Abstract:A polypeptide-DNA hydrogel is prepared by Z. Li, D. Liu, and co-workers, employing the “X”-shaped DNA assembling structure as crosslinker. The hydrogel can be modified with multifunctional components (here fluorescent molecules as a model), and possess excellent self-healing and thixotropic properties, enabling the direct-writing of arbitrary 3D structures. The study on page 1138 provides a simple, universal strategy for the assembly of functionalized hydrogels with biomolecules, especially enzymes.

Abstract:In the past thirty years, DNA molecule has received considerable attention as a promising building material due to its precise base-paring recognition, the programmable sequences, the predictable secondary structures, etc. Based on DNA self-assembly technique, DNA supramolecular hydrogels can be fabricated which are crosslinked networks swollen in an aqueous phase by the supramolecular interaction and also have widely applications in the biomedical field. In this paper, a new type of DNA-based supramolecular hydrogel was prepared by simply mixing two components: Polypeptide-DNA and G-quadruplex. We have successfully synthesized single-stranded DNA (ssDNA) grafted polypeptide via the copper-catalyzed click reaction. By introducing the intermolecular G-quadruplex structure, which is rich in guanine and is capable of forming a four-stranded structure in the presence of potassium, the hydrogel can be formed by DNA hybridization in a fast and in-situ manner and also obtain thermal and ionic responsive gel-sol transition, respectively. In this experiment, we firstly used polyacrylamide gel electrophoresis (PAGE), circular dichroism spectra (CD), UV-Vis spectroscopy to demonstrate the G-quadruplex structure formation, and the assembling process was efficient which are formed the parallel structure and the melting temperature (T-m) was about 43 degrees C. Then by the rheological tests, the shear-storage modulus (G') was obviously higher (up to 2.4 kPa) than the shear-loss modulus (G '') over the entire time range, and a minimum concentration of 0.5 wt% polypeptide-DNA was required to form a gel-like state, and the mechanical strength of the DNA hydrogels increased with increasing concentration of the DNA building blocks. What's more, the hydrogels can switch between the gel and the sol state when changing the temperature from 25 to 50 degrees C many times, indicating that the DNA hydrogels respond to thermal in a reversible way. At last, by using the properties of crown ether complexing potassium ion, the DNA hydrogel can obtain the ion responsiveness properties. Due to its designable responsiveness and good biocompatibility, this supramolecular hydrogel would have great potential applications in tissue engineering and 3D bioprinting field.

Abstract:Supramolecular assembly provides a new strategy for designing various gene carriers for deoxyribonucleic acid (DNA) delivery. In this work, supramolecular pseudo‐block conjugates based on star‐shaped polycation are developed as novel nonviral gene vector. First, cyclodextrin (CD) is selected as core and grafted with low molecular weight polyethylenimine (PEI 1800 ) as arms to obtain high molecular weight star‐shaped polycation CD‐PEI 8 (SCP). Then, azobenzene (Az) modified poly(ethylene glycol) is synthesized and named as azobenzene modified poly(ethylene glycol) (Az‐PEG). Subsequently, the supramolecular pseudo‐block conjugates are prepared via host–guest interaction between CD and Az. The result suggests that SCP/Az‐PEG condenses DNA into spherical nanoparticles with sizes of around 150 nm. The in vitro experiment shows that the cell viability of DNA/SCP/Az‐PEG polyplexes is significantly improved compared with that of PEI 25k /DNA polyplexes. The PEGylation does not decrease the cellular uptake efficiency and transfection efficiency. Furthermore, the cellular trafficking shows that PEGylation does not significantly influence nuclear entry. The star‐shaped architecture of polycation plays an important role in gene delivery. image

Abstract:DNA is a true polymer that stores the genetic information of an organism. With its amazing biological and polymeric characteristics, DNA has been regarded as a universal building block for the construction of diverse materials for real-world applications. Through various approaches including ligation, polymerization, chemical crosslinking, and physical crosslinking, both pure and hybrid DNA gels have been developed as generic materials. This Review discusses recent advances in the construction of DNA-based networks without considering any of DNA's genetic properties. In addition, we highlight the biomedical and non-biomedical applications of DNA as generic materials. Owing to the superb molecular recognition, self-assembly, and responsiveness of DNA, a mushrooming number of DNA materials with various properties have been developed for general utilization.

Abstract:The emergence of hybrid materials combining biomacromolecules and organic polymers has received broad attention based on their potential applications in chemical, biological and materials sciences. Among different coupling strategies, the grafting of oligonucleotides to organic polymers as side chains by covalent bonds provides a novel platform whereby the properties of both oligonucleotides and polymer backbone are integrated, manipulated and optimized for various applications. In this review, we give the perspective on this specific type of DNA polymer hybrid materials , using selected examples with emphasis on bioanalysis, biomedicine and stimuli-responsive materials. It is expected the success of DNA-grafting polymers will not only impact the frabication of novel bimolecule incorporated materials, but also will influence how the properties of synthetic materials are tailored using different functional groups.

