Abstract:The chain effect of polymer is the kernel of the precise synthesis of complex polymer compounds and the development of polymer science. Nucleic acids, a special type of polyester, can form supramolecular polymers via hydrogen bonds among corresponding bases. We focused on the precise control of polymer chains and proposed the concept of "nucleic acid-backboned chimeric polymers " (NCPs). There are three important elements of NCPs, namely nucleic acid backbone, side group and intra-/inter-chain weak interactions. In this review, we expound how to realize the construction of intelligent DNA structures through the regulation of these three elements and the applications of NCPs in various fields.

Abstract:Over the past decade, the field of polymer-oligonucleotide nanomaterials has flourished because of the development of synthetic techniques, particularly living polymerization technologies, which provide access to polymers with well-defined architectures, precise molecular weights, and terminal or side-chain functionalities. Various "living" polymerization methods have empowered chemists with the ability to prepare functional polymer-oligonucleotide conjugates yielding a library of architectures, including linear diblock, comb, star, hyperbranched star, and gel morphologies. Since oligonucleotides are hydrophilic and synthetic polymers can be tailored with hydrophobicity, these amphiphilic polymer-oligonucleotide conjugates are capable of self-assembling into nanostructures with different shapes, leading to many high-value-added biomedical applications, such as drug delivery systems, gene regulation, and 3D-bioprinting. This review aims to highlight the main living polymerization approaches to polymer-oligonucleotide conjugates, including ring-opening metathesis polymerization, atom transfer radical polymerization (ATRP), reversible addition-fragmentation transfer polymerization (RAFT), and ring-opening polymerization of cyclic esters and N-carboxyanhydride. The self-assembly properties and resulting applications of polymer-DNA hybrid materials are highlighted as well.

Abstract:We report a general solid-phase synthesis strategy for a novel class of DNA block copolymer amphiphiles. The assembled spherical micelle structures exhibit recognition properties defined by their DNA sequences and can be used to build higher-ordered structures through hybridization with materials that possess complementary DNA. This work is an important step toward creating DNA modified structures that can act as phase transfer agents and carriers of nanovesicles.

Abstract:Over the past century, synthetic polymers have had a transformative impact on human life, replacing nature‐derived materials in many areas. Yet, despite their many advantages, the structure and function of synthetic polymers still appear rudimentary compared to biological matter: cells use dynamic self‐assembly to construct complex materials and operate sophisticated macromolecular devices. The field of DNA nanotechnology has demonstrated that synthetic DNA molecules can be programmed to undergo predictable self‐assembly, offering unparalleled control over the formation and dynamic properties of artificial nanostructures. Intriguingly, the principles of DNA nanotechnology can be applied to the engineering of soft programmable materials, bringing the abilities of synthetic polymers closer to their biological counterparts. In this perspective, we discuss the unique features of DNA‐functionalized polymer materials. We describe design principles that allow researchers to build complex supramolecular architectures with predictable and dynamically adjustable material properties. Finally, we highlight two key application areas where this biologically inspired material class offers particularly promising opportunities: (1) as dynamic matrices for 3D cell and organoid culture and (2) as smart materials for nucleic acid sequencing and pathogen detection.

Abstract:Poly(α-amino acid)s (PAAs) attract growing attention due to their essential role in the application as biomaterials. To synthesize PAAs with desired structures and properties, scientists have developed various synthetic techniques with respective advantages. Here, different approaches to preparing PAAs are inspected. Basic features and recent progresses of these methods are summarized, including polymerizations of amino acid N-carboxyanhydrides (NCAs), amino acid N-thiocarboxyanhydrides (NTAs), and N-phenoxycarbonyl amino acids (NPCs), as well as other synthetic routes. NCA is the most classical monomer to prepare PAAs with high molecular weights (MWs). NTA polymerizations are promising alternative pathways to produce PAAs, which can tolerate nucleophiles including alcohols, mercaptans, carboxyl acids, and water. By various techniques including choosing appropriate solvents or using organic acids as promoters, NTAs polymerize to produce polypeptoids and polypeptides with narrow dispersities and designed MWs up to 55.0 and 57.0 kg mol-1 , respectively. NPC polymerizations are phosgene-free ways to synthesize polypeptides and polypeptoids. For the future prospects, detail investigations into polymerization mechanisms of NTA and NPC are expected. The synthesis of PAAs with designed topologies and assembly structures is another intriguing topic. The advantages and unsettled problems in various synthetic ways are discussed for readers to choose appropriate approaches for PAAs.

