Abstract:As a new generation of artificial enzymes, nanozymes have the advantages of high catalytic activity, good stability, low cost, and other unique properties of nanomaterials. Due to their wide range of potential applications, they have become an emerging field bridging nanotechnology and biology, attracting researchers in various fields to design and synthesize highly catalytically active nanozymes. However, the thorough understanding of experimental phenomena and the mechanisms beneath practical applications of nanozymes limits their rapid development. Herein, the progress of experimental and computational research of nanozymes on two issues over the past decade is briefly reviewed: (1) experimental development of new nanozymes mimicking different types of enzymes. This covers their structures and applications ranging from biosensing and bioimaging to therapeutics and environmental protection. (2) The catalytic mechanism proposed by experimental and theoretical study. The challenges and future directions of computational research in this field are also discussed.

Abstract:Abstract Nanozyme is a series of nanomaterials with enzyme-mimetic activities that can proceed with the catalytic reactions of natural enzymes. In the field of biomedicine, nanozymes are capturing tremendous attention due to their high stability and low cost. Enzyme-mimetic activities of nanozymes can be regulated by multiple factors, such as the chemical state of metal ion, pH, hydrogen peroxide (H 2 O 2 ), and glutathione (GSH) level, presenting great promise for biomedical applications. Over the past decade, multi-functional nanozymes have been developed for various biomedical applications. To promote the understandings of nanozymes and the development of novel and multifunctional nanozymes, we herein provide a comprehensive review of the nanozymes and their applications in the biomedical field. Nanozymes with versatile enzyme-like properties are briefly overviewed, and their mechanism and application are discussed to provide understandings for future research. Finally, underlying challenges and prospects of nanozymes in the biomedical frontier are discussed in this review. Graphical Abstract

Abstract:Nanozymes are nanomaterials with intrinsic enzyme-like characteristics that have been booming over the past decade because of their capability to address the limitations of natural enzymes such as low stability, high cost, and difficult storage. Along with the rapid development and ever-deepening understanding of nanoscience and nanotechnology, nanozymes hold promise to serve as direct surrogates of traditional enzymes by mimicking and further engineering the active centers of natural enzymes. In 2007, we reported the first evidence that Fe3O4 nanoparticles (NPs) have intrinsic peroxidase-mimicking activity, and since that time, hundreds of nanomaterials have been found to mimic the catalytic activity of peroxidase, oxidase, catalase, haloperoxidase, glutathione peroxidase, uricase, methane monooxygenase, hydrolase, and superoxide dismutase. Uniquely, a broad variety of nanomaterials have been reported to simultaneously exhibit dual- or multienzyme mimetic activity. For example, Fe3O4 NPs show pH-dependent peroxidase-like and catalase-like activities; Prussian blue NPs simultaneously possess peroxidase-, catalase-, and superoxide dismutase-like activity; and Mn3O4 NPs mimic all three cellular antioxidant enzymes including superoxide dismutase, catalase, and glutathione peroxidase. Taking advantage of the physiochemical properties of nanomaterials, nanozymes have shown a broad range of applications from in vitro detection to replacing specific enzymes in living systems. With the emergence of the new concept of "nanozymology", nanozymes have now become an emerging new field connecting nanotechnology and biology. Since the landmark paper on nanozymes was published in 2007, we have extensively explored their catalytic mechanism, established the corresponding standards to quantitatively determine their catalytic activities, and opened up a broad range of applications from biological detection and environmental monitoring to disease diagnosis and biomedicine development. Here we mainly focus on our progress in the systematic design and construction of functionally specific nanozymes, the standardization of nanozyme research, and the exploration of their applications for replacing natural enzymes in living systems. We also show that, by combining the unique physicochemical properties and enzyme-like catalytic activities, nanozymes can offer a variety of multifunctional platforms with a broad of applications from in vitro detection to in vivo monitoring and therapy. For instance, targeting antibody-conjugated ferromagnetic nanozymes simultaneously provide three functions: target capture, magnetic separation, and nanozyme color development for target detection. We finally will address the prospect of nanozyme research to become "nanozymology". We expect that nanozymes with unique physicochemical properties and intrinsic enzyme-mimicking catalytic properties will attract broad interest in both fundamental research and practical applications and offer new opportunities for traditional enzymology.

