Abstract:The synthesis of molecular cages consisting of fully fused, π-conjugated rings is rare due to synthetic challenges including preorganization, large strain, and poor solubility. Herein, we report such an example in which a tris-2-aminobenzophenone precursor undergoes acid-mediated self-condensation to form a truncated tetrahedron, one of the 13 Archimedean solids. Formation of eight-membered [1,5]diazocine rings provides preorganization and releases the strain while still maintains weak π-conjugation of the backbone. Thorough characterizations were performed by X-ray, NMR, and UV–vis analysis, assisted by theoretical calculations. The cage exhibits a rigid backbone structure with a well-defined cavity that confines a magnetically shielded environment. The solvent molecule, o-dichlorobenzene, is precisely encapsulated in the cavity at a 1:1 ratio with multiple π···π, C–H···π, and halogen···π interactions with the cage skeleton, implying its template effect for the cage closing reaction. General and Modular Synthesis of Covalent Organic Cages for Efficient Molecular Recognition

Abstract:Cage-type structures based on coordination and dynamic covalent chemistry have the characteristics of facile and efficient preparation but poor stability. Chemically stable organic cages, generally involving fragment coupling and multi-step reactions, are relatively difficult to synthesize. Herein, we offer a general and modular strategy to customize covalent organic cages with diverse skeletons and sizes. First, one skeleton (S) module with three extension (E) modules and three reaction (R) modules are connected by one- or two-step coupling to get the triangular monomer bearing three reaction sites. Then one-pot Friedel-Crafts condensation of the monomer and linking module of paraformaldehyde produces the designed organic cages. The cage forming could be regulated by the geometrical configuration of monomeric blocks. The S-E-R angles in the monomer is crucial; only 120o (2,4-dimethoxyphen as reaction module) or 60o (2,5-dimethoxyphen as reaction module) angle between S-E-R successfully affords the resulting cages. By the rational design of the three modules, a series of organic cages have been constructed. In addition, the host-guest properties show that the representative cages could strongly encapsulate neutral aromatic diimine guests driven by solvophobic interactions in polar solvents, giving the highest association constant of (2.58 ± 0.18) × 105 M-1.

Abstract:An interlocked covalent organic molecular cage with a trigonal-prismatic structure based on anthracene units and imine bonds was synthesized by a template-free and one-pot reaction in a relatively good yield. By comparing the single-crystal structures of the interlocked cage and two additional monomeric cages as reference compounds, a cause of weak supramolecular interaction-induced synthesis was proposed; π···π interactions induce the triphenylbenzene units as the core and anthracenes as side parts close to each other, which provides a possibility of the preorganization for the formation of the interlocked structure.

Abstract:The interest in shape-persistent organic cages is nearly as old as the interest in supramolecular chemistry. In the beginning, organic cages have often been synthesized in a stepwise manner, which is not only laborious but very often also accompanied by low overall yields. In 1988, MacDowell published the one pot high-yielding synthesis of [2 + 3] imine cages based on TREN and aromatic dialdehydes, exploiting the reversible condensation of amines and aldehydes to imines, which was later used by others to make even larger cages on the basis of resorcinarenes. In 2008, the synthesis and characterization of an adamantoid [4 + 6] imine cage by condensation of a C3 v-symmetric triaminotriptycene and commercially available 4- tert-butyl salicyldialdehyde was introduced by the author, which was the ignition of our group activities in this research area. In 2011, we published the first gas-sorption data for this [4 + 6] imine cage: with a measured specific surface area of SABET = 1377 m2/g according to the model of Brunauer-Emmett-Teller (BET) this was twice as high as for the reported smaller cages of Cooper. For a second desolvated polymorph of the same cage, an even higher SABET = 2071 m2/g was determined; still one of the highest surface areas until date for porous organic molecular materials. Subsequently, the influence of the substituent in 4-position of the salicyldialdehyde for the reaction to [4 + 6] imine cages was investigated as well as the role of the phenolic hydroxyl group. It turned out that the phenolic hydroxyl group is crucial as directing group to increase the formation of the cage as well as stabilize the structure by cyclic six-membered intramolecular hydrogen bonds. The concept was extended to other imine-based cages of different geometry and size. For instance, a [4 + 4] cubic structure from triptycene trissalicylaldehyde and triptycene triamine was accessible as an amorphous insoluble solid, able to adsorb 18.2 wt % CO2 at ambient conditions. To gain further insight into the structural needs of the molecular precursors, rigidity and preorientation of reacting sites were investigated on prismatic [2 + 3] and truncated tetrahedral [4 + 4] imine cages, showing that rigidity and preorientation is beneficial or even crucial for cage formation. Furthermore, chiral self-sorting was studied on the basic of racemic triamines. Besides imine condensation, we explored the reversible formation of boronic esters from boronic acids and diols. Triptycene tetraol with its 120° angle between the aromatic units has been used in the condensation with benzene triboronic acid to achieve a large cuboctahedral [12 + 8] cage with pore dimensions of 2 nm, which are by IUPAC definition mesoporous. After activation the measured specific surface area was SABET = 3758 m2/g, a number rarely achieved even for other porous compounds such as threedimensional framework materials. Smaller tetrahedral [4 + 6] boronic ester cages were synthesized too. These cages show a selective gas sorption with preference of saturated hydrocarbon ethane over ethylene and acetylene. What distinguishes porous materials derived from molecular cages from three-dimensional frameworks or networks the most is their solubility; thus, the cages are soluble porous units (SPUs) in a broader sense. Taking advantage of this, [4 + 6] imine cages were postfunctionalized in solution to change the gas sorption properties in the crystalline state. Furthermore, cage solutions were spray-coated onto quartz crystal microbalances to enhance affinity and selectivity for sensing of airborne analytes. In this Account, the contributions from our lab on porous organic cages are presented.

