Abstract:In this work, the macromolecular design and modular synthesis of degradable and biocompatible copolymers via radical polymerization and click chemistry is highlighted and the resulting systems are evaluated as gene delivery carriers. Poly(ethylene glycol) (PEG) grafted poly[2-methylene-1,3-dioxepane (MDO)-co-propargyl acrylate (PA)-co-2-(dimethyl aminoethyl methacrylate (DMAEMA)] (MPD) is synthesized using radical polymerization and azide-alkyne click chemistry. The polymers are less cytotoxic and are able to condense plasmid DNA into nanosized particles. The low transfection efficiency of polyplexes in HepG2 cells is significantly improved by mixing Tat peptide with polyplexes.

Abstract:A one-step swelling and polymerization technique was used in the synthesis of porous glycidyl methacrylate (GMA) and ethylene glycol dimethacrylate (EGDMA) monodisperse polymeric microspheres. The polystyrene (PS) seed obtained in the dispersion polymerization was used as a shape template. The presence of epoxide rings in the chemical structure of microspheres enables their post-polymerization chemical modifications involving: the Diels-Alder reaction with sodium cyclopentadienide and maleic anhydride, the reaction with 4,4′-(bismaleimido)diphenylmethane, and the thiol-Michael reaction with methacryloyl chloride and 2-mercaptopropionic acid. Changing the reaction mixture composition—the amounts of crosslinking monomer and PS seed as well as the type and concentration of porogen porous microspheres of different porous structures were obtained. Their porous structures were characterized in the dry and swollen states. The copolymers obtained from the equimolar monomers mixture modified in the above way were applied as the column packing materials and tested in the reverse-phase HPLC (High-Performance Liquid Chromatography). A few factors influencing morphology and porous structure of microspheres were studied.

Abstract:Polyglycidyl methacrylate (PGMA) microspheres are widely used in chromatographic fillers because they contain active epoxides and are easy to introduce hydroxyl, amino and carboxyl groups. In this paper, the influence factors on the pore size and structure of PGMA microspheres were discussed and optimized. In this paper, the macroporous PGMA microspheres of about 7μm were synthesized with good monodispersion. The influencing factors of pore size and pore structure were discussed and optimized. Finally, the PGMA microspheres were modified functionally to make their surfaces rich in carboxylic acid groups. When the functional PGMA microspheres were prepared into stationary phase for liquid chromatography. It was found that the PGMA microspheres showed excellent separation effect on basic proteins, nucleosides and basic nucleotides.

Abstract:The paper presents a method of the preparation and functionalization of polymer microspheres consisting of glycidyl methacrylate (GMA) and crosslinking agents: 1,4-dimethacryloyloxybenzene (1,4DMB) and trimethylolpropane trimethacrylate (TRIM). Poly(GMA- co -1,4DMB) and poly(GMA- co -TRIM) microspheres were obtained by seed swelling polymerization. To introduce thiol groups into the microspheres structure, the reaction with thiocarboxylic acids was performed. The chemical structure of parent and modified microspheres was confirmed by FTIR and Raman spectroscopy. Elemental composition of microspheres after functionalization was determined by elemental analysis. The analysis showed the percentage of sulfur in the range of 2.78–4.51%, which corresponds to a concentration of thiol group in the range of 0.87–1.41 mmol g −1 . Additionally, the porous structure of the copolymers was investigated using the low-temperature nitrogen adsorption–desorption method. The starting microspheres are characterized by a specific surface in the range of 150–160 m 2 g −1 , whereas functionalized copolymers indicate slightly lower surface area, of about 130 m 2 g −1 . The thermal stability of the materials was determined by the method of differential scanning calorimetry and thermogravimetric analysis. The course of the thermal degradation under oxidative conditions of modified microspheres is different from the starting copolymers. The functionalized microspheres showed much higher thermal stability (approximately 270 °C) compared to the starting microspheres (230–250 °C).

