Abstract:Peroxidase mimics made of inorganic nanomaterials as alternatives to natural peroxidases have been extensively developed over the past few decades, owing to their superior properties relative to their natural counterparts. Nevertheless, it has been still a challenge to substantially enhance the catalytic efficiency of peroxidase mimics. In this work, we report a type of highly efficient peroxidase mimics that are made of iridium nanoparticles (Ir NPs) with rough surfaces. The Ir NPs possess an ultrahigh catalytic efficiency with a catalytic constant (K-cat) at the regime of 10(10) s(-1), orders of magnitude higher than the K-cat of horseradish peroxidase as a natural peroxidase. As a proof-of-concept demonstration, the Ir NPs were applied to colorimetric lateral flow assay (CLFA). Using carcinoembryonic antigen as a model disease biomarker, the Ir NPs-based CLFA achieved a low limit of detection (LoD) of 39 pg/mL, which was similar to 28 times lower than the LoD of conventional gold nanoparticles-based CLFA using the same antibodies.

Abstract:In this work, we report a peroxidase mimic composed of iridium (Ir) nanoparticles with an average size of only 1.1 nm that are coupled to a tungsten suboxide (WO2.72) nanorod as support. Such a peroxidase mimic exhibited an area-specific catalytic efficiency as high as 1.5 x 10(4) s(-1) nm(-2), outperforming most existing peroxidase mimics of noble metals. As proof-of-concept demonstrations, the peroxidase mimic was applied to colorimetric immunoassays of carcinoembryonic antigen (a cancer biomarker) and aflatoxin B1 (a potent carcinogen), which provided substantially enhanced detection sensitivities compared to conventional enzyme-based assays.

Abstract:The unique enzyme-like properties of metal nanoparticles (NPs) hold great promise for chemical and biomedical applications; implementation relies on improvements in their catalytic efficiency. We have constructed PtCuCo nanostructures using hydrothermal treatment of Co3O4, Cu2+, and Pt2+ in aqueous solutions, resulting in PtCuCo trimetallic alloy NPs having hollow structures, as confirmed by TEM, XRD, XPS, and EDS analyses. These PtCuCo NPs are capable of oxidizing 3,3',5,5'-tetramethyl-benzidine (TMB) in either the absence or presence of hydrogen peroxide, suggesting efficient peroxidase- and oxidase-like activity. These NPs could also catalyze the reduction of 4-nitrophenol by NaBH4. We believe that the combination of Pt and oxidized Cu and Co species results in surfaces with rich active sites possessing a wide range of electronic variation, which promotes both generation of hydroxyl radicals and electron transfer. Consequently, the catalytic capabilities of our PtCuCo NPs are greater than those of either Pt or PtCu NPs. Building upon the peroxidase-like activity of PtCuCo NPs, we developed a colorimetric method not only capable of specifically detecting glucose, but also able to detect rongalite and HS-, on the basis of the inhibitory effect of these substances upon PtCuCo nanoperoxidase.

Abstract:Peroxidase-mimicking nanozymes offer unique advantages in terms of high stability and low cost over natural peroxidase for applications in bioanalysis, biomedicine, and the treatment of pollution. However, the design of high-efficiency peroxidase-mimicking nanozymes remains a great challenge. In this study, we adopted a structural-design approach through hybridization of cube-CeO2 and Pt nanoparticles to create a new peroxidase-mimicking nanozyme with high efficiency and excellent stability. Relative to pure cube-CeO2 and Pt nanoparticles, the as-hybridized Pt/cube-CeO2 nanocomposites display much improved activities because of the strong metal-support interaction. Meanwhile, the nanocomposites also maintain high catalytic activity after long-term storage and multiple recycling. Based on their excellent properties, Pt/cube-CeO2 nanocomposites were used to construct high-performance colorimetric biosensors for the sensitive detection of metabolites, including H2 O2 and glucose. Our findings highlight opportunities for the development of high-efficiency peroxidase-mimicking nanozymes with potential applications such as diagnostics, biomedicine, and the treatment of pollution.

