Abstract:Cascade radical cyclization of <i>N</i>-arylacrylamides is an attractive method to prepare 3,3-disubstituted oxindoles. As the reported methods often require additives and/or photocatalysts, we herein report an additive- and photocatalyst-free deaminative strategy for their synthesis under mild conditions, enabled by photoactivation of an electron donor–acceptor (EDA) complex. Abstract:The electron-withdrawing groups (EWGs) in the electrophilic alkenes employed in the Michael addition reaction are almost only CO2R, CN, COR, NO2, and SO2Ph. Although amides (CONR1R2) are also typical electron-withdrawing groups and are of great importance in organic synthesis, they are scarcely employed as the EWGs of the electrophilic alkenes in the Michael addition reaction. In this work, the Michael reactions of acrylamide and its derivatives with cyclanones were successfully carried out in the presence of enough radical inhibitors. The amide groups play a key role in producing the preferred products. The N-substituted acrylamides, including N-monosubstituted and N,N-disubstituted acrylamides could react with cyclohexanone (CHn) to give the expected 2-carbamoylethyl derivatives; however, acrylamide reacting with cyclohexanone only produced ene-lactam. Cyclanones also have effects on the products, while the ring size of cyclanones influences the reaction yield and the α-substituent decides the ratio of resulting isomeric ene-lactams.

Abstract:A new alkoxycarbonyl-radical-triggered cascade cyclization reaction of N-(o-cyanobiaryl)acrylamides with alkyloxalyl chlorides has been developed. With this method, a broad range of ester-containing phenanthridines are prepared through photoredox-catalyzed alkoxycarbonyl radical addition/nitrile insertion/cyclization with N-(o-cyanobiaryl)acrylamides under mild conditions. This strategy features a wide substrate scope, metal free conditions, and operational simplicity.

Abstract:An efficient silver-catalyzed oxidative cyclization of acrylamides with 1,3-dicarbonyl compounds is described. It proceeds through a tandem radical addition/cyclization process, in which two new C-C bonds were formed. Abstract:Benzoquinone and 9-phenylanthracene barely undergo anticipated cycloaddition under acid catalysis. Instead, 9-anthracenyl aryl ethers are obtained as unexpected products. Mechanistic studies indicate that the reaction likely undergoes an ionic mechanism between protonated anthracene species and nucleophilic oxygen of 1,4-benzoquinone or 1,4-hydroquinone. A variety of 9-anthracenyl aryl ethers are constructed with this method. Produced anthracenyl aryl ethers are potential scaffolds for new fluorescent molecules.

Abstract:A visible-light-mediated radical cascade cyclization of N-(o-cyanobiaryl)acrylamides with oxime esters for the assembly of acyl-containing pyrido[4,3,2-gh]phenanthridines has been developed. The present protocol tolerates a wide range of oxime esters through a single reaction via fragmentation, radical addition, nitrile insertion, and cyclization under mild conditions. Radical cascade reactions represent a fundamental synthetic method to forge chemical bonds and structurally intricate organic molecules in an efficient and straightforward manner from readily available substances.1 Among these, 2-cyano-3-arylaniline derived acrylamides are appealing building blocks in radical cascade cyclization reactions due to their diverse levels of reactivity for the one-pot production of polycyclic architectures. In this context, a series of oxidative promoted radical cascade cyclization of 2-cyano-3-arylaniline derived acrylamides have been established for the synthesis phenanthridine derivatives.2 However, these methods required unfriendly strong oxidants as radical initiators and high temperatures, thereby seriously restricting their applications in synthesis. Meanwhile, photocatalytic radical reactions have been regarded as a better alternative protocol in organic synthesis owning to their efficiency and environmental friendliness.3 A few examples of radical cyclization of 2-cyano-3-arylaniline derived acrylamides enabled by photocatalysis have been documented.4 To extend the applications of this strategy in organic synthesis, handling acrylamide derivatives to execute visible-light-induced cascade cyclization protocols under mild conditions is still highly desirable. Oxime esters have been received numerous attentions in synthetic community because of their unique single-electron redox activities.5-7 Typically, a carbon-centered radical could be generated from oxime esters by a single electron transfer under the photocatalytic conditions to produce highly reactive iminyl radicals, followed by β-C–C bond cleavage. As such, acyl oxime esters are regarded as effective acyl radical precursors for acyl-containing molecules synthesis.7 For example, Wu disclosed an elegant radical cascade reaction between acyl oxime esters and acrylamides in 2019.7a Subsequently, the same group reported a multi-component difunctionalization of styrenes with acyl oxime esters, which provide an efficient synthetic method to produce β-carbonyl imides.7b Inspired by these findings, we envisioned that the possibility of developing a radical cascade cyclization of 2-cyano-3-arylaniline derived acrylamides with acyl oxime esters for the polyheterocycles synthesis. Herein, we reported a photocatalytic radical cascade reaction of 2-cyano-3-arylaniline derived acrylamides with acyl oxime esters to generate acylated phenanthridine derivatives through a tandem acyl radical addition/nitrile insertion/cyclization process (Scheme 1).

