Abstract:Zhemaidong (ZMD) and Chuanmaidong (CMD) are the two genuine cultivation areas of Ophiopogonis Radix which has been widely used as a traditional Chinese medicine in China for treating cardiovascular and pulmonary diseases. Differences between ZMD and CMD in chemical constituents and pharmacological effects have been reported, however, the details remain largely unknown. The aim of this study was to comprehensively characterize the chemical composition of Ophiopogonis Radix from the two producing areas and compare their immunomodulatory activities. An approach of HPLC-MS coupled with multivariate statistical analysis was established to reveal the characteristic constituents of ZMD and CMD. Furthermore, the effects of ZMD and CMD on the macrophage phagocytosis and gastrointestinal peristalsis were also determined in zebrafish models for assessing the immunomodulatory activities of these two strains. The results revealed that the chemical constituents of ZMD and CMD were much different from each other, and 19 constituents could be served as chemical markers to distinguish these two strains. Moreover, ZMD showed higher promoting rates in macrophage phagocytosis and gastrointestinal motility than those of CMD, suggesting ZMD might possess better immunomodulatory activities. Taken together, the results generated from this study indicated that the herbs from different producing areas should be evaluated and considered in preparing TCM prescriptions. (C) 2016 Elsevier B.V. Abstract:In this work, the chemical constituents in Da-Huang-Gan-Cao-Tang, a traditional Chinese formula, were studied by liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry and liquid chromatography coupled with ion trap mass spectrometry for the first time. Among the 146 compounds detected in Da-Huang-Gan-Cao-Tang, 104 compounds were identified unambiguously or tentatively based on their accurate molecular weight and multistage MS data, including one potential novel compound and two reported in Glycyrrhiza genus for the first time. The possible fragmentation pathways were proposed and fragmentation rules of the major types of compounds were concluded. This study provided an example to facilitate the tedious identification of chemical composition in traditional Chinese medicine, and maybe a promising reference approach to research the analogous formulae.
HPLC Determination of Four Components in Qizhiweitong Granules

Abstract:Objective:To establish an HPLC method for the simultaneous determination of albiflorin,paeoniflorin,naringin and glycyrrhizic acid in Qizhiweitong granules.Method:The chromatography was performed on a Zorbax SB-C18 column(250 mm×4.6 mm,5 μm)with a mobile phase of water(containing 0.2% acetic acid)-acetonitrile(containing 0.2% acetic acid)and linear gradient elution.The flow rate was 1.0 mL·min-1.The column temperature was 30 ℃.The detection wavelength was 230 nm from 0 to 15 min,284 nm from 15 to 20 min and 254 nm from 20 to 35 min.Results:The calibration curves of albiflorin,paeoniflorin,naringin and glycyrrhizic acid were linear in the ranges of 15.1-226 μg·mL-1(r=0.9998),25.6-383 μg·mL-1(r=0.9998),27.7-277 μg·mL-1(r=0.9998)and 9.43-47.2 μg·mL-1(r=0.9998)respectively.The mean recoveries of low,middle and high concentrations of the four components were 97.4%-101%(RSD=0.8%-1.7%),96.8%-99.7%(RSD=1.4%-2.6%),97.3%-100%(RSD=1.9%-2.4%).Conclusion:The method is simple and accurate,which can be adopted in the quality control of Qizhiweitong granules.