Abstract:In the past few decades, researchers have developed lots of artificial enzymes with various materials to mimic the structures and functions of natural enzymes. Recently, nanozymes, nanomaterials with enzyme-like characteristics, are emerging as novel artificial enzymes, and attracting researchers' enormous interest. Remarkable advances have been made in the area of nanozymes due to their unique properties compared with natural enzymes and classic artificial enzymes. Until now, lots of nanomaterials have been studied to mimic various natural enzymes for wide applications. To highlight the recent progress of nanozymes (especially in bionanotechnology), here we discuss the diverse applications of nanozymes, which range from sensing, imaging, and therapeutics, to logic gates, pollutant removal, water treatment, etc. Finally, we address the current challenges facing nanozyme research as well as possible directions to fulfill their great potential in future.

Abstract:The manufacture of bionic materials to simulate the natural counterparts has attracted extensive attention. As one of the subcategories of biomimetic materials, the development of artificial enzyme is intensive pursuing. As a kind of artificial enzyme, nanozymes are dedicated to solve the limitations of natural enzymes. In recent years, attributed to the explosive development of nanotechnology, biotechnology, catalysis science, computational design and theory calculation, research on nanozymes has made great progress. To highlight these achievements and help researchers to understand the current investigation status of nanozyme, the state‐of‐the‐art development in nanozymes from fabrication materials to bioapplications are summarized. First different raw materials are summarized, including metal‐based, metal‐free, metal‐organic frameworks‐based, and some other novel matters, which are applied to fabricate nanozymes. The different types of enzymes‐like catalytic activities of nanozymes are briefly discussed. Subsequently, the wide applications of nanozymes such as anti‐oxidation, curing diseases, anti‐bacteria, biosensing, and bioimaging are discussed. Finally, the current challenges faced by nanozymes are outlined and the future directions for advancing nanozyme research are outlooked. The authors hope this review can inspire research in the fields of nanotechnology, chemistry, biology, materials science, and theoretical computing, and can contribute to the development of nanozymes.

Abstract:Using nanomaterials to mimic the function of protein enzymes is an interesting idea. Many nanomaterials have a similar size as enzymes and they also possess catalytic activity. Over the past decade, a surge of nanozyme work has emerged, likely due to the advancement in the synthesis and characterization of inorganic nanoparticles. Many typical enzymatic reactions mimicking oxidases, peroxidases, laccases, superoxide dismutases, and catalases have been realized by simple metal oxide and metal nanoparticles. In addition, small inorganic catalysts have been loaded in nanoparticles to create another type of nanozyme. The applications of nanozymes in biosensor design, environmental remediation, and therapeutics have been demonstrated. In this Topical Review, we briefly summarize the current status of the field and then focus our attention on some important problems faced by the field. These topics include developing better nanozymes with higher activity, better substrate selectivity, and engineering enzyme-like active sites. For practical applications, reliable methods for bioconjugation of nanozymes with affinity ligands need to be achieved, but not at the cost of losing the activity of nanozymes. Finally, fundamental mechanistic studies are needed to rationally design nanozymes and to obtain key insights into a few model systems.

Abstract:Nanozymes are nanomaterials with intrinsic natural enzyme-like catalytic properties. They have received extensive attention and have the potential to be an alternative to natural enzymes. Increasing the atom utilization rate of active centers in nanozymes has gradually become a concern of scientists. As the limit of designing nanozymes at the atomic level, single-atom nanozymes (SAzymes) have become the research frontier of the biomedical field recently because of their high atom utilization, well-defined active centers, and good natural enzyme mimicry. In this review, we first introduce the preparation of SAzymes through pyrolysis and defect engineering with regulated activity, then the characterization and surface modification methods of SAzymes are introduced. The possible influences of surface modification on the activity of SAzymes are discussed. Furthermore, we summarize the applications of SAzymes in the biomedical fields, especially in those of reactive oxygen species (ROS) scavenging and antibacterial. Finally, the challenges and opportunities of SAzymes are summarized and prospected.