Abstract:Functional nucleic acids (FNAs) are garnering tremendous interest owing to their high modularity and unique bioactivity. Three‐dimensional FNAs have been developed to overcome the issues of nuclease degradation and limited cell uptake. We herein developed a new facile approach to the synthesis of multiple three‐dimensional FNA nanostructures by harnessing photo‐polymerization‐induced self‐assembly. Sgc8 aptamer and CpG oligonucleotide were modified as macro chain transfer reagents to mediate in situ polymerization and self‐assembly. Diverse structures, including micelles, rods, and short worms, afford these two FNAs with higher nuclease resistance from serum, greater cellular uptake efficiency, and increased bioactivity.

Abstract:Within the last two decades a series of novel therapeutic nucleic acids entered research and clinical evaluation. Their differences both in biophysical properties as well as in mode and site of biological action provide polymer-based carriers with new delivery challenges. Recent tailor-made designs of polymeric carriers are reviewed that were optimized for nucleic acid cargos such as plasmid DNA, siRNA and micro RNA, messenger RNA or genome-modifying nucleic acids. The specific requirements for the various therapeutic cargos are discussed. Future directions include dynamic bioresponsive polymers as components of nanomachines, multifunctional sequence-defined carriers for evolution-based selective optimization, and organic-inorganic multicomponent nanoassemblies.

Abstract:Bioinspired and biomimetic nucleobase-containing polymers are a series of polydisperse nucleic acid analogs, mainly obtaining through highly efficient and scalable step-growth or chain polymerizations. The combination of pendant nucleobase groups and various backbones endows the polymers/materials with selective multiple H-bonds under distinct conditions, demonstrating the broad applicability of this new family of polymeric materials. In this perspective, we critically summarize recent advances of bioinspired and biomimetic nucleobase-containing polymers and materials in both solution and the bulk. Then, we discuss the effect of multiple H-bonds between complementary nucleobases on the structures and properties of the nucleobase-containing polymers and materials. Selective multiple H-bonds between complementary nucleobases are feasible to modulate the polymer sequence and self-assembly behaviour, achieve templated polymerization, tune nanostructure morphologies and functions, and selectively bind with nucleic acids in various solutions. Meanwhile, bioinspired and biomimetic nucleobase-containing polymers are capable of forming robust polymeric materials such as hydrogels, bioplastics, elastomers, adhesives, and coatings by optimizing the inter- and intramolecular multiple H-bonding interactions. Further, the conclusions and outlook for future development and challenges of bioinspired and biomimetic nucleobase-containing polymers are also presented. This perspective presents useful guidelines for fabricating novel bioinspired and biomimetic polymers and materials through rational design of multiple H-bonds nucleobase interactions.

Abstract:Gene therapy is an important therapeutic strategy in the treatment of a wide range of genetic disorders. Polymers forming stable complexes with nucleic acids (NAs) are non-viral gene carriers. The self-assembly of polymers and nucleic acids is typically a complex process that involves many types of interaction at different scales. Electrostatic interaction, hydrophobic interaction, and hydrogen bonds are three important and prevalent interactions in the polymer/nucleic acid system. Electrostatic interactions and hydrogen bonds are the main driving forces for the condensation of nucleic acids, while hydrophobic interactions play a significant role in the cellular uptake and endosomal escape of polymer-nucleic acid complexes. To design high-efficiency polymer candidates for the DNA and siRNA delivery, it is necessary to have a detailed understanding of the interactions between them in solution. In this chapter, we survey the roles of the three important interactions between polymers and nucleic acids during the formation of polyplexes and summarize recent understandings of the linear polyelectrolyte-NA interactions and dendrimer-NA interactions. We also review recent progress optimizing the gene delivery system by tuning these interactions.

Abstract:Imiquimod, the first member of a new class of immune response modifiers, is approved for the treatment of external genital and perianal warts. Recently, many clinical trials highlighted the potential of imiquimod as a treatment for other viral infections and cutaneous neoplasms. We report two cases of facial keratoacanthomas (KA) treated with topical 5% imiquimod cream. Patients were successfully cleared of KAs after treatment for 8 weeks. No recurrence occurred after a 1‐year follow‐up. Despite the fact that KAs are characterized by the potential for spontaneous regression, it is possible that a faster activation of CD4+ lymphocytes, via interferon release and cytokine secretion takes place after imiquimod application leading to KA regression.

