Abstract:Abstract Nanozyme is a series of nanomaterials with enzyme-mimetic activities that can proceed with the catalytic reactions of natural enzymes. In the field of biomedicine, nanozymes are capturing tremendous attention due to their high stability and low cost. Enzyme-mimetic activities of nanozymes can be regulated by multiple factors, such as the chemical state of metal ion, pH, hydrogen peroxide (H 2 O 2 ), and glutathione (GSH) level, presenting great promise for biomedical applications. Over the past decade, multi-functional nanozymes have been developed for various biomedical applications. To promote the understandings of nanozymes and the development of novel and multifunctional nanozymes, we herein provide a comprehensive review of the nanozymes and their applications in the biomedical field. Nanozymes with versatile enzyme-like properties are briefly overviewed, and their mechanism and application are discussed to provide understandings for future research. Finally, underlying challenges and prospects of nanozymes in the biomedical frontier are discussed in this review. Graphical Abstract

Abstract:Nanozymes are nanomaterials with intrinsic natural enzyme-like catalytic properties. They have received extensive attention and have the potential to be an alternative to natural enzymes. Increasing the atom utilization rate of active centers in nanozymes has gradually become a concern of scientists. As the limit of designing nanozymes at the atomic level, single-atom nanozymes (SAzymes) have become the research frontier of the biomedical field recently because of their high atom utilization, well-defined active centers, and good natural enzyme mimicry. In this review, we first introduce the preparation of SAzymes through pyrolysis and defect engineering with regulated activity, then the characterization and surface modification methods of SAzymes are introduced. The possible influences of surface modification on the activity of SAzymes are discussed. Furthermore, we summarize the applications of SAzymes in the biomedical fields, especially in those of reactive oxygen species (ROS) scavenging and antibacterial. Finally, the challenges and opportunities of SAzymes are summarized and prospected.

Abstract:The manufacture of bionic materials to simulate the natural counterparts has attracted extensive attention. As one of the subcategories of biomimetic materials, the development of artificial enzyme is intensive pursuing. As a kind of artificial enzyme, nanozymes are dedicated to solve the limitations of natural enzymes. In recent years, attributed to the explosive development of nanotechnology, biotechnology, catalysis science, computational design and theory calculation, research on nanozymes has made great progress. To highlight these achievements and help researchers to understand the current investigation status of nanozyme, the state‐of‐the‐art development in nanozymes from fabrication materials to bioapplications are summarized. First different raw materials are summarized, including metal‐based, metal‐free, metal‐organic frameworks‐based, and some other novel matters, which are applied to fabricate nanozymes. The different types of enzymes‐like catalytic activities of nanozymes are briefly discussed. Subsequently, the wide applications of nanozymes such as anti‐oxidation, curing diseases, anti‐bacteria, biosensing, and bioimaging are discussed. Finally, the current challenges faced by nanozymes are outlined and the future directions for advancing nanozyme research are outlooked. The authors hope this review can inspire research in the fields of nanotechnology, chemistry, biology, materials science, and theoretical computing, and can contribute to the development of nanozymes.

Abstract:The prevalence of periodontal and peri-implant diseases has been increasing worldwide and has gained a lot of attention. As multifunctional nanomaterials with enzyme-like activity, nanozymes have earned a place in the biomedical field. In periodontics and implantology, nanozymes have contributed greatly to research on maintaining periodontal health and improving implant success rates. To highlight this progress, we review nanozymes for antimicrobial therapy, anti-inflammatory therapy, tissue regeneration promotion, and synergistic effects in periodontal and peri-implant diseases. The future prospects of nanozymes in periodontology and implantology are also discussed along with challenges.