Abstract:Radix Ophiopogonis and Radix Liriopes are two widely used herbal medicines and functional foods with similar pharmacological activities and clinical efficacies. In order to figure out whether phytoequivalence of species leads to therapeutic equivalence and consistency, in this study, a novel sensitive and selective high performance liquid chromatography-tandem mass spectrometric method (HPLC-MS/MS) has been developed for the simultaneous analysis of 11 bioactive components (6 steroidal saponins and 5 flavonoids) and then was applied for comparing in Radix Ophiopogonis and Radix Liriopes from different habitats. Separation was performed using a C18 analytical column with gradient elution of acetonitrile and 0.3‰ acetic acid (v/v) at a flow rate of 0.8mL/min. Multiple-reaction monitoring (MRM) scanning was employed with switching electrospray ion source polarity between positive and negative modes in a single run. All calibration curves showed good linearity (r2>0.9915) within the test ranges. The intra-day and inter-day precision for 11 analytes were less than 3.7% and 4.2%, respectively. The recoveries were between 95.3% and 106.7%. Fifty-three batches of commercial samples from different sources were analyzed using the developed method. Hierarchical clustering analysis (HCA) and content analysis were performed to differentiate and classify the samples. The results demonstrated that steroidal saponins were the major components in both Liriopes species, and a low content of flavonoids could become an important symbol of distinction between Ophiopogonis and Liriopes. The developed method could successfully differentiate between Ophiopogonis and Liriopes.

Abstract:A simple and accurate analytical method was developed for simultaneous quantification of three steroidal saponins in the roots of Ophiopogon japonicus via high-performance liquid chromatography (HPLC) with mass spectrometry (MS) in this study. Separation was performed on a Tigerkin C18 column and detection was performed by mass spectrometry. A mobile phase consisted of 0.02% formic acid in water (v/v) and 0.02% formic acid in acetonitrile (v/v) was used with a flow rate of 0.5?mL?min-1. The quantitative HPLCMS method was validated for linearity, precision, repeatability, stability, recovery, limits of detection and quantification. This developed method provides good linearity (r?>0.9993), intra- and inter-day precisions (RSD <4.18%), repeatability (RSD <5.05%), stability (RSD <2.08%) and recovery (93.82102.84%) for three steroidal saponins. It could be considered as a suitable quality control method for O. japonicus.
Determination of Homoisoflavonoids in Ophiopogon japonicus by RP-HPLC

Abstract:The homoisoflavonoid content of 14 samples of Ophiopogon japonicus collected from different parts of China have been investigated. Three principal homoisoflavonoids, methylophiopogonanone A, methylophiopogonanone B, and 6-aldehydo-isoophiopogonanone A, were analyzed simultaneously by reversed-phase high-performance liquid chromatography with a binary mixture of acetonitrile and 0.3% aqueous acetic acid as mobile phase. The recovery of the method was 98.5–102.6% and good linearity ( r > 0.9990) was obtained for all the homoisoflavonoids over a relatively wide concentration range. The results indicated that the homoisoflavonoid content of Ophiopogon japonicus varied significantly from species to species, and from locality to locality. The quality and identity of Ophiopogon japonicus from the herb market can be determined from the homoisoflavonoid content.

Abstract:Objective To develop a new HPLC method to generate the fingerprint of Ophiopogonis Radix cultivated in Sichuan province. Methods A total of 10 batches of Ophiopogonis Radix samples were analyzed by HPLC,and 8 reference substances were used for the identification of the common peaks. The fingerprint was further analyzed by similarity analysis,relative retention time analysis and relative peak area analysis. Results The similarities of all the samples tested were higher than 0. 900. The relative peak areas of the common peaks varied in a relatively wide range (RSD=7. 1% -71. 9%,n=10). Conclusion The method is simple,practical,suitable for industrial application,and could be used for the quality evaluation of Ophiopogonis Radix.

