General and Modular Synthesis of Covalent Organic Cages for Efficient Molecular Recognition

Abstract:Cage-type structures based on coordination and dynamic covalent chemistry have the characteristics of facile and efficient preparation but poor stability. Chemically stable organic cages, generally involving fragment coupling and multi-step reactions, are relatively difficult to synthesize. Herein, we offer a general and modular strategy to customize covalent organic cages with diverse skeletons and sizes. First, one skeleton (S) module with three extension (E) modules and three reaction (R) modules are connected by one- or two-step coupling to get the triangular monomer bearing three reaction sites. Then one-pot Friedel-Crafts condensation of the monomer and linking module of paraformaldehyde produces the designed organic cages. The cage forming could be regulated by the geometrical configuration of monomeric blocks. The S-E-R angles in the monomer is crucial; only 120o (2,4-dimethoxyphen as reaction module) or 60o (2,5-dimethoxyphen as reaction module) angle between S-E-R successfully affords the resulting cages. By the rational design of the three modules, a series of organic cages have been constructed. In addition, the host-guest properties show that the representative cages could strongly encapsulate neutral aromatic diimine guests driven by solvophobic interactions in polar solvents, giving the highest association constant of (2.58 ± 0.18) × 105 M-1.

Abstract:Interlocked molecular assemblies constitute a captivating ensemble of chemical topologies, comprising two or more separate components that exhibit remarkably intricate structures. The interlocked molecular assemblies are typically identical, and heterointerlocked systems that comprise structurally distinct assemblies remain unexplored. Here, we demonstrate that metal-templated synthesis can be exploited to afford not only a homointerlocked cage but also a heterointerlocked cage. Treatment of a carboxylated 2,9-dimethyl-1,10-phenanthroline (dmp) or Cu(I) bis-dmp linker with a Ni4-p-tert-butylsulfonylcalix[4]arene cluster affords noninterlocked octahedron and quadruply interlocked double cages consisting of two identical tetragonal pyramids, respectively. In contrast, when a mixture of dmp and Cu(I) bis-dmp linkers is used, a quadruply heterointerlocked cage is produced, consisting of a tetragonal pyramid and an octahedron. With photoredox-active [Cu(dmp)2]+ in the structures, both interlocked cages exhibit remarkable performance as photocatalysts for atom transfer radical addition (ATRA) reactions of trifluoromethanesulfonyl chloride with alkenes or oxo-azidations of vinyl arenes. These interlocked structures serve the dual purpose of stabilizing photocatalytically active components against deactivation and encapsulating substrates within the cavity, resulting in yields comparable to or even surpassing those of their molecular counterparts. This work thus provides a new strategy that combines metal templating and nontemplating approaches to design new types of interlocked assemblies with intriguing architectures and properties.

Abstract:Supramolecular architectures have flowered over the past few decades as an inventive class of functional materials for various applications. Discrete porous molecular cages are anticipated to be potential supramolecular catalysts that mimic enzyme-like catalytic activities. In this context, dynamic covalent chemistry (DCC)-driven organic cages received an enormous research interest due to the easy engineering of these porous cages with desired shapes, sizes, and functionalities. Herein, we have highlighted the role of DCC-driven stable porous cages as catalysts where catalysis occurs via the activation of the encapsulated substrates. As a result, in many cases these hosts result in better yields and stereoselectivity compared to conventional reactions without cages. This perspective presents the use of dynamic covalent bond-driven organic cages as efficient supramolecular catalysts under both homogeneous and heterogeneous reaction conditions. Specifically, the implementation of imine/amine organic cages as solid supports for ultrafine metal nanocatalysts is briefly focused on. Furthermore, various aspects of postsynthetically modified stable organic cages as efficient supramolecular hosts in different organic transformations are emphasized. Overall, we have critically analyzed the potential of porous organic cages as suitable platforms for organic transformations and for generating catalytically active metal nanoparticles.

Abstract:Cage compounds are very attractive structures for a wide range of applications and there is ongoing interest in finding effective ways to access such kinds of complex structures, particularly those possessing dynamic adaptive features. Here we report the accessible synthesis of new type of organic cage architectures, possessing two different dynamic bonds within one structure: hydrazones and disulfides. Implementation of three distinct functional groups (thiols, aldehydes and hydrazides) in the structure of two simple building blocks resulted in their spontaneous and selective self-assembly into aromatic cage-type architectures. These organic cages contain up to ten components linked together by twelve reversible covalent bonds. The advantage provided by the presented approach is that these cage structures can adaptively self-sort from a complex virtual mixture of polymers or macrocycles and that dynamic covalent chemistry enables their deliberate disassembly through controlled component exchange.