Abstract:Cage compounds are very attractive structures for a wide range of applications and there is ongoing interest in finding effective ways to access such kinds of complex structures, particularly those possessing dynamic adaptive features. Here we report the accessible synthesis of new type of organic cage architectures, possessing two different dynamic bonds within one structure: hydrazones and disulfides. Implementation of three distinct functional groups (thiols, aldehydes and hydrazides) in the structure of two simple building blocks resulted in their spontaneous and selective self-assembly into aromatic cage-type architectures. These organic cages contain up to ten components linked together by twelve reversible covalent bonds. The advantage provided by the presented approach is that these cage structures can adaptively self-sort from a complex virtual mixture of polymers or macrocycles and that dynamic covalent chemistry enables their deliberate disassembly through controlled component exchange.

Abstract:By a one-pot reaction strategy and a stepwise fragment-coupling approach, three types of giant cage molecules with a triangular prism shape and an approximate cavity volume of 930 angstrom(3) were synthesized. Through efficient aromatic nucleophilic substitution reactions of chlorotriazine with functionalized amines or aliphatic substitution reactions of the linking imino moieties with allyl bromide, the giant cages were successfully functionalized on the surface with multiple hydroxy, 2-pyridyl, or allyl groups. As evidenced by variable-temperature H-1 NMR spectroscopy, the D-3h- and C-3v-symmetric cage molecules underwent very slow conformational changes or deformation in solution because of their giant size and their flexible building units. Their giant cavities, different electronic interiors, and facile multiple exterior surface functionalization could render the molecular cages powerful three-dimensional macrocyclic hosts that might find wide applications in supramolecular science.

Abstract:Three tetrahedral organic cages have been obtained by condensing a triamino linker with a set of three ostensibly analogous triformyl precursors. Despite the large number of imine bonds formed, the corresponding cages were obtained in exceptionally high yields. Both theory and experimental results demonstrate that intramolecular CH⋅⋅⋅π interactions within all of the cage frameworks play an important role in abetting the condensations and contributing to the near-quantitative synthetic yields. The three cages of this study exhibit high thermodynamic and kinetic stability. A variety of small neutral guest molecules with complementary sizes and geometries may be used as templates in the cage forming reactions. Among the guests that may be used in this way is white phosphorus (P4 ), whose inherent reactivity towards oxygen is almost fully attenuated when bound within one of the cages.

Abstract:Functional nanosized cage complexes with an octahedral framework have been synthesized by the facile self-assembly reactions of six 90 PdII-containing compounds {[Pd(dppf)(OTf)(2)] or [Pd(PPh3)(OTf)(2)], dppf = 1,1'-bis(diphenylphosphanyl) ferrocene, OTf = trifluoromethylsulfonate} and four 120 degrees functional tritopic ligands [2,4,6-tri(4-pyridyl)-1,3,5-triazine] in acetone. Diffusion-ordered NMR spectroscopy (DOSY) and AFM images clarify the formation of 3-4 nm nanoparticles. These highly symmetrical cage complexes are discrete, face-directed, functional nanoparticles with symmetrical octahedral structures. The self-assembled functional cage complexes exhibit interesting electrochemical activities, and thus will potentially be useful in the catalysis of redox reactions, the encapsulation of sizeable guest molecules and the control of organic synthetic reactions.