Abstract:Nanozymes, the catalytic nanomaterials with enzyme-like properties, have attracted enormous interests in recent years. They have been used for wide range applications from biosensing and bioimaging to therapeutics and environmental protection. In this review, we highlighted the recent progress of nanozymes in analytical applications. We first discussed the in vitro applications, which covered the detection of bioactive small molecules, nucleic acids, protein biomarkers, cells, etc. We then discussed the in vivo applications, which included the monitoring of bioactive small molecules in live brains and tumor tissues, the study, of drug efficacy, and the investigation of drug and nanozyme metabolism, etc. Finally, we concluded the potential challenges of nanozymes when applying to analytical chemistry and prospected future directions.

Abstract:Nanozymes are nanomaterials with excellent enzyme-like characteristics. Compared with natural enzymes, nanozymes possess significant advantages, such as low cost, high stability, good durability, controllable activity, easy storage, and excellent reusability, thus they have been widely applied in industrial, medical, and biological fields. In the past decade, nanozymes have made impressive progress in their activity exploration, mechanism study, and advanced applications. In this review, we analyze the source of nanozyme catalytic activities from two aspects, including the structure comparison of natural enzymes and nanozymes and the nano effects of nanomaterials. We also systematically summarize the catalytic mechanism of nanozymes, and discuss the reason for their multiple enzyme-like activities. In addition, RONS regulation-based biological applications are comprehensively reviewed. Finally, we present the current challenges of nanozymes and their future research directions.

Abstract:Since the discovery of the first peroxidase nanozyme (Fe3O4), numerous nanomaterials have been reported to exhibit intrinsic enzyme-like activity toward inorganic oxygen species, such as H2O2, oxygen, and O2•-. However, the exploration of nanozymes targeting organic compounds holds transformative potential in the realm of industrial synthesis. This review provides a comprehensive overview of the diverse types of nanozymes that catalyze reactions involving organic substrates and discusses their catalytic mechanisms, structure-activity relationships, and methodological paradigms for discovering new nanozymes. Additionally, we propose a forward-looking perspective on designing nanozyme formulations to mimic subcellular organelles, such as chloroplasts, termed "nano-organelles". Finally, we analyze the challenges encountered in nanozyme synthesis, characterization, nano-organelle construction and applications while suggesting directions to overcome these obstacles and enhance nanozyme research in the future. Through this review, our goal is to inspire further research efforts and catalyze advancements in the field of nanozymes, fostering new insights and opportunities in chemical synthesis.

Abstract:In the past few decades, researchers have developed lots of artificial enzymes with various materials to mimic the structures and functions of natural enzymes. Recently, nanozymes, nanomaterials with enzyme-like characteristics, are emerging as novel artificial enzymes, and attracting researchers' enormous interest. Remarkable advances have been made in the area of nanozymes due to their unique properties compared with natural enzymes and classic artificial enzymes. Until now, lots of nanomaterials have been studied to mimic various natural enzymes for wide applications. To highlight the recent progress of nanozymes (especially in bionanotechnology), here we discuss the diverse applications of nanozymes, which range from sensing, imaging, and therapeutics, to logic gates, pollutant removal, water treatment, etc. Finally, we address the current challenges facing nanozyme research as well as possible directions to fulfill their great potential in future.

Abstract:Using nanomaterials to mimic the function of protein enzymes is an interesting idea. Many nanomaterials have a similar size as enzymes and they also possess catalytic activity. Over the past decade, a surge of nanozyme work has emerged, likely due to the advancement in the synthesis and characterization of inorganic nanoparticles. Many typical enzymatic reactions mimicking oxidases, peroxidases, laccases, superoxide dismutases, and catalases have been realized by simple metal oxide and metal nanoparticles. In addition, small inorganic catalysts have been loaded in nanoparticles to create another type of nanozyme. The applications of nanozymes in biosensor design, environmental remediation, and therapeutics have been demonstrated. In this Topical Review, we briefly summarize the current status of the field and then focus our attention on some important problems faced by the field. These topics include developing better nanozymes with higher activity, better substrate selectivity, and engineering enzyme-like active sites. For practical applications, reliable methods for bioconjugation of nanozymes with affinity ligands need to be achieved, but not at the cost of losing the activity of nanozymes. Finally, fundamental mechanistic studies are needed to rationally design nanozymes and to obtain key insights into a few model systems.