Abstract:Recent strides in nanotechnology have given rise to nanozymes, nanomaterials designed to emulate enzymatic functions. Despite their promise, challenges such as batch-to-batch variability and limited atomic utilization persist. This study introduces Pt(Glu)2, a platinum glutamic acid complex, as a versatile small-molecule peroxidase mimic. Synthesized through a straightforward method, Pt(Glu)2 exhibits robust catalytic activity and stability. Steady-state kinetics reveal a lower Km value compared to natural enzymes, signifying strong substrate affinity. Pt(Glu)2 was explored for controllable chemical modification and integration into cascade reactions with natural enzymes, surpassing other nanomaterials. Its facile synthesis and seamless integration enhance cascade reactions beyond the capabilities of nanozymes. In biosensing applications, Pt(Glu)2 enabled simultaneous detection of cholesterol and alkaline phosphatase in human serum with high selectivity and sensitivity. These findings illuminate the potential of small molecule mimetics in catalysis and biosensing, paving the way for their broader applications.

Abstract:In this paper, we demonstrated that Au nanorods coated with a shell composed of Pt nanodots (Au@Pt nanostructures) exhibited intrinsic oxidase-like, peroxidase-like and catalase-like activity, catalyzing oxygen and hydrogen peroxide reduction and the dismutation decomposition of hydrogen peroxide to produce oxygen. Based on these findings, we established an Au@Pt nanostructures based enzyme linked immunosorbent assay (ELISA) for the detection of mouse interleukin 2 (IL-2). In comparison with natural enzymes, Au@Pt nanostructures have advantages of low cost, easy preparation, better stability, and tunable catalytic activity (compared with HRP), which make them a promising enzyme mimetic candidate and may find potential applications in biocatalysis, bioassays, and nano-biomedicine such as reactive oxygen species (ROS)-related fields (anti-aging and therapeutics for neurodegenerative diseases and cancers).
Pd-Ir Core-Shell Nanocubes: A Type of Highly Efficient and Versatile Peroxidase Mimic

Abstract:Peroxidase mimics with dimensions on the nanoscale have received great interest as emerging artificial enzymes for biomedicine and environmental protection. While a variety of peroxidase mimics have been actively developed recently, limited progress has been made toward improving their catalytic efficiency. In this study, we report a type of highly efficient peroxidase mimic that was engineered by depositing Ir atoms as ultrathin skins (a few atomic layers) on Pd nanocubes (i.e., Pd-Ir cubes). The Pd-Ir cubes exhibited significantly enhanced efficiency, with catalytic constants more than 20- and 400-fold higher than those of the initial Pd cubes and horseradish peroxidase (HRP), respectively. As a proof-of-concept demonstration, the Pd-Ir cubes were applied to the colorimetric enzyme-linked immunosorbent assay (ELISA) of human prostate surface antigen (PSA) with a detection limit of 0.67 pg/mL, which is ∼110-fold lower than that of the conventional HRP-based ELISA using the same set of antibodies and the same procedure.

Abstract:Due to the limitation that natural peroxidase enzymes can only function in relatively mild environments, nanozymes have expanded the application of enzymology in the biological field by dint of their ability to maintain catalytic oxidative activity in relatively harsh environments. At the same time, the development of new and highly efficient composite nanozymes has been a challenge due to the limitations of monometallic particles in applications and the inherently poor enzyme-mimetic activity of composite nanozymes. The inherent enzyme-mimicking activity is due to Au, Ag, and Pt, along with other transition metals. Moreover, the nanomaterials exhibit excellent enzyme-mimicking activity when composited with other materials. Therefore, this paper focuses on composite nanozymes with simulated peroxidase activity that have been prepared using noble metals such as Au, Ag, and Pt and other transition metal nanoparticles in recent years. Their simulated enzymatic activity is utilized for biomedical applications such as glucose detection, cancer cell detection and tumor treatment, and antibacterial applications.