Abstract:We live in a world full of synthetic materials, and the development of new technologies builds on the design and synthesis of new chemical structures, such as polymers. Synthetic macromolecules have changed the world and currently play a major role in all aspects of daily life. Due to their tailorable properties, these materials have fueled the invention of new techniques and goods, from the yogurt cup to the car seat belts. To fulfill the requirements of modern life, polymers and their composites have become increasingly complex. One strategy for altering polymer properties is to combine different polymer segments within one polymer, known as block copolymers. The microphase separation of the individual polymer components and the resulting formation of well defined nanosized domains provide a broad range of new materials with various properties. Block copolymers facilitated the development of innovative concepts in the fields of drug delivery, nanomedicine, organic electronics, and nanoscience. Block copolymers consist exclusively of organic polymers, but researchers are increasingly interested in materials that combine synthetic materials and biomacromolecules. Although many researchers have explored the combination of proteins with organic polymers, far fewer investigations have explored nucleic acid/polymer hybrids, known as DNA block copolymers (DBCs). DNA as a polymer block provides several advantages over other biopolymers. The availability of automated synthesis offers DNA segments with nucleotide precision, which facilitates the fabrication of hybrid materials with monodisperse biopolymer blocks. The directed functionalization of modified single-stranded DNA by Watson-Crick base-pairing is another key feature of DNA block copolymers. Furthermore, the appropriate selection of DNA sequence and organic polymer gives control over the material properties and their self-assembly into supramolecular structures. The introduction of a hydrophobic polymer into DBCs in aqueous solution leads to amphiphilic micellar structures with a hydrophobic polymer core and a DNA corona. In this Account, we discuss selected examples of recent developments in the synthesis, structure manipulation and applications of DBCs. We present achievements in synthesis of DBCs and their amplification based on molecular biology techniques. We also focus on concepts involving supramolecular assemblies and the change of morphological properties by mild stimuli. Finally, we discuss future applications of DBCs. DBC micelles have served as drug-delivery vehicles, as scaffolds for chemical reactions, and as templates for the self-assembly of virus capsids. In nanoelectronics, DNA polymer hybrids can facilitate size selection and directed deposition of single-walled carbon nanotubes in field effect transistor (FET) devices.
Cooperative Hybridization of Oligonucleotides ' Introduction

Abstract:Nucleic acids have been demonstrated to be versatile nanoscale engineering materials with the construction of dynamic DNA structures, motors, and circuits. These constructions generally rely on the clever use and integration of relatively few reaction mechanisms and design primitives. Here, cooperative hybridization is introduced as a mechanism in which two oligonucleotides of independent sequence can stoichiometrically, simultaneously, and cooperatively hybridize to a DNA complex. Cooperative hybridization is rigorously characterized and modeled and is shown to implement digital concentration comparison with amplification, as well as digital Boolean logic. These designs, based on cooperative hybridization, excel in being robust to impurities and not requiring oligonucleotide purification. The precise temporal and spatial control of molecules is a fundamental goal of both synthetic biology and nanotechnology and is essential for building reliable nanoscale structures and devices. Nucleic acids, by virtue of their well-understood hybrid-ization thermodynamics and kinetics, 1 exponential information content and 0.4 nm addressability, 2 and economy of synthesis and preparation, 3 have emerged as a leading material for nano-scale engineering. 4-9 Furthermore, the biological relevance of nucleic acids 10,11 and the ease of coupling nucleic acids to other materials, such as proteins 12 and carbon nanotubes, 13 facilitate the use of nucleic acids both as synthetic biomaterials and as scaffolds for other nanotechnological applications. Although the first generation of DNA nanotechnology research has focused on the self-assembly of static DNA nanostructures, 14-18 recent works in the field have also expanded into the realm of constructing dynamic nucleic acid devices, in which nucleic acid nanostructures conditionally and programmably reconfigure in solution. 8,9 Examples include cascaded logical and amplification circuits, 19-27 DNA origami boxes that close and open, 28 molecular walkers that traverse predefined landscapes, 29-32 controlled rotating DNA frameworks, 33-35 and chain reaction DNA motors and dendrimers. While some of these constructions relied on functional nucleic acid molecules with innate catalytic activity (known as ribozymes and deoxyribozymes), 19-22,29,30 many others were constructed using purely rational design approaches, based on the well-characterized thermodynamic 1,39 and kinetic 40,41 properties of DNA hybridization, branch migration, and dissociation processes. Abstract:Mendelian genetics spark the century‐old revolution in genetic engineering. DNA structural engineering may well spark a similar revolution in chemistry. The first few man‐made DNA structures are purely DNA but increased interest in hybrid DNA with other molecules is regaining popularity. Nucleic acids can interact with non‐nucleic acid‐based materials, showing their materialistic flexibility and potential and the prospect of forming new future materials. This review provides a deep materialistic background based on the interaction of nucleic acids with these non‐nucleic acid‐based materials covering comprehensive topics. The non‐nucleic acid materials are broadly divided into biological materials, for example, small‐biomolecules, peptides, proteins, and lipids, and non‐biological materials consisting of metal ions, chemical molecules, low‐dimensional nanomaterials, and micro‐scale polymers. Of significance, combining the unique properties of nucleic acids with these non‐genetic materials can bring about surprising synergistic characteristics and drive attractive applications in bio‐imaging, chem/bio‐sensing, disease therapies, antimicrobial works, and even information storage. Nucleic acids when integrated with cell‐exogenous materials can be remarkably new materials. The hybrids are shown to address the key issues arising from varied fields of disease diagnosis and therapy, materials sciences, analytical chemistry, and materials engineering. Abstract:Nucleic acids hold great promise for bottom-up construction of nanostructures via programmable self-assembly. Especially, the emerging of advanced sequence design principles and the maturation of chemical synthesis of nucleic acids together have led to the rapid development of structural DNA/RNA nanotechnology. Diverse nucleic acids-based nano objects and patterns have been constructed with near-atomic resolutions and with controllable sizes and geometries. The monodispersed distribution of objects, the up-to-sub-millimeter scalability of patterns, and the excellent feasibility of carrying other materials with spatial and temporal resolutions have made DNA/RNA assemblies extremely unique in molecular engineering. In this review, we summarize recent advances in nucleic acids-based (mainly DNA-based) near-atomic fabrication by focusing on state-of-the-art design techniques, toolkits for DNA/RNA nano-engineering and related applications in a range of areas.