Abstract:Macroporous styrene-divinylbenzene (St-DVB) co-polymer microspheres can be synthesized through the free-radical cross-linking co-polymerization of styrene and divinylbenzene in the presence of an inert diluent. The research developments such as in preparation methods, synthesis variables and various applications achieved in the field of St-DVB co-polymer microspheres during the last few years is presented in this article. It has been shown that pore structures of St-DVB co-polymer microspheres, such as the pore size, the pore size distribution, the total pore volume, as well as the specific surface area, were strongly depended on their synthesis variables, such as the type and amount of the diluent, the amount of DVB and the volume ratio of monomer and diluent.

Abstract:In this study hyperbranched polyacetals (HBPAs) were used as a new type of porogens to prepare porous biodegradable microspheres. HBPAs were synthesized through the transacetalization process, which was carried out with an AB, type monomer of 4-(2-hydroxyethoxy) -benzaldehyde dimethyl acetal and a B, type core of benzaldehyde dimethyl acetal catalyzed with pyridinium camphorsulfonate (PCS). The results demonstrated that HBPAs were highly pH sensitive. They degraded rapidly at pH 5. 0, and barely degraded at pH 7. 4. The porous biodegradable poly (lactic acid) (PLA) microspheres were prepared by blending HBPAs and PLA with the emulsion-solvent evaporation method and the blending microspheres were then placed in acidic conditions for 2 weeks. As HBPAs hydrolyzed faster than PLA,the HBPAs component was first degraded from the blending microspheres to form small pores in the PLA matrix. DSC results and degradation process of the blending microspheres showed that the phase separation appeared in the blending microspheres of HBPAs and PLA. This phase separation favored the hydrolysis of HBPAs in the blending microspheres. The pore size of the microspheres can be regulated by changing the weight ratio of HBPAs to PLA. With increasing the HBPAs amount, the pore size of the PLA microspheres increased in a linear manner. HBPAs, as a new type of porogens, offer a new method to prepare porous microspheres.

Abstract:Polymer composite microspheres offer several advantages including highly designable structural properties, adjustable micro-nano particle size distribution, easy surface modification, large specific surface area, and high stability. These features make them valuable in various fields such as medicine, sensing, optics, and display technologies, with significant applications in clinical diagnostics, pathological imaging, and drug delivery in the medical field. Currently, microspheres are primarily used in biomedical research as long-acting controlledrelease agents and targeted delivery systems, and are widely applied in bone tissue repair, cancer treatment, and wound healing. Different types of polymer microspheres offer distinct advantages and application prospects. Efforts are ongoing to transition successful experimental research to industrial production by expanding various fabrication technologies. This article provides an overview of materials used in microsphere manufacturing, different fabrication methods, modification techniques to enhance their properties and applications, and discusses the role of microspheres in drug delivery engineering.

Abstract:Suspension-emulsion combined polymerization(SECP) process,in which 2-hydroxyethyl methacrylate(HEMA) emulsion polymerization constituents(EPC) were drop-wisely added to methyl methacrylate(MMA) suspension polymerization system,was applied to prepare PMMA/PHEMA composite microspheres with great size.The particle morphology,swelling,drug absorption and release properties of microspheres were investigated.It is found that the composite microspheres exhibited porous structure in the surface and PHEMA latex particles coagulated on the surface of PMMA particle.The swelling ratios of PMMA/PHEMA composite microspheres in water and benzyl alcohol are larger than that of PMMA microspheres.The absorption capacity of ibuprofen on PMMA/PHEMA composite microspheres was 0.356 g/g polymer in 48h and the equilibrium release time and release percentage of ibuprofen are 360 h and 82%,respectively,while the absorption capacity of ibuprofen on PMMA microspheres is 0.276 g/g polymer,and the equilibrium release time and percentage are 216 h and 60%,respectively.

Abstract:Mesoporosity combined with organic polymer functionality and hydrophobicity could give rise to promising new materials with applications in host–guest chemistry and biosensor devices. The synthesis of polymer–silica hybrid mesoporous materials with pore sizes of 3–6 nm is described, in which materials with large specific surface areas (∼800 m2 g–1) and pore volumes (∼0.7 cm3 g–1) as well as relatively narrow pore size distributions are produced by a sol-gel reaction in the presence of a non-surfactant template or pore-forming agent, which is subsequently removed by solvent extraction.

