Abstract:An efficient synthesis of novel fused heterocycles has been established via microwave-assisted palladium-catalyzed intramolecular direct arylation. The acyclic aryl bromides are readily available from microwave-assisted one-pot annulation of N-(2-bromobenzyl)-2-aminophenois and ethyl 2-bromoalkanoates. The intramolecular direct arylation is then performed in the presence of palladium(II) acetate and dppf (10 mol% each) in toluene using potassium carbonate (2 equiv) as the base under microwave heating (150 degrees C, 1 h) to afford the products in 43-99% yields. Steric effect is observed for Ar-Ar bond formation, giving a substantial amount of the debromination byproduct.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:Abstract Currently, the selective activation of C(sp 3 )–F bonds and C–C bonds constitute one of the most widely used procedures for the synthesis of high-value products that range from pharmaceuticals to agrochemical applications. While numerous examples of these two methods have been reported in their respective fields, the processes which merge the activation of both single C(sp 3 )-F bonds and C–C bonds in one step still remain elusive. Here, we demonstrate the controllable defluoroalkylation–distal functionalization of trifluoromethylarenes with unactivated alkenes via distal heteroaryl migration. This is proposed to proceed via tandem C(sp 3 )–F and C–C bond cleavage using visible-light photoredox catalysis combined with Lewis acid activation. This strategy provides facile and flexible access to multiply functionalized α , α -difluorobenzylic ketones in useful yields (up to 88%) under mild conditions. The products can be further transformed into other valuable compounds, demonstrating the method’s utility.

Abstract:The significance of organofluorine compounds has inspired the establishment of numerousmethods for the functionalization of rather inert C-F bonds. Despite advances achieved in the manipulation of C(sp(2))-F bonds by employing transitionmetal catalysts, such as Pd, Rh, Cu, Ni, Ru, and Ir, strategies that address the paucity of effective pathways for selective activation of multiple C(sp(3))-F bonds remained challenging. In this context, we present an unprecedented coupling-aromatization-cyclization reaction of polyfluorinated ketones with diverse Nand S-nucleophiles that forms regiodefined perfluoroalkylated naphtho[1,2-b] furan/benzofuran derivatives by harnessing Co-promoted distinctive quadruple C(sp(3))-F bonds cleavage relay. This chemistry involving controlled and successive selective defluorination at heteronuclear centers would greatly contribute to the preparation of drug-like heterocycles as well as the late-stage elaboration of bio-relevant compounds. Controlled experiments and DFT theoretical studies revealed that the combination of cheap cobalt salt with Cs2CO3 enable expeditious C-F functionalization.

Abstract:The photoactivation of electron donor-acceptor complexes to obtain fluoroalkyl arenes provides an economical and environmental-friendly means of directly installing fluoroalkyl groups (-CF2H, -CF3, -C4F9, -C6F13, et al.) at late-stage drug discovery. However, recent progress in the field has been constrained to the use of electron-rich arenes as electron donors which is determined by the inherent electronic properties. In comparison, the role reversal strategy of commercial fluoroalkyl reagents as donors and various aromatic substrates as acceptors remains an unexplored frontier. Herein, we proposed a photoactivation of electron donor–acceptor (EDA) complex between fluoroalkyl sulfonates (donor) and in-situ-generated protonated aromatic hydrocarbons (catalytic acceptor) to achieve the C-H fluoroalkylation and tandem cyclization/fluoroalkylation of a series of (hetero)arenes. The process relies on available reagents, avoiding the need for metals, exogenous photocatalysts, and additives.

Abstract:Cyclobutanone is a strained motif with broad applications, while direct assembly of the aromatic ring fused cyclobutanones beyond benzocyclobutenone (BCB) skeletons remains challenging. Herein, we report a Rh-catalyzed formal [3+2] annulation of diazo group tethered alkynes involving a 4-exo-dig carbocyclization process, providing a straightforward access to furan-fused cyclobutanones. DFT calculations disclose that, by comparison to the competitive 5-endo-dig process, 4-exo-dig carbocyclization is mainly due to lower angle strain of the key sp-hybridized vinyl cationic transition state in the cyclization step. Using less reactive catalysts Rh2(carboxylate)4 is critical for high selectivity, which is explained as catalyst-substrate hydrogen bonding interaction. This method is proved successful to direct access previously inaccessible and unknown furan-fused cyclobutanone scaffolds, which can participate in a variety of post-functionalization reactions as versatile synthetic blocks. In addition, preliminary antitumor activity study of these products indicates that some molecules exhibite significant anticancer potency against different human cancer cell lines.