Abstract:An unprecedented tandem radical sulfenylation/cyclization reaction of N-arylacrylamides with sulfonyl hydrazides has been developed in the presence of iodine for the selective synthesis of 3-(sulfenylmethyl)oxindoles and 3-sulfenyl-3,4-dihydroquinolin-2(1H)-ones. Preliminary mechanistic studies showed that sulfonyl hydrazides decomposed completely at an early stage to thiosulfonates and disulfides, both of which underwent tandem radical sulfenylation/cyclization with N-arylacrylamides at a late stage.

Abstract:Abstract A Rh-catalyzed electrochemical vinylic C–H annulation of acrylamides with alkynes has been developed in an undivided cell, affording cyclic products in good to excellent yield. Divergent syntheses of α-pyridones and cyclic imidates are accomplished by employing N-phenyl acrylamides and N-tosyl acrylamides as substrates, respectively. Additionally, excellent regioselectivities are achieved when using unsymmetrical alkynes. This electrochemical process is environmentally benign compared to traditional transition metal-catalyzed C–H annulations because it avoids the use of stoichiometric metal oxidants. DFT calculations elucidated the reaction mechanism and origins of substituent-controlled chemoselectivity. The sequential C–H activation and alkyne insertion under rhodium catalysis leads to the seven-membered ring vinyl-rhodium intermediate. This intermediate undergoes either the classic neutral concerted reductive elimination to produce α-pyridones, or the ionic stepwise pathway to produce cyclic imidates.

Abstract:Decarbonylative and dehydrogenative cascade annulation of aldehydes with N‐(o‐cyanobiaryl)acrylamides was developed for the synthesis of a broad spectrum of alkyl‐ and carbonyl‐substituted pyrido[4,3,2‐gh]phenanthridines. Secondary and tertiary aldehydes can undergo the decarbonylative cascade under DTBP/malonic acid conditions, while aryl, primary, and secondary aldehydes can undergo dehydrogenative reaction under TBHP system. Such transformations were triggered by alkyl and acyl radicals, and the annulations were accomplished through intramolecular additions of carbon‐centered radical to nitrile group and iminyl radical to aromatic ring.

Abstract:An unprecedented use of readily accessible aliphatic acylhydrazides as alkylating agents has been demonstrated in the copper -catalyzed tandem alkylation/cyclization of N-arylacrylamides. A range of aliphatic acylhydrazides smoothly participated in the oxidative 5-exo- trig cyclization of N-arylacrylamides with CuCO3 as the catalyst and DTBP as the oxidant, delivering structurally diverse 3,3-dialkyloxindoles in moderate to good yields. This study paves the way for the synthetic applications of aliphatic acylhydrazides as alkyl radical precursors.

Abstract:Herein, we introduce an electrochemical dehydrogenative [3 + 2]/[5 + 2] annulation of easily available N-arylacrylamides with gamma,sigma-unsaturated malonates through C(sp(3))-H/C(sp(2))-H functionalization. The employment of inexpensive ferrocene as the redox catalyst allows access to diverse benzo[b]azepin-2-ones in moderate to excellent yields without stoichiometric oxidants. This protocol features broad substrate scope and excellent selectivity, and mechanistic studies indicated that the reaction proceeded through the oxidation of a C(sp(3))-H bond to generate an alkyl radical, radical addition across the C=C bond, [3 + 2]/[5 + 2] annulations, and C(sp(2))-H functionalization cascades.

Abstract:Radical cascade difluoroacetamidation of N-(arylsulfonyl)acrylamides with α,α-difluoro-α-(TMS)-acetamides has been achieved for the first time. This CuBr-mediated transformation is easy to perform, generates high yields and shows a good substrate scope. This cascade reaction proceeds via difluoroacetamidation, aryl migration and desulfonylation. The products can be readily transformed into synthetically useful compounds such as difluorofunctionalized esters and alcohols in excellent yields.
