Abstract:New red-emitting conjugated polymers have been successfully synthesized by incorporating classical two-photon absorption (TPA) units, electron-rich units, and a small amount of electron-deficient units along the polymer backbones. Water-dispersible nanoparticles (NPs) based on these polymers were also fabricated for applications in two-photon excitation fluorescence imaging of cell membrane. Through optimization of the polymer/matrix mass ratio and the initial feed concentration of the polymer solution, a high quantum yield (QY) of 24% was achieved for the red-emitting NPs in water. TPA cross section and two-photon action cross section values of these polymers at 750 nm reached up to 1000 GM and 190 GM per repeat unit in aqueous media, 2.5 × 10(5) GM and 4.7 × 10(4) GM per NP, respectively. Furthermore, these NPs displayed excellent photostability and biocompatibility. Their applications as two-photon excitation fluorescence probes for cell membrane imaging have been demonstrated in three different cell lines with excellent imaging contrast. These results demonstrated that these polymer NPs hold great potentials as excellent two-photon excitation fluorescence probes in various biological applications.

Abstract:Highly emissive inorganic-organic nanoparticles with core-shell structures are fabricated by a one-pot, surfactant-free hybridization process. The surfactant-free sol-gel reactions of tetraphenylethene- (TPE) and silole-functionalized siloxanes followed by reactions with tetraethoxysilane afford fluorescent silica nanoparticles FSNP-1 and FSNP-2, respectively. The FSNPs are uniformly sized, surface-charged and colloidally stable. The diameters of the FSNPs are tunable in the range of 45-295 nm by changing the reaction conditions. Whereas their TPE and silole precursors are non-emissive, the FSNPs strongly emit in the visible vision, as a result of the novel aggregation-induced emission (AIE) characteristics of the TPE and silole aggregates in the hybrid nanoparticles. The FSNPs pose no toxicity to living cells and can be utilized to selectively image cytoplasm of HeLa cells.

Abstract:Novel cross-linked copolymers of PEG-IM-PhNH2 are successfully synthesized through PEGylation via radical polymerization of 2-isocyanatoethyl methacrylate and poly(ethylene glycol) monomethyl ether methacylate and subsequent cross-linking with an amino-terminated aggregation-induced emission fluorogen. Such obtained amphiphilic copolymers can self-assemble to form uniform fluorescent polymeric nanoparticles (FPNs) and be utilized for cell imaging. These cross-linked FPNs are demonstrated good water dispersibility with ultralow critical micelle concentration (∼ 0.002 mg mL(-1)), uniform morphology (98 ± 2 nm), high red fluorescence quantum yield, and excellent biocompatibility. More importantly, this novel strategy of fabricating cross-linked FPNs paves the way to the future development of more robust and biocompatible fluorescent bioprobes.

Abstract:Fluorescent carbon nanoparticles (FCNs) have gradually become the most promising alternative candidates to other traditional fluorescent nanomaterials for biological applications on account of their excellent fluorescence property and remarkable biocompatibility. Although many methods have reported on the preparation of FCNs, to date, no studies have reported the preparation of polymers of functionalized FCNs. A high-efficiency method was developed in this work to synthesize high-quality poly(ethylene oxide) (PEG)-functionalized FCNs from cigarette ash and thiol group-containing PEG via a facile one-pot ultrasonic irradiation treatment. A series of characterization techniques demonstrated the uniform nanoscale size, good fluorescence stability, high water dispersibility and remarkable biocompatibility of the generated FCNs. Furthermore, cell imaging was easily achieved at high resolution using the synthetic FCNs as probes, which validates their potential for bioimaging applications. In summary, an efficient one-pot strategy is reported for the first time on the preparation of PEG-functionalized FCNs with the assistance of ultrasonic irradiation. This method should be of great research interest for the fabrication of other polymer-functionalized FCNs with designable properties and functions.
Preparation and Two-Photon Imaging of a Water-Soluble Fluorescent Nanoparticle

Abstract:A water-soluble polymeric fluorescent nanoparticle (PTCN) has been proposed for two-photon fluorescence (TPF) imaging of living cells. PTCN was firstly prepared by chemical crosslinking of polyethyleneimine using a dual carboxyl 1, 8-naphthalimide derivative as the crosslinker. Subsequently, the investigations on its structure, morphology, one- and two-photon fluorescence properties and cytotoxicity were carried out. It was found that PTCN was spherical with the diameters of 5-10 nm. It could emit fluorescence at 536 nm with the excitation wavelengths of 443 nm and 800 nm, respectively. PTCN possessed good pH stability and photostability. Its fluorescence did not change in the pH range of 4.0-9.0. In pH 7.4 buffer solution, when it was detected at lambda(ex)/lambda(em) = 443 nm/536 nm for 12000 times, the decrease in fluorescence intensity was less than 1%. Cytotoxic assays indicated it was low toxic upon interactions with Hela cells for 24 h at a concentration of 15.0 mg/L. Finally, the application of PTCN for imaging live cells was assessed by TPF microscopy technique. After Hela cells were incubated with PTCN for 2 h, the green fluorescence was observed in the cellsap at the excitation wavelength of 800 nm. The proposed nanoparticle is potential in the development of targeting TPF probes.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.

