Abstract:Three new triterpenoid saponins, named julibroside J(16) (2), julibroside J(17) (3), and julibroside J(21) (4), each of which possesses an oleanane triterpenoid aglycone of acacic acid, two monoterpenoid acids, and nine sugar moieties, together with one known saponin, julibroside II (1), were isolated from the stem bark of Albizia julibrissin by chromatographic methods. Their structures were established by chemical and spectroscopic means. Saponins 1, 2, and 4 showed inhibitory activities against Bel 7402 human cancer cell line in vitro.

Abstract:Three new triterpenoid saponins, julibrosides A5-A7 (1-3), together with five known saponins (4-8), were isolated from the stem bark of Albizia julibrissin. Their structures were elucidated on the basis of extensive spectroscopic data analysis of MS, 1D and 2D NMR, and chemical methods. Compounds 7 and 8 were isolated from the genus Albizia for the first time. The new compounds showed no cytotoxicity and anti-inflammatory activity.

Abstract:Two undescribed oleanane-type saponins, julibrosides K-L, along with three undescribed oleanane-type prosapogenins, julibrosides M-O, were isolated from the stem bark of Albizia julibrissin Durazz. and the mild alkaline hydrolysate of the total saponin, respectively. Their structures were established by extensive analysis of 1D and 2D NMR experiments (COSY, TOCSY, HSQC, HMBC, and HSQC-TOCSY) and mass spectrometry. Furthermore, the cytotoxic activities of the isolated compounds against BGC-823, A549, HCT-116, and HepG2 cell lines were evaluated, and julibroside L showed significant cytotoxic activities against the four cancer cell lines with IC50 values of 5.77, 4.80, 4.26, and 4.93 μM, respectively.

Abstract:Bioassay-guided fractionation of the ethanolic extract of the stem bark of Albizia julibrissin led to the isolation of ten new oleanane-type triterpenoid saponins, julibrosides J37-J46 (1-10), along with six known analogues (11-16). In addition, 11 prosapogenins (17-27) were prepared by mild or strong alkaline hydrolysis of the total saponin. The structures of 1-27 were determined by spectroscopic and chemical means, and their cytotoxicities against four human cancer cell lines, BGC-823, A549, HCT-116, and HepG2 were evaluated. Compounds 5-16 exhibited significant inhibitory activity with IC50 values ranging from 2.59 to 9.30μM, and 8 turned out to be the most active compound with all IC50 values <5μM. A preliminary structure-activity relationship of these saponins clearly indicated that the outer monoterpenoid moiety (MT') is a crucial substituent for cytotoxicity, and the linkage sites of the MT' unit greatly influenced the activity. It could also be inferred that the existence of 16-OH of the aglycone almost has no effect on cytotoxicity and the N-acetyl-glucosamine moiety at C-3 seems to enhance activity.

Julibrosides J(25) and J(22) showed marked activities against four kinds of cancer cell lines in a concentration of 100 mu g/mL assayed by SRB method.

[Anti-tumor target identification and molecular mechanism study of total saponins from Albizia julibrissin]

Abstract:Dried stem bark from Albizia julibrissin(AJ) is a common traditional Chinese herb with several therapy effects including insomnia, anxiety and anti-tumor. Recently, the anti-tumor effect and mechanism studies of AJ have drawn much attention; however, there are still some troubles in chemical composition separation, which leads to the difficulties in pharmacological research of AJ. In this study, we firstly confirmed the proliferation inhibitory effect of total saponins from AJ(TSAJ)on human hepatocarcinoma(HepG2) cells, and also tested the apoptosis induction effect of TSAJ. Then, we successfully captured the potential target proteins from HepG2 lysates by using TSAJ-modified solid beads, and identified the target proteins by LC-MS/MS. Finally, we confirmed 5 target proteins including Exportin-2, Beta-actin-like protein 2, Myosin-9, Protein transport protein Sec61 subunit beta,and Cytochrome c oxidase copper chaperone, which are responsible forcell apoptosis, proliferation, differentiation andmigration. In summary, our findings elucidate the potential anti-tumor mechanism of TSAJ from the direct target proteins, and provide a new insight for exploring the pharmacological mechanism of traditional Chinese medicine.
Triterpenoid Saponins from Cortex Albiziae