Abstract:Macromonomer poly( N-vinylformamide) and functional polymer microspheres are synthesized by radical polymerization, respectively.The properties of the macromonomer and the functional polymer microspheres have been studied by GPC, spectrophotometer and SEM.The results indicate that the macromonomer and the microspheres possess definite properties of thermo-sensitivity.Moreover the functional polymer microspheres have monodispersibity and its particle diameter is in the range of 1～5?μm.

Abstract:Poly(ionic liquid) microspheres are a new type of polymer microsphere that has piqued the interest of researchers due to their superior properties and wide range of applications. In this paper, the synthesis and application of poly(ionic liquid) microspheres in recent years are reviewed. The basic classification, structure and properties of poly(ionic liquid) microspheres are briefly described, as well as the characteristics of poly(ionic liquid) microspheres of various kinds and structures, including some novel micro spheres, new synthesis processes, and new application fields. Abstract:Polymeric porous particles are currently used for various applications in biotechnology, tissue engineering and pharmaceutical science, e.g., floating drug delivery systems and inhaled formulations. Particle shape and size depend on variable parameters; among them, polymer type and concentration, stirring speed, pH and type of solvent. In this study, porous poly(lactic-co-glycolic) acid (PLGA) and poly(d,l-lactide) (PLA) microspheres (MPs), with varying sizes and morphologies, were synthesized and optimized using both batch formulation and a flow-focusing microfluidic device. A well-established method of preparation utilizing solvent evaporation and the double emulsion technique was performed. Similar to other batch encapsulation methods, this technique is time and reagent consuming and consists of several steps. Hence, although porous structures provide tremendous opportunity in the design of new applications for tissue engineering and as improved controlled-release carriers, the synthesis of these particles with predefined properties remains challenging. We demonstrated the fabrication of porous MPs using a simple microfluidic device, compared to batch synthesis fabrication; and the effect of solvent, polymer concentration and type, post-hydrolysis treatment, on porosity degree. Moreover, a kinetic release study of fluorescent molecule was conducted for non-porous in comparison to porous particles. An overview of future prospects and the potential of these porous beads in this scientific area are discussed.

Abstract:Polysilsesquioxane (PSQ) microspheres have shown promise in many fields, but previous studies about porous PSQ microspheres are scarce. Herein, we fabricated novel micron-sized thiol-functional polysilsesquioxane (TMPSQ) microspheres with open and interconnected macropores by combining inverse suspension polymerization with two-step solâ€“gel and polymerization-induced phase separation processes, without using phase-separation-promoting additives or sacrificial templates. The chemical composition of the TMPSQ microspheres was confirmed using FTIR and Raman spectroscopy. The morphology of the TMPSQ microspheres was characterized using SEM and TEM. TGA was employed to test the thermal stability of the TMPSQ microspheres. Mercury intrusion porosimetry and nitrogen adsorptionâ€“desorption tests were performed to investigate the pore structure of the TMPSQ microspheres. The results showed that the TMPSQ microspheres had open and interconnected macropores with a pore size of 839 nm, and the total porosity and intraparticle porosity reached 70.54% and 43.21%, respectively. The mechanism of porous generation was proposed based on the morphological evolution observed using optical microscopy. The macropores were formed through the following four steps: phase separation (spinodal decomposition), coarsening, gelation, and evaporation of the solvent. The macropores can facilitate the rapid mass transfer between the outer and inner spaces of the TMPSQ microspheres. The TMPSQ microspheres are promising in various fields, such as catalyst supports and adsorbents.

Abstract:Biodegradable and biocompatibility polymeric microspheres have wide applications in many fields, especially as carriers for drug delivery and form the basis of several therapies approved by the US Food and Drug Administration. This paper describes the preparation controllable and monodisperse microspheres from biodegradable and biocompatibility polylactic acid (PLA) using a single microfluidic approach. We have demonstrated that one could control the size of microspheres from 15 mu m to 150 mu m in size (polydispersity index < 5%) by altering the relative sheath/sample flow rate ratio. Polylactic acid microspheres are ideally potential carrier for drug delivery.

