Abstract:Benzo[b]fluorenes were synthesized via CuI-catalyzed cascade reactions of 2-alkynylbenzaldehyde N-tosylhydrazones and aromatic terminal alkynes in the presence of base including the formation of the benzoenyne-allene intermediate in situ and its Schmittel cyclization. Density functional theory calculation was performed to give the energy difference for the formation of 5phenyl-11H-benzo[b]fluorene and 1,2-diphenyl-1H-cyclobuta[a]indene from the proposed diradical intermediates.

Abstract:We herein disclose a highly efficient one-pot synthetic strategy for dihydrobenzo[a]fluorenes via cascade rhodium(III)-catalyzed ortho-C-H activation/annulation of thiobenzamides with aryl ethynyl ketones and subsequently copper(II)-promoted intramolecular C-H/C-H cross-coupling reactions. Mechanistic investigations suggest that Cu(II) plays two crucial roles by serving as a sulfide scavenger to regenerate the Rh(III) catalyst and promoting the intramolecular C-H/C-H cross-coupling reaction. This protocol greatly streamlines accesses to a variety of appealing tetracyclic benzo[a]fluorene skeletons, which may have potential biological activity and medicinal properties.

Abstract:Here, a general copper-catalyzed radical cascade carbocyclization reaction with 2-arylbenzoimidazoles and a Togni reagent was realized. Structurally diverse CF3-containing tetracyclic core benzimidazo[2,1-a]isoquinoline-6(5H)-ones were obtained in moderate to good yields. The wide substrate scope, good functional group tolerance, and ease of scale-up of this method are expected to promote its potential applications in pharmacy and biotechnology.

Abstract:An efficient approach for the synthesis of 2-trifluoromethyl-benzimidazoles via cascade annulation of tri-fluoroacetimidoyl chlorides and amines using a heterogeneous copper doped g-C3N4 catalyst has been developed. Compared with the previously reported homogeneous catalytic system, the reaction exhibits higher efficiency and the copper doped g-C3N4 catalyst can be readily recycled for three times with slightly decreased catalytic reactivity.

Abstract:A novel cascade reaction was developed for the synthesis of diverse members of a series of benzofuro(3,2-d]pyrimidine derivatives. The process utilizes readily prepared 3-chlorochromenones and various commercially available amidines and their analogues as starting materials. Abstract:A TfOH-promoted synthesis of fluorinated poly-fused heterocycles via the cascade cyclization of azadienes and difluoroenoxysilanes has been developed, leading to the facile construction of fluorinated benzofuro[3,2-b]pyridines, 5H-indeno-[1,2-b]pyridines, and 5,6-dihydrobenzo[h]quinolines. This one-pot formal [4 + 2] approach involves 1,4-difluoroalkylation, desulfo-nylation, cyclization, and dehydrated and dehydrofluorinated aromatization and represents the first application of difluoroenoxysilane in cascade transformations. Furthermore, this methodology is highlighted by the synthesis of three fluoro analogues of bioactive molecules with potent topoisomerase inhibitory activities.

Abstract:A novel method for the synthesis of trifluoromethyl-containing 1'H-spiro[azirine-2,4'-quinolin]-2'(3'H)-ones by a CF3-radicabtriggered tandem reaction of benzene-linked 1,7-enynes is described. This protocol utilizes 1trifluoromethyl-1,2-benziodoxole as the trifluoromethylating reagent and TMSN3 as the aminating reagent. By this method, various potentially bioactive trifluoromethylated 1'H-spiro [azirine-2,4'-quinolin] -2'(3'H) -ones were facilely synthe-sized via a radical cascade process.

Abstract:Intramolecular aromatic substitution and Büchner reaction have been established as powerful methods for the construction of polycyclic compounds. These reactions are traditionally catalyzed by Rh(II) catalysts with α-diazocarbonyl compounds as the substrates. Herein a transition-metal-free intramolecular aromatic substitution/Büchner reaction is presented. These reactions use readily available N-tosylhydrazones as the diazo compound precursors and show wide substrate scope.

Abstract:An efficient cascade method has been developed for the synthesis of aza[2,1-b][1,3]-benzothiazinones from cyclic thiourea and methyl 2-iodobenzoate via the Cu(I)-catalyzed C–S coupling/amidation process. Abstract:A novel Ru-catalyzed redox-neutral [4+2] cyclization of 2-arylbenzimidazoles with α-trifluoromethyl-α-diazoketones has been achieved through sequential C-H activation and defluorinative annulation. This synthetic protocol unlocks modular and expeditious access to 6-fluorobenzimidazo[2,1-a]isoquinolines with high efficiency and excellent functional group compatibility. The resultant monofluorinated heterocyclic products can readily diversified by various nucleophiles.

Abstract:Benzofluorenes are an important class of polycyclic aromatic hydrocarbons (PAHs), which are widely used in materials science for light‐sensitive electronic components. In addition, the benzofluorene skeleton also serves as a key structural unit in various natural products. Therefore, the development of synthetic strategies for the construction of benzofluorenes has been a prominent research topic in synthetic chemistry. Over the past few decades, numerous efforts have been made for the construction of benzofluorenes via cascade cyclization of alkyne units. This review aims to summarize recent advances in the cascade cyclization of alkyne units for the construction of Based on the different reaction mechanisms underlying these transformations, this review can be divided into four categories: (1) thermal‐induced cascade cyclization of alkyne units for the construction of benzofluorenes, (2) metal‐catalyzed cascade cyclization of alkyne units for the construction of benzofluorenes, (3) acid‐promoted/catalyzed cyclization of alkyne units for the construction of benzofluorenes, and (4) base‐promoted cascade cyclization of alkyne units for the construction of benzofluorenes.

Abstract:A facile synthesis of a diverse array of benzo[b]fluorenone derivatives has been developed. The method involves a Cu-0/Selectfluor-catalyzed tandem annulation of 1,6-enynes, which takes place under mild reaction conditions. For tert-butylethynyl-substituted 1,6-enynes, the major reaction pathway was a tandem annulation/C-C-bond-cleavage/fluorination process leading to fluorinated benzo[b]fluorenones as the major products; tert-butyl-substituted benzo[b]fluorenones were obtained as the minor products. For arylethynyl-substituted 1,6-enynes, the substrates underwent tandem annulations to give 5-aryl-substituted benzo[b]fluorenones in moderate to excellent yields. For (trimethylsilyl)ethynyl-substituted 1,6-enynes, the reaction involved a tandem annulation/C-Si-bond-cleavage process to deliver 11H-benzo[b]fluoren-11-ones in moderate yields.

Abstract:An efficient copper-promoted hydration reaction and its application in the synthesis of benzo[b]furan and benzo[b]thiophene derivatives is presented starting from readily available 2-fluorophenylacetylene derivatives. The key annulation step involves the hydration of the C-F bond of 2-fluorophenylacetylene derivatives followed by an intramolecular annulation to afford benzo[b]furan and benzo[b]thiophene derivatives. Moreover, structurally important 2,2'-bisbenzofuran scaffolds are provided in good yields.

Abstract:We developed a method for allowing cascade cyclization of 2-nitrochalcones with pyrazoles, imidazole, and indazole in the presence of CuI catalyst, DBU base, and THF solvent. The conditions were tolerant of an array of useful functionalities including ester, nitro, cyano, halogen groups. A mechanistic consideration was also provided, as H(2)O(2) was presumably a byproduct. Our method appears to be a rare example to directly prepare C3-heterocyclic unprotected indoles.