Abstract:DNA exhibits many attractive properties, such as programmability, precise self-assembly, sequence-coded biomedical functions, and good biocompatibility; therefore, DNA has been used extensively as a building block to construct novel nanomaterials. Recently, studies on oligonucleotide–polymer conjugates (OPCs) have attracted increasing attention. As hybrid molecules, OPCs exhibit novel properties, e.g., sophisticated self-assembly behaviors, which are distinct from the simple combination of the functions of DNA and polymer, making OPCs interesting and useful. The synthesis and applications of OPCs are highly dependent on the choice of the polymer block, but a systematic summary of OPCs based on their molecular structures is still lacking. In order to design OPCs for further applications, it is necessary to thoroughly understand the structure–function relationship of OPCs. In this review, we carefully categorize recently developed OPCs by the structures of the polymer blocks, and discuss the synthesis, purification, and applications for each category. Finally, we will comment on future prospects for OPCs.

Abstract:DNA has gained great attention because of its unique structure, excellent molecular recognition property, and biological functions. When married with versatile synthetic polymers, the DNA conjugated polymer hybrids, known as DNA block copolymers (DBCs), have been launched and well developed for the syntheses of new materials and nanostructures with different functions in the past several decades. Compared to conventional synthetic block copolymers, using DNA as a building block provides several advantages over other polymer candidates, such as molecular recognition, programmable self-assembly, biocompatibility, and sequence-encoded information. In this review, recent developments in this area will be summarized and meaningful breakthroughs will be highlighted. We will discuss representative examples of recent progress in the syntheses, structure manipulations, and applications of DBCs.

The assembled spherical micelle structures exhibit recognition properties defined by their DNA sequences and can be used to build higher-ordered structures through hybridization with materials that possess complementary DNA. This work is an important step toward creating DNA modified structures that can act as phase transfer agents and carriers of nanovesicles.

Abstract:Deoxyribonucleic acid (DNA), a macromolecule that stores genetic information in organisms, has recently been gradually developed into a building block for new materials due to its stable chemical structure and excellent biocompatibility. The efficient preparation and functional integration of various molecular complexes and composite materials based on nucleic acid skeletons have been successfully achieved. These versatile materials possess excellent physical and chemical properties inherent to certain inorganic or organic molecules but are endowed with specific physiological functions by nucleic acids, demonstrating unique advantages and potential applications in materials science, nanotechnology, and biomedical engineering in recent years. However, issues such as the production cost, biological stability, and potential immunogenicity of DNA have presented some unprecedented challenges to the application of these materials in the field. This review summarizes the cutting-edge manufacturing techniques and unique properties of DNA-based complexes and composites and discusses the trends, challenges, and opportunities for the future development of nucleic acid-based materials.

Abstract:Over the past decade, the field of polymer-oligonucleotide nanomaterials has flourished because of the development of synthetic techniques, particularly living polymerization technologies, which provide access to polymers with well-defined architectures, precise molecular weights, and terminal or side-chain functionalities. Various "living" polymerization methods have empowered chemists with the ability to prepare functional polymer-oligonucleotide conjugates yielding a library of architectures, including linear diblock, comb, star, hyperbranched star, and gel morphologies. Since oligonucleotides are hydrophilic and synthetic polymers can be tailored with hydrophobicity, these amphiphilic polymer-oligonucleotide conjugates are capable of self-assembling into nanostructures with different shapes, leading to many high-value-added biomedical applications, such as drug delivery systems, gene regulation, and 3D-bioprinting. This review aims to highlight the main living polymerization approaches to polymer-oligonucleotide conjugates, including ring-opening metathesis polymerization, atom transfer radical polymerization (ATRP), reversible addition-fragmentation transfer polymerization (RAFT), and ring-opening polymerization of cyclic esters and N-carboxyanhydride. The self-assembly properties and resulting applications of polymer-DNA hybrid materials are highlighted as well.

