Abstract:Since the first report in the 1950s, immunosensors and immunoassays with good specificity have got extensive applications in clinical diagnosis, food safety, and environmental monitoring. As a typical type of immunosensors, enzyme-linked immunosorbent assays (ELISAs) are able to offer high sensitivity for target sensing because of the catalytically amplified readouts produced by their enzyme labels (horseradish peroxidase, alkaline phosphatase, et al.). However, the use of these natural enzymes brings some undesired defects to traditional ELISAs, including high cost and short shelflife. Fortunately, the discovery of nanomaterials with enzyme-like catalytic features (nanozymes) provides a potential way to overcome the above shortcomings. By replacing natural enzymes with artificial nanozymes, the developed immunosensors exhibit lower cost, better stability, and easier production along with comparable sensitivity and selectivity. In this regard, the past few years have witnessed the booming development of nanozyme-based immunosensors. Herein, we make a review on currently developed immunosensors and immunoassays where nanozymes are used as promising alternatives to natural enzymes. Progress of nanozyme-based immunosensors made in recent years is summarized according to various signaling modes. Some trends in the field, like the exploration of high-activity single-atom nanozymes to further improve the sensitivity, are discussed as well, hoping to provide some guides for future research. Abstract:Electrochemical immunosensors have attracted immense attention due to the ease of mass electrode production and the high compatibility of the miniature electric reader, which is beneficial for developing point-of-care diagnostic devices. Electrochemical immunosensors can be divided into label-free and label-based sensing strategies equipped with potentiometric, amperometric, or impedimetric detectors. Emerging nanomaterials are frequently used on electrochemical immunosensors as a highly rough and conductive interface of the electrodes or on nanocarriers of immobilizing capture antibodies, electroactive mediators, or catalyzers. Adopting nanomaterials can increase immunosensor characteristics with lower detection limits and better sensitivity. Recent research has shown innovative immobilization procedures of nano-materials which meet the requirements of different electrochemical immunosensors. This review discusses the past five years of advances in nanomaterials (metal nanoparticles, metal nanostructures, carbon nanotubes, and graphene) integrated into the electrochemical immunosensor. Furthermore, the new tendency and endeavors of nanomaterial-based electrochemical immunosensors are discussed.

Abstract:Owing to its advantages such as simplicity, good sensitivity and ease of operation, enzyme-linked immunosorbent assay (ELISA) has been a powerful tool in food safety and verification. However, the relatively low sensitivity and accuracy are the main limitations of traditional ELISA, which have hindered its application in food analysis. Thus, it is of ultra-importance to construct effective strategies to solve the problem. Till now, great progress has been made in boosting the accuracy, sensitivity and stability to improve traditional ELISA. Fortunately, the nanomaterials-based ELISA (nano-ELISA) greatly developed ELISA and behaved superior performance. In this review, recent advances about nano-ELISA are discussed in four basic parts including absorbent substrate, recognition element, enzyme label and chromogenic agent as well as the related application in food analysis, which provides a new train of thought and tactics for other techniques such as biosensors in electrochemistry, optics and magnetism.

Abstract:Immunoassay as a rapid and convenient method for detecting a variety of targets has attracted tremendous interest with its high specificity and sensitivity. Among the commonly used immunoassays, enzyme-linked immunosorbent assay has been widely used as a gold standard method in various fields that consists of two main components including a recognition element and an enzyme label. With the rapid advances in nanotechnology, nanobodies and nanozymes enable immunoassays with enhanced specificity and sensitivity compared with conventional antibodies and natural enzymes. This review is focused on the applications of nanobodies and nanozymes in immunoassays. Nanobodies advantage lies in their small size, high specificity, mass expression, and high stability. Nanozymes with peroxidase, phosphatase, and oxidase activities and their applications in immunoassays are highlighted and discussed in detail. In addition, the challenges and outlooks in terms of the use of nanobodies and the development of novel nanozymes in practical applications are discussed.

Abstract:Nanozyme, a term defined for nanomaterial with enzyme-like properties, has attracted significant research attention owing to its striking merits. Recently, a surge of nanozymes have been demonstrated to catalyze some typical enzymatic reactions mimicking oxidase, peroxidase and catalase. Especially, nanozymes with peroxidase-like activity have grown into a big family due to their broad range of applications in the field of biosensing and immunoassay. Since inorganic nanoparticles possess the advantages of high stability and easy surface modification, nanozymes have been emerging alternatives to natural enzymes to some extent. In this Review, we briefly summarize several typical nanozymes and then focus our attention on their enormous applications with respect to analytical chemistry. Representative examples would be discussed in detail from the literatures of last 10 years. Additionally, the current challenges and future directions about nanozymes are speculated at the end of this review.