Abstract:Nanozymes are nanomaterial-based artificial enzymes. By effectively mimicking catalytic sites of natural enzymes or harboring multivalent elements for reactions, nanozyme systems have successfully served as direct surrogates of traditional enzymes for catalysis. With the rapid development and ever-deepening understanding of nanotechnology, nanozymes offer higher catalytic stability, ease of modification and lower manufacturing cost than protein enzymes. Additionally, nanozymes possess inherent nanomaterial properties, providing not only a simple substitute of enzymes but also a multimodal platform interfacing complex biologic environments. Recent extensive research has focused on designing various nanozyme systems that are responsive to one or multiple substrates by tailored means. Catalytic activities of nanozymes can be regulated by pH, H2O2 and glutathione concentrations and levels of oxygenation in different microenvironments. Moreover, nanozymes can be remotely-controlled via different stimuli, including a magnetic field, light, ultrasound, and heat. Collectively, these factors can be adjusted to maximize the diagnostic and therapeutic efficacies of different diseases in biomedical settings. Therefore, by integrating the catalytic property and inherent nanomaterial nature of nanozyme systems, we anticipate that stimuli-responsive nanozymes will open up new horizons for diagnosis, treatment, and theranostics.

Abstract:Nanozymes are nanomaterials with excellent enzyme-like characteristics. Compared with natural enzymes, nanozymes possess significant advantages, such as low cost, high stability, good durability, controllable activity, easy storage, and excellent reusability, thus they have been widely applied in industrial, medical, and biological fields. In the past decade, nanozymes have made impressive progress in their activity exploration, mechanism study, and advanced applications. In this review, we analyze the source of nanozyme catalytic activities from two aspects, including the structure comparison of natural enzymes and nanozymes and the nano effects of nanomaterials. We also systematically summarize the catalytic mechanism of nanozymes, and discuss the reason for their multiple enzyme-like activities. In addition, RONS regulation-based biological applications are comprehensively reviewed. Finally, we present the current challenges of nanozymes and their future research directions.

Abstract:Nanozymes are nanomaterials with enzyme-like characteristics (Chem. Soc. Rev., 2013, 42, 6060-6093). They have been developed to address the limitations of natural enzymes and conventional artificial enzymes. Along with the significant advances in nanotechnology, biotechnology, catalysis science, and computational design, great progress has been achieved in the field of nanozymes since the publication of the above-mentioned comprehensive review in 2013. To highlight these achievements, this review first discusses the types of nanozymes and their representative nanomaterials, together with the corresponding catalytic mechanisms whenever available. Then, it summarizes various strategies for modulating the activity and selectivity of nanozymes. After that, the broad applications from biomedical analysis and imaging to theranostics and environmental protection are covered. Finally, the current challenges faced by nanozymes are outlined and the future directions for advancing nanozyme research are suggested. The current review can help researchers know well the current status of nanozymes and may catalyze breakthroughs in this field.

Abstract:The research on nanozymes has increased dramatically in recent years and a new interdiscipline, nanozymology, has emerged. A variety of nanomaterials have been designed to mimic the characteristics of natural enzymes, which connects an important bridge between nanotechnology and biological science. Unlike natural enzymes, the nanoscale properties of nanozymes endow them with the potential to regulate their enzymatic-like activity from different perspectives. The mechanisms behind those methods are intriguing. In this Review, we introduce these mechanisms from the aspects of surface chemistry, surface modification, molecular imprinting, and hybridization and then focus attention on some specific catalytic mechanisms of several representative nanozymes. The applications of nanozymes ranging from bioassay, imaging, to disease therapy are also discussed in detail to prove the fact that the inherent physicochemical properties of nanomaterials not only make nanozymes the analogues of biological enzymes, but also endow them with incomparable advantages and broad prospects in biomedical fields. Finally, four characteristics and some challenges of nanozymes are summarized.

