Abstract:An unprecedented trifunctionalization of CC bonds in 2-(1-azidovinyl)-1,1 '-biphenyls has been successfully achieved using the NCS/AgNO2 system, leading to the preparation of 6-(dichloro(nitro)methyl)phenanthridines in moderate to good yields. In this process, the NCS/AgNO2 system serves as a NO2 radical initiator as well as a chloro group source. The present protocol is a rare example of the selective construction of densely functionalized phenanthridine derivatives in a one-pot manner.

Abstract:A convenient and efficient protocol for the synthesis of phenanthridine-6-carbonitriles has been successfully developed involving a copper(II)-catalyzed tandem annulation of vinyl azides and NaN3 in the presence of PhI(OAc)(2). A variety of functionalized phenanthridine-6-carbonitriles were synthesized in moderate to good yields under mild reaction conditions. Mechanistic study revealed that a radical pathway might be involved in this annulation reaction.

Abstract:Azetidines, being four-membered N-heterocycles, possess significant potential in contemporary medicinal chemistry owing to their favorable pharmacokinetic properties. Regrettably, the incorporation of functionalized azetidines into pharmaceutical lead structures has been impeded by the absence of efficient synthetic methods for their synthesis. In this study, a Rh-catalyzed one-carbon ring expansion of aziridines with vinyl-N-triftosylhydrazones is presented, which facilitates the synthesis of high value-added 2-alkenyl azetidine products. This research represents the first example of ring expansion of aziridines enabled by vinyl carbenes. Additionally, a one-pot two-step protocol, initiated from cinnamaldehyde, was successfully achieved, offering a step-economical and facile approach for the synthesis of these compounds. The pivotal aspect of this successful transformation lies in the in-situ formation of an alkenyl aziridinium ylide intermediate. Experimental investigations, coupled with computational studies, suggest that a diradical pathway is involved in the reaction mechanism.

Abstract:An efficient strategy assisted by visible-light-promoted iminyl radical formation has been developed for the synthesis of 6-(fluoro)alkylated phenanthridine derivatives. In the reactions, addition of alkyl and trifluoromethyl radicals onto vinyl azides gives iminyl radicals, which then undergo intramolecular homolytic aromatic substitution leading to phenanthridines. These reactions can be carried out under mild conditions with high chemical yields and broad substrate scope.

Abstract:Vinyl azides have emerged as important synthons in recent years and a variety of useful synthetic methodologies have been achieved in the construction of the nitrogen heterocycles, such as indoles, pyrroles, imidazoles, pyrazoles, pyridines and pyrimidines. The unstable azide group and the presence of neighboring unsaturated double bond accentuate the high reactiveity of vinyl azides with other electrophiles and nucleophiles in various manners. One of the important features of vinyl azides is that they can decompose to vinyl nitrenes or highly strained three-membered cyclic imines, 2H-azirines. These two three-atom intermediates can easily undergo tandem reactions with 1,3-dipoles to generate the nitrogen-containing heterocycles. Vinyl azide can also act as a two-atom synthon while azide group serves as a leaving group in the cycloaddition reactions. What's more, when vinyl azide is connected with a carbonyl group in the alpha-position of azide, the resulted alpha-azidovinylketone can react with the dinucleophiles, leading to the amino substituted ring. The purpose of this review is to present the synthetic approaches achieved both in our group and other groups.

Abstract:An 2,2'-azobis(2-methylpropionitrile) (AIBN) initiated tandem reaction of vinyl azides with isopropylxanthic disulfide to construct C-S/C-N bonds was disclosed. A range of functionalized 6-sulfanylmethyl phenanthridines could be easily accessed in 50%similar to 84% yields with a good regioselectivitv. The mechanism study indicates a free radical pathway in this reaction.

Abstract:A number of variously substituted phenanthridines have been synthesized using the newly developed methodology under mild conditions. The Hendrickson reagent initiated cascade annulation, which is composed of a mild conversion of stable amide precursor to highly reactive imido-carbonium intermediate and a subsequent intramolecular Friedel-Crafts reaction, successfully served as the key method.