Abstract:Organic and polymer fluorescent nanomaterials have been a frontier research focus. Here in this work, a series of fluorinated zwitterionic random copolymers end‐attached with a quasi‐chromophoric group of pyrene or tetraphenylethylene (TPE) have been well synthesized via atom transfer radical polymerization with activators regenerated by electron transfer (ARGET ATRP). Those random copolymers with total degree of polymerization 100 or 200 are able to produce fluorescent single‐chain nanoparticles (SCNPs) through intra‐chain self‐folding assembly with quite uniform diameters in the range of 10∼20 nm as characterized by dynamic light scattering and transmission electron microscopy. By virtue of the segregation or confinement effect, both SCNPs functionalized with pyrene or TPE group are capable of emitting fluorescence, with pyrene tethered SCNPs exhibiting stronger fluorescence emission reaching the highest quantum yield around 20%. Moreover, such kind of fluorescent SCNPs manifest low cytotoxicity and good cell imaging performance for Hela cells. The creation of fluorescent SCNPs through covalently attached one quasi‐chromophore to the end of one fluorinated zwitterionic random copolymer provides an alternative strategy for preparing polymeric luminescence nanomaterials, promisingly serving as a new type of fluorescent nanoprobes for biological imaging applications. Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) property have received increasing attention and possess promising biomedical application potential in the recent years. Many efforts have been devoted to the fabrication methodologies of FPNs and significant advance has been achieved. In this contribution, a novel strategy for the fabrication of AIE-active amphiphilic copolymers is reported for the first time based on the Ce(IV) redox polymerization. As an example, ene group containing AIE-active dye (named as Phe-alc) is directly grafted onto a water soluble polymer polyethylene glycol (PEG) in H2 O/THF system under low temperature. Thus-obtained amphiphilic fluorescent polymers will self-assemble into FPNs with ultra-low critical micelle concentration, ultra-brightness, and great water dispersibility. Biological evaluation results suggest that the PEG-poly(Phe-alc) possess excellent biocompatibility and can be used for tracing their behavior in cells using confocal laser scanning microscope. These features make PEG-poly(Phe-alc) FPNs promising candidates for many biomedical applications, such as cell imaging, drug delivery vehicles, and targeted tracing. More importantly, many other functional groups can also be incorporated into these AIE-active FPNs through the redox polymerization. Therefore, the redox polymerization should be a facile and effective strategy for fabrication of AIE-active FPNs.

Abstract:Fluorescent nanoparticles used to detect important biological events in living cells or animals are in increasing demand in the biological and biomedical fields, and have attracted much attention from chemists and biologists in the past decade. Here, one aggregation-induced emission (AIE) bioconjugate, tetraphenylethene labelled chitosan (TPE-CS), is synthesized, which could be strongly emissive in the solid state. TPE-CS is used as a coating agent for negatively charged hydroxyapatite (HA) nanoparticles, and TPE-CS/HA nanocomposites with positive charges are well dispersed in their aqueous solution with a diameter of 111.9 nm. MTT assay indicates that the fluorescent TPE-CS/HA nanoparticles have good cytocompatibility. 293T cells are imaged by TPE-CS/HA nanoparticles. First, the nanoparticles are adhered to the cell membrane, and then many more particles are endocytosed through phagocytotic vesicles by culturing for a long time, resulting in a much stronger fluorescence emission. TPE-CS/HA bioprobes could strongly bind the cell cytoplasmic region, and might have promising applications in tumor diagnosis, long-term cell tracing, drug metabolism and drug delivery systems.