Abstract:Nanozymes with enzyme-like catalytic performances have served as ideal surrogates for nature enzymes in diverse applications due to their low cost and excellent catalytic stability in harsh environments. However, the nanozymes’ intrinsic catalytic activity remains poor compared to their counterpart natural enzymes, limiting their broader development. To date, extensive efforts have been made to improve nanozyme catalytic properties, including activity modulation strategies focused on size, morphology, and facet regulation; ligand modification regulation; heteroatom doping regulation; composition regulation; and external regulation. The heteroatom doping strategy has proven to be an effective method, accelerating electron transfer to improve nanozyme mimic activity. Herein, we summarize current progress on heteroatom-doped nanozymes for biomedical applications. Moreover, we discuss their synthesis methods and influencing factors. Finally, we discuss current challenges and future perspectives for heteroatom-doped nanozymes to highlight some novel and advanced heteroatom-doped nanozymes that establish a bridge between heteroatom-doped nanozymes and natural enzymes for preclinical translations.

Abstract:Because of the high catalytic activities and substrate specificity, natural enzymes have been widely used in industrial, medical, and biological fields, etc. Although promising, they often suffer from intrinsic shortcomings such as high cost, low operational stability, and difficulties of recycling. To overcome these shortcomings, researchers have been devoted to the exploration of artificial enzyme mimics for a long time. Since the discovery of ferromagnetic nanoparticles with intrinsic horseradish peroxidase-like activity in 2007, a large amount of studies on nanozymes have been constantly emerging in the next decade. Nanozymes are one kind of nanomaterials with enzymatic catalytic properties. Compared with natural enzymes, nanozymes have the advantages such as low cost, high stability and durability, which have been widely used in industrial, medical, and biological fields. A thorough understanding of the possible catalytic mechanisms will contribute to the development of novel and high-efficient nanozymes, and the rational regulations of the activities of nanozymes are of great significance. In this review, we systematically introduce the classification, catalytic mechanism, activity regulation as well as recent research progress of nanozymes in the field of biosensing, environmental protection, and disease treatments, etc. in the past years. We also propose the current challenges of nanozymes as well as their future research focus. We anticipate this review may be of significance for the field to understand the properties of nanozymes and the development of novel nanomaterials with enzyme mimicking activities.

Abstract:Nanozyme, a term defined for nanomaterial with enzyme-like properties, has attracted significant research attention owing to its striking merits. Recently, a surge of nanozymes have been demonstrated to catalyze some typical enzymatic reactions mimicking oxidase, peroxidase and catalase. Especially, nanozymes with peroxidase-like activity have grown into a big family due to their broad range of applications in the field of biosensing and immunoassay. Since inorganic nanoparticles possess the advantages of high stability and easy surface modification, nanozymes have been emerging alternatives to natural enzymes to some extent. In this Review, we briefly summarize several typical nanozymes and then focus our attention on their enormous applications with respect to analytical chemistry. Representative examples would be discussed in detail from the literatures of last 10 years. Additionally, the current challenges and future directions about nanozymes are speculated at the end of this review.

Abstract:Single-atom nanozyme (SAzyme) is the hot topic of the current nanozyme research. Its intrinsic properties, such as high activity, stability, and low cost, present great substitutes to natural enzymes. Moreover, its fundamental characteristics, i.e., maximized atom utilizations and well-defined geometric and electronic structures, lead to higher catalytic activities and specificity than traditional nanozymes. SAzymes have been applied in many biomedical areas, such as anti-tumor therapy, biosensing, antibiosis, and anti-oxidation therapy. Here, we will discuss a series of representative examples of SAzymes categorized by their biomedical applications in this review. In the end, we will address the future opportunities and challenges SAzymes facing in their designs and applications.

