Abstract:N-aryltriazole nucleosides are new chemical entities with potential biological activity. The two phosphor ligands, Synphos and Xantphos, had a selective and effective impact on Pd-catalyzed C-N coupling with the 5- and 3-bromotriazole acyclonucleoside isomers, affording the corresponding and otherwise difficult to achieve N-aryltriazole nucleosides with good to excellent yields. In addition, two of the synthesized nucleosides showed superior anticancer activity against drug-resistant pancreatic cancer, compared to the reference drug gemcitabine.

Abstract:Five new tetradentate ligands and their corresponding palladium complexes, [Pd(L)]Cl2 (L = N,N′-dimethyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-diethyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-dipropyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-ditert-butyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-dicyclohexyl-1,10-phenanthroline-2,9-dimathanamine) have been synthesized. The ligands and their complexes have been characterized by elemental analysis, IR, and 1H NMR. The complexes have been assayed for antitumor activity in vitro against the mouse leukemia L1210 and the mouse liver carcinoma Bel7402 cell lines. The results showed that the activities of these complexes are significantly dependent on the nature of the alkyl groups on the coordinated amine moieties, and three of these palladium complexes showed lower ID50 values against the two cell lines than cisplatin.

Abstract:Novel N-aryltriazole nucleosides were synthesized via Cu-mediated C–N cross-coupling reaction starting with 3-aminotriazole ribonucleoside and various boronic acids. Two of them exhibited potent apoptosis-related antiproliferative activity against the drug-resistant pancreatic cancer cell line MiaPaCa-2, with an increased potency compared to gemcitabine, the reference treatment for pancreatic cancer. A preliminary SAR study suggests that the appended N-aryl moiety and the substituent at its para-position, as well as the ribose sugar component, contribute considerably to the observed antiproliferative activity.

This transformation is achieved by utilizing simple and readily available base as promoter via C–O bond cleavage, offering a new amination strategy to access several anticancer-active compounds. Abstract:An SAR study revealed that NHC‐Pd complexes bearing C^N ligands with stronger electron donation and moderate steric hindrance are more cytotoxic towards HepG2 and Caco2 cancer cells. A series of mixed N‐heterocylic carbene (NHC) palladium complexes bearing C^N bidentate ligands derived from benzylamine, phenethylamine, and biphenylamine were prepared and fully characterized by NMR spectroscopy, ESI‐MS spectrometry, and X‐ray diffraction analyses. The complexes were potent in inhibiting proliferation of HepG2 and Caco2 cancer cells, and a SAR (structure–activity relationship) study revealed C^N ligands with stronger electron donation and moderate steric hindrance are more cytotoxic. DNA binding study through UV–vis absorption titration indicated that such complexes bound stronger to CT‐DNA molecules with higher binding constants. Additional molecular docking of the best performer suggested a groove binding mode through hydrogen bonding and π‐π stacking interactions. The apoptosis induced by the two best performers of α,α‐dimethylbenzylamine and S‐α‐methylbenzylamine ligated complexes was further analyzed by morphology change, cell cycle distribution, ROS production, and mitochondrial membrane potential studies.

Abstract:Catellani reactions of aryl iodides allow the double functionalization at the ortho and ipso positions in one pot. Catellani reactions involving a nitrene intermediate are not yet known. In this paper, a few palladium amino complexes were prepared from PdCl2, anthranil and iodoarenes, and their structures were determined by X-ray single-crystal diffraction. These complexes are supported by a pincer C,N,O tridentate ligand forming fused six-membered palladacycles. The high stability of the palladium complexes inhibits their reductive elimination.

Abstract:A series of novel pyridine stabilized Pd–NHC complexes were developed, which revealed high activities and broad substrates tolerance in the amination of various (hetero)-aryl chlorides. Besides various secondary amines, a wide range of primary anilines and aliphatic amines were also well tolerated. The results highlight us a new strategy to increase catalyst activity in the future catalyst design by alternating the σ-donor property and flexibility of NHC ligands.

Abstract:Novel N-aryltriazole nucleosides were synthesized via a Cu-mediated C-N cross-coupling reaction, using 3-aminotriazole acyclonucleosides and various boronic acid reagents. Interestingly, N-arylation proceeded much more rapidly on the amide group than oil the amine group, leading to selective N-arylation of the amide functionality on nucleosides containing both groups on the triazole nucleobase.

Abstract:A novel, green, efficient, and stable magnetically heterogeneous nanocatalyst was developed by immobilizing butanesulfonic acid (BuSO3H) onto the surface of MFe2O4 magnetic nanoparticles (MNPs). The resulting core-shell structure of the MFe2O4@PDA@BuSO3H nanocatalyst was thoroughly characterized using various analytical techniques, including Fourier-Transform Infrared Spectroscopy (FT-IR), X-ray Diffraction (XRD), Energy-Dispersive X-ray Spectroscopy (EDS), Field Emission Scanning Electron Microscopy (FESEM), Transmission Electron Microscopy (TEM), Vibrating Sample Magnetometry (VSM), and Brunauer-Emmett-Teller (BET) analysis. A nanocatalyst was used to synthesize 2-benzopyrazine-aminoimidazole hybrid derivatives through a domino multicomponent Knoevenagel-condensation-cyclization reaction (5a-p) in an environmentally friendly manner. The resulting compounds were then tested for their anticancer activity against three types of human cancer cells (MCF-7, A549, and U87) using the MTT assay. The experiment showed that the nanocatalyst had excellent catalytic activity, and the synthesized compounds exhibited promising antiproliferative activity. Notably, compounds 5g and 5h, containing a 2-naphthyl ring, showed the highest antiproliferative effects against MCF-7 cells, with IC50 values of 0.03 and 0.32 μM, respectively. Additionally, the activity of compounds 5g and 5h in tubulin polymerization, apoptosis induction, and cell cycle arrest in MCF-7 cells were investigated. The results demonstrated that these compounds effectively induced apoptosis and cell cycle arrest. The binding of representative compounds to the colchicine binding site of tubulin was confirmed through molecular modeling studies.