Abstract:Glycosylated cross-linked red fluorescent amphiphilic polymer has been facilely synthesized in one pot, which would readily self-assemble into nanoparticles with high water dispersibility in aqueous media. The resulted nanoparticles have surplus carboxyl groups and glycosyl groups on the surface, which can be further functionalized, and they can demonstrate strong bio-favorable red fluorescence with a fluorescence quantum yield of 18% due to the aggregation induced emission (AIE) dyes aggregated in the core. Moreover, the nanoparticles also revealed great photostability and structure stability with an ultralow critical micelle concentration of 0.0018 mg mL(-1). Abstract:Aggregation-induced emission (AIE) dyes based fluorescent polymeric nanoparticles (FNPs) have been intensively explored for biomedical applications. However, many of these AIE-active FNPs are relied on the self-assembly of amphiphilic copolymers, which are not stable in diluted solution. Therefore, the introduction of cross-linkages into these micelles has demonstrated to be an efficient route to overcome this stability problem and endow ultra-low critical micelle concentrations (CMC) of these AIE-active FNPs. In this work, we reported the fabrication of cross-linked AIE-active FNPs through controllable reversible addition fragmentation chain transfer polymerization by using commercially available octavinyl-T8-silsesquioxane (8-vinyl POSS) as the cross-linkage for the first time. The resultant cross-linked amphiphilic copolymers (named as PEG-POSS-PhE) are prone to self-assemble into stable core-shell nanoparticles with well water dispersity, strong red fluorescence and low CMC (0.0069 mg mL(-1)) in aqueous solution. More importantly, PEG-POSS-PhE FNPs possess some other properties such as high water dispersity, uniform morphology and small size, excellent biocompatibility and cellular internalization, providing great potential of PEG-POSS-PhE FNPs for biological imaging application. Abstract:Aggregation-induced emission (AIE) polymeric nanoparticles (NPs) with a size of several tens of nanometers were successfully prepared by miniemulsion copolymerization of methyl methacrylate (MMA) with an AIE monomer, 1-allyl-1-methyl-2,3,4,5-tetraphenylsilole (AMTPS). The prepared AIE poly(MMA-co-AMTPS) NPs exhibited strong photoluminescence with high quantum yields. Promisingly, the brightness of AIE poly(MMA-co-AMTPS) NPs, which increased linearly with an increase in AMTPS content, could be accurately modulated via the AMTPS content in the range of 0-20 wt%. In vitro cell experiments indicated that the AIE poly(MMA-co-AMTPS) NPs were promising cell staining reagents with good biocompatibility and high photostability.

Abstract:Fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) characteristics have attracted much attention for biomedical applications due to their remarkable AIE feature, high water dispersity and desirable biocompatibility. The development of facile and effective strategies for fabrication of these AIE-active FPNs therefore should be of great importance for their biomedical applications. In this work, we reported that a catalyst-free thiol-yne click reaction can be utilized for fabrication of AIE-active FPNs in short reaction time and even without protection of inert gas. The results indicated that the obtained AIE-active amphiphilic copolymers (PEGMA-PhE) can readily self-assemble into luminescent nanoparticles (PEGMA-PhE FPNs) with high water dispersity, uniform size and morphology, red fluorescence. Cell viability examination and cell uptake behavior of PEGMA-PhE FPNs confirmed that these AIE-active FPNs possess low toxicity towards cells and can be easily internalized by cells through non-specific route. Therefore the remarkable properties of PEGMA-PhE FPNs such as high water dispersity, AIE-active fluorescence and nanoscale size as well as excellent biocompatibility make them promising for biomedical applications.

Abstract:In recent years, aggregation-induced emission (AIE) dyes based fluorescent organic nanoparticles (FONs) have achieved significant progress in various biomedical applications. In this work, we developed a covalent strategy to prepare biocompatible AIE-active dyes based cross-linked copolymers (MPC-POSS-PhE) via controllable reversible addition fragmentation chain transfer (RAFT) polymerization using zwitterionic 2-methacryloyloxyethyl phosphorylcholine (MPC), polymerizable AIE dye (named as PhE) and 8-vinyl polyoctahedral silsesquioxanes (POSS) as monomers. Due to the existence of hydrophilic MPC and hydrophobic PhE, the resultant copolymers will self-assemble into core-shell nanoparticles in aqueous solution with ultra-low critical micelle concentration (CMC). This could effectively overcome the drawbacks of non-crosslinked micelles and show more attractive properties and better performance for biomedical applications. Furthermore, the characterization results and biological assays demonstrated that the final MPC-POSS-PhE FONs show stable aqueous stability, uniform size and morphology, high water dispersity, desirable optical properties and low cytotoxicity. These remarkable properties make the resultant AIE-active nanoprobes great potential for biomedical applications.

Abstract:Application of organic dyes is limited in biomedical fields due to the rapid self-quenching, poor stability and water solubility. In this work, polymer modified organic dyes is developed through the dyes initiating polymerization. The polymer length is studied by tuning the amount of monomer. With an optimal molecule weight of the polymer, the pi-pi stacking from pi-conjugated organic fluorescent dyes and fluorescence quenching are inhibited, resulting in an enhancement of fluorescence intensity and photostability. Nanoparticles are further fabricated to be used for cell uptake based on the optimized organic dyes. Then, long term cellular fluorescence imaging is realized. This work highlights the potential of polymer modification to improve the performance of organic dyes and expand their applications.
Fluorescent non-conjugated polymer dots for targeted cell imaging

Abstract:Through the chemical crosslinking of the sub-fluorophore, linear non-conjugated polymers can possess strong photoluminescence (PL), which is a very important fluorescence behavior and the non-conjugated polymer dots (PDs) are efficient bio-fluorophores for bio-based applications. Herein, the new type of non-conjugated polyethyleneimine (PEI) PDs was further modified by targeting molecules (folic acid) for a new generation of bio-fluorophores. The free folic acid can quench the PL of PDs by energy transfer, while the conjugated folic acid@PDs (FA@PDs) can still maintain their PL properties to a certain degree. The FA@PDs also possess lower toxicity compared with free PDs, which is possibly due to blocking of the amino groups. Moreover, we investigated the targeted bioimaging applications of the FA@PDs, which gave a very important direction for application of these types of materials.
