The Protective Effects Of Prenatal Exercise On Kidney In Offspring Mice Under High-fat Diet Exposure

Abstract:Prenatal aerobic exercise has beneficial effects on the health of offspring which could persist into adulthood. However, it is largely unknown whether prenatal exercise could protect kidney health of offspring fed with high-fat diet (HFD) and the mechanisms that underlie the effects are unclear. PURPOSE: To investigate the effects of prenatal exercise on the renal morphology and function in the offspring treated with HFD, and whether it is related to AMPK-SIRT1-PGC-1α pathway. METHODS: pregnant C57BL/6 N mice were separated into a sedentary group and an exercise group at random. The exercise group was given a moderate-intensity swimming training (60 min/day, 6 day/week, gestation day 1.5-16.5). Male offspring at weaning were subjected to 8 weeks of HFD (p-SED-HFD, p-EX-HFD) while the control group received a standard diet (p-SED-NFD, p-EX-NFD). Renal function, morphology and protein expressions of male offspring were evaluated after 8-week HFD. RESULTS: 1) Prenatal exercise decreased the body weight (30.13 ± 0.47 vs. 26.52 ± 0.80 g, p < 0.05) and kidney coefficients (7.34 ± 0.95 vs. 6.29 ± 0.26, p < 0.05) in the offspring of HFD treatment. 2) Serum creatinine and urea nitrogen concentration in exercise groups showed no significantly differences compared to the sedentary groups (p > 0.05), demonstrating that renal function was not declined after 8 weeks of HFD. 3) HE staining showed that p-SED-HFD group had increased glomerular area (1.42 ± 0.02 vs. 1.00 ± 0.01) compared with NFD groups. However, there was a significant decrease in p-EX-HFD group (1.16 ± 0.02, both p < 0.05). 4) MASSON and Oil red staining showed that prenatal exercise decreased collagen area (2.08 ± 0.07 vs. 1.58 ± 0.03) and lipid accumulation area (7.39 ± 0.19 vs. 5.91 ± 0.07, both p < 0.05) of kidney in HFD groups. 5) The protein expression levels of p-AMPK/AMPK (0.88 ± 0.05 vs. 1.24 ± 0.09, p < 0.05), SIRT1 (0.68 ± 0.08 vs. 0.88 ± 0.11, p < 0.05) and PGC-1α (0.74 ± 0.08 vs. 0.90 ± 0.13, p > 0.05) in p-EX-HFD were higher than those in the p-SED-HFD group. CONCLUSION: Prenatal exercise can decrease kidney coefficients and improve renal morphology by shrinking collagen and lipid accumulation in kidney of HFD offspring, which probably relates to AMPK-SIRT1-PGC-1α pathway.