Abstract:The root tuber of Ophiopogon japonicus (Thunb.) Ker-Gawl ("Maidong" in Chinese), with steroidal saponins and homoisoflavonoids as its representative chemical compositions, is a representative medicinal herbs with multiple major producing areas. This study aimed to distinguish the O. japonicas samples from Zhejiang and Sichuan by using an ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF MS)-based metabolome analysis. Firstly, a global chemical constituent identification of O. japonicas was carried out by using both automatic and manual methods. An integrated steroidal saponins structural identification strategy in O. japonicas based on exact mass information, fragmentation characteristics and retention time was developed. Overall, 135 steroidal saponins, 47 homoisoflavonoids and 9 other metabolites were quickly identified or tentatively identified from the MSE continuum data. Furthermore, multivariate statistical analysis revealed that O. japonicas from Zhejiang and Sichuan can clearly be separated and some markers were screened. Moreover, some major active components including total soluble sugar, total soluble polyphenol, total flavonoid, total saponin and 10 specific compounds were analyzed quantitatively. In general, these results showed that there were many differences between the metabolic profile data of O. japonicas from different producing areas, O. japonicas from Sichuan showed higher level steroidal saponins and samples from Zhejiang had higher contents of homoisoflavonoids specifically, and indicated that metabolite profiling by UPLC/Q-TOF MS is an effective approach for the discrimination of medicinal herbs from different geographical origins.

Abstract:Four compounds were isolated from the tubers of Ophiopogon japonicus for the first time. They were elucidated as cyclo-(Phe-Tyr) (1), cyclo-(Leu-ILe) (2), N-(2-(4-hydroxyphenyl) ethyl)-4-hydroxycinnamide (3), and tianshic acid (4) on the basis of spectral analysis. Among them, the cyclodipeptides of 1 and 2 were structure type firstly isolated from the genus Ophiopogon.
Qualitative and quantitative studies on two commercial specifications of Polygonatum odoratum

Abstract:The rhizoma of Polygonatum odoratum (PO) is used to treat yin injuries of the lung and stomach in traditional Chinese medicine. The chemical constituents of this herb are steroidal saponins, homoisoflavanones, and alkaloids. Xiangyuzhu (XPO) and Guanyuzhu (GPO) are available in the market as two specifications of the commodity. Nonetheless, systematic research on the identification and comparison of chemical constituents of these two commercial specifications is yet lacking. Herein, an integrated method combing ultra-high-performance liquid chromatography-quadruple time-of-flight mass spectrometry (UHPLC-Q-TOF/MS) with ultra-high-performance liquid chromatography-charged aerosol detection (UHPLC-CAD) was employed for the comprehensively qualitative and quantitative analyses of PO. A total of 62 compounds were identified by UHPLC-Q-TOF/MS, among which 13 potential chemical markers were screened out to distinguish two commercial specifications. Subsequently, the absolute determination method for polygodoraside G, polygonatumoside F, and timosaponin H1 was established and validated by UHPLC-CAD. The contents of the three compounds were 13.33-236.24 μg/g, 50.55-545.04 μg/g, and 13.34-407.83 μg/g, respectively. Furthermore, the ratio of timosaponin H1/polygodoraside G could be applied to differentiate the two specifications. Samples with a ratio <2 are considered XPO and >5 are considered GPO. Therefore, the above results provide a valuable means for the quality control of PO.

Abstract:Taxilli Herba (TAXH) is an important traditional Chinese medicine with a long history, dating from the Eastern Han Dynasty to the present times. However, the active constituents in it that parasitize different hosts vary, affecting its clinical efficacy. Given the complexity of the host origins, evaluating the quality of TAXH is critical to ensure the safety and effectiveness of clinical medication. In the present study, a quantitative method based on ultra-fast liquid chromatography tandem triple quadrupole mass spectrometry (UFLC-QTRAP-MS/MS) was established, which simultaneously determined the content of 33 active constituents, including 12 flavonoids, 4 organic acids, 12 amino acids, and 5 nucleosides in 45 samples. Orthogonal partial least squares discriminant analysis (OPLS-DA) was employed to classify and distinguish between TAXH and its adulterants, Tolypanthi Herba (TOLH). A hierarchical clustering analysis (HCA) was conducted combined with a heatmap to visually observe the distribution regularity of 33 constituents in each sample. Furthermore, gray relational analysis (GRA) was applied to evaluate the quality of samples to get the optimal host. The results demonstrated that TAXH excelled TOLH in quality as a whole. The quality of TAXH parasitizing Morus alba was also better, while those that were parasitic on Cinnamomum camphora and Glyptostrobus pensilis had relatively poor quality. This study may provide comprehensive information that is necessary for quality control and supply a scientific basis for further exploring the quality formation mechanism of TAXH.

