Abstract:Iron oxide magnetic nanoparticles (MNPs) are widely used as diagnostic and therapeutic agents for biomedical applications. Quantitatively analyzing biodistribution, pharmacokinetics and organ clearance of MNPs in mouse models is important for understanding their in vivo behavior. In this study, we developed a novel histochemical method for visualizing unlabeled MNPs in mouse tissues by employing their intrinsic peroxidase-mimicking activity, regarding which we reported previously that MNPs could catalyze the oxidation of peroxidase substrates to produce a color reaction at the site of MNPs (Gao et al. Nat. Nanotechncd. 2007, 2, 577-583(1)). Based on this MNPs-peroxidase approach, we determined the biodistribution and organ clearance of MNPs by visualizing and quantifying the localization of MNPs within the main organs. Compared to traditional Prussian blue assay, this novel MNPs-peroxidase approach has higher sensitivity. In conclusion, the developed MNPs-peroxidase approach based on intrinsic peroxidase activity of iron oxide nanoparticles was used effectively for quantitative detection of MNPs in mice by histochemical staining. Presumably, other nanoparticles having intrinsic peroxidase activity could also be considered.

Abstract:A novel magnetic-enhanced colorimetric assay was constructed based on aptamer conjugated PtCo bimetallic nanoparticles (NPs) with high oxidase-like catalytic activity, high water solubility, low cell toxicity, and superparamagnetic properties. It was found that the incorporation of magnetic metal Co atoms into NPs could not only be facilitated for magnetic separation, but also resulted in the significantly improved oxidase-like catalytic activity of the nanoparticles for cancer-cell detection without the destructive H2O2. The present work demonstrates a general strategy for the design of multifunctional materials based on bimetallic nanoparticles for different applications, such as biosensors, nanocatalysts and nanomedicine.

In this paper, we report the physical and magnetic characterizations on iron oxide nanoparticle products from Ocean NanoTech. Standard characterization tools such as vibrating-sample magnetometry, X-ray diffraction, dynamic light scattering, transmission electron microscopy, and zeta potential analysis are used to provide MNP customers and researchers with an overview of these iron oxide nanoparticle products. In addition, the dynamic magnetic responses of these iron oxide nanoparticles in aqueous solutions are investigated under low- and high-frequency alternating magnetic fields, giving a standardized operating procedure for characterizing the MNPs from Ocean NanoTech, thereby yielding the best of MNPs for different applications.This article has not yet been cited by other publications.

Abstract:To demonstrate their applications in biological and medical fields such as in immunoassays, magnetic separation of cells or proteins, drug or gene delivery, and magnetic resonance imaging, the template-free syntheses of water-soluble and surface functionalized magnetic nanomaterials have become essential and are challenging. Herein, we developed a facile one-pot template-free method for the preparation of amine-functionalized magnetite nanoparticles and hollow nanospheres by using FeCl(3)(.)6H(2)O as single iron source. These magnetic nanomaterials were characterized by TEM, SEM, XRD, and FTIR technologies. Their magnetic properties were also studied by using a superconducting quantum interference device (SQUID) magnetometer at room temperature. Then the amine-functionalized magnetite nanoparticles were applied to immunoassays and magnetic resonance imaging in live mice.