Abstract:Dynamic covalent imine reactions between 2′,3′-dimethoxy-[1,1′:4′,1′'-terphenyl]-3,3′',5,5′'-tetracarbaldehyde (DMTT) and cyclohexanediamine, p-phenylenediamine, and benzidine, respectively, generate a porous organic cage (DMPOC) and two covalent organic frameworks (COFs), USTB-29, and USTB-30. DMPOC shows a [3 + 6] topological cage-like structure according to single crystal X-ray diffraction result. In contrast, both microcrystalline USTB-29 and USTB-30 exhibit two-dimensional monoporous structures in an eclipsed AA stacking style based on powder X-ray diffraction and theoretical simulations. In addition, DMPOC is capable of efficiently absorbing the iodine vapor with an outstanding uptake of 5.10 g/g, much higher than that of USTB-29 (3.07 g/g) and USTB-30 (3.16 g/g). Cage to COFs transformations have been realized from DMPOC to USTB-29 and USTB-30 via the imine bond exchange with slightly increased iodine vapor uptake. Mechanism investigations uncover that both nitrogen and oxygen atoms of POC and COFs contribute to iodine vapor capture due to the formation of charge transfer matter, and loose interaction introducing adaptive expanding voids of DMPOC is suggested to capture more iodine vapor than that of COFs with strong π-π interactions.

Abstract:A novo triangular covalent organic cage with reduced symmetry was synthesized employing 2,4,6‐triethylbenzene derivatives as caps and 8‐amino‐2‐quinoline carboxyl group as edges. The molecular cage could bind nitrate, acetate, and fluoride anions with moderate affinity. Organic molecular cages with low symmetry are promising to generate complex chemical environments for molecular recognition and catalysis. Herein, a new triangular prism cage with three rigid 8‐aminoquinoline‐2‐carboxylic acid derivatives as edges were synthesized. This molecular cage possesses reduced symmetry and contains both secondary amine and amide as hydrogen donating groups for molecular recognition. Abstract:Covalently linked molecular cages can provide significant advantages (including, but not limited to enhanced thermal and chemical stability) over metal-linked coordination cages. Yet, while large coordination cages can now be created routinely, it is still challenging to create chemically robust, covalently linked molecular cages with large internal cavities. This fundamental challenge has made it difficult, for example, to introduce endohedral functional groups into covalent cages to enhance their practical utility ( e.g. , for selective guest recognition or catalysis), since the cavities would have simply been filled up with such endohedral functional groups in most cases. Here we now report the synthesis of some of the largest known covalently linked molecular tetrahedra. Our new covalent cages all contain 12 peripheral functional groups, which keep them soluble. They are formed from a common vertex, which aligns the hydrazide functions required for the hydrazone linkages with atropisomerism. While we previously reported this vertex as a building block for the smallest member of our hydrazone-linked tetrahedra, our original synthesis was not feasible to be carried out on the larger scales required to successfully access the larger tetrahedra. To overcome this synthetic challenge, we now present an improved synthesis of our vertex, which only requires a single chromatographic step (compared to 3 chromatographic purification steps, which were needed for the initial synthesis). Our new synthetic route enabled us to create a whole family of molecular cages with increasing size (all linked with hydrolytically stable hydrazone bonds), with our largest covalent cage featuring p -quarterphenyl linkers and the ability to encapsulate a hypothetical sphere of approximately 3 nm in diameter. These results now open up the possibility to introduce functional groups required for selective recognition and catalysis into chemically robust covalent cages (without blocking the cavities of the covalent cages).

Abstract:Zirconium-based porous coordination cages have been widely studied and have shown to be potentially useful for many applications as a result of their tunability and stability, likely as a result of their status as a molecular equivalent to the small 8 Å tetrahedral pores of UiO-66 (Zr6(μ3-O)4(μ2-OH)4(C8O4H4)6). Functional groups attached to these molecular materials endows them with a range of tunable properties. While so-called multivariate MOFs, structures containing multiple types of functional groups on different bridging ligands within a structure are common, incorporating multiple functional moieties in permanently microporous molecular materials has proved challenging. By applying a mixed-ligand, or heteroleptic, synthesis strategy to cage formation, we have designed a straight-forward, one-pot synthesis of 10 Å zirconium-based molecular cages in a basket-shaped, or Zr12L6, geometry containing 3:3 ratios of combinations of two types of functional moieties from 11 different ligand options. Additionally, using more sterically hindered ligands, such as 5-benzyloxybenzene dicarboxylate, we show that ligand geometry governs the resulting cage geometry. This method allows for multiple functional groups to be incorporated in molecular cages and the ratio of moieties incorporated can be easily controlled. With this strategy in hand, we show that ligands for which zirconium cage syntheses have been elusive, such as 2,5-dihydroxybenzene dicarboxylate, have now been successfully incorporated into porous structures.