Platinum Janus Nanoparticles as Peroxidase Mimics for Catalytic Immunosorbent Assay

Abstract:The catalytic efficiency of peroxidase mimic enzymes has not been substantially improved despite the wide variety of peroxidase mimic nanozymes. Here, we report a class of highly efficient peroxidase mimics, platinum Janus nanoparticles (Pt Janus NPs), which are synergy assembled from silica NPs and single-component Pt NPs. Pt Janus NPs exhibit ultrahigh catalytic efficiency, with a catalytic constant (K cat) as high as 5.6 × 105 s–1, which is about 50-fold greater than the K cat value of traditional Pt NPs and about 140-fold higher than that of HRP. Compared with the single-component Pt NPs, silica components in Pt Janus NPs could effectively adsorb the intermediates during the catalysis process, which weakened adsorption on the surface of Pt NPs. Moreover, we reveal that the silica component could be avoided by aggregation of Pt NPs, which protects the catalytic sites and improves the catalytic efficiency. We conclude that Janus NPs have a stable, flexible, and efficient structure that protects the catalytic activity of nanozymes to enhance the catalytic efficiency. The relative signal strength is 3.4-fold higher than that of Pt ELISA when Pt Janus NPs were applied to direct immunoassay.

Designed inorganic nanomaterials for intrinsic peroxidase mimics: A review

Abstract:Over the past decade, a wide range of nanomaterials have attracted extensive attention for artificial enzymes (nanozymes) to meet task-specific applications because of their intrinsic enzyme-like catalytic activities. Artificial enzymes based on inorganic nanomaterials are now envisioned as an increasingly relevant alternative to traditional natural enzymes due to their structure robustness, unusual high catalytic performance, facile synthesis from inexpensive and abundant raw materials. This tutorial review summarizes and highlights latest progress in design and construction strategies of inorganic nanomaterials to mimic the structure and functionality of natural peroxidase with a focus on optical technique, both the existing research hotspots and future prospects are outlined and discussed. Furthermore, methodologies for tailoring nanomaterials-based intrinsic peroxidase mimics to enhance their catalytic activity including morphology control, surface modulation and heterostructure coupling are also summarized. Finally, we put forward our personal insights into future research challenges and opportunities, hoping to motivate more researchers to participate in the research in this emerging field.

Abstract:In this work, bimetallic Bi/Pt nanoparticles in bovine serum albumin biomolecular scaffold (BSA-Bi/PtNPs) were synthesized through a facile and green method. As compared with BSA-PtNPs, the BSA-Bi/PtNPs possess enhanced peroxidase-like catalytic activity. Moreover, the BSA-Bi/PtNPs are stable in harsh conditions such as high temperature, extreme pH environments, and high ionic strength, as well as in common biological matrixes. These prominent advantages enable the BSA-Bi/PtNPs to be applied to a wide range of fields. Bioassays, such as serum glucose detection, extracellular hydrogen peroxide (H2O2) monitor, and cancer cells labeling, have been realized with satisfying results. The linear range of glucose determination was from 1 to 100 μM and the limit of detection (LOD) was 0.2 μM. The H2O2 released from each MCF-7 cell after stimulation was calculated to be 2.66 × 10-16 mol/s. By utilizing folic acid as a recognition element, tumor cell could be readily distinguished by BSA-Bi/PtNPs and the LOD for MCF-7 cell detection was 90 cells.

Abstract:Among different nanozymes, peroxidase-like nanozymes have attracted extensive attention because of their great prospects in biosensing, bioimaging and disease treatment. By combining experiments with computational studies, several reports have elucidated the catalytic mechanism and structure-activity relationship of peroxidase-like nanozymes. Furthermore, to enable the rational design of highly active peroxidase-like nanozymes, several pioneering studies have developed numerous descriptors that can be used to predict their catalytic activity. These rationally designed highly active nanozymes have been used for in vitro and in vivo assays. This review first focused on the progress in the rational design of peroxidase-like nanozymes, and then introduced some typical examples of the analytical applications of peroxidase-like nanozymes. In addition, the key issues and challenges faced by peroxidase-like nanozymes were summarized and their further development directions were prospected.

Abstract:As alternatives to natural peroxidase, peroxidase mimics made of inorganic nanomaterials have been extensively developed for a variety of biosensing applications in recent years. In this work, we report an innovative concept to design and synthesize peroxidase mimics for sensitive biosensing–engineering thin metallic nanowires with narrow diameters (<5 nm), high aspect ratios, and branched structures. The nanowire has a significantly larger total surface area than conventional three-dimensional peroxidase mimics, ensuring a high catalytic efficiency. Moreover, the thin nanowires with good mechanical flexibilities induce minimal steric effects during biosensing. Using iridium (Ir) nanowires as a model example, we demonstrate that the nanowires could achieve an ultralow detection limit of 0.92 pg/mL in immunoassay of carcinoembryonic antigen (a biomarker for recurrence of colorectal cancer), which is 42- and 370-fold lower than the detection limits of conventional Ir nanoparticle- and enzyme-based immunoassays, respectively. The concept of crafting thin nanowires as peroxidase mimics was successfully extended from Ir to platinum- (Pt) and Ir-Pt-alloyed nanowires.