Abstract:The Cu-catalyzed regioselective synthesis of phenazine N-oxides was realized from benzoxadiazoles and diaryliodonium salts. The process was initiated by the electrophilic arylation of benzoxadiazoles with diaryliodonium salts and followed by benzocyclization reactions. The further reduction of N-oxides in situ to phenazine scaffolds and deviation to organic fluorescent materials were readily accomplished.

Abstract:Over the past several years, rhodium-azavinylcarbene (Rh-AVC) has grown into an enabling synthon for the synthesis of diverse nitrogen heterocycles. Herein we present an overview of our recent achievements in this field, including the Rh-AVC-promoted formal [4+3], [3+3], and [3+2] cycloadditions. These reactions allow for the efficient synthesis of several classes of important nitrogen heterocycles, such as azepines, pyrroles, and pyrazines. Abstract:Catalyst-controlled divergent intermolecular cycloadditions of vinylaziridines with alkynes have been developed. By using [Rh(NBD)2]BF4 as the catalyst, a [3 + 2] cycloaddition reaction was achieved with broad substrate scope and high stereoselectivity under mild reaction conditions. Moreover, the chirality of vinylaziridines can be completely transferred to the [3 + 2] cycloadducts. When the catalyst was changed to [Rh(η(6)-C10H8) (COD)]SbF6, the alternative [5 + 2] cycloadducts were selectively formed under otherwise identical conditions.

Abstract:Vinylcarbenoids are used as three-carbon components in Rh(III)-catalyzed C-H activation/[4 + 3] cycloaddition with benzamides to access azepinones. This transformation makes a feature of simple starting materials, mild reaction conditions and high efficiency. A kinetic isotope effect study was conducted and a plausible mechanism is proposed.

Abstract:A general concise method for the synthesis phenanthridin‐6(5H)‐ones via photoinduced intramolecular annulation of N‐phenylbenzamides was developed. Under argon atmosphere and room temperature, phenanthridin‐6(5H)‐ones were obtained via irradiation N‐phenylbenzamides with a 280 nm UV lamp in the presence of methanesulfonic acid in toluene. The mechanism is illustrated and believed to proceed in the order of amides tautomerization, 6π‐electric cyclization, [1,5]‐H shift, amide‐imidine tautomerization, keto‐enol tautomerism and evolution hydrogen.

Abstract:An efficient one-pot three-component palladium-catalyzed domino reaction of aryl iodide, 2-bromophenylboronic acid, and norbornadiene to produce phenanthrenes has been developed. Norbornadiene serves both as the activator of ortho-C-H bond and the source of ethylene via a retro-Diels-Alder reaction. The method features inexpensive and readily available substrates, a broad range of functional groups, and good yields.

Abstract:The transmetalation triggered rhodium‐catalyzed C‒H bond activation and tandem annulation of 2‐biphenylboronic acids with sulfoxonium ylides or iodonium ylides has been developed. Various products of phenanthrenes were constructed under redox‐neutral conditions in 34−86% yields. Several mechanism exploration experiments and derivatization reactions were conducted in sequence to gain a deeper understanding of the process and the potential of this transformation. It offers an alternative approach for the synthesis of phenanthrene derivatives.

Abstract:The synthesis, properties, and application of carbonyl-bridged phenanthrene, namely phenanthrenone are described. A new synthetic route involving palladium-catalyzed annulation between 4,5-dibromo-9-fluorenone and internal alkynes was developed to simplify the synthesis of phenanthrenone, which is a type of undeveloped building blocks. The reaction tolerates a variety of substituents at the alkynes and proceeds in moderate yields. Phenanthrenone derivatives possess the advantages of both fluorenone and phenanthrene backbones, such as extended conjugation, good planarity and rigidity, and tunable energy gap. These attractive features make phenanthrenone an ideal electron-deficient π-building block for developing novel polycyclic aromatic hydrocarbons. In addition, the phenanthrenone derivatives can be served as organic photoredox catalysts for organocatalytic atom transfer radical polymerization and oxidation of alcohol.