Abstract:An effective method for the synthesis of furans is developed via a tandem 1,2-acyloxy migration/intramolecular [3 + 2] cycloaddition/aromatization of enol ether-tethered propargylic esters. The reaction exhibits excellent functional group tolerance, broad substrate scope, and excellent chemoselectivity. The isolation of dihydrofuran intermediates in some cases gives more insight into the [3 + 2] cycloisomerization process.

Abstract:Alkynyloxiranes have been employed for the first time as effective coupling partners in Cp*Rh-III-catalyzed C-H functionalization reactions. Their annulation with N-aryloxyamides then offers a redox-neutral and efficient synthesis of functionalized 2,3-dihydrobenzofurans bearing an exocyclic E-allylic alcohol and a tetrasubstituted carbon center in good yields with a broad substrate scope. The products can be easily converted to molecules with more complexity, which provides an opportunity for rapid assembly of structurally diverse heterocycles.

Abstract:The Pd-catalyzed annulative π-extension of 1,8-dibromonaphthalene for the preparation of fluoranthenes in a single operation has been investigated. With specific arenes such as fluorobenzenes, the Pd-catalyzed double functionalization of C–H bonds yields the desired fluoranthenes. The reaction proceeds via a palladium-catalyzed direct intermolecular arylation, followed by a direct intramolecular arylation step. As the C–H bond activation of several benzene derivatives remains very challenging, the preparation of fluoranthenes from 1,8-dibromonaphthalene via Suzuki coupling followed by intramolecular C–H activation has also been investigated to provide a complementary method. Using the most appropriate synthetic route and substrates, it is possible to introduce the desired functional groups at positions 7–10 on fluoranthenes.

Abstract:A high‐valent copper‐catalyzed regioselective fluoroarylation of gem‐DFCPs with simple arenes as the C─H nucleophiles and NFSI as the electrophilic fluororeagent is reported. The activation of the congested proximal C─C bond is proposed via a concerted σ‐bond metathesis with a high‐valent Cu─F species. Transition‐metal (TM) catalyzed reaction of gem‐difluorinated cyclopropanes (gem‐DFCPs) has drawn much attention recently. The reaction generally occurs via the activation of the distal C─C bond in gem‐DFCPs by a low‐valent TM through oxidative addition, eventually producing mono‐fluoro olefins as the coupling products. However, achieving regioselective activation of the proximal C─C bond in gem‐DFCPs that overcomes the intrinsic reactivity via TM catalysis remains elusive. Here, a new reaction mode of gem‐DFCPs enabled by high‐valent copper catalysis, which allows exclusive activation of the congested proximal C─C bond is presented. The reaction that achieves fluoroarylation of gem‐DFCPs uses NFSI (N‐fluorobenzenesulfonimide) as electrophilic fluoro reagent and arenes as the C─H nucleophiles, enabling the synthesis of diverse CF3‐containing scaffolds. It is proposed that a high‐valent copper species plays an important role in the regioselective activation of the proximal C─C bond possibly via a σ‐bond metathesis.

Abstract:The activation and functionalization of C-F bonds for the construction of C-C and C-X bonds have drawn significant research attention in recent years. However, the chemo-and stereoselective control of dual C-F bond functionalization of gem-difluoroalkenes remains a formidable challenge. Herein, a Ni-catalyzed reductive cross-coupling of gem-difluoroalkenes with alkenyl-electrophiles and D2O that allowed the generation of C(sp2)-C(sp2) bonds and C(sp2)-D bonds in one pot by successive defluorination is described. This methodology offers facile access to various deuterated 1,3-dienes with broad functional group compatibility and (E)-selectivity under mild conditions. Preliminary mechanistic studies indicate that alpha-alkenyl-substituted monofluoroalkenes might be intermediates in this protocol.