Abstract:Polymeric microspheres (PMs) have attracted great attention in the field of biomedicine in the last several decades due to their small particle size, special functionalities shown on the surface and high surface-to-volume ratio. However, how to fabricate PMs which can meet the clinical needs and transform laboratory achievements to industrial scale-up still remains a challenge. Therefore, advanced fabrication technologies are pursued. In this review, we summarize the technologies used to fabricate PMs, including emulsion-based methods, microfluidics, spray drying, coacervation, supercritical fluid and superhydrophobic surface-mediated method and their advantages and disadvantages. We also review the different structures, properties and functions of the PMs and their applications in the fields of drug delivery, cell encapsulation and expansion, scaffolds in tissue engineering, transcatheter arterial embolization and artificial cells. Moreover, we discuss existing challenges and future perspectives for advancing fabrication technologies and biomedical applications of PMs.

Abstract:Macroporous copolymer particles have a broad range of applications such as ion exchange resins and sorbents, catalyst supports, and carriers of biologically active species. Many of these applications require precise control of the dimensions of microbeads in the range from 50 to 100 mu m and predetermined size of pores. IN This paper reports semicontinuous photoinitiated microfluidic synthesis of macroporous polymer particles with the designated dimensions and a range of internal structures. Comparison with microspheres synthesized by conventional suspension polymerization shows,that microfluidic synthesis provides better control over the porous structure of the microbeads.

Abstract:Polymer porous microspheres with large specific surface areas and good fluidity have promising important applications in the biomedical field. However, controllable fabrication of porous microspheres with precise size, morphology, and pore structure is still a challenge, and phase separation caused by the instability of the emulsion is the main factor affecting the precise preparation of porous microspheres. Herein, a method combining the iso-density emulsion (IDE) template and microfluidics was proposed to realize the controllable preparation of polymer porous microspheres. The IDE exhibited excellent stability with minimal phase separation within 4 h, thus showing potential advantages in the large-scale preparation of porous microspheres. With the IDE template combined microfluidics technique and the use of a customized amphoteric copolymer, PEG-b-polycaprolactone, polycaprolactone (PCL) porous microspheres with porosity higher than 90% were successfully prepared. Afterwards, the main factors, including polymer concentration, water–oil ratio and homogenization time were investigated to regulate the pore structure of microspheres, and microspheres with different pore sizes (1–30 μm) were obtained. PCL porous microspheres exhibited comparable cell viability relative to the control group and good potential as cell microcarriers after surface modification with polydopamine. The modified PCL porous microspheres implanted subcutaneously in rats underwent rapid in vivo degradation and tissue ingrowth. Overall, this study demonstrated an efficient strategy for the precise preparation of porous microspheres and investigated the potential of the as-prepared PCL porous microspheres as cell microcarriers and micro-scaffolds.

Abstract:Abstract Core–shell structured magnetic mesoporous polymer or carbon‐based microspheres not only possess the combined merits of magnetic particles and stable mesoporous shell but also provide various organic functional groups for further modification and immobilization of active sites, thus opening up more possibility for various applications. Herein, a bottom‐up soft‐templating strategy is developed to controllably synthesize core–shell magnetic mesoporous polydopamine microspheres (MMP) and their derivative magnetic mesoporous carbon (MMC) microspheres via an amphiphilic block copolymer‐directed interface assembly and polymerization (denoted as abc‐DIAP) approach. The obtained uniform MMP microspheres have a well‐defined structure consisting of magnetic core, silica middle layer and mesoporous PDA shell, uniform mesopores of 11.9 nm, high specific surface areas (235.6 m2 g−1) and rich functional groups. They show fast magnetic separation speed and superior performance in selective adsorption of Cyt.C from complex biosample solutions. Moreover, they can be in situ converted into core–shell magnetic mesoporous carbon (MMC) for efficient in‐pore immobilization of ultrafine Au nanoparticles for high‐efficiency catalytic epoxidation of styrene with high conversion (88.6%) and selectivity (90.1%) toward styrene oxide.