Abstract:Nanozymes are nanomaterials with intrinsic enzyme-like properties. Since the landmark research report on nanozymes in 2007, numerous scientists focus on the topic of nanozyme. As an artificial enzyme, nanozymes take advantage of good stability, easy modification, designability, ease of preparation, and low cost. In recent years, due to the explosion of nanotechnology, biomimetic science, catalytic science, and theoretical computing, a great number of nanozymes have been fabricated. To highlight these achievements and allow the related researchers to grasp the current research status of structure-dependent nanozymes, the state-of-the-art in nanozymes from different spatial dimensions (0D, 1D, 2D, 3D) to the bioanalysis applications were reviewed. For different spatial dimensions nanozymes, the 0D nanodot, nanosphere, nanocluster; 1D nanorod, nanotube, 2D nanosheet, lamellar structure, and 3D metal-organic frameworks, covalent organic frameworks, hierarchical structure, and hydrogel were all discussed. Furthermore, the nanozymes applied for disease diagnosis, tumor microenvironment sensing, pathogen detection, drug detection, food detection, and environmental sensing were all discussed. Finally, the current challenges faced in nanozymology are outlined and the future directions for advancing nanozyme research are outlooked. We hope this review will inspire related research on nanozymes and contribute to developing nanozymes in bioanalysis.

Abstract:Emerging as cost-effective potential alternatives to natural enzymes, nanozymes have attracted increasing interest in broad fields. To exploit the in-depth potential of nanozymes, rational structural engineering and explicit catalytic mechanisms at the molecular scale are required. Recently, impressive progress has been made in mimicking the characteristics of natural enzymes by constructing metal active sites, binding pockets, scaffolds, and delicate allosteric regulation. Ingenious in-depth studies have been conducted with advances in structural characterization and theoretical calculations, unveiling the "black box" of nanozyme-catalytic mechanisms. This review introduces the state-of-art synthesis strategies by learning from the natural enzyme counterparts and summarizes the general overview of the nanozyme mechanism with a particular emphasis on the adsorbed intermediates and descriptors that predict the nanozyme activity The emerging activity assessment methodology that illustrates the relationship between electrochemical oxygen reduction and enzymatic oxygen reduction is discussed with up-to-date advances Future opportunities and challenges are presented in the end to spark more profound work and attract more researchers from various backgrounds to the flourishing field of nanozymes.

Abstract:As a new generation of artificial enzymes, nanozymes have the advantages of high catalytic activity, good stability, low cost, and other unique properties of nanomaterials. Due to their wide range of potential applications, they have become an emerging field bridging nanotechnology and biology, attracting researchers in various fields to design and synthesize highly catalytically active nanozymes. However, the thorough understanding of experimental phenomena and the mechanisms beneath practical applications of nanozymes limits their rapid development. Herein, the progress of experimental and computational research of nanozymes on two issues over the past decade is briefly reviewed: (1) experimental development of new nanozymes mimicking different types of enzymes. This covers their structures and applications ranging from biosensing and bioimaging to therapeutics and environmental protection. (2) The catalytic mechanism proposed by experimental and theoretical study. The challenges and future directions of computational research in this field are also discussed.