Abstract:Although enzyme-like nanomaterials have been extensively investigated over the past decade, most research has focused on the peroxidase-like, catalase-like, or SOD-like activity of these nanomaterials. Identifying nanomaterials having oxidase-like activities has received less attention. In this study, we demonstrate that platinum nanoparticles (Pt NPs) exhibit catechol oxidase-like activity, oxidizing polyphenols into the corresponding o-quinones. Four unique approaches are employed to demonstrate the catechol oxidase-like activity exerted by Pt NPs. First, UV–vis spectroscopy is used to monitor the oxidation of polyphenols catalyzed by Pt NPs. Second, the oxidized products of polyphenols are identified by ultrahigh-performance liquid chromatography (UHPLC) separation followed by high-resolution mass spectrometry (HRMS) identification. Abstract:In past decade, Pt-based nanomaterials as peroxidase mimics have attracted much attention for H2O2 and glucose detection. However, easy aggregation of Pt nanoparticles (Pt NPs) greatly decreases their peroxidase-like activity. In this work, novel Pt/EMT nanocomposites were prepared by uniformly loading Pt NPs (5-8 nm) onto the support of ultrasmall EMT zeolite (15-20 nm), a kind of low-silica microporous aluminosilicate material. The hybrid Pt/EMT nanomaterials could be well dispersed in water to form a homogeneous suspension, and were then utilized as a superior peroxidase-like catalyst for oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2). The optimal catalyst of 2.6Pt/EMT nanocomposite exhibited excellent catalytic performance toward H2O2 and TMB than natural enzyme of horseradish peroxidase (HRP) by using a steady-state kinetic analysis based on the typical Michaelis-Menten kinetics theory. The peroxidase-like catalyst showed a promising activity in a wide pH and temperature range as well as the long-term stability. A facile and reliable colorimetric assay based on the peroxidase mimic of Pt/EMT nanocomposite was constructed for precise detection of H2O2 and glucose in a wide linear range, with low limits of detection of 1.1 μM and 13.2 μM, respectively. Due to high selectivity to glucose against other sugars on the catalyst, the method was demonstrated to accurately measure the concentration of glucose in real samples including human blood serum and fruit juices, indicating a potential application of the Pt/EMT nanocomposites as a robust peroxidase mimic and a reliable biosensor in the fields of clinical diagnosis, pharmaceutical, food research and so on.
Galvanic Replacement Synthesis of VOx@EGaIn-PEG Core-Shell Nanohybrids for Peroxidase Mimics

Abstract:The development of high-performance biosensors is a key focus in the nanozyme field, but the current limitations in biocompatibility and recyclability hinder their broader applications. Herein, we address these challenges by constructing core-shell nanohybrids with biocompatible poly(ethylene glycol) (PEG) modification using a galvanic replacement reaction between orthovanadate ions and liquid metal (LM) (VOx@EGaIn-PEG). By leveraging the excellent charge transfer properties and the low band gap of the LM surface oxide, the VOx@EGaIn-PEG heterojunction can effectively convert hydrogen peroxide into hydroxyl radicals, demonstrating excellent peroxidase-like activity and stability (Km = 490 μM, vmax = 1.206 μM/s). The unique self-healing characteristics of LM further enable the recovery and regeneration of VOx@EGaIn-PEG nanozymes, thereby significantly reducing the cost of biological detection. Building upon this, we developed a nanozyme colorimetric sensor suitable for biological systems and integrated it with a smartphone to create an efficient quantitative detection platform. This platform allows for the convenient and sensitive detection of glucose in serum samples, exhibiting a good linear relationship in the range of 10-500 μM and a detection limit of 2.35 μM. The remarkable catalytic potential of LM, combined with its biocompatibility and regenerative properties, offers valuable insights for applications in catalysis and biomedical fields.

