Abstract:An efficient catalytic enantioselective hetero-Diels - Alder reaction of Danishefsky's diene with aldehydes using the magnesium binaphthoxide system has been developed, affording a variety of 2-substituted 2,3-dihydro-4H-pyran-4-ones in high yields and with excellent ee values. The aggregation behavior and nonlinear effect of the catalytic system, as well as the remarkable stereoelectronic effects of ligands on the catalysis, have also been investigated. On the basis of the structure of the isolated reaction intermediate, the stereochemistry of the products, and the information attained from a study of the nonlinear effect, a plausible asymmetric induction model has been proposed. Abstract:Combinatorial coordination chemistry strategy combined with high-throughput screening techniques has been successfully applied to engineering practical enantioselective catalysts for asymmetric hetero-Diels-Alder reaction. The reaction of Danishefsky's diene with a variety of aldehydes can be carried out with 0.1-0.005 mol % of H4-BINOL/Ti/H4-BINOL or H4-BINOL/Ti/H8-BINOL catalysts at room temperature under solvent- and MS-free conditions to afford dihydropyrone derivatives with up to quantitative yield and 99.8% ee.

Abstract:This paper describes the successful development of a series of chiral zinc catalysts containing (R)-3,3′-Br2-BINOL ligand and various diimine activators for enantioselective HDA reaction of Danishefsky's diene with aldehydes through a combinatorial approach, affording the corresponding 2,3-dihydro-4H-pyran-4-one derivatives in excellent yields and enantioselectivities. The application of this type of catalysts was also extended to the diethylzinc addition to the benzaldehyde, affording the corresponding secondary alcohol with up to 94.5% ee under optimized conditions. On the basis of these facts, the integration of two distinct enantioselective reactions, HDA and diethylzinc addition reactions, has been realized in one-pot with the promotion of a single chiral zinc catalyst in a sequential manner. The impact of diimine additive on the catalytic system of HDA reaction was also investigated by probing the nonlinear effect of reaction system. The positive nonlinear effect exhibited in the catalytic system could be attributed to the poor solubility of the heterochiral zinc species. On the basis of various experimental findings disclosed in this research, a possible mechanism for the asymmetric induction in the 3,3′-Br2-BINOL/Zn/diimine catalyzed enantioselective HDA reaction of Danishefsky's diene with aldehydes was outlined.

Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Abstract:Cu-catalyzed enantioselective Cu-B(pin) addition to 1,1-disubstituted alkenes generating tertiary alkylcopper complexes that contain a quaternary stereogenic center, followed by the protonation of the complexes are presented. The resulting formal hydroboration products are formed in up to 98% yield and 99:1 enantiomeric ratio. The C-B(pin) moiety can be converted to a variety of useful functional groups. The method is applied to the enantioselective formal synthesis of (R)-ketoprofen. Detailed stereochemical models for enantioselective induction were elucidated through DFT calculations.

Abstract:AbstractA highly enantioselective catalytic system for exo‐Diels–Alder reactions was developed based on the newly discovered bispyrrolidine diboronates (BPDB). Activated by various Lewis or Brønsted acids, BPDB can catalyze highly stereoselective asymmetric exo‐Diels–Alder reactions of monocarbonyl‐based dienophiles. When 1,2‐dicarbonyl‐based dienophiles are used, the catalyst can sterically distinguish between the two binding sites, which leads to highly regioselective asymmetric Diels–Alder reactions. BPDB can be prepared as crystalline solids on a large scale and are stable under ambient condition. Single‐crystal X‐ray analysis of the structure for acid‐activated BPDB indicated that its activation involves cleavage of a labile B←N bond.

Abstract:A class of chiral self-supported catalysts prepared from bis-BINOL ligands and diethyl zinc were successfully applied in the hetereogeneous enantioselective epoxidation of a range of (E)-α,β-unsaturated ketones, affording the corresponding epoxy ketones in high enantioselectivity. The self-supported catalysts were easily recovered from the reaction mixture and were reusable for several consecutive runs.

Abstract:An efficient synthesis of the enantiomerically pure 3,3'-bis-arylated BINOL derivatives is accomplished through the palladium-catalyzed Suzuki-Miyaura coupling of the unprotected 3,3'-dibromo-BINOL with complete retention of enantiopurity. The active catalyst system Pd(OAc)2/BI-DIME has enabled mild reaction conditions at palladium loads as low as 500 ppm.

Abstract:Chiral bis(oxazoline) complexes of Cu(II), Zn(II) and Mg(II) have been immobilized on silica support via hydrogen-bonding interactions. Up to 93% ee is obtained in the Diels–Alder reaction between 3-((E)-2-butenoyl)-1,3-oxazolin-2-one and cyclopentadiene at room temperature with the heterogeneous bis(oxazoline) complexes, and the catalysts can be recycled without losing enantioselectivity. Experimental and theoretical studies show that the reversal of the absolute product configuration upon immobilization of the PhBOX-Cu(II) catalyst is triggered by the anion dissociation from Cu(II) onto the surface of the support.

Abstract:The hetero-Diels–Alder reactions of 1-alkyl-3-(tert-butyldimethylsilyl)oxy-1,3-butadienes with ethyl glyoxylate catalyzed by chiral salen–metal complexes have been studied. With a cobalt(II) complex as the catalyst, the reaction of 1-(2-benzyloxyethyl)-3-(tert-butyldimethylsilyl)oxy-1,3-butadiene with ethyl glyoxylate gave the Diels–Alder product in 75% isolated yield with the endo:exo ratio >99:1 and the enantiomeric excess ≥52%. The effects of temperature, catalyst and solvent are also discussed.

Abstract:Four analogous binaphthyl compounds (R)-3a-3d containing (R)-3,3'-bis(2-pyridyl) groups were synthesized by the conjugation of (R)-2,2'-dimethoxy-1,1'-binaphthyl-3,3'-diboronic acid [(R)-2] with 2-bromopyridine, 2-bromo-5-methylpyridine, 2-chloro-4-fluoropyridine and 2-chloro-3-(trifluoromethyl)pyridine via Pd-catalyzed Suzuki reactions, respectively. The application of the four chiral ligands in combination with Et2Zn and Ti(Oi-Pr)(4) to the asymmetric addition of phenylacetylene to various aldehydes has been studied. The results show that (R)-3a and (R)-3b are not good catalysts for the alkynylzinc addition to aldehydes, (R)-3d shows good enantioselectivity only for the alkynylzinc addition to aliphatic aldehydes, and (R)-3c exhibits excellent enantioselectivity for phenylethynylzinc addition to both aromatic and aliphatic aldehydes. All the four chiral ligands produced the opposite configuration of the propargylic alcohols to that of the chiral ligands.

Abstract:The first chiral Bronsted acid-catalyzed asymmetric direct aza hetero-Diels-Alder reaction has been described. The phosphoric acids, prepared from binol and H-8-binol derivatives, have shown catalytic ability for the reaction of cyclohexenone with N-PMP-benzaldimine. A chiral phosphoric acid, derived from 3,3-di(4-chloropheneyl)-H-8-binol, exhibited superior enantioselectivity, affording fairly good yields and enantioselectivities for the reaction of a range of aromatic aldimines with cyclohexenone.
