression of NPC1L1 and SR - B1 in the jejunum and increased the expression of ABCG5, further supporting its protective effect on intestinal structure and function. 5. **The preventive effect of β - estradiol on cholesterol - induced dysfunction in Caco - 2 cells**: - In vitro experiments, Caco - 2 cells showed lipid accumulation and dysfunction under the action of 100 μM cholesterol, and 10 nM β - estradiol intervention could significantly reduce cholesterol accumulation, inhibit the inflammatory response, and increase the expression of tight - junction - related proteins. In conclusion, through in vivo and in vitro experiments, this paper systematically explores the mechanism by which β - estradiol regulates intestinal function by activating the ERβ and GPR30 - mediated PI3K/AKT signaling pathway, thereby alleviating dyslipidemia in post - menopausal women.
