Abstract:Reported is the palladium-catalyzed dehydrogenative annulation of indole-2- and 3-carboxamides with symmetrical alkynes leading respectively to β- and γ-carbolinones. The reaction is carried out using Pd(OAc)2 as a catalyst and TBAB as a base at 50 ˚C under air atmosphere for β-carbolinones, whereas it requires a 100 ˚C heating under O2 atmosphere for the γ-ones. Free (NH)-indoles do not undergo this reaction. Both primary and a variety of alkyl-substituted amides were successfully coupled with symmetrical alkyl and aryl acetylenes. When unsymmetrical alkynes were used, better regioselectivities (79-89%) were obtained for the β-carbolinones than for the γ-carbolinones (57-67%). A mechanism was suggested without evidence.

This protocol provides a facile and straightforward approach to access diverse difluoromethylated carbinols in good to excellent yields under mild conditions. Abstract:Ethyl and phenyl oxanilates were readily prepared from N-(2-bromobenzyl)anilines and oxalyl chloride monoethyl and monophenyl esters, respectively. It was found that the ethyl oxanilate survived in the presence of K2CO3 in DMA at 120 degrees C and underwent an intramolecular direct arylation using Pd(OAc)(2)-dppf, furnishing the 5,6-dihydrophenanthridine derivative. In contrast, the corresponding phenyl oxanilates decomposed upon exposure to K2CO3 in DMA at 120 degrees C and were transformed into N-arylisoindolin-1-ones via Pd(OAc)(2)-dppf-catalyzed intramolecular decarbonylative coupling. Except for the 4-methoxy-substituted oxanilic acid phenyl ester, other phenyl oxanilates possessing electron-withdrawing (NO2, Cl) and weak electron-donating (Me) substituents provided the N-arylisoindolin-1-ones in 43-80% yields.

Abstract:A general synthesis of 4-hydroxylcarbazoles by domino vinylogous conjugate addition/cyclization/elimination/aromatization of easily prepared 3-nitroindoles with alkylidene azlactones under mild and transition-metal-free conditions has been developed. This method was also applicable to other nitrosubstituted benzofused heterocycles such as 3-nitrobenzothiophene, 2-nitrobenzothiophene, and 2-nitrobenzofuran. The valuable tetracyclic carbazole derivatives, such as 6H-oxazolo[4,5-c]carbazole and 3,6-dihydro-2H-oxazolo[4,5-c]carbazol-2-one, were readily prepared from the product, demonstrating synthetic utility of this method.

Abstract:Palladium-catalyzed [4+2] annulation of aryl and alkenyl carboxamides with 1,3-dienes via C–H functionalization is developed using air as the terminal oxidant. The method demonstrates good functional group tolerance and high stereoselectivity, affording a series of 3,4-dihydroisoquinolones and 5,6-dihydropyridinones in yields of up to 99%.

Abstract:The condensation of 3/2‐carbonyl‐2/3‐vinylindoles with phenyl hydrazine in the presence of diphenyl phosphate followed by thermal electrocyclization and elimination of aniline afforded γ, β‐carbolines. A facile synthesis of 3‐alkyl‐2‐aryl substituted γ‐carboline derivatives is reported involving diphenyl phosphate catalyzed condensation of 3‐(acetyl/propionyl)‐2‐(nitroaryl)vinyl‐1‐phenylsulfonylindoles with phenyl hydrazine followed by concurrent electrocyclization and aromatization through elimination of aniline. This strategy could be successfully explored with 1‐phenylsulfonylindole‐3/2‐carboxaldehydes containing vinyl unit at 2/3‐position to afford the respective γ‐and β‐carboline derivatives. The domino reaction of easily accessible 2/3‐carbonyl indoles containing a vinyl unit at the adjacent positions with phenyl hydrazine as an amine source afforded a wide variety of carboline derivatives in good to acceptable yields. The synthetic utility of a representative γ‐carboline could be demonstrated via transformation into the corresponding indolo[a]carboline framework.

Abstract:A rapid, microwave-assisted synthesis of β-carbolines via a successive condensation/cyclization/dehydrogenation approach is described. This methodology involves the coupling of various tryptamines with aromatic aldehydes/glyoxals. The product imine undergoes a Pictet–Spengler cyclization followed by a final dehydrogenation to yield β-carbolines in a three-step domino reaction. The use of the bifunctional catalyst Pd/C/K-10 combined with microwave irradiation enabled the synthesis of β-carbolines in short reaction times and in good to excellent yields.

Abstract:In this work, carbazoles, one of the most important types of nitrogenous compounds, were used as substrates to synthesize dicarbazolyl(aryl)methanols in moderate to excellent yields (up to 95%). This simple and efficient strategy can regioselectively form an sp 3 quaternary carbon with two large sterically hindered carbazole rings, one benzene ring, and an active hydroxy group in a one-pot reaction. The resulting triarylmethanes are all new products, and have high potential as building blocks for medicinal chemistry or materials science. Abstract:In this study, we present our findings on the synthesis of α-, β-, γ-carbolines via PIFA/BF3·OEt2-mediated intramolecular cyclization of isomeric azidomethyl(indolyl)acrylates. Alternately, 1,5,7-triazabicyclo[4.4.0]dec-5-ene/1,8-diazabicyclo(5.4.0)undec-7-ene (TBD/DBU)-mediated Michael addition followed by intramolecular cyclization of isomeric 2/3-(azidomethyl)indol-3/2-yl)acrylates also furnished the respective β- and γ-carboline derivatives. Unexpectedly, the thermolysis of 2-(azidomethyl)-3-(indol-3-yl)acrylates led to the formation of ethyl 5-(2-(phenylsulfonamido)aryl)nicotinates along with γ-carbolines, via nitrene insertion followed by rearrangement and cyclization. The isomeric 2-(azidomethyl)-3-(indol-2-yl)acrylate upon thermolysis led to the expected δ-carboline. Thermal intramolecular nitrene insertion reaction could also be extended for accessing pyrido benzothiophene, pyrido thiophene, and quinoline.