Abstract:Nanozymes are nanomaterials with intrinsic enzyme-like properties. Since the landmark research report on nanozymes in 2007, numerous scientists focus on the topic of nanozyme. As an artificial enzyme, nanozymes take advantage of good stability, easy modification, designability, ease of preparation, and low cost. In recent years, due to the explosion of nanotechnology, biomimetic science, catalytic science, and theoretical computing, a great number of nanozymes have been fabricated. To highlight these achievements and allow the related researchers to grasp the current research status of structure-dependent nanozymes, the state-of-the-art in nanozymes from different spatial dimensions (0D, 1D, 2D, 3D) to the bioanalysis applications were reviewed. For different spatial dimensions nanozymes, the 0D nanodot, nanosphere, nanocluster; 1D nanorod, nanotube, 2D nanosheet, lamellar structure, and 3D metal-organic frameworks, covalent organic frameworks, hierarchical structure, and hydrogel were all discussed. Furthermore, the nanozymes applied for disease diagnosis, tumor microenvironment sensing, pathogen detection, drug detection, food detection, and environmental sensing were all discussed. Finally, the current challenges faced in nanozymology are outlined and the future directions for advancing nanozyme research are outlooked. We hope this review will inspire related research on nanozymes and contribute to developing nanozymes in bioanalysis.

Abstract:<p>Nanozymes, the nanomaterials with catalytic function similar to natural enzymes, are well-known for their distinct merits such as low cost and high stability. Among them, metal-organic frameworks (MOFs) based nanozymes have attracted more attention. It is not only because of its large surface area, adjustable pore structure and good biocompatibility, but also due to its inorganic-organic hybrid structure which can be designed to own different mimic enzyme activities. In addition, MOFs with the above structural characteristics can be used to construct multi-enzyme cascade system to better simulate natural enzymes. In this review, MOFs-based single and multienzyme systems have been introduced. Besides, we summarized some strategies to enhance their catalytic activity. Moreover, the applications in analytical chemistry, environmental treatment and diseases treatment have been reviewed. Finally, the current challenges and future perspective on the development of MOFs-based nanozymes are discussed.</p>

Abstract:Abstract Nanozymes are nanomaterials with enzyme-like characteristics. As a new generation of artificial enzymes, nanozymes have the advantages of low cost, good stability, simple preparation, and easy storage, allowing them to overcome many of the limitations of natural enzymes in enzymatic therapy. Currently, most reported nanozymes exhibit oxidoreductase-like activities and can regulate redox balance in cells. Nanozymes with superoxide dismutase and catalase activity can be used to scavenge reactive oxygen species (ROS) for cell protection, while those with peroxidase and oxidase activity can generate ROS to kill harmful cells, such as tumor cells and bacteria. In this review, we summarize recent progress in nanozyme-based medicine for enzymatic therapy and highlight the opportunities and challenges in this field for future study.

Till now nanozymes have been used in biomedical sensing. diagnosis and therapeutics. as well as environment protection. Despite of the great success achieved in the past several decades. how to efficiently design nanozymes is still one of the bottlenecks in the field. which is originated from the complicated composition and ambiguity in the active sites of nanomaterials. To tackle these challenges. this insight first summarizes the current efficient design strategies of nanozymes. such as computation-aided high throughput screening. rational design. and biomimetic design. And then. the development of bio-inspired metal-organic framework (MOF) nanozymes. particularly the structure-activity relationship study. is highlighted. At the end. combined with current research trend. several directions and inspirations for the future study are suggested to advance the nanozymes research.

Abstract:Single-atom nanozymes (SANs) are one of the newest generations of nanozymes, which have been greatly developed in the past few years and exploited widely for many applications, such as biosensing, disease diagnosis and therapy, bioimaging, and so on. SANs, possessing dispersed single-atom structures and a well-defined coordination environment, exhibit remarkable catalytic performance with both high activity and stability. In this paper, the most recent progress in SANs is reviewed in terms of their advanced synthesis, characterization, functional mechanisms, performance validation/optimization, and biomedical applications. Several technical challenges hindering practical applications of SANs are analyzed, and possible research directions are also proposed for overcoming the challenges.