Abstract:Artificial enzyme mimetics are a current research interest because natural enzymes bear some serious disadvantages, such as their catalytic activity can be easily inhibited and they can be digested by proteases. A very recently study reported by Yan et al. has proven that Fe(3)O(4) magnetic nanoparticles (MNPs) exhibit an intrinsic enzyme mimetic activity similar to that found in natural peroxidases, though MNPs are usually thought to be biological and chemical inert (Gao, L. Z.; Zhuang, J.; Nie, L.; Zhang, J. B.; Zhang, Y.; Gu, N.; Wang, T. H.; Feng, J.; Yang, D. L.; Perrett, S.; Yan, X. Y. Nat. Nanotechnol. 2007, 2, 577-583). In the present work, we just make use of the novel properties of Fe(3)O(4) MNPs as peroxidase mimetics reported by Yan et al. to detect H(2)O(2). The Fe(3)O(4) MNPs were prepared via a coprecipitation method. The as-prepared Fe(3)O(4) MNPs were then used to catalyze the oxidation of a peroxidase substrate 2,2'-azino-bis(3-ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt (ABTS) by H(2)O(2) to the oxidized colored product (see eq 1) which provides a colorimetric detection of H(2)O(2). As low as 3 x 10(-6) mol/L H(2)O(2) could be detected with a linear range from 5 x 10(-6) to 1 x 10(-4) mol/L via our method. More importantly, a sensitive and selective method for glucose detection was developed using glucose oxidase (GOx) and the as-prepared Fe(3)O(4) MNPs. The detection platforms for H(2)O(2) and glucose developed in the present work not only further confirmed that the Fe(3)O(4) MNPs possess intrinsic peroxidase-like activity but also showed great potential applications in varieties of simple, robust, and easy-to-make analytical approaches in the future.

Abstract:To a certain extent, uneven coating of nanoparticles may lead to exposure of their contents. In this work, a simulation system was established and exposed Fe3O4 magnetic nanoparticles (MNPs) were used to investigate the influence of the exposed case in a chemiluminescence immunoassay. Some factors such as the concentration of MNPs, coexistence of horseradish peroxidase (HRP), different particle sizes, and various treatment methods were investigated. It was confirmed that both the bodies of nanoparticles and metallic oxides on the exposed surface have associations with the detection signal. This conclusion may provide a new way to explain the deviation emerged in a chemiluminescence immunoassay. In addition, an exposed Fe3O4 MNPs based chemiluminescence immunoassay was developed to detect human chorionic gonadotropin (hCG). Carboxyl-functionalized MNPs were modified with anti-alpha-hCG antibody, and further used as solid-phase supports for sandwich-type immunoassay. The detection limit for hCG under optimal conditions was as low as 1.184 ng/mL with a linear response range of 2-100 ng/mL and a correlation coefficient of 0.989. The proposed method could be used for the determination of hCG in serum samples, which demonstrated their application potential in rapid and cost-effective detection of biological samples. Abstract:A rapid ligand-exchange method was developed to transfer high quality hydrophobic magnetite nanocrystals into water-soluble NPs by using protocatechuic acid as a ligand via homogenous reaction. After ligand exchange, the magnetite nanocrystals not only exhibited outstanding stability in water, but also maintained high crystallinity and saturation magnetization. Cell viability experiments demonstrated good biocompatibility of the NPs. For 12 nm magnetite nanoparticles (NPs), the small hydrodynamic size of 14 nm enabled a high T-1 relaxivity of 17.8 mM(-1) s(-1) while high saturation magnetization of 77.8 emu g(-1) enabled the NPs to exhibit a high T-2 relaxivity of 220 mM(-1) s(-1) in MRI phantom experiments. In vivo MR imaging experiments further confirmed that the NPs were eminent T-1 and T-2 contrast agents. Moreover, the high quality NPs can be used as excellent magnetic heating agents under an alternating magnetic field. With all those features, including multimodality imaging and magnetic hyperthermia, the NPs can be used as single compound multifunctional agents for various biomedical applications, especially for cancer diagnosis and therapy.

Abstract:Ferroferric oxide nanoparticle (denoted as Nano-Fe3O4) has low toxicity and is biocompatible, with a small particle size and a relatively high surface area. It has a wide range of applications in many fields such as biology, chemistry, environmental science and medicine. Because of its superparamagnetic properties, easy modification and function, it has become an important material for addressing a number of specific tasks. For example, it includes targeted drug delivery nuclear magnetic resonance (NMR) imaging in biomedical applications and in environmental remediation of pollutants. Few articles describe the preparation and modification of Nano-Fe3O4 in detail. We present an evaluation of preparation methodologies, as the quality of material produced plays an important role in its successful application. For example, with modification of Nano-Fe3O4, the surface activation energy is reduced and good dispersion is obtained.