Abstract:The present study investigated a robust method for the preparation of four flavone C-glycosides, i.e. orientin, homoorientin, vitexin and isovitexin, which were prepared from an ethanol aqueous extract, i.e. antioxidant of bamboo leaves (AOB), by AB-8 resin-based column chromatography and preparative high-performance liquid chromatography (HPLC) using a mobile phase consisting of 10% and 15% (v/v) of acetonitrile and 1% acetic acid. These flavone C-glycosides were further purified by the drowning-out crystallization method using methanol and water as drowning-out anti-solvents and salting-out agents, respectively. The purity was assessed by analytical HPLC and the confirmation of chemical structures was performed by IR, MS, NMR and UV spectroscopy. Orientin (49mg), homoorientin (142mg), vitexin (15mg) and isovitexin (62mg) were prepared from 6.5g of crude column chromatography fraction obtained from 5L of AOB concentrated solution. The present method is robust and suitable for preparing available quantities of pure flavone C-glycosides and the quantification of orientin, homoorientin, vitexin and isovitexin in bamboo leaves.
Analysis of Ochratoxin A and Ochratoxin B in Traditional Chinese Medicines by Ultra-High-performance Liquid Chromatography–tandem Mass Spectrometry Using [13C20]-Ochratoxin A As an Internal Standard

Abstract:The paper reported a reliable analytical method for simultaneous determination of ochratoxin A (OTA) and ochratoxin B (OTB) in traditional Chinese medicines (TCMs) by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The development of the method and investigations on the matrix influence were described in particular. The matrix effects were thereby minimized by using a reliable internal standard and a simple sample pretreatment. The established method was further validated by determining the linearity (R(2) > or = 0.9990), average recovery (86.3-114.2%), sensitivity (limit of quantitation 0.03-0.19 ng mL(-1)) and precision (relative standard deviation < or = 13.1%). It was shown to be a suitable method for simultaneous determination of OTA and OTB in various TCMs. Finally, a total of 51 TCMs widely used in China were screened for OTA and OTB with the proposed method. The results showed that only 4 samples were contaminated with ochratoxins at low levels, indicating that it was low risk of ochratoxins to consumers who occasionally used TCMs.

Abstract:Ophiopogon japonicus (Linn. f.) Ker-Gawl (O. japonicas), mainly cultivated in Sichuan and Zhejiang province in China, has different bioactive components and therefore their pharmacological activities. To explain the different clinical efficacy of O. japonicas derived preparations, herein we report differences of pharmacological activities between Sichuan and Zhejiang O. japonicas and behind them the exact differences of bioactive components. Based on a LC/MS-IT-TOF method, the differences of bioactive components between Sichuan and Zhejiang O. japonicas extracts were analyzed and respective characteristic components were picked out. We determined 39 ophiopogonones and 71 ophiopogonins compounds in Sichuan and Zhejiang O. japonicas extracts and found the contents of these compositions have several times difference. Evidenced by experimental data of pharmacological activities in inhibiting cardiomyocyte damage induced by H2O2, mouse macrophage cell inflammation induced by lipopolysaccharide and cytotoxicity in vitro, Zhejiang O. japonicas extract had a stronger antioxidant and anti-inflammatory capacity than Sichuan O. japonicas extract, and the two O. japonicas extracts exhibited selective cytotoxicity on different cancer cell lines in vitro. These data shed light on the links between bioactive components and pharmacological activities of O. japonicas derived preparations. Thus, geographical origin of O. japonicas should be considered to be a key factor in efficacy studies and further clinical application.