Abstract:Colorimetric immunoassays for tumor marker detection have attracted considerable attention due to their simplicity and high efficiency. With the achievements of nanotechnology and nanoscience, nanomaterials-based colorimetric immunoassays have been demonstrated to be promising alternatives to conventional colorimetric enzyme-linked immunoassays. This review is focused on the progress in colorimetric immunoassays with the signal amplification of nanomaterials, including nanomaterials-based artificial enzymes to catalyze the chromogenic reactions, analyte-induced aggregation or size/morphology change of nanomaterials, nanomaterials as the carriers for loading enzyme labels, and chromogenic reactions induced by the constituent elements released from nanomaterials.

Abstract:Abstract Immunoassay is a powerful technique that uses highly specific antigen‐antibody interactions to detect biochemical targets such as proteins and toxins. As a diagnostic tool, immunoassay is employed in the screening, diagnosis, and prognosis of diseases, which are crucial for the grasp and control of patient conditions in clinical practice. With the rapid development of nanotechnology, immunoassays based on nanoprobes have attracted more and more attention due to the advantages of high sensitivity, specificity, stability, and versatility. These nanoprobes are nanoscale particles that can act as signal carriers or targeting agents for immunoassays. In this paper, we review the recent advances in various types of nanoprobes for immunoassays, such as colloidal gold, quantum dots, magnetic nanoparticles, nanozymes, aggregation‐induced emission, and up‐conversion nanoparticles. The effect of the nanoprobe construction and synthesis methods on their detection performance deserves to be studied in depth. We also compare their detection ranges and limits in different immunoassay methods, such as lateral flow immunoassays, fluorescent immunoassays, and surface‐enhanced Raman scattering immunoassays. Moreover, we discuss the benefits and challenges of nanoprobes in immunoassays and provide insights into their future development. This study aims to offer a comprehensive and critical perspective on the role of nanoprobes in the field of immunoassays.

Abstract:Nanozymes are nanomaterials with enzyme-like catalytic activities. The unique features of nanozymes (such as high stability, low cost, large surface area for bioconjugation, ease of storage, and multi-functionalities) offer unprecedented opportunities for designing electrochemical biosensors. Recent years have witnessed the rapid development of nanozyme-based electrochemical biosensors. To highlight these achievements, this review first discusses the representative nanozymes including peroxidase mimics, oxidase mimics, hydrolase mimics, and superoxide dismutase mimics used in electrochemical biosensors. Then, it summarizes the bioanalytical applications for the detection of various analytes. Finally, current challenges and future research directions are summarized.

Abstract:Molecularly imprinted polymer (MIP) is illustrated as an analogue of a natural biological antibody-antigen system. MIP is an appropriate substrate for electrochemical sensors owing to its binding sites, which match the functional groups and spatial structure of the target analytes. However, the irregular shapes and slow electron transfer rate of MIP limit the sensitivity and conductivity of electrochemical sensors. Nanomaterials, famous for their prominent electron transfer capacity and specific surface area, are increasingly employed in modifications of MIP sensors. Staying ahead of traditional electrochemical sensors, nanomaterials-based MIP sensors represent excellent sensing and recognition capability. This review intends to illustrate their advances over the past five years. Current limitations and development prospects are also discussed.

Abstract:The novel nanomaterials (e.g., gold nanoparticles (AuNPs), silver nanoparticles (AgNPs), platinum nanoparticles (PtNPs), quantum dots (QDs), carbon nanotubes (CNTs), graphene oxides (GOs), and metal organic frameworks (MOFs)) with unique characteristics have been widely applied for the construction of in vitro and in vivo nanosensors. However, the detection of extremely low abundant biomarkers remains a great challenge. To solve this issue, different kinds of nucleic acid amplification approaches (e.g., hybridization chain reaction (HCR), Exo III-assisted target recycling (EATR), strand displacement amplification (SDA), rolling circle amplification (RCA), and ligase chain reaction (LCR)) have been introduced, endowing the nanosensors with improved sensitivity and good performance. In this review, we summarize the recent advance in the development of nanosensors based on the integration of nanomaterials with nucleic acid amplification approaches for sensitive detection of DNAs, RNAs, enzymes, proteins, and small biomolecules. Moreover, we give a new insight into their future directions.

