Abstract:Significant amount of studies demonstrated that the abnormal expression of microRNAs were involved in the carcinogenesis and progression of various kinds of human cancers, also including osteosarcoma. Therefore, understanding the physiological and molecular mechanisms of miRNAs in osteosarcoma may provide novel therapeutic strategies for treatments of patients with osteosarcoma. In this study, we investigated the expression, biological roles of microRNA-486 (miR-486) on osteosarcoma and its underlying mechanism. The results showed that miR-486 was significantly down-regulated in osteosarcoma tissues and osteosarcoma cell lines. In addition, overexpression of miR-486 inhibited cell proliferation, migration and invasion of osteosarcoma. Moreover, SIRT1 was identified as a direct target gene of miR-486 in osteosarcoma. SIRT1 knockdown could mimics the functions of miR-486 over-expression in osteosarcoma cells. In conclusion, the data of the present study provides compelling evidences that miR-486 acted as a tumor suppressor in osteosarcoma through targeting SIRT1 and could serve as a novel therapeutic target.
MicroRNA-211 inhibits tumor growth and invasion through directly targeting RUNX 2 in osteosarcoma

Abstract:Osteosarcoma is the most common primary neoplasm of the bone and mainly occurs in adolescents and young adults. Despite great progress in current treatments, approximately one-third of patients with OS will not survive for more than five years, and fewer than 50% will live more than ten years. Aberrantly expressed microRNAs contribute to the carcinogenesis and progression of human cancers, including osteosarcoma. Increasing studies suggested that microRNA-211 (miR-211) might play important roles in the development of various cancer types; however, the expression patterns, roles and its underlying mechanism of miR-211 in osteosarcoma remain largely unexplored. In this study, we demonstrated that miR-211 was significantly downregulated in osteosarcoma tissues and cell lines compared with their adjacent normal tissues and human normal osteoblastic cell line, respectively. Functional studies showed that resumption of miR-211 inhibited cell proliferation, migration and invasion in osteosarcoma. Runt-related transcription factor 2 (Runx2) was validated as a direct target gene of miR-211. In addition, RUNX2 mRNA expression was upregulated in osteosarcoma tissues and inversely correlated with miR-211 expression. Furthermore, upregulation of RUNX2 could partially rescue the antitumor effects of miR-211 in OS cells. Taken together, these findings indicated the essential roles of miR-211 in suppressing osteosarcoma progression, suggesting miR-211 as a potential therapeutic target for the treatment of osteosarcoma.

Abstract:Osteosarcoma (OS) is the most common type of primary bone tumor and accounts for similar to 60% of all malignant bone tumors in children and adolescents. A large number of studies have proposed that the dysregulated and dysfunctional microRNAs may serve important roles in the occurrence, progression and metastasis of various types of human cancer, including OS. MicroRNA-493 (miR-493) has been identified to act as a tumor suppressor in several types of human cancer. However, little is known regarding the expression pattern and clinical significance of miR-493 in OS. In the present study, reverse transcription-quantitative polymerase chain reaction analysis revealed that miR-493 was markedly downregulated in OS tissues and cell lines and a low miR-493 level were associated with distant metastasis and clinical stage. Furthermore, functional experiments demonstrated that enforced expression of miR-493 led to a significant decrease in OS cell proliferation and invasion in vitro. Furthermore, through bioinformatics analysis, specificity protein 1 (SP1) was identified as a direct target gene of miR-493 in OS. Its expression was upregulated in OS tissues and was negatively associated with miR-493 expression levels. Inhibition of SP1 expression also suppressed the proliferation and invasion of OS, exerting a similar effect to that induced by miR-493 overexpression. These results suggested that miR-493 inhibited OS cell proliferation and invasion through negative regulation of SP1. Therefore, miR-493/SP1 may represent a potential therapeutic target for the treatment of OS.

Abstract:Background/Aims: MicroRNAs (miRNAs) are a class of small noncoding RNAs that regulate gene expression by repressing translation or cleaving RNA transcripts in a sequence-specific manner. Downregulated microRNAs and their roles in cancer development have attracted much attention. A growing body of evidence showed that microRNA-133a (miR-133a) has inhibitory effects on cell proliferation, migration, invasion, and metastasis of osteosarcoma. Methods: MiR-133a expression in human osteosarcoma cell lines and human normal osteoblastic cell line hFOB was investigated by real-time PCR (RT-PCR). The role of miR-133a in human osteosarcoma growth and invasion was assessed in cell lines in vitro and in vivo. Then, luciferase reporter assay validated IGF-1R as a downstream and functional target of miR-133a, and functional studies revealed that the anti-tumor effect of miR-133a was probably due to targeting and repressing of IGF-1R expression. Results: MiR-133a was lower expressed in human osteosarcoma cell lines than human normal osteoblastic cell line hFOB and its effect on inhibiting proliferation, invasion and metastasis is mediated by its direct interaction with the IGF-1R. Furthermore, the tumour-suppressive function of miR-133a probably contributed to inhibiting the activation AKT and ERK signaling pathway. Conclusion: MiR-133a suppresses osteosarcoma progression and metastasis by targeting IGF-1R in human osteosarcoma cells, providing a novel candidate prognostic factor and a potential anti-metastasis therapeutic target in osteosarcoma.

Abstract:Objective: This study was conducted to dissect the role and potential mechanism of microRNA (miR)-455-3p on osteosarcoma (OS) development.Methods: miR-455-3p and HSF1 expression in OS tissues were detected by RT-qPCR and western blot. Later, gainand loss-of-function assays were implemented in OS cells U-2OS and MNNG. The expression of apoptosis-related genes was measured by RT-qPCR and western blot. MTT, Transwell, scratch test, and flow cytometry were utilized to test OS cell viability, invasion, migration, and apoptosis. The targeting relationship between miR-455-3p and HSF1 was assessed with a dual-luciferase reporter gene assay. The transplantation tumor experiment in nude mice was utilized for in vivo confirmation.Results: Downregulated miR-455-3p and upregulated HSF1 were displayed in OS tissues and cells. Mechanistically, miR-455-3p negatively targeted HSF1. MiR-455-3p inhibition or HSF1 overexpression increased MNNG and U-2OS cell proliferative, invasive, and migrating capabilities, while diminishing U-2OS cell apoptosis. Moreover, HSF1 overexpression negated the impacts of miR-455-3p upregulation on OS cell proliferative, invasive, migrating, and apoptotic abilities. Likewise, overexpressing miR-455-3p curtailed the growth of transplanted OS tumors through HSF1 repression.Conclusion: MiR-455-3p inhibits the development of OS cells by downregulating HSF1, highlighting the possibility of miR-455-3p as an innovative indicator of prognosis and a therapeutic target for OS.& COPY; 2022 The Japanese Orthopaedic Association.