Abstract:Magnetic-optical nanocomposites, which consist of iron oxide nanoparticles and quantum dots, have aroused considerable research interest owing to their unique magnetic and luminescent properties. However, the luminescence of quantum dots tends to be quenched by the magnetic component. Resonant Raman scattering (RRS) of semiconductor nanocrystals, which originates from a strong electron-phonon interaction, provides a new candidate property to be exploited as an analytical tool. Herein, Fe3O4/ZnS nanocomposites were prepared with the seed-mediated growth approach. The nanocomposites exhibited superparamagnetism and characteristic multi-order phonon resonant Raman lines. By using biofunctionalized Fe3O4/ZnS nanocomposites as RRS probes, a femtomolar immunoassay of a model protein, human IgG, was realized with the assistance of an external magnetic field. The results provide guidance for the broad applications of magnetic-optical nanocomposites.

Abstract:Immune magnetite nanoparticles (MNPs) are prepared by four successive reactions, which are MNPs preparation, silica-coating, surface modification with amino group, and conjugation with bio-molecule, respectively. The crystal structure and morphology of intermediate products are characterized by XRD, TEM and AFM. Qualitative and quantitative assays for amino group on the MNPs’ surface are made by FTIR and Organic Element Assay. Ultraviolet–visible absorption spectrum can indirectly illustrate the quantity of bio-molecule conjugated with MNPs. In addition, specific combination and nonspecific combination of immune MNPs are measured by commercial RIA box. The results show that the size of MNPs prepared is 10 ± 5 nm, and silica-coated MNPs with spinel structure have quasi-spherical morphology. Infrared absorption bands of –NH 2 are appeared around 3380–3200 cm −1 and 1650–1510 cm −1 , and the amino group content is 0.5 μmol –NH 2 per mg MNPs. The specific immune combination of immune MNPs is up to 75%, and nonspecific combination is under 5%.

Abstract:Fe3O4 based magnetic polymer nanoparticles (MPNPs) are densely studied for several decades. These Fe3O4 based MPNPs can be used in wastewater treatment and biological field such as magnetic resonance imaging contrast agents, hyperthermia therapy and protein separation. The Fe3O4 based MPNPs are attractive because they combine the advantages of magnetism and polymers together. In order to obtain the practical application in the above mentioned areas, the bare Fe3O4 needs to be functionalised with different kinds of molecules like organic small molecules and polymers and some inorganic molecules like silica, metals and carbon. In this review, the chemical preparation methods, different modification methods and various applications of the Fe3O4 based MPNPs are introduced.

Abstract:Magnetic materials are featured by their arrangement in dipoles in the absence and presence of external magnetic field, and the applications of magnetic materials are designed accordingly. Iron oxide nanoparticles (IONPs) have been widely studied in recent decades due to their promise in a wide range of applications such as data storage and in the biomedical and catalytic fields. Among the various phases of IONPs, Fe3O4 nanoparticles (NPs) were the most typical and widely used. This chapter introduces some established colloidal chemical synthetic protocols toward Fe3O4 NPs, which mainly include co-precipitation process, thermal decomposition of organometallic precursors, hydrothermal synthesis, chemical reduction of metal salts, sol-gel methods, and polyol process. Some promising biological applications of IONPs are subsequently illustrated, especially in cancer diagnostics and therapy, including magnetic resonance imaging (MRI), magnetic hyperthermia (MH), magnetic targeting, and cell tracking.

Abstract:Diverse magnetite (Fe3O4) nanostructures have been fabricated by a facile, one-pot, economic and environment-friendly solvent thermal process simply using ethanol as the solvent, including regular magnetite sub-micrometer spheres composed of partially ordered array of magnetite nanoparticles, hollow magnetite aggregates and monodispersed magnetite nanoparticles. The prepared magnetite nanoparticles possess distinctively superparamagnetic behavior, showing great potential in biotechnological applications. Importantly, this general synthetic strategy could be extended to prepare other spinel ferrite nanoparticles such as CoFe2O4. A novel hybrid micelle self-assembly mechanism was introduced and demonstrated to explain the formation procedure of these special nanostructures. Furthermore, the magnetic resonance imaging capability of prepared magnetite nanoparticles was investigated both in vitro and in vivo.