Abstract:A variety of organic cages with different geometries have been developed during the last decade, most of them exhibiting a single cavity. In contrast, the number of organic cages featuring a pair of cavities remains scarce. These structures may pave the way towards novel porous materials with emergent properties and functions.We herein report on rational design of a three-dimensional hexaformyl precursor 1 , which exhibits two types of conformers, i.e . Conformer-1 and -2, with different cleft positions and sizes. Aided by molecular dynamics simulations, we select two triamino conformation capturers (denoted CC). Small-sized CC-1 selectively capture Conformer-1 by matching its cleft size, while the large-sized CC-2 is able to match and capture both conformers. This strategy allows the formation of three compounds with twin cavities, which we coin diphane. The self-assembly of diphane units results in superstructures with tunable proton conductivity, which reaches up to 1.37×10 -5 S cm -1 .

Abstract:Here, we present a versatile modular strategy for crafting novel covalent organic cages (para-cage[n]arenes and meta-cage[n]arenes, n = 3,4) and bimacrocycles (meta-bimacrocyclic-arenes) with stable backbones and modifiable rims. These structures can be synthesized from commercially available aromatic multialdehydes in a three-step process: quantitative bromination, Suzuki-Miyaura reaction (yielding over 60%), and a rapid one-pot Friedel-Crafts reaction with paraformaldehyde. Notably, the cage[n]arenes exhibit a well-defined prismatic shape, and the bimacrocyclic-arenes display both dimeric and monomeric configurations.

Abstract:Different types of multinuclear half-sandwich rhodium macrocycles and cages were designed and synthesized by using two similar multifunctional hydroxamate ligands (pyrazine-2-hydroxamic acid (NaHL1) and 4,4-bipyridine-2-hydroxamic acid (KHL2)) featuring one monodentate site and two pairs of chelating sites. The Rh-III-Pd-II heterometallic macrocycles were constructed by using the semi-open palladium(II) source [Pd(en)Cl-2] with two free acceptor sites. However, only one kind of macrocycle was found when the shorter ligand L-1 was used, while in for the larger ligand, various spectroscopic techniques demonstrated the coexistence of hexanuclear and octanuclear macrocycles in solution and the proportions of both components depended on concentration and temperature. The palladium salt Pd(NO3)(2), as a source of naked Pd2+, was introduced to assemble the cuboid-shaped cage composed of two types of metal ions and three types of organic ligands. In addition, two silver(I)-containing mixed-metal complexes bridged by pyrazine were obtained, in which two forms of decanuclear complex with C-2v and C-2h point symmetry cocrystallizedone is a polymeric structure and the other is a discrete cage. However, the third form, with D-2 point symmetry, was found in the larger cage.

The chemistry and applications of boron clusters have inspired researchers ever since their discovery several decades ago, and the selective modification of the cage positions has remained a major synthetic challenge. A rhodium(III)‐catalyzed B−H functionalization–cyclization cascade of closo ‐dodecaborate amides is reported. The transformations occur chemoselectively at B−H positions in the presence of C−H bonds prone to competitive cyclometalation. Previously inaccessible cage derivatives with B−C(sp 2 ) and B−C(sp 3 ) bonds as well as a fused diboraoxazole ring are obtained in a one‐pot process. The reactions proceed under mild conditions and exhibit complete cage regioselectivity with broad functional group tolerance. These cluster derivatives enable a largely extended investigation of the application of anionic boron clusters in research areas such as photoluminescent materials and medicinal chemistry.

Abstract:The controlled synthesis of organometallic supramolecular macrocycles cages remains interesting and challenging work in the field of supramolecular chemistry. Here, two tetranuclear rectangular macrocycles and an octuclear cage were designed and synthesized utilizing a rigid and functionalized pillar linker, 2,6-bis(pyridin-4-yl)-1,7-dihydrobenzo [1,2-d:4,5-d′]diimidazole (BBI4PY) based on three half-sandwich rhodium building blocks bearing different sizes. X-ray crystallography in combination with 1H NMR spectroscopy elucidated that the two building blocks with shorter spacers only result in rectangular macrocycles. However, the building block of bulkier size to avoid the π-π stacking interactions between two ligands BBI4PY led to the formation of an octuclear cage complex. The latter cage contains two types of metal ions, namely Rh3+ and Cu2+, showing significant characteristics of heterogeneous metal-assembling compounds. In addition, the cage accommodates two free isopropyl ether solvent molecules, thus displaying host–guest behavior.

