Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:Construction of a chiral methanamine unit at the C3 position of pyrrole is highly desirable; nevertheless, it remains challenging due to its intrinsic electronic properties. Herein, we present an operationally straightforward and direct asymmetric approach to access -(3-pyrrolyl)methanamines under benign organocatalytic circumstances for the first time. The one-pot transformation proceeds smoothly through an amine-catalyzed direct Mannich reaction of succinaldehyde with various endo-cyclic imines, followed by a Paal-Knorr cyclization with a primary amine. Several N-H/alkyl/Ar α-(3-pyrrolyl)methanamines with an aza-quaternary center have been synthesized with good yields and excellent enantioselectivity.

Abstract:The first enantioenriched synthesis of 1,1,3-trisubstituted (trifluoromethyl)indene derivatives, bearing a quaternary stereogenic carbon center, is reported using a simple chiral sulfinamide phosphine-catalyzed asymmetric allylic alkylation of a mixture of indenes with Morita–Baylis–Hillman carbonates. The resulting derivatives can serve as a valuable synthetic building block for some drugs and natural products. Broad substrate scope and high regio- and enantioselectivity of this reaction were particularly remarkable.

Abstract:Highly enantioselective synthesis of acyclic α-tertiary amines through asymmetric desymmetrization is reported. This approach is based on chiral phosphoric acid mediated, enantioselective, oxidative desymmetrization of 2-substituted 2-nitro-1,3-diolbenzylidine acetals in the presence of DMDO as an oxidant. The method allows for the formation of a wide variety of chiral 2-nitro-1,3-diols in high enantioselectivity, which could be transformed into optically pure, unnatural α-alkyl series. The synthetic utility of this method has been further demonstrated by the expedient construction of the core structure of natural products manzacidins enantioselectively.

Abstract:We present herein an enantioselective protocol for the chiral phosphoric acid-catalyzed addition of 3-arylisoxazol-5-amines to highly reactive 3-methide-3-pyrroles to provide a diverse range of heterotriarylmethanes bearing an amino acid moiety in good yields (up to 97%) and high enantioselectivities (up to 93% ee) under mild conditions. The chiral spirocyclic phosphoric acid is crucial in converting the initial 1-pyrrol-3-yl carbinols into reactive 3-methide-3-pyrroles and obtaining the good enantiocontrol, thereby facilitating the desired enantioselective transformation.

Abstract:The catalytic enantioselective alkynylation of isatins, catalyzed by a Zn(OCOCF3)(2)/chiral bis(sulfonamide)-diamine (BSDA) catalyst under mild condition, has been developed. The desired chiral 3-alkynyl-3-hydroxy-2-oxindoles were obtained in moderate to excellent yields (up to 96%) and good to excellent enantioselectivites (up to 97% ee). Abstract:The synthesis of unnatural α-amino acid derivatives has attracted considerable interest in recent years, as they are ubiquitous in protein synthesis and peptide-based drug discovery. Herein, we reported the chiral phosphoric acid catalyzed asymmetric reaction of indoles with β,γ-alkynyl-α-imino esters for the enantioselective synthesis of unnatural indole-based α-amino acid derivatives. This asymmetric organocatalysis protocol enables efficient synthesis of unnatural α-amino acid derivatives with a tetrasubstituted stereogenic center and an alkyne moiety with up to 99% yield and 98% ee, resulting in operationally simple conditions, short reaction time, and broad substrate scope.

Abstract:An enantioselective synthesis of unnatural pyrazole-based alpha-chiral amino acid derivatives from the asymmetric reaction of N-aryl-5-aminopyrazoles with beta,gamma-alkynyl-alpha-imino esters using a chiral spirocyclic phosphoric acid catalyst was developed. Using the established methodology, various pyrazole-based alpha-amino acid derivatives with tetrasubstituted carbon stereocenters were obtained in 67-98% yields and with 73-99% enantioselectivities. The NH2 functionality in the corresponding products enables further transformations to a chiral thiourea and a lactam.

Abstract:A chiral amine (DHQD)2PHAL catalyzed 3‐allylation of indolin‐2‐imines and MBH carbonates has been established, affording enantioenriched 3‐allylindoles in high yields. Importantly, based on our previous work, we were able to obtain each enantiomer of chiral 3‐allylindoles by fine‐tuning the catalytic system. With chiral amine (DHQD)2PHAL as nucleophilic catalyst, asymmetric synthesis of enantioenriched 3‐allylindoles was achieved from indolone‐2‐imines and MBH carbonates. Compared with our previous chiral phosphine catalysis, the chiral amine had exactly the opposite effect in terms of enantioselectivity. Importantly, we could obtain each enantiomer by adjusting the catalyst system.

Abstract:An efficient organocatalytic atroposelective three-component cascade reaction of 2,3-diketoesters, aromatic amines, and 1,3-cyclohexanediones has been developed for the highly enantioselective synthesis of axially chiral N-arylindoles. The success of this method derives from the use of a newly developed second-generation chiral spirocyclic phosphoric acid as the catalyst. In addition, this protocol was extended to the synthesis of an axially chiral monophosphorus ligand.

Abstract:With the aid of chiral phosphoric acid, enantioselective 1,6-addition of tryptamines to in situ formed alkynyl 7-methylene-7H-indoles from tertiary α-(7-indolyl)methanols has been established for the first time, furnishing a broad range of axially chiral tetrasubstituted allenes featuring indole and hexahydropyrrolo[2,3-b]indole unit in good to high yields and asymmetric induction. Importantly, this protocol not only enriches the chemistry of α-(7-indolyl)methanols, but also provides an efficient access for preparation of structurally diversified axially chiral tetrasubstituted allenes.

Abstract:We have accomplished an efficient asymmetric synthesis of (-)-pentazocine and (-)-metazocine from the readily available d-tyrosine, featuring a ring-closing metathesis (RCM) reaction for the formation of the C ring and an intramolecular Friedel-Crafts reaction for the assembly of the B ring. The new strategy established herein should be applicable to enantioselective synthesis of a broad range of chiral benzomorphan analogues, thereby facilitating the biological and medicinal chemistry studies of these clinically important molecules.

Abstract:An efficient method for the synthesis of five-membered chiral propargylic amines from 2-aryl-3H-indol-3-one and alkynylsilanes has been developed. The reaction proceeded under the catalytic system of PtCl(4), oxazoline-based ligand L11, Zn(CF(3)COO)(2), and AcOH in DCE at 95 °C via in situ desilylation of TMS-alkynes. This methodology also highlights a new protocol for the in situ desilylation of alkynylsilanes. The reaction showed a broad substrate scope with good yields and enantioselectivity.