Structural transformations of metal-organic cages through tetrazine-alkene reactivity

Abstract:The assembly of metal-organic cages follows rules based on the coordination preferences of metal ions and the geometries of their typically rigid and planar precursor ligands. PdnL2n metal-organic cages are amongst the most structurally diverse with subtle differences in the metal-ligand coordination vectors giving rise to drastically different assemblies, however al-most all rely on the rigidity of aromatic linker groups to avoid the formation of intractable mixtures of structures. Here we exploit the inverse electron-demand Diels-Alder (IEDDA) reaction between tetrazine linker groups and alkene reagents to trigger structural changes induced by post-assembly modification. The structure of the 1,4-dihydropyridazine produced by IEDDA (often an afterthought in click chemistry) is crucial, with the presence of two sp3 centers leading to a greater degree of flexibility and non-planarity in this ligand. This drastically changes the range of accessible metal-ligand coordination vec-tors, triggering an initial Pd4L8 tetrahedral cage to transform into different Pd2L4 lantern cages, with both the extent (thermo-dynamics) and rate (kinetics) of this transformation dependent on the alkene dienophile selected. With cyclopentene, the unsymmetrical 1,4-dihydropyridazine ligands undergo integrative sorting in the solid state, with both head-to-tail orientation selection and enantiomer selection, leading to a single lantern isomer from the 39 possible. This preference is rationalized through entropy, symmetry, and the extent of hydrogen bonding. Subsequent oxidation of the 1,4-dihydropyridazine to the aromatic pyridazine rigidifies the ligands and imparts planarity again. The oxidized ligands can no longer fit in the lantern structure, inducing further structural transformations into Pd4L8 tetrahedral cages and Pd3L6 double-walled triangles. The concept of controllable addition of limited additional flexibility and then its removal through well-defined reactivity we en-visage being of great interest for those interested in structural transformations of any class of supramolecular architecture.

Abstract:Supramolecular cages, featuring well-defined molecular cavities, have gained interest in material chemistry owing to their adjustable shapes and sizes, selective host-guest chemistry, and dynamic bonding behavior. This study explored our attempts to synthesize supramolecular cages based on dynamic secondary bonding in hypervalent iodine systems. Hypervalent iodine-based cage precursors consisting of two iodine centers were linked together by di-yne pillars were successfully synthesized in multiple steps. Unfortunately, preliminary attempts to convert the cage precursor into a desired cage material have resulted in insoluble materials that are difficult to characterize. As an attempt to address solubility issues, an alternative alkoxy derived cage precursor was synthesized successfully in multiple steps. Upon attempted formation of the cage compound, these soluble chain-containing materials also suffered from low solubilities. The general design strategy and synthesis of the precursor materials is provided.

Organometallic macrocycles and cages based on bis(amidinate) ligands

Abstract:A series of half-sandwich iridium-based complexes bearing 1,4-benzenebis (N,N'-dicyclohexylamidine) (H2L1) or 4,4'-bisphenylbis(N,N'-dicyclohexylamidine) (H2L2) has been synthesized and characterized. Treatment of [Cp*IrCl2](2) (Cp* = eta(5)-pentamethyl-cyclo pentadienyl) with deprotonated H2L1 or H2L2 at low temperature resulted in formation of binuclear complexes [Cp*IrCl](2)(L-1) and [Cp*IrCl](2)(L-2). They were chosen as precursors to construct macrocycles and cages. Tetranuclear metallamacrocycles [(Cp*Ir)(4)(L-1)(2) (pyrazine)(2)][OTf](4) and [(Cp*Ir)(4)(L-2)(2)(pyrazine)(2)][OTf](4) were assembled by the reaction of complex [Cp*IrCl](2) (L-1) or [Cp*IrCl](2)(L-2) with pyrazine after the removal of chloride from the iridium centers with silver salt AgOTf (OTf = O2SCF3). When 2,4,6-tri(4-pyridyl)-1,3,5-triazine (tpt) is used instead of pyrazine under the same conditions, hexanuclear metallacages [(Cp*Ir)(6)(L-1)(3)(tpt)(2)][OTf](6) and [(Cp*Ir)(6)(L-2)(3)(tpt)(2)][OTf](6) were formed. Single-crystal X-ray analysis indicated that the cation of complex [(Cp*Ir)(4)(L-1)(2)(pyrazine)(2)][OTf](4) is a rectangular structure, while the cation of complex [(Cp*Ir)(4)(L-2)(2)(pyrazine)(2)][OTf](4) is seriously distorted, nearly to a tetrahedral state. Moreover, complexes [(Cp*Ir)(6)(L-1)(3)(tpt)(2)][OTf](6) and [(Cp*Ir)(6)(L-2)(3)(tpt)(2)][OTf](6) are prism-like cages with large cavity size, which can be prepared in very high yield in the absence of templates. Surprisingly, solutions of multinuclear complexes are very sensitive to carbon monoxide, in which the frameworks were destroyed, giving the binuclear complexes [(Cp*Ir)(2)(CO)(2)(L-1)][OTf](2) and [(Cp*Ir)(2)(CO)(2)(L-2)][OTf](2).
