Abstract:New red-emitting conjugated polymers have been successfully synthesized by incorporating classical two-photon absorption (TPA) units, electron-rich units, and a small amount of electron-deficient units along the polymer backbones. Water-dispersible nanoparticles (NPs) based on these polymers were also fabricated for applications in two-photon excitation fluorescence imaging of cell membrane. Through optimization of the polymer/matrix mass ratio and the initial feed concentration of the polymer solution, a high quantum yield (QY) of 24% was achieved for the red-emitting NPs in water. TPA cross section and two-photon action cross section values of these polymers at 750 nm reached up to 1000 GM and 190 GM per repeat unit in aqueous media, 2.5 × 10(5) GM and 4.7 × 10(4) GM per NP, respectively. Furthermore, these NPs displayed excellent photostability and biocompatibility. Their applications as two-photon excitation fluorescence probes for cell membrane imaging have been demonstrated in three different cell lines with excellent imaging contrast. These results demonstrated that these polymer NPs hold great potentials as excellent two-photon excitation fluorescence probes in various biological applications.

Abstract:Glycosylated cross-linked red fluorescent amphiphilic polymer has been facilely synthesized in one pot, which would readily self-assemble into nanoparticles with high water dispersibility in aqueous media. The resulted nanoparticles have surplus carboxyl groups and glycosyl groups on the surface, which can be further functionalized, and they can demonstrate strong bio-favorable red fluorescence with a fluorescence quantum yield of 18% due to the aggregation induced emission (AIE) dyes aggregated in the core. Moreover, the nanoparticles also revealed great photostability and structure stability with an ultralow critical micelle concentration of 0.0018 mg mL(-1). Abstract:Chitosan with tetraphenylethene pendants (TPE-CS) are synthesized by reaction between amine and isothiocyanate groups of chitosan and tetraphenylethene (TPE), respectively. Nanoparticles of TPE-CS (TPE-CS NPs) are fabricated by ionic gelation method. The NPs are uniform in size, spherical in shape, monodispersed, and positive in surface charge. The suspension of TPE-CS NPs emits strong blue fluorescence under photoexcitation due to the aggregation-induced emission characteristics of the TPE moieties. The NPs can be internalized into cytoplasm through endocytosis pathway and retain inside the live cells to image the cells. Cytotoxicity assay reveals that TPE-CS NPs are cytocompatible and thus can be used for long-term live cell imaging.

Abstract:Fluorescent nanoparticles used to detect important biological events in living cells or animals are in increasing demand in the biological and biomedical fields, and have attracted much attention from chemists and biologists in the past decade. Here, one aggregation-induced emission (AIE) bioconjugate, tetraphenylethene labelled chitosan (TPE-CS), is synthesized, which could be strongly emissive in the solid state. TPE-CS is used as a coating agent for negatively charged hydroxyapatite (HA) nanoparticles, and TPE-CS/HA nanocomposites with positive charges are well dispersed in their aqueous solution with a diameter of 111.9 nm. MTT assay indicates that the fluorescent TPE-CS/HA nanoparticles have good cytocompatibility. 293T cells are imaged by TPE-CS/HA nanoparticles. First, the nanoparticles are adhered to the cell membrane, and then many more particles are endocytosed through phagocytotic vesicles by culturing for a long time, resulting in a much stronger fluorescence emission. TPE-CS/HA bioprobes could strongly bind the cell cytoplasmic region, and might have promising applications in tumor diagnosis, long-term cell tracing, drug metabolism and drug delivery systems.

Abstract:Aggregation‐induced emission (AIE) dye‐based fluorescent glycopolymers nanoparticles (FGNs) are synthesized with abundant carbohydrate groups on the surface for the first time. This is carried out firstly by free radical polymerization between AIE monomer (PhE) and a renewable biobased monomer itaconic anhydride (ITA), then by the ring‐opening reaction between ITA and glucosamine hydrochloride, to obtain glycopolymers with plenty of glycosyl groups on them. The resulted amphiphilic glycopolymer is prone to self‐assemble into nanoparticles with high water dispersibility due to the surplus carboxyl groups and glycosyl groups covered on the surface, which can also be further functionalized. The obtained nanoparticles demonstrate strong fluorescence emission, owning to the AIE dyes in the core of nanoparticles. Biocompatibility evaluation and cell uptake behavior of the nanoparticles are further investigated to explore their potential biomedical applications; the demonstrated excellent biocompatibility makes them promising for cell imaging. image

Abstract:Water dispersion aggregation-induced emission (AIE) dyes based nanomaterials have recently attracted increasing attention in the biomedical fields because of their unique optical properties, outstanding performance as imaging and therapeutic agents. The methods to conjugate hydrophilic polymers with AIE dyes to solve the hydrophobic nature of AIE dyes and makeS them widely used in biomedicine, which have been extensively explored and paid great effort previously. Although great advance has been made in the fabrication and biomedical applications of AIE-active polymeric nanoprobes, facile and efficient strategies for fabrication of biodegradable AIE-active nanoprobes are still high desirable. In this work, amphiphilic biodegradable fluorescent organic nanoparticles (PLL-TPE-O-E FONs) have been fabricated for the first time by conjugation of AIE dye tetraphenylethene acrylate (TPE-O-E) with Poly-l-Lysine (PLL) through a facile one-step Michael addition reaction, which was carried out under rather mild conditions, included air atmosphere, near room temperature and absent of metal catalysts or hazardous reagents. Due to the unique AIE properties, these amphiphilic copolymers tend to self-assemble into high luminescent water dispersible nanoparticles with size range from 400 to 600nm. Laser scanning microscope and cytotoxicity results revealed that PLL-TPE-O-E FONs can be internalized into cytoplasm with negative cytotoxicity, which implied that PLL-TPE-O-E FONs are promising for biological applications.