Abstract:Magnetic nanoparticles (MNPs) have been extensively used in drug/gene delivery, hyperthermia therapy, magnetic particle imaging (MPI), magnetic resonance imaging (MRI), magnetic bioassays, and so forth. With proper surface chemical modifications, physicochemically stable and nontoxic MNPs are emerging contrast agents and tracers for in vivo MRI and MPI applications. Herein, we report the high magnetic moment, irregularly shaped gamma'-Fe4N nanoparticles for enhanced hyperthermia therapy and T-2 contrast agent for MRI application. The static and dynamic magnetic properties of gamma'-Fe4N nanoparticles are characterized by a vibrating sample magnetometer (VSM) and a magnetic particle spectroscopy (MPS) system, respectively. Compared to the gamma-Fe2O3 nanoparticles, gamma'-Fe4N nanoparticles show at least three times higher saturation magnetization, which, as a result, gives rise to the stronger dynamic magnetic responses as proved in the MPS measurement results. In addition, gamma'-Fe4N nanoparticles are functionalized with an oleic acid layer by a wet mechanical milling process. The morphologies of as-milled nanoparticles are characterized by transmission electron microscopy (TEM), dynamic light scattering (DLS), and nanoparticle tracking analyzer (NTA). We report that with proper surface chemical modification and tuning on morphologies, gamma'-Fe4N nanoparticles could be used as tiny heating sources for hyperthermia and contrast agents for MRI applications with minimum dose.

Abstract:Magnetic nanospheres have numerous applications in biomedicine, biotechnology and wastewater treatment, due to their high surface area, tunable sphere size and superparamagnetic properties. Magnetic nanoparticles can be designed and endowed with optical, electronic and fluorescent properties, allowing a wide range of functionality. Multifunctional magnetic particles with heterodimer structures allow various kinds of target molecules to be attached onto their specific parts via affinity or coordinate bonding, etc. The abilities of these nanodevices, including the encapsulation of target molecules in magnetic hybrid nanostructures and easy magnetic separation in the presence of external magnetic fields, show much promise for magnetic imaging, magnetic separation and drug delivery. Consequently, magnetic particles offer excellent potential future uses in disease diagnosis, hyperthermia, immunoassays, electrochemical biosensors, contaminated water treatment and optical detection. In this article, we review the preparation and application of inorganic and organic magnetic composite spheres in the fields of magnetic separation, drug delivery, hyperthermia, magnetic resonance imaging, and others. The size, specific surface area, structure, magnetic properties and surface functional groups of nanospheres have a great influence on their effectiveness in these applications. Facile Synthesis of Iron Oxide Nanozymes for Synergistically Colorimetric and Magnetic Resonance Detection Strategy

Abstract:Iron oxide nanomaterials with mimic enzymes activity have been paid more attention in the clinical diagnosis field. The modified surface molecules would influence the catalytic activity of nanozyme, which is worth studying. Furthermore, the traditional detection strategy is based on colorimetric change of substrates, however, the optical signal is easy to be interfered in complex biological applications. In our research, an efficient and facile preparation strategy was developed to obtain functional artificial nanozymes. Herein, three kinds of surfactants, including citrate acid, poly(ethylene glycol) bis (carboxymethyl) ether and tannic acid have been applied to modify these nanomaterials that showed uniform size, high soluble dispersity and stability. Furthermore, these nanozymes exhibited different peroxidase-like activity to catalyze the hydrogen peroxide and 3,3',5,5'-tetramethylbenzidine. More importantly, magnetic relaxation effect of iron oxide nanozymes was found to be changed during the catalytic reaction. In addition, the relationship between the magnetic signal of nanozymes and the substrate concentration showed a good linear dependence. Combined with the natural enzymes, the magnetic detection of iron oxide nanozymes also exhibited excellent substrate specificity. On these bases, a dual-function specific assay was constructed and further used for glucose detection. In conclusion, this study demonstrated an efficient iron oxide nanozymes preparation method and constructed a new synergistically colorimetric-magnetic diagnosis strategy.