Abstract:Zhejiang Ophiopogonjaponicus (ZOJ) is a specific variety of Ophiopogon japonicus with characteristic steroidal saponins and homoisoflavonoids, which are also main pharmacodynamic constituents with clinical effects, including curing inflammation and cardiovascular diseases. However, few analysis methods were applied to simultaneously and quantitatively determine two kinds of its constituents, and hazardous organic solvents are mostly used for extraction. In this study, a new validated simultaneous extraction and determination method for four characteristic steroidal saponins and homoisoflavonoids in ZOJ was established by ionic liquid–ultrasonic extraction (IL-UAE) combined with HPLC-DAD-ELSD analysis, which can be used for the quality control of ZOJ. Chromatographic separation was performed with a DAD wavelength at 296 nm, and the ELSD parameters of the drift tube temperature (DTT), atomizer temperature (AT), and nitrogen gas pressure (NGP) were set at 20% heating power, 70 °C, and 25 psi, respectively. The optimal IL-UAE conditions were 1 mol/L [Bmim]CF3SO3 aqueous solution, a liquid–material ratio of 40 mL/g, and an ultrasonic time of 60 min. The proposed method is reliable, reproducible, and accurate, which were verified with real sample assays. Consequently, this work will be helpful for the quality control of ZOJ. It can also present a promising reference for the simultaneous extraction and determination of different kinds of constituents in other medicinal plants.

Abstract:This study sought to determine the primary components (isoquinoline alkaloids, diterpenoids and steroids) in crude extracts of the Chinese herb Jin-Guo-Lan, prepared from the roots of Tinospora sagittata and T. capillipes, by liquid chromatography/electrospray ionization multistage mass spectrometry coupled with diode-array detection (LC-DAD/ESI-MS(n)). After separation on a reversed-phase C(18) column using gradient elution, positive and negative ESI-MS experiments were performed. In positive ion mode, the three types of compounds showed very different characteristic ions: strong [M](+) or [M+H](+) ions were observed for isoquinoline alkaloids; [M+NH(4)](+) and/or [M+H-CO(2)](+) for diterpenoids; [M+H-nH(2)O](+) (n=1-3) for steroids. These adduct ions and/or fragments were used to deduce the mass and categories of known and unknown components in crude extracts, and their structures were further confirmed by ESI-MS(n) in positive ion mode. Moreover, UV absorption peaks obtained from DAD provided useful functional group information to aid the MS(n)-based identification. As a result, 11 compounds were unambiguously identified by comparing with standard compounds and 13 compounds were tentatively identified or deduced according to their MS(n) data. Two of these compounds (13-hydroxycolumbamine and 13-hydroxyjatrorrhizine) were found to be new compounds and another one (13-hydroxypalmatine) was detected for the first time as a natural product. In addition, a [M-*CH(3)-H(2)O](*+) ion in MS(2) of [M](+) after in-source collision-induced dissociation was used to differentiate positional isomers of protoberberine alkaloids, columbamine and jatrorrhizine. Although the roots of T. sagittata and T. capillipes contain almost identical compounds, the content of the compounds in them is dramatically different, suggesting the necessity for further comparison of the bioactivities of the two species.
Comparative Studies on Fingerprints of Root of Ophiopogon japonicus Cultivated in Different Areas