Abstract:We live in a world full of synthetic materials, and the development of new technologies builds on the design and synthesis of new chemical structures, such as polymers. Synthetic macromolecules have changed the world and currently play a major role in all aspects of daily life. Due to their tailorable properties, these materials have fueled the invention of new techniques and goods, from the yogurt cup to the car seat belts. To fulfill the requirements of modern life, polymers and their composites have become increasingly complex. One strategy for altering polymer properties is to combine different polymer segments within one polymer, known as block copolymers. The microphase separation of the individual polymer components and the resulting formation of well defined nanosized domains provide a broad range of new materials with various properties. Block copolymers facilitated the development of innovative concepts in the fields of drug delivery, nanomedicine, organic electronics, and nanoscience. Block copolymers consist exclusively of organic polymers, but researchers are increasingly interested in materials that combine synthetic materials and biomacromolecules. Although many researchers have explored the combination of proteins with organic polymers, far fewer investigations have explored nucleic acid/polymer hybrids, known as DNA block copolymers (DBCs). DNA as a polymer block provides several advantages over other biopolymers. The availability of automated synthesis offers DNA segments with nucleotide precision, which facilitates the fabrication of hybrid materials with monodisperse biopolymer blocks. The directed functionalization of modified single-stranded DNA by Watson-Crick base-pairing is another key feature of DNA block copolymers. Furthermore, the appropriate selection of DNA sequence and organic polymer gives control over the material properties and their self-assembly into supramolecular structures. The introduction of a hydrophobic polymer into DBCs in aqueous solution leads to amphiphilic micellar structures with a hydrophobic polymer core and a DNA corona. In this Account, we discuss selected examples of recent developments in the synthesis, structure manipulation and applications of DBCs. We present achievements in synthesis of DBCs and their amplification based on molecular biology techniques. We also focus on concepts involving supramolecular assemblies and the change of morphological properties by mild stimuli. Finally, we discuss future applications of DBCs. DBC micelles have served as drug-delivery vehicles, as scaffolds for chemical reactions, and as templates for the self-assembly of virus capsids. In nanoelectronics, DNA polymer hybrids can facilitate size selection and directed deposition of single-walled carbon nanotubes in field effect transistor (FET) devices.

Abstract:This review concentrates on significant development in an important biomacromolecule, nucleic acid, especially deoxyribonucleic acid (DNA), and its biomedical application in cell uptake, drug delivery, gene regulation, and therapeutics. Herein, DNA molecules as nanomaterials are coarsely categorized into linear DNA, DNA aptamer, and programmable DNA origami structures, both of which are easily conjugated with other popular nanomaterial, i.e., carbon nanotube, metallic nanoparticles, quantum dots, proteins, etc., via π-π stacking, base-pair hybridization or chemical modification. This review not only provides mechanism and perspective on biomedical applications using hybrids of DNA-based nanocomposites, but also integrates an ideal strategy on designing and inventing the state-of-the-art targeted drug delivery systems.

Abstract:The chain effect of polymer is the kernel of the precise synthesis of complex polymer compounds and the development of polymer science. Nucleic acids, a special type of polyester, can form supramolecular polymers via hydrogen bonds among corresponding bases. We focused on the precise control of polymer chains and proposed the concept of "nucleic acid-backboned chimeric polymers " (NCPs). There are three important elements of NCPs, namely nucleic acid backbone, side group and intra-/inter-chain weak interactions. In this review, we expound how to realize the construction of intelligent DNA structures through the regulation of these three elements and the applications of NCPs in various fields.

Abstract:With the advent of DNA nanotechnology, DNA-based biomaterials have emerged as a unique class of materials at the center of various biological advances. Owing to DNA's high modification capacity via programmable Watson–Crick base-pairing, DNA structures of desired design with increased complexity have been designed. However, the lack of inherent functionality, along with low mechanical properties, high synthesis costs, and poor stability, limited the adaptability of DNA-based material toward complex biological systems. To impart functionality, DNA-based hybrid biomaterials synthesized by conjugating DNA with functional materials have been realized. Today, DNA-based hybrid materials have attracted significant attention in biological engineering with broad applicative prospects in biomedicine, clinical diagnosis, and nanodevices. Here, we summarize the recent advances in DNA-based hybrid materials with an in-depth understanding of general molecular design principles, functionalities, and applications. Finally, the challenges and prospects associated with DNA-based hybrid materials are discussed at the end of the review.

Abstract:<p>A novel kinetically interlocking multiple-units (KIMU) supramolecular polymerization system with DNA double crossover backbone is designed. The rigidity of DX endows the polymer with high molecular weight and stability. The observed concentration of the formed polymers is insensitive and stable under ultralow monomer concentration owing to the KIMU interactions, in which multiple noncovalent interactions are connected by the phosphodiester bonds. Furthermore, a pH-responsive DNA supramolecular hydrogel is constructed by introducing a half <i>i</i>-motif domain into the DNA monomer. The rigidity of DNA polymer endows the hydrogel with high mechanical strength and low gelation concentration. This study enriches the KIMU strategy and offers a simple but effective way to fabricate long and stable supramolecular polymers by balancing the reversibility and stability. It also shows great potentials to construct next generation of smart materials, such as DNA nanostructures, DNA motors, and DNA hydrogels.</p>