Abstract:With the ever increasing interest in biomedical applications of luminescent probes, the development of novel luminescent nanoparticles which showed high water dispersibility, strong luminescence and excellent biocompatibility has been extensively pursued. In this work, cross-linked luminescent polymeric nanoparticles based on aggregation induced emission (AIE) dyes were fabricated through combination of free radical polymerization and ring-opening reaction using AIE dye as the fluorogen and linker simultaneously. Our results demonstrated that these AIE dye based cross-inked polymeric nanoparticles showed high water dispersibility, remarkable luminescent properties and excellent biocompatibility, is highly potential for biomedical applications.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.

Abstract:Fluorescence probes play a crucial role in optical imaging for visualization of complex biological processes. As compared with conventional organic fluorogens, the probes with aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics show significant advantages in high quantum yield at concentrated and aggregated state, large Stokes shift and low cytotoxicity. However, the synthesis of AIE-active fluorescent probes through the ESIPT mechanism has received only very limited attention. On the other hand, the preparation of biodegradable fluorescent probes through the ESIPT mechanism has not been demonstrated thus far. In this work, we reported for the first time that water dispersible and biodegradable fluorescent polymeric nanoparticles with AIE and ESIPT characteristics could be facilely obtained through conjugation of 2,4-Dihydroxybenzophenone based benzophenone azine (BPA) and polyethylene glycol (PEG) using hexamethylene diisocyanate. The final copolymers contained hydrophilic and biocompatible PEG and biodegradable urethane linkage are readily self-assembled into core-shell nanostructures. Moreover, the self-assembled BPA-PEG2000 fluorescent organic nanoparticles (FONs) displayed obvious AIE feature, high water dispersibility, superb biocompatibility, biodegradability and excellent cell dyeing performance. All of the above properties implied that BPA-PEG2000 FONs are promising candidates for a variety of biomedical applications.

Abstract:Fluorescent water-dispersible conjugated polymeric nanoparticles (WDCPNs) were synthesized for cellular imaging. Sonogashira polycondensations of various combinations of symmetric monomers (A(x) + B-y, x > 2, y >= 2) were firstly conducted in confined nanoreactors to produce soluble conjugated polymeric nanoparticles (SCPNs) with narrow size distribution and tunable fluorescences emission. The subsequent click reaction between the terminal alkyne groups in the SCPNs and azide terminated PEG(1000) endowing good dispersity of the WDCPNs in aqueous solutions and stable fluorescence emission. The WDCPNs exhibited bright fluorescence in cellular imaging and showed good biocompatibility as revealed by cell variability assay, demonstrating them as ideal fluorescent probes for biological imaging and detection.

Abstract:The development of novel fluorescent nanoprobes has attracted great current research interest over the past few decades due to their superior optical properties and multifunctional capability as compared with small organic dyes. Although great advance has been made in the utilization of fluorescent nanoprobes for biomedical applications, development of novel fluorescent nanoprobes that possess good fluorescent properties, biocompatibility, biodegradability and water dispersibility through a convenient and effective route is still highly desirable. In this work, we reported for the first time that novel fluorescent organic nanoparticles (FONs) can be conveniently fabricated via self-polymerization of dopamine and polyethyleneimine at room temperature and in an air atmosphere within 2 h. These FONs exhibited strong green fluorescence, high water stability and excellent biocompatibility, making them highly potential for biological imaging applications. More importantly, due to the high reactivity of polydopamine, these FONs might also be further functionalized with other functional components through Michael addition or Schiff base reaction. Therefore the method described in this work would open new avenues for the fabrication of fluorescent nanoprobes for various biomedical applications.

Abstract:Application of organic dyes is limited in biomedical fields due to the rapid self-quenching, poor stability and water solubility. In this work, polymer modified organic dyes is developed through the dyes initiating polymerization. The polymer length is studied by tuning the amount of monomer. With an optimal molecule weight of the polymer, the pi-pi stacking from pi-conjugated organic fluorescent dyes and fluorescence quenching are inhibited, resulting in an enhancement of fluorescence intensity and photostability. Nanoparticles are further fabricated to be used for cell uptake based on the optimized organic dyes. Then, long term cellular fluorescence imaging is realized. This work highlights the potential of polymer modification to improve the performance of organic dyes and expand their applications.

Abstract:Fluorescent nanoparticles with a tunable emission show a good potential for usage in biological imaging. Exciplex emission usually appears with a large red shift from the normal emission peak. The integration of exciplex emission into nanoparticles offers a rational strategy to designing fluorescent nanoparticles with a tunable emission. In this work, we doped electron acceptors into the electron donor poly(N-vinylcarbazole) (PVK) to develop novel fluorescent nanoparticles with a conveniently modulated PVK emission. Through careful design of the molecular structures of the electron acceptors, we demonstrated that controlled donor-acceptor spatial stacking and electron transitions could regulate the exciplex emission of the PVK/ acceptor nanoparticles. Thus, the structurally controlled exciplex formation allowed for the preparation of multicolored fluorescent nanoparticles. Moreover, further modifications with the cyclic peptide RGD showed little disruption to the structure of the PVK/acceptor nanoparticles and the corresponding exciplex emission. Hence, the nanoparticles showed the ability to be used for targeted cellular imaging. On the basis of the RGD-integrin alpha(nu)beta(3) (ligand-receptor) interaction, the nanoparticles were effectively endocytosed by target cancer cells. We anticipate that this research could provide a new strategy for the fabrication of fluorescent nanoparticles with a tunable emission, leading to useful materials for fluorescent imaging.