Abstract:AIM:To establish the fingerprints for characterization of the chemical components of maidong(root of Ophiopogon japonicus)in sichuan(Chuanmaidong) and hangzhou(Hangmaidong).METHODS:HPLC-ELSD and ESI/TOF-MS analytical methods were applied to detect n-butanol extracts respectively,and HPLC-UV method was employed to study ether extracts.RESULTS:HPLC-ELSD and ESI/TOF-MS methods provided a wide range of the chemical information of n-butanol extracts.HPLC-ELSD fingerprint patterns,as a whole,were different between Chuanmaidong and Hangmaidong although they shared several mutual peaks.However,ESI/TOF-MS analysis showed close similarity between Chuanmaidong and Hangmaidong in the MS~(1) scan.On the other hand,HPLC-UV fingerprint revealed similarity in the second half of the fingerprint pattern.CONCLUSION:Most saponins and flavonoids contained in the two cultivating areas are similar.Meanwhile,the fingerprint developed by HPLC-ELSD and HPLC-UV methods could differentiate the cultivating areas of the two maidong.
Qualitative Analysis and Componential Differences of Chemical Constituents in Taxilli Herba from Different Hosts by UFLC-Triple TOF-MS/MS

Abstract:Taxilli Herba (TH) is a well-known traditional Chinese medicine (TCM) with a wide range of clinical application. However, there is a lack of comprehensive research on its chemical composition in recent years. At the same time, Taxillus chinensis (DC) Danser is a semi parasitic plant with abundant hosts, and its chemical constituents varies due to hosts. In this study, the characterization of chemical constituents in TH was analyzed by ultra-fast liquid chromatography coupled with triple quadrupole-time of flight tandem mass spectrometry (UFLC-Triple TOF-MS/MS). Moreover, partial least squares discriminant analysis (PLS-DA) was applied to reveal the differential constituents in TH from different hosts based on the qualitative information of the chemical constituents. Results showed that 73 constituents in TH were identified or tentatively presumed, including flavonoids, phenolic acids and glycosides, and others; meanwhile, the fragmentation pathways of different types of compounds were preliminarily deduced by the fragmentation behavior of the major constituents. In addition, 23 differential characteristic constituents were screened based on variable importance in projection (VIP) and p-value. Among them, quercetin 3-O-β-D-glucuronide, quercitrin and hyperoside were common differential constituents. Our research will contribute to comprehensive evaluation and intrinsic quality control of TH, and provide a scientific basis for the variety identification of medicinal materials from different hosts.

Abstract:Zang, Jialin ; Li, Gang ; Zhao, Chaoyang ; Lv, Zhen ; We created a fast and effective thin-layer chromatography method to detect homoisoflavones in Polygonatum odoratum (Mill.) Druce (POD), demonstrating high sensitivity, specificity, and stability. Utilizing this method, we assessed the quality of 20 commercial batches of POD. Our results indicated that homoisoflavones (III, IV, and V) were present in 8 samples, absent in 7, and detected in 5. This method facilitates the effective evaluation of POD germplasm resources, laying a strong foundation for enhancing quality standards. This advancement aids in the standardization and quality control of POD, ensuring the consistency of its active ingredients and thereby supporting its therapeutic applications.

Determination of ochratoxin A in traditional Chinese medicinal plants by HPLC-FLD

Abstract:Traditional Chinese medicinal plants (TCMPs), commonly used as spices, additives or foods, are also widely used in China to prevent and cure human disease. Due to their provenance, TCMPs may be contaminated by various fungal species, including ochratoxigenic fungi, during growth, collection, transportation and, especially, storage. A reliable method for the determination of ochratoxin A (OTA) in TCMPs of different origins was developed to monitor OTA levels in China. Analyses involved the extraction of OTA by methanol/water, immunoaffinity column (IAC) clean-up and HPLC quantification with fluorescence detection (FLD). Positive results were further confirmed by LC-ESI-MS/MS. The limit of detection (LOD) was 0.3 microg kg(-1), based on a signal-to-noise ratio of 3 : 1. Among the total of 57 TCMPs samples collected from six different areas, 31 were visibly moldy due to inappropriate storage; 26 sample, purchased from local TCMPs markets, were not visibly moldy. The results showed that 23 of the visibly moldy samples and two of the not visibly moldy were contaminated with OTA at levels ranging 1.2-158.7 and 2.5-5.6 microg kg(-1), respectively. This is the first report of the natural occurrence of OTA in TCMPs.
