Abstract:A series of chiral tridentate ligands derived from readily available enantiopure l-proline were designed and synthesized. The ligands together with Cu(OAc)2 were successfully used in asymmetric Henry reactions. Various structurally divergent aldehydes and nitromethane were converted into versatile β-nitro alcohols in MeOH at room temperature with very good yields (up to 85%) and enantioselectivities (up to 86%).

Abstract:Copper-catalyzed asymmetric Henry reactions are described. Using a new chiral amidophosphine ligand, the Henry reaction of nitromethane and various aldehydes proceeded smoothly to provide chiral β-nitroalcohols in reasonable yields (up to 98%) with high enantioselectivity (up to 97% ee).

Abstract:This article is a continuation of our previous research on the catalytic capability of a chiral copper complex based on commercially available (S)-2-aminomethylpyrrolidine and 3,5-di-tert-butylsalicylaldehyde with various counter-anions in the asymmetric Henry reaction. Our findings indicate that depending on the type of base used, chiral nitroalcohols with yields up to 98% and ee values up to 77%, as well as β-nitrostyrenes with yields up to 88%, can be produced. Additionally, it has been found that the outcome of the reaction and the catalytic properties of copper (II) complexes (S)-Cu1 and (S)-Cu2 are influenced by the structure of the aldehyde used.

Abstract:Several new chiral N,N'-dimethylated salan ligands with bulky substituents were synthesized and their in-situ generated Cu(II) complexes were evaluated in the asymmetric Henry reaction. Substituents on the aryloxide moieties of these ligands were found to show remarkable effect on the enantioselectivity. Cu(II) complex generated from the ligand with 1,1-diphenylethyl groups at the ortho-position of the aryloxide moieties and Cu(OAc)(2)center dot H2O was found to show good catalytic performance, giving the 2-nitro1-phenylethanol product in 85% yield with 94% ee in the presence of TEA in THF at -20 degrees C. The catalyst systems were examined with different aldehydes and the corresponding products were obtained in good yields (up to 94%) with 85% to 95% ee in the presence or absence of TEA. Diastereoselective reactions using nitroethane as the nucleophile afford syn-beta-nitroalcohols in good yields (48%-66%) with good dr (up to 11.5:1 syn/anti) and high ee values (92%-96%). Abstract:Several novel chiral tridentate ligands containing thiazoline were efficiently synthesized from commercially available l=cysteine in high yield. These ligands were subsequently applied to the asymmetric Henry reaction of nitromethane and various aldehydes. It was found that the structures of the thiazoline ligands had a significant influence on the enantioselectivity. It was shown that the optimal catalyst for this reaction was a ligand complexed with CuCl, which was formed from chiral thiazoline with chiral aminoalcohol. At −20°C, with 10 mol% of this ligand, a product with (S)-configuration was isolated in 93% yield and 98% enantiomeric excess. Copyright © 2015 John Wiley & Sons, Ltd.

Abstract:A general asymmetric deconstructive alkynylation of cyclic oximes with terminal alkynes was reported using copper/chiral cinchona alkaloid-based N,N,P-ligand catalysts and used to produce more than 60 examples of enantioenriched γ- and δ-alkynyl nitriles in a highly functional group-compatible manner. A wide variety of low-cost terminal alkynes and easily available cyclic oximes with substituent diversity were found to be well tolerated with the current asymmetric transformation with good functional group tolerance. Abstract:A highly anti-selective asymmetric Henry reaction has been developed, affording synthetically versatile beta-nitroalcohols in a predominately anti-selective manner (mostly above 15:1) and excellent ee values (mostly above 95?%). Moreover, the anti-selective Henry reaction was carried out in the presence of water for the first time with up to 99?% ee. The catalytic mechanism was proposed based on the detection of the intermediates by extractive electrospray ionization mass spectrometry (EESI-MS). Furthermore, the anti adducts have been successfully transformed into the biochemically important (+)-spisulosine and a pyrroloisoquinoline derivative.

Abstract:A great number of reports have described asymmetric catalytic Henry reactions using nitromethanes as pronucleophiles, but far more challenging is diastereoselective catalytic Henry reactions using substituted higher nitroalkanes instead of nitromethane to generate chiral β-nitro alcohol scaffolds with four adjacent stereogenic centers in a one-pot operation. This review summarizes the current state and applications of such reactions involving complex nitroalkane coupling with various carbonyl compounds for resolving double chiral centers with high enantio- and diastereoselectivities.

Abstract:A new type of chiral tertiary amine ligand was designed and derived from l-proline and (R)-BINOL. These new chiral ligands chelated with Cu(II) showed highly catalytic efficiency in enantioselective Henry reactions. Excellent yields (up to 99%) and high enantioselectivities (up to 96% ee) were achieved for aromatic, hetero-aromatic and aliphatic aldehyde substrates, without an additional base additive or the need for air or moisture exclusion.