Abstract:Various kinds of nanomaterials have been described in recent years that represent stable and low-cost alternatives to biomolecules (such as enzymes) for use in (bio)analytical methods. The materials typically include, metal/metal oxides, metal complexes, nanocomposites, porphyrins, phthalocyanines, smart polymers, and carbonaceous nanomaterials. Due to their biomimetic and other properties, such nano-materials may replace natural enzymes in chemical sensors, biosensors, and in various kinds of bioassays. This overview (with 252 references) highlights the analytical potential of such nanomaterials. It is divided into sections on (a) the types of nanomaterials according to their intrinsic nature, (b) non-enzymatic sensor designs (including electrochemical, colorimetric, fluorescent and chemiluminescent methods), and (c), applications of non-enzymatic sensors in the biomedical, environmental and food analysis fields. We finally address current challenges and future directions. Graphical abstract This review discusses different types of nanomaterials, which are explored as a potential biomimetic material to replace the natural enzyme in the field of biosensors, and have found widespread applications in biomedical, food and environmental analysis.

Abstract:Biosensor technology is experiencing an unprecedented momentum that is driven by rapid advances in material science for utilization in various fields, and is constantly evolving to develop fast, simple and sensitive sensors. Here, we present a comprehensive and concise review on electrochemiluminescent (ECL) nanomaterial-based aptasensors including challenges and applications. ECL aptasensor combines the merits of a biosensor, selectivity of an aptamer, and sensitivity of ECL to construct sensitive and selective sensors. Signal amplification processes through the incorporation of nanomaterials are essential to further enhance the sensitivity of ECL sensing systems. Nanomaterials are commonly utilized in sensor fabrication as (i) electrode modifiers, (ii) nanocarriers, (iii) nanocatalysts, and (iv) luminophores. The review highlights the effects of integrating nanomaterials such as gold nanoparticles, metal nanoparticles, metal oxides, carbon nanomaterials, metal organic frameworks, and quantum dots using various research strategies that address signal amplification to enhance the ECL signal. Furthermore, various concepts have been demonstrated with relevant figures and explanations to serve as a general overview for the design of ECL aptasensors. Lastly, we discuss the underlying problems and possibilities for ECL aptasensor research and development.

Abstract:As nanoscale materials with the function of catalyzing substrates through enzymatic kinetics, nanozymes are regarded as potential alternatives to natural enzymes. Compared to protein-based enzymes, nanozymes exhibit attractive characteristics of low preparation cost, robust activity, flexible performance adjustment, and versatile functionalization. These advantages endow them with wide use from biochemical sensing and environmental remediation to medical theranostics. Especially in biomedical diagnosis, the feature of catalytic signal amplification provided by nanozymes makes them function as emerging labels for the detection of biomarkers and diseases, with rapid developments observed in recent years. To provide a comprehensive overview of recent progress made in this dynamic field, here an overview of biomedical diagnosis enabled by nanozymes is provided. This review first summarizes the synthesis of nanozyme materials and then discusses the main strategies applied to enhance their catalytic activity and specificity. Subsequently, representative utilization of nanozymes combined with biological elements in disease diagnosis is reviewed, including the detection of biomarkers related to metabolic, cardiovascular, nervous, and digestive diseases as well as cancers. Finally, some development trends in nanozyme-enabled biomedical diagnosis are highlighted, and corresponding challenges are also pointed out, aiming to inspire future efforts to further advance this promising field.

Abstract:Using chitosan-modified magnetite nanoparticles (CS-MNPs) as a replacement of enzymes in conventional ELISA configurations, a magnetic nanoparticle-linked immunosorbent assay is developed. The CS-MNPs are synthesized by using a one-step solvothermal process, where chitosan in the reaction system is used both as a ligand and as a surface modification agent. The as-synthesized CS-MNPs have amine groups on their surface which provide good dispersibility in aqueous solutions and convenient sites for covalent linking of antibodies with the MNPs. They also possess catalytic properties that are able to catalyze color reactions in immunoassays and magnetic properties that can be used to capture, separate, and enrich antigens prior to the assay procedure. By employing both the catalytic and magnetic properties of the CS-MNPs, a capture-detection immunoassay is developed, where antigens can be captured, separated, and enriched prior to the assay procedure.

Abstract:Over the past decade, artificial nanomaterials that exhibit properties similar to those of enzymes are gaining attraction in electrochemical biosensing as highly stable and low-cost alternatives to enzymes. This review article discusses the main features of the various nanomaterials (metal oxide, metal, and carbon-based materials) explored so far to mimic different kinds of enzymes. The unprecedented opportunities imparted by these functional nanomaterials or their nanohybrids, mostly providing peroxidase-like activity, in electrochemical affinity biosensing are critically discussed mainly in connection with their use as catalytic labels or electrode surface modifiers by highlighting representative strategies reported in the past 5 years with application in the food, environmental, and biomedical fields. Apart from outlining the pros and cons of nanomaterial-based enzyme mimetics arising from the impressive development they have experienced over the last few years, current challenges and future directions for achieving their widespread use and exploiting their full potential in the development of electrochemical biosensors are discussed.