Abstract:With the advent of DNA nanotechnology, DNA-based biomaterials have emerged as a unique class of materials at the center of various biological advances. Owing to DNA's high modification capacity via programmable Watson–Crick base-pairing, DNA structures of desired design with increased complexity have been designed. However, the lack of inherent functionality, along with low mechanical properties, high synthesis costs, and poor stability, limited the adaptability of DNA-based material toward complex biological systems. To impart functionality, DNA-based hybrid biomaterials synthesized by conjugating DNA with functional materials have been realized. Today, DNA-based hybrid materials have attracted significant attention in biological engineering with broad applicative prospects in biomedicine, clinical diagnosis, and nanodevices. Here, we summarize the recent advances in DNA-based hybrid materials with an in-depth understanding of general molecular design principles, functionalities, and applications. Finally, the challenges and prospects associated with DNA-based hybrid materials are discussed at the end of the review.

Abstract:Polypeptides are fascinating materials with unique properties for various biological materials. We highlight here recent advances in amino acid N-carboxyanhydrides (NCAs) and synthetic polypeptides from the aspects of chemistry, self-assembly and biological applications. New synthetic methodologies, mechanistic studies and optimization of polymerization conditions for the preparation of well-defined novel polypeptides are comprehensively reviewed and evaluated. Functional polypeptides, mostly prepared from novel NCA monomers, with ultra-stable helical conformation, stimuli-sensitive properties, or glycoprotein mimetics are summarized. We also highlight a number of interesting self-assembled structures of polypeptides in solid state and solution, with particular emphasis on those structures other than amphiphilic self-assembly. The biological applications of polypeptides in drug and gene delivery are also reviewed. Future directions and perspectives are discussed in the conclusion.

Abstract:The translation of DNA into synthetic molecules enables their manipulation by powerful evolution-based methods previously available only to proteins and nucleic acids. The development of increasingly sophisticated DNA-templated small-molecule syntheses is crucial to broadening the scope of this approach. Here, we report the translation of DNA templates into monocyclic and bicyclic N-acyloxazolidines using multistep DNA-templated organic synthesis. Second-generation template architectures, used for the first time in a multistep DNA-templated synthesis, together with reactions and linker cleavage strategies not previously described in a DNA-templated format, were crucial to the successful translation. The products generated in this work represent the most complex small molecules to date synthesized in a DNA sequence-programmed manner and provide the basis for DNA-templated synthetic heterocycle libraries.

Abstract:Sequence-defined polymer is one of the most promising alternative media for high-density data storage. It could be used to alleviate the problem of insufficient storage capacity of conventional silicon-based devices for the explosively increasing data. To fulfil the goal of polymer data storage, suitable methods should be developed to accurately read and decode the information-containing polymers, especially for those composed by a combination of the natural and unnatural monomers. Nanopore-based approaches have become one of the most competitive analysis and sequencing techniques, which are expected to read both natural and synthetic polymers with single-molecule precision and monomeric resolution. Herein, this work emphasizes the advances being made in nanopore reading and decoding of information stored in the man-made polymers and DNA nanostructures, and discusses the challenges and opportunities towards the development and realization of high-density data storage.