Abstract:Nanozymes, nanomaterial-based artificial enzymes, exhibit potential for emulating the catalytic functions inherent in enzymes. Nanozymes have advantages such as low cost, facile synthesis, high stability and adjustable activities. As a promising approach for healthcare, nanozymes have sparked considerable interest, and have been chosen as one of the 2022 Top Ten Emerging Technologies in Chemistry by the International Union of Pure and Applied Chemistry (IUPAC). This Primer provides theranostic insights from the nanozyme toolbox, encompassing the design of nanoparticles, evaluation of activities and applications. We focus on rational strategies to enhance nanozyme activities, emphasizing standardized evaluations across different activities, and outline specific details for their practical applications. The selection of candidates for diagnosis, as well as those for in vivo theranostic applications, is carefully considered based on appropriateness. We also acknowledge current challenges and limitations, presenting future perspectives and positioning nanozymes as an alternative and effective choice in theranostics. This Primer aims to contribute to the understanding and advancement of nanozyme applications in healthcare, offering a comprehensive guide for researchers in this dynamic field. Abstract:Single-atom nanozymes (SANs) are the new emerging catalytic nanomaterials with enzyme-mimetic activities, which have many extraordinary merits, such as low-cost preparation, maximum atom utilization, ideal catalytic activity, and optimized selectivity. With these advantages, SANs have received extensive research attention in the fields of chemistry, energy conversion, and environmental purification. Recently, a growing number of studies have shown the great promise of SANs in biological applications. In this article, we present the most recent developments of SANs in anti-infective treatment, cancer diagnosis and therapy, biosensing, and antioxidative therapy. This text is expected to better guide the readers to understand the current state and future clinical possibilities of SANs in medical applications.

Abstract:The research on nanozymes has increased dramatically in recent years and a new interdiscipline, nanozymology, has emerged. A variety of nanomaterials have been designed to mimic the characteristics of natural enzymes, which connects an important bridge between nanotechnology and biological science. Unlike natural enzymes, the nanoscale properties of nanozymes endow them with the potential to regulate their enzymatic-like activity from different perspectives. The mechanisms behind those methods are intriguing. In this Review, we introduce these mechanisms from the aspects of surface chemistry, surface modification, molecular imprinting, and hybridization and then focus attention on some specific catalytic mechanisms of several representative nanozymes. The applications of nanozymes ranging from bioassay, imaging, to disease therapy are also discussed in detail to prove the fact that the inherent physicochemical properties of nanomaterials not only make nanozymes the analogues of biological enzymes, but also endow them with incomparable advantages and broad prospects in biomedical fields. Finally, four characteristics and some challenges of nanozymes are summarized.

Abstract:In recent years, with the development of nanomaterials, a slice of nanomaterials has been demonstrated to possess high catalytic activity similar to natural enzymes and counter the dilemmas including easy inactivation and low yield natural of enzymes, which are labeled as nanozymes. The catalytic activity of nanozymes could be easily regulated by size, structure, surface modification and other factors. In comparison with natural enzymes, nanozymes featured with a more stable structure, economical preparation and preservation, diversity of functions and adjustable catalytic activity, thus becoming the potentially ideal substitute for natural enzymes. Generally, the are mainly three types containing metal oxide nanozymes, noble metal nanozymes and carbon-based nanozymes, owing various applications in biomedical, energy and environmental fields. In this review, to summarize the recent representative applications of nanozymes, and potentially explore the scientific problems in this field at the same time, we are going to discuss the catalytic mechanisms of diverse nanozymes, with the emphasis on their applications in the fields of tumor therapy, anti-inflammatory and biosensing, hoping to help and guide the future development of nanozymes.

Recently, a surge of nanozymes have been demonstrated to catalyze some typical enzymatic reactions mimicking oxidase, peroxidase and catalase. Especially, nanozymes with peroxidase-like activity have grown into a big family due to their broad range of applications in the field of biosensing and immunoassay. Since inorganic nanoparticles possess the advantages of high stability and easy surface modification, nanozymes have been emerging alternatives to natural enzymes to some extent. In this Review, we briefly summarize several typical nanozymes and then focus our attention on their enormous applications with respect to analytical chemistry. Representative examples would be discussed in detail from the literatures of last 10 years. Additionally, the current challenges and future directions about nanozymes are speculated at the end of this review.