Abstract:The development of three-dimensional (3D) supramolecular coordination complexes is of great interest from both fundamental and application points of view because these materials are useful in molecular catalysis, separation and purification, sensing, etc. Herein, we describe the synthesis of two Klärner's molecular-clip-based tetrapyridyl donors, which possess a C-shaped structure as shown by X-ray analysis, and subsequently use them to prepare four convex trigonal-prismatic cages via coordination-driven self-assembly with two 180° diplatinum(II) acceptors. The cages are fully characterized by multinuclear NMR (31P and 1H) analysis, diffusion-ordered spectroscopy, electrospray ionization time-of-flight mass spectrometry, and UV/vis absorption spectroscopy. Moreover, the incorporation of molecular-clip-based ligands provides these cages with free cavities to encapsulate fullerene C60 via aromatic interactions, which may be useful for fullerene separation and purification. The studies described herein enlarge the scope of the platinum(II)-based directional bonding approach in the preparation of curved 3D metallacages and their host-guest chemistry.
C(3i)-symmetric Octanuclear Cadmium Cages: Double-Anion-templated Synthesis, Formation Mechanism, and Properties.

Abstract:A series of C3i-symmetric bicapped trigonal antiprismatic Cd8 cages [2X@Cd8L6(H2O)6].n?Y.solvents (X=Cl-, Y=NO3-, n=2: MOCC-4; X=Br-, Y=NO3-, n=2: MOCC-5; X=NO3-, Y=NO3-, n=2: MOCC-6; X=NO3-, Y=BF4-, n=2: MOCC-7; X=NO3-, Y=ClO4-, n=2: MOCC-8; X=CO32-, n=0: MOCC-9), doubly anion templated by different anions, were solvothermally synthesized by means of a flexible ligand. Interestingly, the CO32- template for MOCC-9 was generated in situ by two-step decomposition of DMF solvent. For other MOCCs, spherical or trigonal monovalent anions could also play the role of template in their formation. The template abilities of these anions in the formation of the cages were experimentally studied and are discussed for the first time. Anion exchange of MOCC-8 was carried out and showed anion-size selectivity. All of the cage-like compounds emit strong luminescence at room temperature.

Abstract:A [2 + 3] chiral covalent organic cage is produced through a dynamic covalent chemistry approach by mixing two readily available building units, viz. an enantiopure 3,3'‐diformyl 2,2'‐BINOL compound (A) with a triamino spacer (B). The two enantiomeric (R,R,R) and (S,S,S) forms of the cage C are produced nearly quantitatively thanks to the reversibility of the imine linkage. The X‐ray diffraction analysis of cage (S,S,S)‐C highlights that the six OH functions of the BINOL fragments are positioned inside the cage cavity. Upon reduction of the imine bonds of cage C, the amine cage D is obtained. The ability of the cage D to host the 1‐phenylethylammonium cation (EH+) as a guest is evaluated through UV, CD and DOSY NMR studies. A higher binding constant for (R)‐EH+ cation (Ka = 1.7 106 ± 10 % M‐1) related to (S)‐EH+ (Ka = 0.9 106 ± 10 % M‐1) is determined in the presence of the (R,R,R)‐D cage. This enantiopreference is in close agreement with molecular dynamics simulation.

Abstract:Making oppositely charged metal–organic cages (MOCs) into a tightly ordered structure may bring interesting functions. Herein, we report a novel structure composed of anionic (Zr4L6)8− (L=embonate) tetrahedral cages and in-situ-formed cationic [Zn4(Bim)4]4+ (Bim=[BH(im)3]−; im=imidazole) cubic cages. Chiral transfer is observed from enantiopure (Zr4L6)8− cage to enantiopure [Zn4(Bim)4]4+ cage. A pair of enantiomers (PTC-373(Δ) and PTC-373(Λ)) are formed. PTC-373 exhibits high chemical and thermal stabilities, affording an interesting single-crystal-to-single-crystal transformation. More importantly, the combination of ionic pair cages significantly enhances its third-order nonlinear optical property, and its thin-film exhibits an excellent optical limiting effect.

Abstract:The electronic effect arising from the different bidentate chelating sites on hydroxamate ligands allows selective coordination modes for the half-sandwich Ir/Rh metal centers. Despite the potential for obtaining a random statistical mixture of diastereoisomers as a consequence of differences in connectivity, only D-2 symmetric octanuclear cages were formed selectively by using asymmetric pyridyl-carboxylate bridging ligands.