Abstract:A facile approach is proposed for the growth of platinum nanoparticles on graphene oxide (PtNPs/GO). The resulting PtNPs/GO hybrid has been proved to function as peroxidase mimics that can catalyze the oxidation of peroxidase substrates in the presence of hydrogen peroxide (H2O2). Kinetic studies indicate that the PtNPs/GO nanocomposite has a considerably higher affinity for both 3,3', 5,5'-tetramethyl-benzidine (TMB) and H2O2 than those of other platinum-based peroxidase mimics. Furthermore, colorimetric recognition and sensing of L-cysteine with high sensitivity and selectivity is presented based on target-induced shielding against the peroxidase-like activity of PtNPs/GO. We envision that this material will be an ideal candidate for a wide range of potential applications in the fields of biomedicine and environmental chemistry.

To overcome the challenges in creating enzymatic nanomaterials that present integrated advantages of high efficiency, selectivity, stability, and biocompatibility for visual colorimetric biosensors, here, we have constructed a new nanosized Ru clusterzyme supported by TiO 2 () via a facile two-step method to serve as peroxidase (POD)-like enzymatic nanomaterials for colorimetrical biomarker detection. Our morphology and chemical/electronic structure analysis prove that the consists of abundant surface-decorated Ru clusterzyme centers via Ru-O coordination. Benefiting from the unique enzymatic properties of Ru clusterzymes, the displays the highest V max (135.14 M s −1 ) and turnover number (210.8 × 10 -3 s −1 ) for H 2 O 2 among the state-of-the-art POD-mimics, manifesting the exceptional biosensing properties of compared to recently reported POD-like nanomaterials. Colorimetric studies of have displayed a relatively larger range and lower detection limit for the H 2 O 2 , L-cysteine, and glucose, thus exhibiting comparable detecting sensitivity with previous reports. Moreover, the presents extraordinary resistance to extreme environments, including low pH and high temperature. This work will create a new path to design efficient colorimetrical biosensors and offer tremendous potential for future biocatalysis, bioassays, and therapeutic applications.

Abstract:In recent decades, scholarly investigations have predominantly centered on nanomaterials possessing enzyme-like characteristics, commonly referred to as nanozymes. These nanozymes have emerged as viable substitutes for natural enzymes, offering simplicity, stability, and superior performance across various applications. Inorganic nanoparticles have been extensively employed in the emulation of enzymatic activity found in natural systems. Nanoparticles have shown a strong ability to mimic a number of enzyme-like functions. These systems have made a lot of progress thanks to the huge growth in nanotechnology research and the unique properties of nanomaterials. Our presentation will center on the kinetics, processes, and applications of peroxidase-like nanozymes. In this discourse, we will explore the various characteristics that exert an influence on the catalytic activity of nanozymes, with a particular emphasis on the prevailing problems and prospective consequences. This paper presents a thorough examination of the latest advancements achieved in the domain of peroxidase mimetic nanozymes in the context of cancer diagnosis and treatment. The primary focus is on their use in catalytic cancer therapy, alongside chemotherapy, phototherapy, sonodynamic therapy, radiation, and immunotherapy. The primary objective of this work is to offer theoretical and technical assistance for the prospective advancement of anticancer medications based on nanozymes. Moreover, it is anticipated that this will foster the investigation of novel therapeutic strategies aimed at achieving efficacious tumor therapy.

Abstract:Nanozymes, as a new generation of artificial enzymes, exhibit similar chemical properties, catalytic efficiency, and reaction kinetics to natural enzymes. Nanozymes can offer several advantages over natural enzymes, including the decreased cost, the increased stability, and the enhanced catalytic activity. These advantages have positioned nanozymes as a research focus in the fields of chemistry, materials and biomedicine. Polyoxometalates (POMs) and their composites have been found to possess excellent catalytic capabilities as peroxidase mimics. Given this, this review aims to provide a comprehensive overview of the POM-based nanozymes, covering their structural categorization, evolution, and various applications over the past decade. The dynamic nature of this field would promise the intriguing challenges and opportunities in the future. Additionally, we address the existing issues with the POM-based peroxidase-like enzymes and suggest the potential directions for future research. This review would serve as a valuable resource for researchers seeking to develop the improved therapeutic and diagnostic technologies using the POM-based nanozymes, thereby advancing the fields of biochemistry and materials science.
