Abstract:Recently, the asymmetric reduction of imines catalyzed by Lewis-basic organocatalyst has been received much attention as the cheap hydrogen source and the simple post treatment. Based on the different functional groups of organocatalysts, this paper introduced formamide, pyridine amide, sulfonamide, supported and other Lewis basic organocatalysts. The structural characteristics, catalytic activity and mechanism of the Lewis-basic organocatalysts were summarized.

Abstract:In order to ameliorate the asymmetric inductivity and catalytic activity of the salen ligands with pyrrolidine diamine collar,Lewis acid and Lewis base group have been introduced at different positions of the salen unit.These new bifunctional N-oxide salen-Ti catalysts were successfully synthesized and applied to the catalytic asymmetric cyanosilylation of ketones in mild conditions.The products were obtained generally with moderate enantiomeric excesses and excellent yields.

Abstract:Magnetic nano-Fe3O4-supported organocatalysts were synthesized by anchoring valine-derived formamide onto the surface of Fe3O4 magnetic nanoparticles, which were applied in the asymmetric reduction of imines with trichlorosilane at room temperature in toluene. The high level of yield and enantioselectivity catalyzed by magnetic nano-Fe3O4-supported organocatalysts was obtained. In the immobilization process, CuI-catalyzed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) “click chemistry” was used as the anchored bridge. The magnetic nanoparticles can simplify the recovery of the organocatalyst and its separation from the reaction system. By an external magnet, the catalyst can be recycled and reused five times without a remarkable activity decline.

Abstract:The backbone of D-glucosamine hydrochloride was fine-tuned and modified by protecting the hydroxyl groups. In order to reduce imines with trichlorosilane, the carbohydrate-derived organocatalysts were prepared and screened. Methyl-4,6-O-benzylidene-2-amino-2-deoxy-α-D-glucopyr anoside was found as the best catalyst. The reduction was proceeded under CHCl3 as solvent at 40?C, affording 68% - 94% yield.

Abstract:Novel organocatalysts combining carbohydrate and N-formyl-l-valine derivatives were prepared by CuII-catalyzed diazo transfer and CuI-catalyzed azide–alkyne 1,3-dipolar cycloaddition CuAAC click chemistry. It was found that the carbohydrate-based valine-derived formamide organocatalyst had high catalytic activity for the asymmetric reduction of imines with trichlorosilane. The reduction can proceed at room temperature in toluene in high yield (up to 98%) and with excellent enantioselectivity (up to 94%). ‘CuAAC’ click chemistry is a bridge to link N-formyl-l-valine derived organocatalysts with carbohydrates.

Abstract:The highly regio- and enantioselective iron-catalyzed anti-Markovnikov hydroboration of 1,1-disubstituted aryl alkenes is reported by using a novel chiral iminopyridine oxazoline (IPO) ligand, in which the iminopyridine group is proposed to stabilize the iron and chiral oxazoline group to control enantioselectivity. This distinct class of reactive IPO ligands will likely be of high value for a large variety of asymmetric transformations using first-row transition metals.

Abstract:The asymmetric reduction of N-unsubstituted β-enamino esters is the key step for catalytic asymmetric construction of sitagliptin, a prevalent drug for the treatment of type 2 diabetes. However, it is still lack of environment friendly, low-cost, nontoxic, and highly selective catalysts. Herein, we report a series of new silyl ether substituted l-pipecolinic formamides organocatalysts for the asymmetric reduction of N-unsubstituted β-enamino esters with trichlorosilane as the reducing agent. The steric hindrance of silyl ether substitutes can remarkably influence the catalytic performance. The optimized triphenylsilyl substituted catalyst 2k can efficiently catalyze the reduction reaction, affording enantioenriched β-amino ester with excellent yield (95 %) and high ee (82 %).

Abstract:A cobalt-catalyzed asymmetric reduction of beta,beta-dialkyl alpha,beta-unsaturated esters with polymethylhydrosiloxane (PMHS) was reported to deliver the corresponding esters containing a chiral trialkyl carbon center at beta-position with up to 97 % yield and 98 % ee. The chiral tridentate ligand oxazoline iminopyridine (OIP) could perform well for the asymmetric reduction instead of chiral bidentate ligands. This operationally simple protocol shows a broad scope of substrates using one equivalent of readily available PMHS as a cheap and easy-to-handle reductive reagent.

Abstract:A diverse array of bidentate P-containing ligands has been developed for Ir-catalyzed asymmetric hydrogenation, but chiral monodentate phosphine ligands remained comperatively underexplored. We herein report a novel iridium catalyst with XuPhos as a chiral monodentate phosphine ligand for the direct asymmetric reductive amination of ketones with secondary amines. This catalytic system tolerates a wide range of substrates, providing a series of chiral tertiary amines efficiently with high enantioselectivities. The high catalytic activity attributed to the presence of the aliphatic cyclohexyl group on the P-atom and the relative configuration of the ligand, which is crucial for the XuPhos to act as a monodentate P-ligand for iridium.

Abstract:The development of catalyst based on porous crystalline materials (PCM) constructed from metal ions or clusters and multidentate organic ligands is a topic of great interest. In view of the Lewis acidic and basic properties of PCMs, we report for the first time that MIL-101(Cr) works as a synergistic catalyst for the reduction of imines with trichlorosilane as the hydrogen source. Both ketimines and aldimines were tolerated with this protocol, giving the corresponding amines in moderate to high yields. The operational simplicity as well as mild reaction conditions renders this protocol an attractive approach for the synthesis of amines. Furthermore, a chiral MOF, CMIL-101, was also realized by grafting chiral N-formyl proline derivatives to the open metal sites. Abstract:Starting from diverse alkene-tethered aryl iodides and O-benzoyl-hydroxylamines, the enantioselective reductive cross-electrophilic 1,2-carboamination of unactivated alkenes was achieved using a chiral pyrox/nickel complex as the catalyst. This mild, modular, and practical protocol provides rapid access to a variety of beta-chiral amines with an enantioenriched aryl-substituted quaternary carbon center in good yields and with excellent enantioselectivities. This process reveals a complementary regioselectivity when compared to Pd and Cu catalysis.