Abstract:Cortex Albiziae, the dried stem bark of a leguminous plant, Albizia julibrissin Durazz, was specified in Chinese Pharmacopoeia (1995 edit.) as a traditional Chinese medicine to be used to relieve melancholia and uneasiness of body and mind, to invigorate the circulation of blood and subside a swelling. In a course of our quality assessment of traditional Chinese medicines, the n-BuOH soluble part of 95% EtOH extracts from the stem barks of Albizia julibrissin was subjected to a series of solvent treatment and chromatographic separations, including Rp-HPLC methods, giving rise to 28 pentacyclic triterpenoid saponins. Based on chemical methods and spectroscopic evidences, the structures of saponins were identified, which included six pairs of diastereoisomers, five pairs of position isomers, and 26 of them were new compounds. All the saponins are trisdesmosidic saponins, which are composed of an acacic acid moiety, 7-9 monosaccharide moieties, 1-2 monoterpenic acid moieties, and exhibited their molecular ion peaks around m/z 2 000. Their carbon-13 signals and most of proton signals were assigned based on 1D and 2D NMR experiments. The proton and carbon signals of several known saponins were revised and reassigned. The spectroscopic properties related to these saponins were analyzed and summarized. The cytotoxic activity and other activities of these saponins and their analogues were discussed also.

Abstract:OBJECTIVE:To separate chemical and active constituents from the stem bark of Albiza julibrissin.METHODS:Chromatography and spectral methods were used.RESULTS:3-O-beta-D-xylopyranosyl-(1-->2)-alpha-L-arabinopyranosyl-(1-->6)-beta-D-glucopyranosyl-21-O-[(6S)-2-trans-2-hydroxymethyl-6-methyl-6-O-[3-O-(6S)-2-trans-2-hydroxymethyl-6-methyl-6-hydroxy-2,7-octadienoyl)-beta-D-xylopyranosyl]-2,7-octadienoyl]-acacic acid-28-O-beta-D-glucopyranosyl-(1-->3)-[alpha-L-arabinofuranosyl-(1-->4)]-alpha-L-rhamnopyranosyl-(1-->2)-beta-D-glucopyranosyl ester (1) was separated and identified.CONCLUSION:Compound 1 is a new saponin named as Julibroside J(24).

Abstract:Twenty-six naturally occurring oleanolic acid saponins and their derivatives, 16 of which were synthesized in this study, were preliminarily evaluated against human cancer cells. From SAR studies, the presence of alpha-L-rhamnosyl residue at the terminal of both C-3 and C-28 position for oleanolic acid bidesmosides was important to enhance cytotoxicity, and introducing more sugar residues at C-3-OH of compound 12 with C-28 carboxylic acid is a favorable modification to ameliorate the anticancer activity. Furthermore, alpha-L-rhamnosyl moiety linked to C-2-OH of the first monosaccharide (alpha-L-alabinose, beta-D-xylose, beta-n-galactose or beta-D-glucose) in C-3-OH of oleanolic acid was helpful to improve the cytotoxicity. According to the predicted log P values, lipophilicity of the synthesized saponins was not an important factor for cytotoxicity. (C) 2013 Elsevier Masson SAS. All rights reserved.

Abstract:Three new minor oleanane triterpenoid saponins, cylindrosides B (1), C (2), and D (3), were isolated from the seed of Cylindrokelupha dalatensis using chromatographic method. Their structures were established on the basis of the chemical and spectroscopic evidences. They displayed significant antitumor activity in vitro against HL60 cancer cell lines and IC50 values were 7.15 ± 0.63, 10.07 ± 0.97, and 4.74 ± 0.57 μM, respectively, by MTT method.
