Abstract:The cyclization of nitroalkyne catalyzed by Hg(OTf)2 to produce the corresponding benzo[c]isoxazole in excellent yields with high selectivity is reported. On the basis of this strategy, a one-pot method to synthesize indole derivatives has been developed. In this transformation, two Hg-carbene intermediates are proposed to be involved.

Abstract:Concise and efficient four-component tandem approaches to polysubstituted indolizines have been developed under metal-free and mild aerobic conditions in refluxing acetonitrile. The mechanism of the novel reactions was proposed involving the formation of pyridinium ylides and α,β-unsaturated ketones with subsequent 1,3-dipolar cycloaddition and aromatization reaction.

Abstract:Convenient and regioselective syntheses of 1,2-annulated, and 1,2-, 5,6- and 1,2-, 7,8-bisannulated polycyclic indolizine derivatives have been achieved by one-pot tandem reactions of cyclic N-ylides derived from the corresponding N-substituted pyridinium, quinolinium, and isoquinolinium salts 1–3 with dichloro substituted α,β-unsaturated carbonyl compounds 4–7. The reactions of the N-ylides with 2,3-dichloroindenone 4, 3,4-dichlorocoumarin 5, and 4a,6,7,8a-tetrachloro-1,4-methanonaphthalene-5,8-dione 6 proceed by sequential [3+2] cycloaddition and elimination of hydrogen chlorides from the cycloadducts. On the other hand, reactions of the N-ylides with 2,3-dichloro-1,4-naphthoquinone 7 take place via a novel reaction sequence to give the products 15–17.

Abstract:Indole-containing polyaromatic scaffolds are widely found in natural products, pharmaceutical agents, and π-conjugated functional materials. Often, the synthesis of these highly valuable molecules requires a multistep sequence. Therefore, a simple, one-step protocol to access libraries of polyaromatic indole scaffolds is highly desirable. Herein we describe the direct synthesis of polysubstituted indolo[2,1-a]isoquinoline analogues via a double C-H annulation cascade using triazene as an internally cleavable directing group. Evidence from HRMS and theoretical calculations suggests that an unprecedented 1,2-alkyl migration might be responsible for the in situ cleavage of the directing group. Both kinetic isotope effects and DFT calculations suggested that the alkyne insertion step is rate-limiting for the second C,N annulation reaction.

Abstract:One-pot synthesis of 2-(N-sulfonylimino)indolines has been developed. The procedure combines the copper-catalyzed three-component reaction of sulfonyl azides, o-bromophenylacetylenes, and amines and the copper-catalyzed intramolecular C–N coupling in one sequence, which afforded the products in moderate to good yields. The resulting 2-(N-sulfonylimino)indolines could be easily transformed to pharmaceutically valuable oxindoles (indolin-2-ones).

Abstract:An efficient method for the construction of 1,2,3-triarylindoles through one-pot multicomponent reactions using calcium carbide, 3 mol of iodoarenes, and aromatic amines as starting materials was described. A series of target products were obtained by the simultaneous formation of five bonds in one step. The main advantages for this protocol are the use of a convenient alkyne source, wide scope of substrates, and simple workup procedure. The method can be extended to the gram scale.

Abstract:One-pot synthesis of cyclohexene derivatives from two alkynes and one maleic acid derivative could be achieved. Two alkynes were treated with [Cp2ZrBu2] to give zirconacyclopentadienes. Addition of 2 equiv of H2O followed by the reaction with maleic anhydride at 100 degrees C in toluene for 12 h afforded the cyclohexene derivatives in high yields. The corresponding titanacyclopentadiene did not give the desired product under the same conditions. In the case of Hf, only 8% of the product was obtained. Dienylzirconocene, which could be prepared by the addition of one equiv of EtOH or t-BuOH to the zirconacyclopentadiene, also reacted with maleic anhydride to give the cyclohexene derivative in 82 and 62% yield, respectively.

Abstract:A one-pot synthesis of pyrrolo[3,2-d]pyridazine derivatives via zirconocene-mediated four-component coupling of one Si-tethered diyne, two nitriles, and one azide is reported. When TMSN3 was used, pyrrole-2,3-diones were formed in good yields. Further condensation of these highly functionalized pyrrole-2,3-diones with hydrazine and hydroxylamine afforded useful pyrrole-fused heterocycles.
